Abstract:Asymmetric expansion: A catalytic asymmetric ring-expansion reaction of the title compounds occurs in the presence of a Sc(OTf)(3) catalyst bearing an N,N'-dioxide-based ligand. Highly functionalized 2-quinolone derivatives containing a chiral C4-quaternary stereocenter were obtained in high yields and high levels of selectivity under mild reaction conditions (see scheme; Tf=trifluoromethanesulfonyl).

Abstract:Herein, a general strategy for the regioselective dearomative 1,2-hydrocyanoalkylation of chromium-bound (hetero)arenes with simple alkylnitriles as pronucleophiles was disclosed, providing rapid access to 1,3-cyclohexadienes possessing useful alkylnitrile groups. The versatility of this methodology further enabled a selective dearomative deuteration reaction. Finally, synthetic applications of the method in the formal synthesis of natural products, including erysotramidine, demethoxyerythratidinone, and morphine, were demonstrated.

Abstract:α-Pyridones and α-pyrones are ubiquitous structural motifs found in natural products and biologically active small molecules. Here, we report an Rh-catalyzed electrochemical vinylic C–H annulation of acrylamides with alkynes, affording cyclic products in good to excellent yield. Divergent syntheses of α-pyridones and cyclic imidates are accomplished by employing N- phenyl acrylamides and N- tosyl acrylamides as substrates, respectively. Additionally, excellent regioselectivities are achieved when using unsymmetrical alkynes. This electrochemical process is environmentally benign compared to traditional transition metal-catalyzed C–H annulations because it avoids the use of stoichiometric metal oxidants. DFT calculations elucidated the reaction mechanism and origins of substituent-controlled chemoselectivity. The sequential C–H activation and alkyne insertion under rhodium catalysis leads to the seven-membered ring vinyl-rhodium intermediate. This intermediate undergoes either the classic neutral concerted reductive elimination to produce α-pyridones, or the ionic stepwise pathway to produce cyclic imidates.

Abstract:A novel Pd-catalyzed intermolecular dehydrogenative annulation of aryl iodides and aryl carbamic chlorides for the efficient synthesis of phenanthridinone derivatives was developed. Simple aryl iodides and carbamic chlorides readily made from various anilines, a broad substrate scope with hetero/polycycles, as well as high-value products, make this direct dehydrogenative annulation approach very practical and attractive.

Abstract:Two novel and synthetically useful reaction patterns of organolithium compounds with nitriles are reported to afford pyridine derivatives as the final products. Both 1-lithio-1,3-dienes and 1,4-dilithio-1,3-dienes, which can be easily generated in situ by lithiation of their corresponding iodo compounds, react with nitriles in the presence of HMPA to form substituted pyridines including 2,2'-bipyridines and tetrahydroisoquinolines in high yields.

Abstract:Transition-metal-catalyzed hydrofunctionalization of methylenecyclopropanes presents a useful but challenging transformation due to the complex selectivity and multiple reaction pathways. We describe herein an unprecedented highly efficient and selective palladium-catalyzed hydrocyanation of methylenecyclopropanes to give various 2-substituted allylic nitriles. Mechanistic studies demonstrated that the transformation may undergo Markovnikov-type hydrometalation and β-carbon elimination.

Abstract:Catalytic ring-opening/functionalization of unactive cyclopropanes has proven to be a significant but challenging task in organic synthesis. Herein, we disclose the photoredox and cobalt cocatalyzed ring-opening/acceptorless dehydrogenative functionalization of monodonor cyclopropanes, which provides a promising platform to achieve a sustainable and atom-economic approach to assemble allylic N -acyl-acetal derivatives. The reaction features mild conditions, broad substrate scopes, and excellent functional group compatibilities. The optimized conditions accommodate various cycloalkylamides and primary, secondary, and tertiary alcohols, with applications in late-stage functionalization of pharmaceutically relevant compounds, stimulating the further utility in medicinal chemistry. Abstract:An unprecedented silver-promoted regioselective (4 + 1) annulation of isocyanoacetates with pyridinium salts is reported. The established protocol provides controlled, facile, and modular access to a range of synthetically useful N-fused heterocyclic scaffolds containing indolizines, pyrrolo[1,2-a]quinolines, pyrrolo[2,1-a]isoquinolines, and 1H-imidazo[4,5-a]indolizin-2(3H)-ones. A mechanistic pathway involving nucleophilic addition/protonation/elimination/cycloisomerization is proposed.

Abstract:We have developed a Au-promoted Pd-catalyzed cyclization/cross-coupling of N,N-dimethyl-o-alkynylaniline with aryl iododes to synthesize 2,3-diarylindoles under mild and base-free conditions. A related vinyl-Au species has been isolated through Au-promoted cyclization of N,N-dimethyl-o-alkynylaniline and structurally characterized. Further study on its reactivity suggests the vinyl-Au species might be out of catalytic cycle, and PhPd(OTf)(PPh3)(2) is probably the reaction intermediate. (C) 2020 Published by Elsevier Ltd.

Abstract:A Pd‐catalyzed synthesis of 5‐iminopyrrolones from propiolamides was developed, which featured wide substrate scope and good yields. This efficient conversion involved a tandem hydrocyanation, E to Z isomerization of alkenes, and nitrile/amide cyclization. Control experiments show the intermediacy of β‐cyanation products, and the Z‐isomers was efficiently converted to the target products by Pd/L2 catalysis only after the cyanation reagent was completely consumed.

Abstract:A useful synthesis of five-membered N-heterocycles has been developed through an iridium-catalyzed domino-ring-opening cyclization of vinylaziridines with β-ketocarbonyls. α-Substituted 1,3-dicarbonyls reacted with vinylaziridines to give 2-methylenepyrrolidines bearing two adjacent sp3-carbon centers with moderate to excellent diastereoselectivity, while the reaction of α-unsubstituted 1,3-dicarbonyls afforded 2-pyrrolines in good yield.

Abstract:Rh-catalyzed C-H activation of phenacyl phosphoniums in coupling with alpha-diazocarbonyl compounds has been realized with the assistance of a mutifunctional phosphonium ylidic directing group, providing expedient accesses to 1-naphthols and isocoumarins. Switchable synthesis of 1-naphthols and isocoumarins was realized by substrate control, where these transformations were enabled by initial C-H activation and subsequent intramolecular Wittig reaction or nucleophilic C-O formation.

Abstract:Multisubstituted 2-pyridones are prevalent in pharmaceuticals and bioactive molecules. We herein report an efficient and regioselective approach for the synthesis of trisubstituted 2-pyridone derivatives by a rhodium-catalyzed C–H activation/Lossen rearrangement/cyclization cascade reaction between acrylamides and propargyl alcohols. The desirable features of this protocol include a reusable catalytic system, high regioselectivity, uncommon Lossen rearrangement, good functional group tolerance, operation at room temperature, simple purification by filtration in most cases, and scale-up synthesis with as low as 1 mol% catalyst loading. Additionally, deuterium labeling and KIE assays were performed to investigate the reaction mechanism. The vital effect of the hydroxyl group on propargyl alcohols in determining the regioselectivity was demonstrated by control experiments and DFT calculations. In addition, Mulliken atomic charge analysis of the key intermediates was also carried out to probe the origin of the observed preference for the Lossen rearrangement.

Abstract:We presented a cascade transformation of N, N-diallylamines and fluoroalkyl iodides into various functionalized fluoroalkylated pyrrolidines through a visible light-induced synthetic process in the solvent-free conditions. In this reaction system, the substrate N, N-diallylamine acted both as the base and the electron donor. We further demonstrated the practicality of this protocol by the direct modification of amino acids and pharmaceutical molecules.
