Abstract:Application of proteins and peptides as human therapeutics is expanding rapidly as drug discovery becomes more prevalent. Conjugation of polymers to proteins can circumvent many problems and pegylation of proteins is now emerging as acceptable practice. This paper describes the synthesis of alpha-aldehyde-terminated poly(methoxyPEG)methacrylates from Cu(I) mediated living radical polymerization (Mn = 11 000, 22 000 and 32 000; PDi < 1.15), and their efficient conjugation to lysozyme, as a model protein. This offers an attractive and flexible alternative to linear poly(ethylene glycol) opening up the possibility of using the full power of living radical polymerization.
Ligand-free Palladium-Mediated Site-Specific Protein Labeling Inside Gram-Negative Bacterial Pathogens.

Abstract:Palladium, a key transition metal in advancing modern organic synthesis, mediates diverse chemical conversions including many carbon-carbon bond formation reactions between organic compounds. However, expanding palladium chemistry for conjugation of biomolecules such as proteins, particularly within their native cellular context, is still in its infancy. Here we report the site-specific protein labeling inside pathogenic Gram-negative bacterial cells via a ligand-free palladium-mediated cross-coupling reaction. Two rationally designed pyrrolysine analogues bearing an aliphatic alkyne or an iodophenyl handle were first encoded in different enteric bacteria, which offered two facial handles for palladium-mediated Sonogashira coupling reaction on proteins within these pathogens. A GFP-based bioorthogonal reaction screening system was then developed, allowing evaluation of both the efficiency and the biocompatibilty of various palladium reagents in promoting protein-small molecule conjugation. The identified simple compound-Pd(NO3)2 exhibited high efficiency and biocompatibility for site-specific labeling of proteins in vitro and inside living E. coli cells. This Pd-mediated protein coupling method was further utilized to label and visualize a Type-III Secretion (T3S) toxin-OspF in Shigella cells. Our strategy may be generally applicable for imaging and tracking various virulence proteins within Gram-negative bacterial pathogens.

Abstract:Abstract The immense application potential of amphiphilic protein-polymer conjugates remains largely unexplored, as established “grafting from” synthetic protocols involve time-consuming, harsh and disruptive deoxygenation methods, while “grafting to” approaches result in low yields. Here we report an oxygen tolerant, photoinduced CRP approach which readily affords quantitative yields of protein-polymer conjugates within 2 h, avoiding damage to the secondary structure of the protein and providing easily accessible means to produce biomacromolecular assemblies. Importantly, our methodology is compatible with multiple proteins (e.g. BSA, HSA, GOx, beta-galactosidase) and monomer classes including acrylates, methacrylates, styrenics and acrylamides. The polymerizations are conveniently conducted in plastic syringes and in the absence of any additives or external deoxygenation procedures using low-organic content media and ppm levels of copper. The robustness of the protocol is further exemplified by its implementation under UV, blue light or even sunlight irradiation as well as in buffer, nanopure, tap or even sea water.

A simple methodology for the synthesis of heterotelechelic protein-polymer-biomolecule conjugates

Abstract:A synthetic protocol for the preparation of hetero-biofunctional protein-polymer conjugates is described. A chain transfer agent, SS-bis (alpha,alpha'-dimethyl-alpha "-acetic acid) trithiocarbonate was functionalized with alpha,omega-pyridyl disulfide (PDS) groups, Subsequently, one of the PDS groups was covalently attached to bovine serum albumin (BSA) at the specific free thiol group on the cysteine residue through a disulfide linkage. The second PDS group remained intact, as it was found to be inaccessible to further BSA functionalization. The BSA-macro-reversible addition-fragmentation chain transfer (RAFT) agent was then used to prepare BSA-polymer conjugates via in situ polymerization of oligo (ethyleneglycol) acrylate and N-(2-hydroxypropyl) methacrylamide using an ambient temperature initiator, 4,4'-azobis [2,9-imidazolin-2-ethyl)propane] dihydrochloride in an aqueous medium. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) confirmed that the in situ polymerization occurred at the protein surface where the RAFT agent was attached and the molecular weights of the BSA-polymer conjugates were found to increase concomitantly with monomer conversion and polymerization time. After polymerization the remaining terminal PDS groups were then utilized to attach thiocholesterol and a flurophore, rhodamine B to the protein-polymer conjugates via disulfide coupling. UV-Vis and fluorescence analyses revealed that similar to 80% of the protein conjugates were found to retain integral PDS end groups for further attachment to free thiol-tethered precursors. Abstract:Various polymers have been tested for protein conjugation with a goal of bridging the complementary advantages of both components. However, many of these polymers, including the most well-established PEG, are nondegradable, which raises potential concerns on their cumulative chronic toxicity. Moreover, the immunogenicity of PEG has recently evoked considerable controversy. Synthetic polypeptides, on the other hand, are biomimetic polymers with tunable degradability, versatile side chain functionalities, unique secondary structures, and fascinating self-assembly behaviors. These properties have made them promising materials in protein modification for various applications. In this Topical Review, we summarize recent advances and list a number of interesting future directions in protein polypeptide conjugation, which we termed protein PEPylation.

Abstract:Protein-polymer conjugates combine properties of biopolymers and synthetic polymers, such as specific bioactivity and increased stability, with great benefits for various applications from catalysis to biomedicine. Furthermore, polymer conjugation can mimic important posttranslational modifications of proteins such as glycosylation. There are typically two approaches to create protein-polymer conjugates: the protein is functionalized in advance with an initiator for a grafting-from method or a previously produced polymer is conjugated to the protein via a grafting-to method. In this study, we present a new approach that uses native proteins and allows for direct grafting-from using a thiol-induced, light-activated controlled radical polymerization (TIRP) that is initiated at thiols from specific cysteine residues of the protein. This straightforward method is employed to introduce polymers onto proteins and enzymes without any prior protein modifications, it works in aqueous buffer and maintains the protein's native structure and activity. The resulting protein-polymer conjugates exhibit high molar masses and low dispersities. We demonstrate the versatility of this approach by introducing different types of polymers such as hydrophilic poly(2-hydroxyethyl acrylate) (pHEAA), temperature-responsive poly(N-isopropylacrylamide) (pNIPAM) as well as glycopolymers mimicking the natural protein glycosylation and enabling selective interactions. We present successful combinations of the protein and polymer functions e.g., temperature-induced aggregation leading to an increase in enzyme activity and the introduction of artificial glycosylation inducing specific protein-protein cluster formation and giving straightforward access to glycosurfaces. Based on this straightforward, potentially scalable yet highly controlled synthesis of protein-polymer conjugates, various areas of applications are envisioned ranging from biomedicine to material sciences.

Abstract:Modifications at polypeptide side chains have been found to affect conformational and properties,which is a vital strategy to prepare functional polypeptides.This contribution describes the synthesis of homo and co-polypeptides containing unsaturated bonds and thioether moieties,poly(S-allylcysteine)(PAC).The pendant vinyl groups provide an active binding site which are able to be further modified with cysteine by thiol-ene click reaction.The sulfoxide from thioether oxidation bestows the polypeptide water solubility and enzymatic catalytic sites.The self-assembly micelles of PEG-b-PAC are ruptured along with oxidation to release the Nile red payloads.Moreover,poly(S-allyl-cysteine sulfoxide),aka.polyalliin,is the oxidation product of PAC which undergoes side chain cleavage catalyzed by alliinase enzyme.The polypeptides are biocompati-ble as confirmed by cell viability assays and may find applications in drug delivery,biosensing and nanoreactor.

Abstract:This review describes recent changes and progress in the field of polymer bioconjugation (i.e. covalent attachment of synthetic polymers to biological entities such as nucleic acids, oligopeptides, proteins, enzymes, carbohydrates, viruses or cells). This important field of research has evolved radically during the last decade. Polymer bioconjugates have been initially developed by biochemists and have been for many years studied practically exclusively for bio-medical applications. However, within the last few years, these novel class of macromolecules have been shown to be useful in many other emerging areas of materials science. Therefore, the synthesis of polymer bioconjugates became rather suddenly a central topic in polymer chemistry. For instance, modern synthetic methods such as controlled radical polymerization, ring opening metathesis polymerization or “click” chemistry have been recently proven to be extremely versatile tools for preparing tailor-made polymer bioconjugates. These novel synthetic trends are discussed in details in the present manuscript.

Abstract:Cobalt-mediated radical polymerizations (CMRPs) have been initiated by the radical decarboxylation of tetrachlorophthalimide activated esters. This allows for the controlled radical polymerization of activated monomers across a broad temperature range with a single cobalt species, with the incorporation of polymer end groups derived from simple carboxylic acid derivatives and termination with an organozinc reagent. This method has been applied to the synthesis of a polymer/graphene conjugate and a water-soluble protein/polymer conjugate, demonstrating the first examples of CMRP in graphene and protein conjugation.

Abstract:To avoid the use of metal catalysts used in conventional approaches, an Eosin Y/TEMED mediated, photoinduced polymerization of vinyl monomers was optimized for the synthesis of protein-polymer conjugates. This oxygen tolerant, photoinduced approach allowed to graft a series of hydrophobic, hydrophilic and responsive polymers with quantitative protein macroinitiator consumption. CALB bioconjugates were also synthesized and found to retain part of the parent protein activity for extended periods of time. Notably, when BSA was used in the absence of initiator, protein-coated nanoparticles were shown to form during emulsion polymerization.

Abstract:A series of alpha-functional maleimide polymethacrylates (M(n) = 4.1-35.4 kDa, PDi = 1.06-1.27) have been prepared via copper-catalyzed living radical polymerization (LRP). Two independent synthetic protocols have been successfully developed and the polymers obtained in multigram scale, with an 80-100% content of maleimide reactive chain ends, depending on the method employed. A method for the synthesis of amino-terminated polymers, starting from Boc-protected amino initiators, has also been developed, as these derivatives are key intermediates in one of the two processes studied in the present work. The alternative synthetic pathway involves an initiator containing a maleimide unit "protected" as a Diels-Alder adduct. After the polymerization step, the maleimide functionality has been reintroduced by retro-Diels-Alder reaction, by simply refluxing those polymers in toluene for 7 h. These maleimido-terminated materials, poly(methoxyPEG((475))) methacrylates and poly(glycerol) methacrylates, differ for both the nature and size of the polymer side branches and showed an excellent solubility in water, a property that made them an ideal candidate for the synthesis of new polymer-(poly)peptide biomaterials. These functional polymers have been successfully employed in conjugation reactions in the presence of thiol-containing model substrates, namely, reduced glutathione (gamma-Glu-Cys-Gly) and the carrier protein, bovine serum albumin (BSA), in 100 mM phosphate buffer (pH 6.8-7.4) and ambient temperature.
C-Terminal Peptide Modification: Merging the Passerini Reaction with Chemo-Enzymatic Synthesis

Abstract:Over the past few decades, peptide/protein synthesis and bio-conjugation has gained increasing interest in the research community owing to the high demand for strategies that provide modified protein adducts in a site-selective fashion. Herein, we report a novel approach that combines the Passerini multicomponent reaction and chemo-enzymatic peptide synthesis (CEPS) for the selective bio-conjugation of peptide C-termini. The Passerini utilizes aqueous acidic buffer conditions to establish chemoselectivity for the carboxylic acids, while the subsequent enzymatic ligation selectively targets the formed C-terminal substrates. We functionalized a diverse set of pentapeptides utilizing numerous isocyanide and car-bonyl compounds and successfully performed subsequent ligations. This combined multicomponent chemoenzymatic method therefore represents a valuable novel technology for future research into site-selective C-terminal modification of peptides/proteins.

Abstract:Construction of polymer-protein nanoassemblies is a challenge as reactions between macromolecules, especially those involving proteins, are inherently inefficient owing to the sparse reactive functional groups and low concentration requirements. We address this challenge using an ultrafast and reversible click reaction, which forms the basis for a novel covalent self-assembly strategy between side-chain functionalized polymers and surface-modified proteins. The linkers in the assembly have been programmed to release the incarcerated proteins in its native form, only when subjected to the presence of a specific trigger. The generality and the versatility of the approach have been demonstrated by showing that this strategy can be used for proteins of different sizes and isoelectric points. Moreover, simple modifications in the linker chemistry offers the ability to trigger these assemblies with various chemical inputs. Efficient formation of nanoassemblies based on polymer-protein conjugates has implications in a variety of areas at the interface of chemistry with materials and biology, such as in the generation of active surfaces and in delivery of biologics. As a demonstration of utility in the latter, we have shown that these conjugates can be used to transport functional proteins across cellular membranes.

Abstract:The capability to graft synthetic polymers onto the surfaces of live cells offers the potential to manipulate and control their phenotype and underlying cellular processes. Conventional grafting-to strategies for conjugating preformed polymers to cell surfaces are limited by low polymer grafting efficiency. Here we report an alternative grafting-from strategy for directly engineering the surfaces of live yeast and mammalian cells through cell surface-initiated controlled radical polymerization. By developing cytocompatible PET-RAFT (photoinduced electron transfer-reversible addition-fragmentation chain-transfer polymerization), synthetic polymers with narrow polydispersity (Mw/Mn < 1.3) could be obtained at room temperature in 5 minutes. This polymerization strategy enables chain growth to be initiated directly from chain-transfer agents anchored on the surface of live cells using either covalent attachment or non-covalent insertion, while maintaining high cell viability. Compared with conventional grafting-to approaches, these methods significantly improve the efficiency of grafting polymer chains and enable the active manipulation of cellular phenotypes.

In Situ Growth Of A Stoichiometric Peg-Like Conjugate At A Protein'S N-Terminus With Significantly Improved Pharmacokinetics

Abstract:The challenge in the synthesis of protein-polymer conjugates for biological applications is to synthesize a stoichiometric (typically 1: 1) conjugate of the protein with a monodisperse polymer, with good retention of protein activity, significantly improved pharmacokinetics and increased bioavailability, and hence improved in vivo efficacy. Here we demonstrate, using myoglobin as an example, a general route to grow a PEG-like polymer, poly(oligo(ethylene glycol) methyl ether methacrylate) [poly(OEGMA)], with low polydispersity and high yield, solely from the N-terminus of the protein by in situ atom transfer radical polymerization (ATRP) under aqueous conditions, to yield a site-specific (N-terminal) and stoichiometric conjugate (1:1). Notably, the myoglobin-poly(OEGMA) conjugate [hydrodynamic radius (R-h): 13 nm] showed a 41-fold increase in its blood exposure compared to the protein (R-h: 1.7 nm) after IV administration to mice, thereby demonstrating that comb polymers that present short oligo(ethylene glycol) side chains are a class of PEG-like polymers that can significantly improve the pharmacological properties of proteins. We believe that this approach to the synthesis of N-terminal protein conjugates of poly(OEGMA) may be applicable to a large subset of protein and peptide drugs, and thereby provide a general methodology for improvement of their pharmacological profiles.

Abstract:We here found that while Meldrum's acid as the reactive warhead allows for the attachment of a single chemical modification on aldehyde-containing proteins, pyrazolone derivatives in combination with a phosphine nucleophile enable protein dual site-specific conjugation with the same or distinct moieties. These reactions are efficient and convergent under biocompatible conditions and allow access to protein bioconjugates with superior stability, homogeneity and flexibility. Our work expands the repertoire of bioconjugation chemistries and offers opportunities to construct bioconjugates with defined structure that have potential for medical and biomaterial applications.
Aqueous Protein–Polymer Bioconjugation via Photoinduced RAFT Polymerization Using High Loading Heterogeneous Catalyst

Abstract:Light-driven polymerization, such as photoinduced electron/energy transfer–reversible addition–fragmentation chain transfer (PET-RAFT) polymerization, enables biological benign conditions and versatile functional polymer structure design, which is readily used in protein–polymer bioconjugates. However, conventional metalloporphyrinic homogeneous catalysts for PET-RAFT polymerization suffer from limited aqueous solubility and tedious purification. Here we demonstrate the design of PET-RAFT photocatalyst from the reticular assembled Zr–porphyrinic metal–organic frameworks (MOFs), along with a biomacromolecule-based chain transfer agent, as efficient bioconjugation tools in water. Our methodology offers manufacturing advantages on bioconjugates under mild conditions such that MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) and cytotoxicity assays have shown the preservation of the protein integrity, bioactivity, and high cell viability after PET-RAFT polymerization. We find that the fast kinetics are benefiting from the ultrahigh loading of metalloporphyrins in MOF-525-Zn. This heterogeneous catalyst also allows us to maintain living characteristics to incorporate myriads of monomers into block copolymers. Other advantages like easy postreaction purification, reusability, and high oxygen tolerance even in an open system are demonstrated. Abstract:A series of well defined semitelechelic and mid-functionalized poly(poly(ethylene glycol) methyl ether methacrylate)s (poly(PEGMA)s) were synthesized through reversible addition-fragmentation chain transfer (RAFT) polymerization using thiazolidine-2-thione-functionalized chain transfer agents (CTAs). The thiazolidine-2-thione group was located either at the end or in the middle of polymer chains depending on the different structural CTAs. All polymers were fully analyzed by (1)H NMR spectroscopy and GPC, confirming their well-defined structures, such as predesigned molecular weights, narrow polydispersity indices, and high yield chain-end or chain-middle functionalization. The thiazolidine-2-thione functionality located at the end of or at the middle of the polymer chains can react with amine residues on protein surfaces, forming protein-polymer conjugates via amide linkages. The bioactivity of protein conjugates were subsequently tested using micrococcus lysodeikticus cell as substitute. The protein conjugations from the mid-functionalized polymer remained much more protein bioactivity comparing to their semitelechelic counterpart with similar molecular weights, indicating the steric hindrance of the mid-functionalized poly(PEGMA) s lead to the better selective conjugation to protein. The number of polymer chains on the protein surface was additionally evaluated by TNBS analysis, exhibiting that there are less mid-functionalized poly(PEGMA) s linked on the protein surface than the semitelechelic polymers, also supporting the hypothesis that the steric hindrance from branch-structural polymers results in the better reaction selectivity. This synthetic methodology is suitable for universal proteins, seeking a balance between the protein bioactivity and the protein protection by the covalent linkage with polymer, and exhibits promising potential for pharmaceutical protein conjugation.

Abstract:The efficient bioconjugation of functional groups/molecules to targeted matrix and bio-related species drives the great development of material science and biomedicine, while the dilemma of metal catalysis, uneasy premodification, and limited reaction efficiency in traditional bioconjugation has restricted the booming development to some extent. Here, we provide a strategy for metal-free click bioconjugation at diverse levels based on activated alkynes. As a proof-of-concept, the abundant native groups including amine, thiol, and hydroxyl groups can directly react with activated alkynes without any modification in the absence of metal catalysis. Through this strategy, high-efficient modification and potential functionalization can be achieved for natural polysaccharide, biocompatible polyethylene glycol (PEG), synthetic polymers, cell penetrating peptide, protein, fast whole-cell mapping, and even quick differentiation and staining of Gram-positive bacteria, etc. Therefore, current metal-free click bioconjugation strategy based on activated alkynes is promising for the development of quick fluorescence labeling and functional modification of many targets and can be widely applied towards the fabrication of complex biomaterials and future in vivo labeling and detection.
