Abstract:The tactics of targeting copolymer composition (CC) and molecular weight (MW) via semibatch controlled radical polymerization (CRP) have been extensively studied. However, little effort is made to target copolymer sequence length (CSL) and its triad sequence, which are among the key microstructures determining material properties of the copolymers. This work presents a method to design and synthesize targeted CSL and copolymer triads in a reversible addition-fragmentation chain transfer radical mini-emulsion copolymerization system (RAFT) through a semibatch chain and sequence model coupled with Alfrey Mayo model. The kinetic model was first derived and correlated with experimental results of the batch St/BA copolymerizations. The semibatch RAFT model was then employed to calculate the feed-rate to target CSL and their triads. St/BA copolymers with different uniform CSLs and their triads are synthesized through the programmed semibatch RAFT mini-emulsion copolymerization processes.
A simple methodology for the synthesis of heterotelechelic protein-polymer-biomolecule conjugates

Abstract:A synthetic protocol for the preparation of hetero-biofunctional protein-polymer conjugates is described. A chain transfer agent, SS-bis (alpha,alpha'-dimethyl-alpha "-acetic acid) trithiocarbonate was functionalized with alpha,omega-pyridyl disulfide (PDS) groups, Subsequently, one of the PDS groups was covalently attached to bovine serum albumin (BSA) at the specific free thiol group on the cysteine residue through a disulfide linkage. The second PDS group remained intact, as it was found to be inaccessible to further BSA functionalization. The BSA-macro-reversible addition-fragmentation chain transfer (RAFT) agent was then used to prepare BSA-polymer conjugates via in situ polymerization of oligo (ethyleneglycol) acrylate and N-(2-hydroxypropyl) methacrylamide using an ambient temperature initiator, 4,4'-azobis [2,9-imidazolin-2-ethyl)propane] dihydrochloride in an aqueous medium. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) confirmed that the in situ polymerization occurred at the protein surface where the RAFT agent was attached and the molecular weights of the BSA-polymer conjugates were found to increase concomitantly with monomer conversion and polymerization time. After polymerization the remaining terminal PDS groups were then utilized to attach thiocholesterol and a flurophore, rhodamine B to the protein-polymer conjugates via disulfide coupling. UV-Vis and fluorescence analyses revealed that similar to 80% of the protein conjugates were found to retain integral PDS end groups for further attachment to free thiol-tethered precursors. Abstract:The product from free radical and coordination polymerization generally are homopolymer or random copolymer with broad molecular weight distribution.In the past two decades,the breakthroughs in controlled/living radical and coordination polymerization and shuttling polymerization make it possible to use almost all vinyl monomers,particularly those cheap monomers,to produce more complex chain structures like di-,tri-block copolymer and gradient copolymers.With rational design,these copolymers might be high performance and high added value synthetic materials.The mechanisms and the up-to-date progress of these polymerization methods were introduced,and their possible applications were discussed.

Abstract:We explore a kinetically controlled strategy to suppress the gelation in the homopolymerization of multi-vinyl monomers (MVMs) via RAFT polymerization. We report the generation of 3D single cyclized polymer structures from the RAFT process, which significantly contradicts the classic F-S theory. This approach enables synthesis of a new generation of nanosize macromolecular architectures.

Abstract:Inspired by nature's two-stage strategy to efficiently synthesize numerous proteins using limited amino acids, a two-stage polymer preparation method has been successfully developed via the combination of ultra-fast RAFT polymerization (stage 1) and post polymerization modification (PPM) through the tricomponent Biginelli reaction (stage 2). Only using 3 monomers, 6 polymer precursors with different main-chain sequences have been quickly prepared in stage 1. In stage 2, by combinatorial synthesis, these 6 polymer precursors underwent the Biginelli reaction in a high-throughput (HTP) manner to rapidly generate 60 derivatives with precisely-controlled structures and various molecular diversities, suggesting the straightforward promotion of polymer synthesis efficiency by learning nature's strategy. Furthermore, HTP-analyses have been attempted to quickly screen some distinctively functional polymers, such as the possible polymeric radical scavengers, metal chelating agents, CT imaging agents, etc., realizing the benefit of HTP in polymer chemistry to efficiently synthesize and analyze a large number of samples. We believe that current research opens a new way to effectively prepare and characterize new libraries of polymers with abundant diversity and functions, and might promote a broader study of multicomponent reactions, combinatorial synthesis and HTP technologies in polymer science.

Abstract:Covalent conjugation of a biologically stable polymer to a therapeutic protein, e.g., an antibody, holds many benefits such as prolonged plasma exposure of the protein and improved tumor uptake. Generation of defined conjugates is advantageous in many applications, and a range of site-selective conjugation methods have been reported. Many current coupling methods lead to dispersity in coupling efficiencies with subsequent conjugates of less-well-defined structure, which impacts reproducibility of manufacture and ultimately may impact successful translation to treat or image diseases. We explored designing stable, reactive groups for polymer conjugation reactions that would lead to conjugates through the simplest and most abundant residue on most proteins, the lysine residue, yielding conjugates in high purity and demonstrating retention of mAb efficacy through surface plasmon resonance (SPR), cell targeting, and in vivo tumor targeting. We utilized squaric acid diesters as coupling agents for selective amidation of lysine residues and were able to selectively conjugate one, or two, high-molecular-weight polymers to a therapeutically relevant antibody, 528mAb, that subsequently retained full binding specificity. Water-soluble copolymers of N-(2-hydroxypropyl) methacrylamide (HPMA) and N-isopropylacrylamide (NIPAM) were prepared by Reversible Addition-Fragmentation chain-Transfer (RAFT) polymerization and we demonstrated that a dual-dye-labeled antibody-RAFT conjugate (528mAb-RAFT) exhibited effective tumor targeting in model breast cancer xenografts in mice. The combination of the precise and selective squaric acid ester conjugation method, with the use of RAFT polymers, leads to a promising strategic partnership for improved therapeutic protein-polymer conjugates having a very-well-defined structure.

Abstract:Protein-polymer conjugates combine properties of biopolymers and synthetic polymers, such as specific bioactivity and increased stability, with great benefits for various applications from catalysis to biomedicine. Furthermore, polymer conjugation can mimic important posttranslational modifications of proteins such as glycosylation. There are typically two approaches to create protein-polymer conjugates: the protein is functionalized in advance with an initiator for a grafting-from method or a previously produced polymer is conjugated to the protein via a grafting-to method. In this study, we present a new approach that uses native proteins and allows for direct grafting-from using a thiol-induced, light-activated controlled radical polymerization (TIRP) that is initiated at thiols from specific cysteine residues of the protein. This straightforward method is employed to introduce polymers onto proteins and enzymes without any prior protein modifications, it works in aqueous buffer and maintains the protein's native structure and activity. The resulting protein-polymer conjugates exhibit high molar masses and low dispersities. We demonstrate the versatility of this approach by introducing different types of polymers such as hydrophilic poly(2-hydroxyethyl acrylate) (pHEAA), temperature-responsive poly(N-isopropylacrylamide) (pNIPAM) as well as glycopolymers mimicking the natural protein glycosylation and enabling selective interactions. We present successful combinations of the protein and polymer functions e.g., temperature-induced aggregation leading to an increase in enzyme activity and the introduction of artificial glycosylation inducing specific protein-protein cluster formation and giving straightforward access to glycosurfaces. Based on this straightforward, potentially scalable yet highly controlled synthesis of protein-polymer conjugates, various areas of applications are envisioned ranging from biomedicine to material sciences.

Abstract:Janus polymerization is a novel and efficient way to synthesize diblock and multiblock copolymers in one step by using Lu(OTf)(3) and propylene epoxide as a catalytic system. By modifying the epoxide initiator, which is located at the block junction with various functional groups, the possibility for future topological design is provided. Herein, 2-bicyclo[2.2.1]hept-5-en-2-yl oxirane (NB-EO) is used as an alternative for propylene epoxide to synthesize a poly(THF-co-CL)-b-PCL diblock copolymer featuring a norbornene group at this position. This also results in multiblock [poly(THF-co-CL)-b-PCL](m) copolymers carrying multiple norbornene moieties through Janus polymerization. Subsequent ring-opening metathesis copolymerization (ROMP) of the resulting poly(THF-co-CL)-b-PCL macromonomers with norbornenyl-terminated polysarcosine (PSar-NB) allows the facile preparation of heterograft molecular polymer brushes (MPBs). The MPBs feature three heterografts of PCL, P(CL-co-THF) and PSar, potentially equipping these structures with biocompatibility, biodegradability, semi-crystallinity, and amphiphilicity. A phase separation is observed after annealing in both TEM and AFM analysis. Due to their amphiphilic nature, the MPBs also undergo self-assembly into micelles in aqueous solution. Such materials combining polypeptoids, polyesters, and polyethers segments are expected to attract wide attention in drug delivery applications.

The polymer molecular weight, architecture, and polydispersity index (PDI) were verified by gel permeation chromatography (GPC) dynamic light scattering gel permeation chromatography (DLS-GPC), and matrix-assisted laser desorption/ ionization time-of-flight (MALDI-TOF) mass spectrometry. This approach afforded well-defined polymers (PDI's < 1 06) and the ability to target various molecular weights. Malcimide functional groups were introduced at the chain ends by heating the polymers in the presence of a furna-protected azo-intiator. This allowed for site-specific conjugation of V131CT4 lysozyme to the polymers to generate multimeric protein- polymer conjugates MALDI-TOF mass spectrometry, electrospray ionization gas-phase electrophoretic mobility macromolecule analysis (ESI-GEMMA), gel electrophoresis, and liquid chromatogrpahy tandem mass spectrometry (LC-MS/MS) of the trypsin digests demonstrated that multimeric protein-polymer conjugates had formed. Abstract:Covalently conjugating proteins with synthetic polymers, particularly poly(ethylene glycol) (PEG) is widely used as a means to improve protein solubility and stability, prolong their circulating half-lives, and lower their immunogenicity. Conventionally, these polymers are attached to random locations on the protein surfaces through the modification of the reactive side chains of amino acid residues such as lysine and cysteine. The “grafting to” polymer conjugation usually leads to heterogeneous products with reduced activity and low yield, which may not be compatible with the intended applications. Therefore, it is highly desirable to synthesize well-defined protein–polymer conjugates by site-specific polymer conjugation. Recently, in situ growth of polymer conjugates from proteins (“grafting from”) has emerged as an alternative to the “grafting to” method. Particularly, site-specific in situ growth of polymer bioconjugates (SIP) is promising in overcoming the limitations of the “grafting to” method. In this review, we introduce the chemistry for synthesis of well-defined protein–polymer conjugates, and emphasize the SIP method as the next-generation platform for synthesis of well-defined protein–polymer conjugates. Furthermore, we exemplify biomedical applications of well-defined protein–polymer conjugates. In the end, we come up potential directions in this research field.
Aqueous Protein–Polymer Bioconjugation via Photoinduced RAFT Polymerization Using High Loading Heterogeneous Catalyst

Abstract:Light-driven polymerization, such as photoinduced electron/energy transfer–reversible addition–fragmentation chain transfer (PET-RAFT) polymerization, enables biological benign conditions and versatile functional polymer structure design, which is readily used in protein–polymer bioconjugates. However, conventional metalloporphyrinic homogeneous catalysts for PET-RAFT polymerization suffer from limited aqueous solubility and tedious purification. Here we demonstrate the design of PET-RAFT photocatalyst from the reticular assembled Zr–porphyrinic metal–organic frameworks (MOFs), along with a biomacromolecule-based chain transfer agent, as efficient bioconjugation tools in water. Our methodology offers manufacturing advantages on bioconjugates under mild conditions such that MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) and cytotoxicity assays have shown the preservation of the protein integrity, bioactivity, and high cell viability after PET-RAFT polymerization. We find that the fast kinetics are benefiting from the ultrahigh loading of metalloporphyrins in MOF-525-Zn. This heterogeneous catalyst also allows us to maintain living characteristics to incorporate myriads of monomers into block copolymers. Other advantages like easy postreaction purification, reusability, and high oxygen tolerance even in an open system are demonstrated. Abstract:Protein‐polymer conjugates and polymeric nanomaterials hold great promise in many applications including biomaterials, medicine or nanoelectronics. In this work, we present the first PISA (polymerization‐induced self‐assembly) approach performed in aqueous medium enabling protein‐polymer conjugates and nanoparticles entirely composed of amino acids by using ring‐opening polymerization (ROP). We indeed show that aqueous ROPISA can be used with protein or peptidic macroinitiators without prior chemical modification and afford the simple preparation of nanomaterials with protein‐like property, for example to implement biomimetic thermoresponsivity in drug delivery. Abstract:Abstract The immense application potential of amphiphilic protein-polymer conjugates remains largely unexplored, as established “grafting from” synthetic protocols involve time-consuming, harsh and disruptive deoxygenation methods, while “grafting to” approaches result in low yields. Here we report an oxygen tolerant, photoinduced CRP approach which readily affords quantitative yields of protein-polymer conjugates within 2 h, avoiding damage to the secondary structure of the protein and providing easily accessible means to produce biomacromolecular assemblies. Importantly, our methodology is compatible with multiple proteins (e.g. BSA, HSA, GOx, beta-galactosidase) and monomer classes including acrylates, methacrylates, styrenics and acrylamides. The polymerizations are conveniently conducted in plastic syringes and in the absence of any additives or external deoxygenation procedures using low-organic content media and ppm levels of copper. The robustness of the protocol is further exemplified by its implementation under UV, blue light or even sunlight irradiation as well as in buffer, nanopure, tap or even sea water.

Efficient Synthesis and Application of Protein-Poly(alpha-amino Acid) Conjugates

Abstract:Protein-polymer conjugates are important therapeutics for various diseases.There are currently two major challenges in this field:one is the search of new biodegradable polymers beyond traditional PEGylation,and the other is to develop highly efficient and site-specific conjugation strategy.Poly(α-amino acid)s (PαAAs) are biodegradable and biocompatible polymers with tunable properties and numerous functions,making them promising candidates for protein modification.In this review,we summarize our recent progresses in protein-PαAAs conjugates.Specifically,we discuss our developments in:(1) Recent developments in the controlled ring-opening polymerization (ROP) of α-amino acid N-carboxyanhydrides (NCAs),including amine-based initiators,organometallic initiators,organosilicon amines initiators and sulfide-based initiators.For instance,trimethylsilyl phenylsulfide (PhSTMS) is a novel initiator for controlled ROP of NCAs.It exhibits higher nucleophilicity than conventional amine-based initiator,and thus affords considerably higher chain initiation rate to ensure a more controlled polymerization.Moreover,this initiator is well-tolerated to various functional groups.(2) In situ functionalization of PαAAs for site-specific protein conjugation,and construction of various topological structures.Using PhSTMS initiator,it in situ generates a reactive phenyl thioester group at one end of the PαAAs,which can be used for protein N-terminus conjugation via native chemical ligation (NCL);moreover,ROP of glycine NCA yields oligoglycine at the other end of PαAAs,which can be used for C-terminus protein conjugation via sortase-A mediated ligation (SML).More interestingly,combinatory use of the two methods can construct various topological protein-PαAA conjugates including the head-to-tail circular conjugates.(3) Development of functional PαAAs for potential protein conjugation.Various functional PαAAs have been developed as delivery materials or hydrogels.To further expand the arsenal of PαAAs for potential modulation of protein functions,PαAAs that mimic protein post-translational modifications (PTM) are synthesized;On the other hand,a series of multiple stimuli-responsive PαAAs are also produced.These PαAAs show interesting enzyme,light,and/or thermal responsiveness,which could be potentially harnessed for modulation of protein functions in the future.

This approach has quickly become a powerful and versatile technique for the synthesis of a wide range of bespoke organic diblock copolymer nano-objects of controllable size, morphology, and surface functionality. Given its potential scalability, such environmentally-friendly formulations are expected to offer many potential applications, such as novel Pickering emulsifiers, efficient microencapsulation vehicles, and sterilizable thermo-responsive hydrogels for the cost-effective long-term storage of mammalian cells.

Abstract:Conventional methods for synthesizing protein/peptide-polymer conjugates, as a means to improve the pharmacological properties of therapeutic biomolecules, typically have drawbacks including low yield, non-trivial separation of conjugates from reactants, and lack of site- specificity, which results in heterogeneous products with significantly compromised bioactivity. To address these limitations, the use of sortase A from Staphylococcus aureus is demonstrated to site-specifically attach an initiator solely at the C-terminus of green fluorescent protein (GFP), followed by in situ growth of a stealth polymer, poly(oligo(ethylene glycol) methyl ether methacrylate) by atom transfer radical polymerization (ATRP). Sortase-catalyzed initiator attachment proceeds with high specificity and near-complete (≈95%) product conversion. Subsequent in situ ATRP in aqueous buffer produces 1:1 stoichiometric conjugates with >90% yield, low dispersity, and no denaturation of the protein. This approach introduces a simple and useful method for high yield synthesis of protein/peptide-polymer conjugates.
