The Advances of Boron Neutron Capture Therapy for Glioblastoma

Abstract:Boron neutron capture therapy (BNCT) kills the tumor cells specifically while sparing the normal tissue in theory, even if the cells have spread to the surrounding normal brain microscopically. The tumor cell selective irradiation is dependent on the nuclear reaction between the isotope boron (10B) and thermal neutrons. It releases α particle and 7Li particle, which has a damage scope of 9 μm length. The reason that the research on boron neutron capture therapy (BNCT) has progressed relatively slowly is the lack of new boron delivering vechicles. The development of the best dosing paradigms for both boronophenylalanine (BPA) and sodium borocaptate (BSH) still have not been optimized. Another major problem is the full dependence on nuclear reactors as neutron sources for BNCT. The therapeutic effectiveness of BNCT will be enhanced in the future according to advanced researches of tumor targeting boron compounds, optimized administration methods, improved boron drug delivery systems, development of a hospital neutron source, and other combination modalities.
