Safety Assessment of Two Chemotype Rosemary Essential Oils

Abstract:Both two types of Rosemary essential oils, chemotype of verbenone and 1,8-cineole, are now being widely used, although their safety have not been reported. In this current study, targeting their safety, we carried out our experimental assessment, including acute oral toxicity test, acute skin irritation test and multiple skin irritation, as well as acute eye irritation test. The gavages method operated under the equal concentration ratio, and LD50 was calculated by Korbor. The experimental results indicated that, LD50 of chemotype of verbenone and 1, 8-cineole are 7 220.35 and 8 088.24 (mg·kg-1) respectively, and their confidence interval on 95 percent level are 5 172.53 to 8 124.41 (mg·kg-1), and 6 982.70 to 9 842.84 (mg·kg-1) respectively. In the acute skin irritation test, Rosemary essential oil with chemotype of verbenone is mild irritant at concentration of 2.3%, chemotype of 1, 8-cineole belong to moderate irritant at concentration of 100% , and the former irritation was significantly higher than the latter under the same concentration. As far as the multiple skin irritation, both of the two chemotypes belong to mild irritant, and the former irritation was significantly higher than the latter under the same concentration. With regard to the acute eye irritation, both chemotypes are nonirritant under the concentration of 50 percent.
