Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.

Abstract:To improve the properties of chitosan,this research used chitosan and chloroacetic acid to synthesize a new type chitosan derivative-N,O-carboxymethyl chitosan.The experiment results(orthogonal test)showed that the optimum condition for making N,O-carboxymethyl chitosan was:reaction temperature 70℃,the mole ratio of sodium hydroxide and chloroacetic acid was 2.4,reaction time was 4h,amount of isopropyl alcohol was 50ml.Analyzed the chemical and physical properties of N,O-carboxymethyl chitosan,the result showed that the isoelectric point of N,O-CMC migrated toward the acid with the increasing degree of substitution;N,O-carboxymethyl chitosan with high degree of substitution had a better moisture content and superior moisture absorption than hyaluronic acid.In the condition of high humidity(RH81%),the hygroscopicity of N,O-CMC with 1.36 degree of substitution was 1.8 times of hyaluronic acid.

Abstract:Chitosan, which is derived from chitin, is the only known natural alkaline cationic polymer. Chitosan is a biological material that can significantly improve the living standard of the country. It has excellent properties such as good biodegradability, biocompatibility, and cell affinity, and has excellent biological activities such as antibacterial, antioxidant, and hemostasis. In recent years, the demand has increased significantly in many fields and has huge application potential. Due to the poor water solubility of chitosan, its wide application is limited. However, chemical modification of the chitosan matrix structure can improve its solubility and biological activity, thereby expanding its application range. The review covers the period from 1996 to 2022 and was elaborated by searching Google Scholar, PubMed, Elsevier, ACS publications, MDPI, Web of Science, Springer, and other databases. The various chemical modification methods of chitosan and its main activities and application research progress were reviewed. In general, the modification of chitosan and the application of its derivatives have had great progress, such as various reactions, optimization of conditions, new synthetic routes, and synthesis of various novel multifunctional chitosan derivatives. The chemical properties of modified chitosan are usually better than those of unmodified chitosan, so chitosan derivatives have been widely used and have more promising prospects. This paper aims to explore the latest progress in chitosan chemical modification technologies and analyze the application of chitosan and its derivatives in various fields, including pharmaceuticals and textiles, thus providing a basis for further development and utilization of chitosan.

Preparation and Characterization of N-succinyl-chitosan

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.

Abstract:Chitosan oligosaccharide (COS), which represent the positively charged basic amino oligosaccharide in nature, is the deacetylated and degraded products of chitin. COS has become the focus of intensive scientific investigation, with a growing body of practical and clinical studies highlighting its remarkable health-enhancing benefits. These effects encompass a wide range of properties, including antibacterial, antioxidant, anti-inflammatory, and anti-tumor activities. With the rapid advancements in chemical modification technology for oligosaccharides, many COS derivatives have been synthesized and investigated. These newly developed derivatives possess more stable chemical structures, improved biological activities, and find applications across a broader spectrum of fields. Given the recent interest in the chemical modification of COS, this comprehensive review seeks to consolidate knowledge regarding the preparation methods for COS derivatives, alongside discussions on their structural characterization. Additionally, various biological activities of COS derivatives have been discussed in detail. Lastly, the potential applications of COS derivatives in biomedicine have been reviewed and presented.

Abstract:Chitosan is a natural biopolymer with a modifiable structure and abundant function groups, moreover, it can be processed into various shapes and sizes, making it suitable for various applications. This review analyzed and summarized the recent advances in chitosan-based materials, particularly focusing on the modification of chitosan by physical, chemical and biological methods, which provided plentiful information on the preparation, characterization and application of chitosan-based materials. In this review, various preparation and modification methods for improving chitosan performances, as well as the application of chitosan and its derivatives, were critically analyzed and summarized in detail. Chitosan-based materials can be fabricated into different shapes and sizes, such as nanofiber, nanoparticles, microspheres, membranes, and 3D printed scaffolds. In addition, chitosan can be modified through physical, chemical, and biological modifications, to obtain versatile chitosan-based materials. For physical modification, crosslinking and blending are commonly used., while chemical modification is based on chemical reactions (e.g. acylation, esterification, etherification, N-alkylation, graft copolymerization, and degradation), to prepare the corresponding derivatives (e.g. chemically cross-linked chitosan, grafted chitosan, low molecular weight chitosan, and oligo-chitosan) through crosslinking, grafting, and degradation. The biological modification is associated with various enzymes to realize the hydrolysis or grafting. With improved performances through modification, as well as various shapes and sizes, chitosan-based materials show promising applications in versatile fields, such as water treatment, food processing, pharmaceutical/biomedicine, textiles and agriculture. This review will deepen the understanding the state of the art of chitosan-based materials, including their preparation, characterization and application.

Abstract:Because of the unique chemical structure, chitosan and its derivatives have been paid close extensive attention as the potential bio-functional material. This review presents recent synthesis of modified chitosan via N-substitution, O-substitution, free radical graft copolymerization and other modification methods and properties of the modified chitosan. The applications of the modified chitosan in metal ions adsorption, dye removal and pharmaceutical fields are illustrated as well. The rapid development in the modification of chitosan describes broad perspectives of the modified chitosan.

Abstract:Two kinds of chitosans, namely N-acetylated and N-deacetylated chitosan were prepared by the modified processes. They can dissolve in both acid and alkali solution. 13C NMR was used to study the basic solution of chitosan, and XRD, FT-IR and SEM were used to study the structure of N-acetylated and N-deacetylated chitosan. The result from X-ray diffraction showed that a transformation of crystal structure occurred during the N-acetylation or N-deacetylation process with the decrease of crystallinity and expansion of crystal lattices. FT-IR spectra revealed that the intermolecular and intramolecular hydrogen bonds were destroyed by both treatments and a looser structure was observed by the SEM. The lower crystallinity, the decreased intermolecular interactions, the more disordered and looser structure were easy for the permeation of LiOH/urea aqueous solution and coordinated with the breakage of intermolecular and intramolecular hydrogen bond by LiOH at low temperature, the prepared chitosans dissolved in LiOH/urea/H2O mixture.

Abstract:In view of rapidly growing interest in the amino polysaccharide chitosan/chitin and its oligosaccharides as functional biopolymer, a recent progress of basic and application studies in chitosan/chitin oligosaccharides chemistry is reviewed as well as some basic aspects of this specialty biomass resource. A special emphasis is placed on the controlled modification reactions to prepare chitin derivatives with well-defined structures and thereby to construct sophisticated molecular architecture having various advanced functions. The reactions discussed here include acylation, quaternary ammonium salt, carboxyalkylation, graft copolymerization, quaternary salt formation, Schiff base formation, reductive alkylation, microwave modification. For conducting modification reactions in a facile and controlled manner, some soluble chitosan/chitin oligosaccharides derivatives are convenient. To fully explore the high potential of these specialty biopolymers, basic and application researches are being made extensively.

Abstract:Chitosan was prepared by alkaline N-deacetylation of squid cartilage β-chitin and carboxymethylated derivatives with different degrees of substitution (DS) were synthesized. The DS of the derivatives calculated by pH titration were 0.64, 0.81, 1.0, 1.33 and 1.59. The antioxidant properties and bile acid binding capacity of the derivatives were studied in vitro. The carboxymethylation of chitosan caused enhancement of bile acid binding capacity. At 1mg/mL, carboxymethyl chitosan (CMCS) showed a stronger scavenging effect than chitosan towards DPPH radicals. The CMCS scavenging effect on superoxide radicals was stronger than that of chitosan, and EC50 values were below 5.6mg/mL. The effectiveness of reducing power correlated with the DS of CMCS. At 1.2mg/mL, the ability of CMCS to chelate ferrous iron was 100%, whereas that of chitosan was only 22.34%. This suggested that carboxymethylation is a possible approach to obtain chitosan derivatives with desirable biological properties.

Abstract:Chitosan, a natural and abundant polysaccharide, offers several advantageous properties, such as excellent biocompatibility, biodegradability, and antimicrobial activity, making it a highly versatile material. However, its extensive application is hindered by its insolubility in water and most organic solvents, due to numerous intramolecular/intermolecular hydrogen bonds. To overcome this limitation and optimize its benefits, chitosan is often chemically modified to enhances its utility in various industries. Biomedical materials, environmental management, biological papermaking, food preservation, and the daily chemical industry are a few examples. Modified chitosan retains the inherent properties of chitosan and acquires new physicochemical characteristics as a result of introduced functional groups. Hydrophilic modifications typically include carboxylation, phosphorylation, and quaternary ammonium modification reactions. The specific properties and applications of chitosan derivatives depend on the grafted groups. This review summarizes the reaction conditions, properties, and applications of hydrophilic chitosan derivatives in drug delivery and food preservation, serving as a reference for the development and use of chitosan-based biomaterials. Bibliography includes 128 references.

Abstract:Chitin is a natural polymeric polysaccharide extracted from marine crustaceans, and chitosan is obtained by removing part of the acetyl group (usually more than 60 %) in chitin's structure. Chitosan has attracted wide attention from researchers worldwide due to its good biodegradability, biocompatibility, hypoallergenic and biological activities (antibacterial, immune and antitumor activities). However, research has shown that chitosan does not melt or dissolve in water, alkaline solutions and general organic solvents, which greatly limits its application range. Therefore, researchers have carried out extensive and in-depth chemical modification of chitosan and prepared a variety of chitosan derivatives, which have expanded the application field of chitosan. Among them, the most extensive research has been conducted in the pharmaceutical field. Abstract:Chitosan is a product of the deacetylation of chitin, which is widely found in nature. Chitosan is insoluble in water and most organic solvents, which seriously limits both its application scope and applicable fields. However, chitosan contains active functional groups that are liable to chemical reactions; thus, chitosan derivatives can be obtained through the chemical modification of chitosan. The modification of chitosan has been an important aspect of chitosan research, showing a better solubility, pH-sensitive targeting, an increased number of delivery systems, etc. This review summarizes the modification of chitosan by acylation, carboxylation, alkylation, and quaternization in order to improve the water solubility, pH sensitivity, and the targeting of chitosan derivatives. The applications of chitosan derivatives in the antibacterial, sustained slowly release, targeting, and delivery system fields are also described. Chitosan derivatives will have a large impact and show potential in biomedicine for the development of drugs in future.

Abstract:Chitosan is a cationic polysaccharide that supports numerous living organisms and can be promptly depolymerized with better functional properties, leading to its extensive applications in pharmaceutics, food, agriculture and environmental protection. The Schiff reaction between chitosan and aldehydes or ketones yields the corresponding aldimines or ketimines, which are converted to N-alkyl derivatives upon hydrogenation with borohydride. The present paper outlined the recent advances in the chemical modification of chitosans with the emphasis on sugarmodified chitosan and trimethylated, N-succinylated, thiolated, and azidated chitosans, as well as cyclodextrin-linked chitosan, crown-ether-bound chitosan and chemical grafting of chitosan.

Abstract:Encapsulation and immobilization technology is important for the food processing and bioengineering industries. Chitosan is a natural polysaccharide prepared by the N - deacetylation of chitin. It has been widely used in food and bioengineering industries, including the encapsulation of active food ingredients, in enzyme immobilization, and as a carrier for controlled drug delivery, due to its significant biological and chemical properties such as biodegradability, biocompatibility, bioactivity, and polycationicity. In this work, chitosan nanoparticles and nanofibers used to encapsulate bioactive substances and immobilize enzymes were reviewed. Preparation of chitosan nanoparticles and nanofibers, including the work achieved in our group on chitosan nanoparticles for enzyme immobilization, were also introduced. Some problems encountered with nano - structured chitosan carriers for bioactive substance encapsulation and enzyme immobilization were discussed, together with the future prospects of such systems.

Abstract:Chitosan was prepared by alkaline N-deacetylation of β-chitin from squid pens, and N-carboxyethylated derivatives (N-CECS) with different degrees of substitution (DS) were synthesized. The carboxyethylation of the polysaccharide was identified by Fourier transform infrared, (1)H and (13)C nuclear magnetic resonance (NMR), and X-ray diffraction analyses. The DS of the derivatives was calculated by (1)H NMR and elemental analysis. All three N-CECS samples showed good water solubility at pH > 6.5. The antioxidant properties and bile acid binding capacity of the derivatives were studied in vitro. The highest bile acid binding capacity of all N-CECS reached 36.9 mg/g, which was 2.6-fold higher than that of chitosan. N-CECS showed a stronger scavenging effect on 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical ability, and EC50 values were below 2 mg/mL. The scavenging ability of N-CECS against superoxide radicals correlated well with the DS and concentration of N-CECS. These results indicated that N-carboxyethylation is a possible approach to prepare chitosan derivatives with desirable in vitro biochemical properties.

Abstract:As one of the most abundant natural polysaccharides that possess good biological activity, chitosan is extracted from chitin. Its application in the food field is being increasingly valued. However, chitosan extraction is difficult, and its poor solubility limits its application. At present, the extraction methods include the acid-base method, new chemical methods, and biological methods. The extraction rates of chitin/chitosan are 4-55%, 13-14%, and 15-28%, respectively. Different chemical modifications have different effects on chitosan, making it applicable in different fields. This article reviews and compares the extraction and chemical modification methods of chitosan, emphasizing the importance of green extraction methods. Finally, the application prospects of chitosan in the food industry are discussed. This will promote the understanding of the advantages and disadvantages of different extraction methods for chitosan as well as the relationship between modification and application, providing valuable insights for the future development of chitosan.
