Abstract:Our research focused on assessing essential oils (MSEO) and aqueous extracts (MSAE) derived from M. subtomentella leaves, with a primary focus on evaluating their properties. From 1 kg of leaves, we successfully obtained 18 mL of essential oil. Upon conducting GC/MS analysis, we identified eleven compounds within the oil, collectively accounting for 100% of the constituents identified. Notably, the predominant compounds in the leaf oil were p-Menth-48) -en-3-one (50.48%), 9-Ethylbicyclo (3.3.1) nonan-9-ol (10.04%) (E)-3,3-Dimethyl-delta-1, alpha-cyclohexaneacetaldehyde (8.53%), and D-Limonene (7.22%). Furthermore, utilizing HPLC/DAD, we explored the phenolic profile of MSAE, extracted through decoction. This analysis revealed the presence of fifty-eight compounds, with five major components collectively constituting 61% of the total compounds identified, rosmarinic acid as the major one. We evaluated the antimicrobial effectiveness of the MSEO against ten different strains, observing its notable efficacy against A. Niger (MIC = 0.09%), P. digitatum (MIC = 0.5%), and G. candidum (MIC = 1%). However, the essential oil demonstrated comparatively lower efficacy against bacteria than fungi. In contrast, the MSAE did not exhibit any antimicrobial activity against the tested strains. Regarding antioxidant activity, the aqueous extract displayed a significantly higher antioxidant capacity than the essential oil, which exhibited relatively lower antioxidant activity. The IC 50 values were determined to be 0.04 ± 0.01 mg/mL, 0.17 ± 0.01 mg/mL, and 13% ± 0.01% (V/V), for ascorbic acid MSAE and MSEO, respectively. We used a computational method called molecular docking to investigate how certain plant compounds affect antioxidant, antibacterial, and antifungal activities. This involved analyzing the interactions between these compounds and specific protein targets known for their roles in these activities.

Abstract:The present work was carried out to evaluate the phenolic compounds, the antibacterial, antifungal and antioxidant activities of essential oil (EO) and aqueous extract (AE) of Lavandula angustifolia L. The extracts were obtained by hydrodistillation, and by decoction method using water, then, total phenolics, flavonoids and condensed tannins were determined using Folin-Ciocalteu, Aluminium chloride, and Bate-Smith reaction methods, respectively. Afterward, the antibacterial activity was evaluated using the borth microdilution and, the antifungal activity was discussed using dilution technique in solid medium. To estimate the antioxidant effect of these extracts, the Total Antioxidant Capacity and DPPH scavenging methods were performed. Results of phytochemical analysis of AE showed that total phenolics, flavonoids and condensed tannins were 67.967 ± 0.139 mg GAE/g DW, 53.578 ± 0.418 mg QE/g DW and 9.194 ± 0.323 mg/g DW, respectively. In addition, spectral analysis by Gas Chromatography of EO showed the presence of four main components in oil lavender: Linalool (29.95%), Linalyl acetate (18.86%), ρ-cymene (14.68%), and α-Campholenal (10.26%). Furthermore, the antibacterial test showed that Staphylococcus aureus and Pseudomonas aeruginosa were the most resistant compared to the other tested strains. Also, it appears that EO had greater antifungal activity compared to AE, and the both showed significant antioxidant potential. These eminent results showed that the use of L. angustifolia L. as an anti-infective in traditional environments is justified and that it should be studied more widely in order to explore its potential.
Comparison of quantity, quality and antibacterial activity of essential oil Mentha longifolia (L.) L. under different traditional and modern extraction methods

Abstract:Extraction is the first and most important step in obtaining the effective ingredients of medicinal plants. Mentha longifolia (L.) L. is of considerable economic importance as a natural raw material for the food and pharmaceutical industries. Since the effect of different extraction methods (traditional and modern methods) on the quantity, quality and antimicrobial activity of the essential oil of this plant has not been done simultaneously; the present study was designed for the first time with the aim of identifying the best extraction method in terms of these features. For this purpose, extracting the essential oil of M. longifolia with the methods of hydrodistillation with Clevenger device (HDC), steam distillation with Kaiser device (SDK), simultaneous distillation with a solvent (SDE), hydrodistillation with microwave device (HDM), pretreatment of ultrasonic waves and Clevenger (U+HDC) and supercritical fluid (SF) were performed. Chemical compounds were identified by gas chromatography coupled with mass spectrometer (GC-MS). Antimicrobial activity of essential oils against various clinical microbial strains was evaluated by agar diffusion method and determination of the minimum inhibitory concentration and minimum bactericidal concentration (MIC and MBC). The results showed that the highest and lowest yields of M. longifolia leaf essential oil belonged to HDC (1.6083%) and HDM (0.3416%). The highest number of compounds belonged to SDK essential oil and was equal to 72 compounds (with a relative percentage of 87.13%) and the lowest number of compounds was related to the SF essential oil sample (7 compounds with a relative percentage of 100%). Piperitenone (25.2-41.38%), piperitenone oxide (22.02-0%), pulegone (10.81-0%) and 1,8-cineole (5-35.0%) are the dominant and main components of M. longifolia essential oil were subjected to different extraction methods. Antimicrobial activity results showed that the lowest MIC value belonged to essential oils extracted by HDM, SDK, SDE and U+HDC methods with a value of 1000 μg/mL was observed against Gram-negative bacteria Shigella dysenteriae, which was 5 times weaker than rifampin and 7 times weaker than gentamicin. Therefore, it can be concluded that in terms of efficiency of the HDC method, in terms of the percentage of compounds of the HDM method, and in terms of microbial activity, the SDK, HDM and U+HDC methods performed better.
Evaluation of Antimicrobial Potential and Comparison of HPLC Composition, Secondary Metabolites Count, and Antioxidant Activity of Mentha rotundifolia and Mentha pulegium Extracts

Abstract:In the present study, the relationship between the phenolic counts, chemical composition, and biological activities of two Mentha species (Mentha rotundifolia (MR) and Mentha pulegium (MP)) was analyzed. The characterization of the action mode against pathogenic bacteria and the inhibition of spore germination of two fungal species using prepared methanolic extracts were studied here for the first time. The obtained data highlighted the presence of positive correlation between the secondary metabolites contents and the biological activities of the investigated extracts. In fact, HPLC analysis showed that the major components in both the extracts were eriocitrin and rosmarinic acid (25 and 20 mg/ml and 12 and 8 mg/ml in methanolic extracts of MR and MP, respectively). Moreover, the MR extract was rich in polyphenols and presents the highest antioxidant activity than MP ones. In addition, both extracts possess an antimicrobial activity against four Gram-positive and five Gram-negative bacteria and one yeast species (Candida albicans) and were able to inhibit the spore germination of two fungi species (Aspergillus niger and Aspergillus flavus). But, the significant activity was observed in the presence of MR methanolic extract. The effect of time on cell integrity of E. coli and L. monocytogenes determined by time-kill and bacteriolysis assays showed that the MR extract had a rapid bacteriolytic effect compared to the MP extract, and their capacities were significant against Gram-negative bacteria than positive ones. Based on the obtained data, it can be concluded that Saudi Mentha species have high pharmacological and industrial importance and they can be used in preparation of food or drugs.

Abstract:Abstract The present research work investigated antioxidant, antibacterial, and antifungal properties of essential oils from the leaves of Mentha rotundifolia var. apodysa Briq. (EOR). Hydro-distillation was used to extract EOR before being subjected to the chemical characterization by the use of GC/MS. Antioxidant activity was assessed by the use of three bioassays namely 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and total antioxidant capacity (TAC). Antimicrobial potency was tested against gram-negative and gram-positive bacteria namely Staphylococcus aureus, Streptococcus pneumoniae , Escherichia coli, Acinetobacter baumannii, and Klebsiella pneumonia , while antifungal activity was tested against Aspergillus niger, Candida albicans, Aspergillus flavus, and Fusarium oxysporum . EOR yield was determined to be 1.31%, with 20 compounds wherein Menthol (31.28%) and Isomenthol (14.28%) constituted the greatest amount. Regarding antioxidant activity, EOR exhibited potent antioxidant power: DPPH (IC 50 value of 0.36 ± 0.03 mg/mL), FRAP (EC 50 value of 0.35 ± 0.03 mg/mL), and TAC (697.45 ± 1.07 mg EAA/g). Antibacterial activity results showed that EOR had broad antibacterial activity on the tested strains. Eventually, EOR resulted in the greatest inhibition zone diameters vs S. aureus (18.20 ± 0.41 mm) followed by E. coli (17.02 ± 0.5 mm). Antifungal activity results showed that EOR exhibited potent antifungal activity and resulted in the greatest inhibition zone diameters up to 51.32 ± 1.32 mm against Aspergillus flavus , and 34.51 ± 1.07 mm against Aspergillus niger.

Abstract:Our study evaluated the in vitro antioxidant properties, antibacterial and antifungal activities, anti-inflammatory properties, and chemical composition of the essential oils (EOs), total phenol, and total flavonoid of wild Mentha pulegium L. This study also determined the mineral (nutritional and toxic) elements in the plant. The EOs were extracted using three techniques—hydro distillation (HD), steam distillation (SD), and microwave-assisted distillation (MAD)—and were analyzed using chromatography coupled with flame ionization (GC-FID) and gas chromatography attached with mass spectrometry detector (GC-MS). The antioxidant effects of the EOs were tested with 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ABTS (2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), while the antibacterial and antifungal activities of the EO and methanolic extract were tested using Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Escherichia coli, and Candida albicans. Twenty-six compounds were identified in the essential oil, representing 97.73% of the total oil, with 0.202% yield. The major components were pulegone (74.81%), menthone (13.01%) and piperitone (3.82%). Twenty-one elements, including macro- and micro-elements (Ba, Br, Ca, Cl, Co, Cr, Cs, Eu, Fe, K, Mg, Mn, Mo, Na, Rb, Sb, Sc, Sr, Th, U and Zn), were detected using neutron activation analysis (INAA) and inductively coupled plasma optical emission spectrometry (ICP-OES), with the concentration of mineral element close to the FAO recommendation. The results show that the EOs and extracts from Mentha pulegium L. had significant antimicrobial activities against the microorganisms, including five human pathogenic bacteria, one yeast (Candida albicans), and one phytopathogenic fungi. The in vivo anti-inflammatory activities of the leaf extracts were confirmed. The results indicate that the EOs and extracts from Mentha pulegium L. have promising applications in the pharmaceutical industries, clinical applications, and in medical research.

Abstract:The chemical composition and antibacterial, insecticidal, and antioxidant properties of the essential oil from Mentha pulegium L. (M. pulegium) growing in Morocco were investigated in this work. To achieve this goal, the oils were obtained by using hydrodistillation before being characterized by GC-MS. The antibacterial and antifungal activities were conducted against pathogenic strains using the disc diffusion and MICS bioassays. The insecticidal activity was carried out versus C. maculatus using contact and inhalation tests. The antioxidant activity was performed by using DPPH and total antioxidant capacity bioassays. The chemical analysis of the oil showed that 20 compounds were identified, which represented 98.91% of the total oil. In the oil, the main components detected were R-(+)-pulegone (76.35%), carvone (5.84%), dihydrocarvone (5.09%), and octanol-3 (2.25%). The essential oil has moderate-to-strong broad-spectrum antibacterial and antifungal properties; the results showed that B. subtilis was the most sensitive strain to M. pulegium oil, with the largest inhibition diameter (25 ± 0.33). For the antifungal activity, the results obtained indicated that Aspergillus niger was the most sensitive fungal strain to M. pulegium oil with an inhibition percentage up to 100%. Regarding the insecticidal activity, the inhalation test showed a high efficacy (100% mortality), and a lethal concentration of LC50 = 1.41 + 0.48 μL/L air was obtained after 24 hours of exposure. Moreover, the contact test showed that a total reduction in fertility and emergence was obtained with a dose of 20 μL/mL of acetone. Regarding the antioxidant activity, the sample concentration necessary to inhibit 50% of HE radicals (IC50) was 7.659 mg/mL (DPPH) and 583.066 57.05 mg EAA/g EO (TAC).

Abstract:Mentha piperita and Lavandula multifida are widely used in Moroccan traditional medicine for the treatment of diabetes and infectious diseases. The aims of this work were the determination of the chemical composition of Mentha piperita (MPEO) and Lavandula multifida (LMEO) essential oils and the evaluation of their antibacterial, antioxidant, and antidiabetic activities. The chemical composition was determined by GC-MS analysis. The antibacterial effects were evaluated against several bacterial strains using disc diffusion, MIC, and MBC methods. The antioxidant activity was evaluated in vitro using DPPH, H2O2, and xanthine oxidase, and the antidiabetic activity was estimated by the inhibitory effects of α-amylase, α-glucosidase, and lipase activities. GC-MS results showed that the main compounds of MPEO were menthone (29.24%), levomenthol (38.73%), and eucalyptol (6.75%). However, eucalyptol (28.11%), 2-bornanone (11.57%), endo-borneol (7.82%), and linalyl acetate (5.22%) are the major compounds of LMEO. The results exhibited important inhibitory effects against some bacterial strains with MIC = MBC = 0.39 mg/mL for MPEO against Staphylococcus aureus ATCC. However, LMEO exhibited remarkable antioxidant and antidiabetic activities compared to MPEO. Indeed, LMEO inhibited DPPH, H2O2, and xanthine oxidase with concentrations of 15.23, 21.52, and 8.89 μg/mL, respectively. Moreover, LMEO exhibited α-amylase and α-glucosidase at IC50 = 85.34 and IC50 = 59.36 μg/mL, respectively. The findings showed that both MPEO and LMEO exhibit promising biological properties. However, the application of these species or their main bioactive compounds requires further investigation.

Abstract:The volatile profile of Mentha longifolia was observed using GC-MS, GC-FID, FT-IR and 13 CNMR. Twenty-two constituents were found to be present in this essential oil, accounting 96.04 (%) of total essential oil and oxegenated monoterpenes were major class of compounds. The key constituents of this essential oil were pieritenone oxide (45.9%), piperitone (17.5%), beta caryophyllene (10.2%), and Germacrene D (5.0%). FT-IR showed peak at 1669 and 1707 cm −1 which may be due to the presence of carbonyl groups. Among the tested compounds, Germacrene D showed highest binding affinity value of −6.8 kcal mol −1 and a pKi value of 6.01. The dsc studies revealed that boiling point of this EO is above 200 °C. Microplate Alamar Blue assay (MABA) was carried out for the assessment of antimycobacterial activity using isoniazid and nicotinic as reference compound and oil was found to be active within conc. range of 0.8–1.6 μg/mL against mycobacterium tuberculosis, hence can act as a potential candidate against antituberculosis

Abstract:The species chosen for this study is sweet mint, Mentha spicata L, harvested in the wilaya of El Bayadh in Algeria. The aim of this work is to highlight the plant's identification criteria, its yield in essential oil, the various physico-chemical parameters and the composition of the essential oil using different techniques (determination of several chemical indices and establishment of the list of constituents of the essential oil using chromatographic methods). The botanical study showed the presence of two types of "Lamiaceae" type secretory hairs. These secretory hairs are the histological support for the production and storage of essential oils. A study of the kinetics of the extraction yield shows that distillation is rapid from 0 to 120 minutes, when the yield reaches an optimum (1.08%). The organoleptic characteristics and physico-chemical indices obtained are in line with the standards given in the bibliography. Thin layer chromatography revealed the presence of several constituents through the appearance of several spots of different colour and Rf, three of which were identified by comparison with controls, namely menthol, cineol and carvone.
Stage-dependent restoration of sensory dorsal columns following spinal cord transection in anuran tadpoles

Abstract:In frogs sensory axons from the lumbar dorsal roots ascend in the dorsal column of the spinal cord to terminate in the medulla and cerebellum. The response of these axons to complete transection of the thoracic spinal cord has been analysed in Rana temporaria tadpoles at different stages of development. The presence and position of dorsal column axons were assessed by using the anterograde transport of horseradish peroxidase or by electrophysiological methods. Before developmental stage VIII, dorsal column axons can grow across the transection and reach their normal areas of termination in the brainstem. Axons that do cross the transection follow their normal pathways. From stage VIII onwards this capacity for growth is largely lost. These results are discussed in terms of the relation between neurogenesis, axon growth and axonal regeneration.
Variability in Chemical Composition and Antimicrobial Activity of Mentha pulegium L. Essential Oil, Cultivated under Different Plant Growth Promoting Rhizobacteria

Abstract:The aim of present work was to study the essential oil chemical composition and antimicrobial activity of cultivated Mentha pulegium L. under different plant growth promoting rhizobacteria (Pseudomonas fluorescens, Bradyrhizobium sp. and Sinorhizobium meliloti) individually and in consortium. Yield, in plants inoculated with Bradyrhizobium sp. and S. meliloti in consortium, increase significantly relative to control plants. GC and GC/MS analyses pointed to a qualitative and quantitative variability of components. The investigated essential oils were clustered into three chemotypes: piperitenone/1,8-cineol (40.9/29.4 %) chemotype in plants inoculated with Bradyrhizobium sp. individually, S. meliloti individually, and Bradyrhizobium sp. and S. meliloti in consortium, piperitone/menthone (41.8/33.8 %) chemotype in plants inoculated with P. fluorescens individually, P. fluorescens and Bradyrhizobium sp. in consortium, and P. fluorescens and S. meliloti in consortium and pulegone/menthol (47.9/31.5 %) chemotype in control plants. The antimicrobial activity, carried out by the disc diffusion method and the determination of the Minimum Inhibitory Concentration (MIC) against ten microorganisms, varied significantly according to the tested microorganism and the rhizobacterial species used individually or in consortium (inhibition zone: 8.5-33.5 mm; MIC: 0.25-2.5 μL/mL). Our findings provided useful indications to select interesting chemotype within M. pulegium, especially in perspective of its cultivation.
Separation and Qualitative Study of Mentha asiatica Boriss. Essential Oil Components by prep-GC, GC-QTOF-MS, NMR and Prediction of Bioactive Compounds

Abstract:In order to understand the material basis of wild Mentha asiatica Boris. in Xinjiang, the chemical constituents of essential oil extracted from aerial parts of this plant were studied. A total 52 components were detected and 45 compounds were identified. First of all, the essential oil was separated by silica gel column chromatography, and divided into several parts according to the results of thin layer chromatography. Eight fractions were obtained, and then each fragment was preliminarily screened for antibacterial activity. It was found that all eight fragments had certain antibacterial activity in different level. Then the fractions were subjected to preparative gas chromatography (prep-GC) for further isolation. Ten compounds were identified by 13 C-NMR, 1 H-NMR and gas chromatography-quadrupole time of flight-Mass spectrometry (GC-QTOF-MS). They are sabinene, limonene and β-caryophyllene, (1R*,3S*,5R*)-sabinyl acetate, piperitone oxide, rotundifolone, thymol, piperitone, 4-hydroxypiperiditone, cedrol. After screened by bioautography, 4-hydroxypiperone and thymol were showed best antibacterial activity. The inhibitory effects of the two isolated compounds on Candida albicans and their related mechanisms were studied. The results showed that, 4-hydroxypiperone and thymol significantly reduced ergosterol content on the surface of Candida albicans cell membrane in a dose-dependent manner. Abstract:This work aims to add value to the Lavandula genus by identifying the chemical composition, antioxidant, and antimicrobial activities of two species lavender from Oulmès in Morocco; Lavandula abrialis and Lavandula stoechas . The uniqueness lies in the integrated approach that combines in vitro and in silico analyses to assess the biological properties of the essential oils (EO). The objective of this study is to enhance the significance of the Lavandula genus by analyzing the chemical composition, antioxidant properties, and antimicrobial effects of two lavender species found in Oulmès, Morocco: Lavandula abrialis and Lavandula stoechas. The distinctiveness is in the comprehensive methodology that merges in vitro and in silico investigations to evaluate the biological characteristics of the essential oils (EO). The extraction of essential oils (EO) by hydrodistillation from the aerial parts of Lavandula abrialis gave a high yield of essential oils (2.9%) compared to Lavandula stoechas (2.3%). A GC-MS analysis of the chemical composition revealed 56 chemical compounds, with some variation in the predominant components, representing between 99.98% and 100% of the EOs of the studied lavenders. Their antioxidant activity was assessed using the DPPH test. This method revealed that L. stoechas EO has a higher percentage of free radical inhibition than L. abrialis. The IC 50 values demonstrate that the antioxidant activity of ascorbic acid is higher (1.62 g/mL) than the EOs of tested plants. Noteworthy, the EO of L. stoechas is more potent (12.94 g/mL) than that of Lavandula tibialis (34.71 g/mL). Regrading, the antibacterial tests, the EO of L. abrialis was particularly active against Staphylococcus aureus BLACT, which is inhibited at a concentration of 6.25 g/mL, while L. stoechas EO has a strong effect on Escherichia coli , with a MIC of 1.56 g/mL. Concerning the antifungal activity of the EOs, yeasts showed sensitivity toward EOs extracted from both L. tibialis and L. stoechas . Moreover, an in silico study was conducted targeting sarA protein of S. aureus (PDB ID: 2fnp) and NADPH oxidase from Lavandula sanfranciscensis (PDB: 2CDU) and results showed that Ishwarone and Selina-3,7 (11)-diene exhibited highest binding energy with −9.8 and −10.8 kcal/mol respectively. Therefore, these two compounds could be used as an antibacterial and antioxidant agents however more experimental and molecular study should be required.

Abstract:Nowadays, antibacterial resistance to synthetic antibiotics has become a big issue worldwide. Mentha piperita essential oil (EO) and its bioactive components with antibacterial, antibiofilm, and antioxidant properties are recommended for the treatment of microbial infections in traditional medicine. This study aimed to detect phytochemical components, antioxidant and cytotoxic activity; to evaluate antibacterial and antibiofilm effects of M. piperita EO on strains. Gas chromatography/mass spectrometry (GC-MS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests were used to determine chemical composition, antioxidant and cytotoxic potentials of EO, respectively. Antibacterial activity was carried out through disc diffusion and minimum inhibitory concentration (MIC) assays. The biofilm biomass of bacteria was determined by the crystal violet (CV) staining method. Menthone (27.28%), L-menthol (23.71%), and limonene (7.48%) were the main components. M. piperita EO was found to have strong antibacterial activity with the MIC-MBC values in the range of 0.125-16.00% (v/v) and moderate antioxidant activity. The peppermint EO showed an antibiofilm effect on all strains and a cytotoxic effect on NIH 3T3 cells. M. piperita EO may be used alone or in combination with antibiotics or carrier oils as a therapeutic agent against bacterial infections. Further in vivo experiments should be carried out to detect a safe dosage of EO.

Abstract:This study aimed to evaluate the effect of extraction with ultrasound-assisted hydrodistillation (UAHD) and hydrodistillation (HD) extraction, on the yield, chemical composition, and biological activities of essential oil (EO) from Mentha rotundifolia . The yield, the composition, and the biological activities of the obtained essential oils were evaluated. The chemical composition of EOs was established by GC-MS. The antibacterial activity was conducted against various pathogenic and beneficial bacteria. The antioxidant activity of EOs was evaluated using DPPH, ABTS, galvinoxyl radical scavenging, reducing power, and CUPRAC assays. Anti-acetylcholinesterase and anti-butyrylcholinesterase inhibitory effects were examined. The results indicated that the extraction yield of 63.83% has been recorded using the UAHD. The GC/MS analysis showed that piperitenone oxide (21.53%) and camphor (26.52%) were the major compounds of the EO extracted by UAHD, while piperitenone oxide (22.60%) and trans-caryophyllene (21.79%) were found to be the predominant compounds of the EO extracted by HD. M. rotundifolia EO extracted by UAHD presented an eminent potency on pathogenic bacteria with inhibition diameters and MIC values ranging from 11.5 to 29.33 mm and from 3.9 to 125 μg/mL, respectively, while exhibiting the lowest activity on beneficial bacteria. The EO from UAHD showed a high acetylcholinesterase and butyrylcholinesterase inhibitory activity (IC 50 = 96.64±0.64 and 23.47±0.12, respectively). EO from UAHD disclosed a potent antioxidant ability compared with the EO obtained by HD. These data showed the possibility of using UAHD to enhance the extraction yield from plants.

Abstract:In the current study, the lipophilic solvent (n‐hexane) extraction, steam distillation (SD), and supercritical CO2 fluid extraction (SC–CO2) methods were used to extract the volatile components of Mentha asiatica Boriss. Comparisons were made between the extraction yield, chemical component and biological activity. The essential oil extracted by the SD method has a high total content of identified active components, good free radical scavenging ability and high‐antibacterial activity. Therefore, optimizing the extraction conditions of SD is of great significance for the extraction and application of Mentha asiatica Boriss. essential oil. In the current study, the lipophilic solvent (n‐hexane) extraction, steam distillation (SD) and supercritical CO2 fluid extraction (SC–CO2) methods were used to extract the volatile components of Mentha asiatica Boriss. Comparisons were made between the extraction yield, chemical component and biological activity. Gas chromatography quadrupole‐time‐of‐flight mass spectrometry (GC–QTOF–MS) and semi‐quantitative analysis of chemical contents by flame ionization detector (GC–FID) were used to study the chemical ingredients in Mentha asiatica Boriss. aromatic extract. The oil extract obtained from lipophilic solvent (n‐hexane) extraction (1.27 ± 0.03%, w/W) showed the highest yield, followed by supercritical CO2 fluid extraction (1.15 ± 0.04%, w/W), the extraction rate of SD was (0.37 ± 0.01%,w/W). The oil extract of Mentha asiatica Boriss. contained a total of 70 components that were identified. In the following, the primary volatile compounds of the essential oil produced using the three extraction techniques: α‐thujene, camphene, sabinene, β‐pinene, β‐cymene, limonene, β‐terpinene, camphol, α‐terpinene, carvenone, carvone, piperitone oxide, thymol, myrtenyl acetate, dihydrojasmone, caryophyllene, germacrene D and caryophyllene oxide. Findings from this study indicate that the three methods of essential oil extraction produced oils with varying antioxidant capacities against DPPH and ABTS free radicals. Against Staphylococcus aureus, Candida albicans, Bacillus subtilis and Escherichia coli, all of the essential oils simultaneously showed clear inhibitory actions. Response surface methodology (RSM) and the Box–Benhnken design with three factors and three levels were both utilized to optimize the distillation process of essential oils. The appropriate extraction parameters were a 3 h extraction period, a solid–liquid ratio of 1:10 and a 3 h soaking time. The highest yield of essential oil was 0.38%. The impact of extraction parameters on essential oil yield was examined using the SD method. The findings indicated that the main factors affecting the yield of volatile oil extracted by SD were soaking time > ratio of solid to liquid > extraction time.

Abstract:The aim of the present study was to investigate chemical composition and antibacterial activity of essential oil from the leaf of Mentha spicata plant against common food-borne pathogenic bacteria ( Staphylococcus aureus, Bacillus subtilis, Bacillus cereus, Listeria monocytogenes, Salmonella typhimurium , and Escherichia coli O157:H7). Chemical composition of the essential oil was identified by gas chromatography coupled with mass spectrometer detector (GC-MS). The antibacterial activity of the essential oil was evaluated by broth microdilution method and agar disk diffusion assay. According to the result of GC-MS analysis, 18 components were identified, accounting for 99.89% of the whole essential oil. The main components were carvone (78.76%), limonene (11.50%), β -bourbonene (11.23%), cis -dihydrocarveol (1.43%), trans -caryophyllene (1.04%), menthone (1.01%), menthol (1%), and terpinen-4-ol (0.99). The essential oil exhibited moderate level of antibacterial activity against all test microorganisms. In general, Gram-positive bacteria were more susceptible to M. spicata essential oil than Gram-negative bacteria. L. monocytogenes was the most sensitive of the microorganisms to the antibacterial activity of M. spicata essential oil (inhibition zone = 22 mm and MIC and MBC = 2.5 μ L/mL). Based on our results, the essential oil of M. spicata plant collected from Kermanshah province, west of Iran, has a potential to be applied as antibacterial agent.
