Abstract:Chitooligosaccharides (COS) has attracted increasing attention due to the various promising bioactivities, tremendous potential in agricultural, environmental nutritional and functional food fields. COS as the major degradation product from chitosan or chitin is prepared via enzymatic, chemical and physical methods. Further obtained COS generally possesses different structural characteristics, such as molecular weight, degree of acetylation and degree of polymerization. Innovations into COS modification has also broadened application of COS in nutrition as well as in agricultural safety. Due to the affinity between structure and bioactivity, diversity of structural characteristics endows COS with various bioactivities like antitumor, antioxidant and anti-inflammatory effects, especially hepatoprotective activity. Therefore, the present review narrates the recent developments in COS physicochemical properties, while paying considerable attention to preparation strategies of COS and their advantages and disadvantages. Moreover, the modification of COS is also discussed including alkylation, quaternization and sulfation, herein the structure-activity relationship of COS was highlighted. Additionally, we summarize the latest research on hepatoprotective activity and mechanisms of COS. Eventually, the future directions of research on COS were discussed, which would provide a new appreciation for the future use of COS.

Abstract:Chitosan oligosaccharides (COS), hydrolysed products of chitosan, are low-molecular weight polymers with a positive charge and good biocompatibility. COS have recently been reported to possess various biological activities, including hypoglycaemic, hypolipidaemic, anti-oxidantantioxidant, immune regulation, anti-inflammatory, antitumour, antibacterial, and tissue engineering activities, exhibiting extensive application prospects. Currently, the biological processes and mechanisms of COS are attractive topics of study, ranging from the genetic, molecular and protein levels. This article reviews the recent discoveries about COS, especially in metabolic regulation, immune function and tissue repair, providing important insights into their multiple biological activities, medical benefits, and therapeutic mechanisms.

The impact of chitooligosaccharides and their derivatives on the in vitro and in vivo antitumor activity: A comprehensive review

Abstract:Chitooligosaccharides (COS) are the degraded products of chitin or chitosan. COS is water-soluble, non-cytotoxic to organisms, readily absorbed through the intestine, and eliminated primarily through the kidneys. COS possess a wide range of biological activities, including immunomodulation, cholesterol-lowering, and antitumor activity. Although work on COS goes back at least forty years, several aspects remain unclear. This review narrates the recent developments in COS antitumor activities, while paying considerable attention to the impacts of physicochemical properties (such as molecular weight and degrees of deacetylation) and chemical modifications both in vitro and in vivo. COS derivatives not only improve some physicochemical properties, but also expand the range of applications in drug and gene delivery. COS (itself or as a drug carrier) can inhibit tumor cell proliferation and metastasis, which might be attributed to its ability to stimulate the immune response along with its anti-angiogenic activity. Further, an attempt has been made to report limitations and future research. The potential health benefits of COS and its derivatives against cancer may offer a new insight on their applications in food and medical fields.

Abstract:Chitin is the most abundant natural polysaccharide and chitosan is its most important derivative. Regardless of having various bioactivities, the water insolubilities of chitin and chitosan limit their applications in many industries. The physical, chemical or enzymatic depolymerization of chitin and chitosan deliver chitooligosaccharides (COS): water-soluble and low molecular weight derivatives, superior to the parent polymers in multiple aspects. COS exhibit an enormously wide range of biological activities and a remarkable potential to be applied in various industries. This review has fully addressed the latest research on the biological activities of COS and the molecular mechanism behind these activities in a correlation with their physicochemical properties. Furthermore, an attempt has been made to report the commercially available COS products. The bioactivities discussed here may offer new understanding of the applications of COS in numerous sectors.

Abstract:Chitosan oligosaccharides (COS), derived from chitin, have garnered considerable attention owing to their diverse biological activities and potential applications. Previous investigations into the bioactivity of COS often encountered challenges, primarily stemming from the use of COS mixtures, making it difficult to discern specific effects linked to distinct degrees of polymerization (DP). Recent progress underscores the significant variation in the biological activities of COS corresponding to different DPs, prompting dedicated research towards synthesizing COS with well-defined polymerization. Among the available methods, enzymatic preparation stands out as a viable and environmentally friendly approach for COS synthesis. This article provides a comprehensive overview of emerging strategies for the enzymatic preparation of single COS, encompassing protein engineering, enzymatic membrane bioreactors, and transglycosylation reactions. Furthermore, the bioactivities of single COS, including anti-tumor, antioxidant, antibacterial, anti-inflammatory, and plant defense inducer properties, exhibit close associations with DP values. The potential applications of single COS, such as in functional food, food preservation, and crop planting, are also elucidated.

Abstract:Chitosan and its oligosaccharides exhibit numerous biochemical characteristics, which includes immune enhancing, antimicrobial and anti-viral properties. Chitosan and its products have drawn much attention of both food and pharmaceutical industry due to their peculiar physiochemical characteristics and effectiveness in cancer therapies. Current study describes the different physiological and chemical properties along with its usefulness in different biological processes. According to the novel studies, the focus of this study will be the anti-cancerous activities of COS. This study will summarize the whole mechanism related to the anti-cancerous functioning of COS along with the deep insight into the future perspectives of COS, especially at molecular level. Finally, future developments regarding the application of this biopolymer will be highlighted.

Abstract:Chitosan is a natural biopolymer with a modifiable structure and abundant function groups, moreover, it can be processed into various shapes and sizes, making it suitable for various applications. This review analyzed and summarized the recent advances in chitosan-based materials, particularly focusing on the modification of chitosan by physical, chemical and biological methods, which provided plentiful information on the preparation, characterization and application of chitosan-based materials. In this review, various preparation and modification methods for improving chitosan performances, as well as the application of chitosan and its derivatives, were critically analyzed and summarized in detail. Chitosan-based materials can be fabricated into different shapes and sizes, such as nanofiber, nanoparticles, microspheres, membranes, and 3D printed scaffolds. In addition, chitosan can be modified through physical, chemical, and biological modifications, to obtain versatile chitosan-based materials. For physical modification, crosslinking and blending are commonly used., while chemical modification is based on chemical reactions (e.g. acylation, esterification, etherification, N-alkylation, graft copolymerization, and degradation), to prepare the corresponding derivatives (e.g. chemically cross-linked chitosan, grafted chitosan, low molecular weight chitosan, and oligo-chitosan) through crosslinking, grafting, and degradation. The biological modification is associated with various enzymes to realize the hydrolysis or grafting. With improved performances through modification, as well as various shapes and sizes, chitosan-based materials show promising applications in versatile fields, such as water treatment, food processing, pharmaceutical/biomedicine, textiles and agriculture. This review will deepen the understanding the state of the art of chitosan-based materials, including their preparation, characterization and application.

Abstract:A concise review of the main current natural sources used to produce chitin—the starting material to produce chitooligosaccharides (COS)—is presented, including algae, arthropods, birds, fish, fungi, mollusks, and, possibly, plants. The principal approaches addressed to produce COSs, grouped as physical, chemical, and biological processes, are also outlined. Subsequently, the COS more relevant applications related to agriculture are briefly outlined, i.e., induction of innate immunity in plants, growth biostimulation, soil amending, biocidal activity, etc. Some interesting findings of this review are: (a) A clear relationship has been undoubtedly established between the low molecular weights (MWs) of these chitinous materials and their striking bioactivities (b) There is no universal consensus about the limit MW below which a substance can be considered a COS and some of the proposed limit values are supported in works that have not proposed them (c) The preparation and application of COS is an active field of research due to the accessibility of chitin sources anywhere and the variety of preparation methods available, as well as the multiple possibilities of modification that these materials offer for the preparation of bioactive derivatives (d) The chemical modification of the great number of existing COS, by a wide range of agents and approaches, including computer simulation studies, is a virgin field that could generate products with powerful elicitor proper-ties (e) Biocidal activities of COSs, advantaged with their greater water solubilities than chitin and chitosan, are remarkably attractive due to the possibility of replacing, partial or completely, injurious synthetic products currently in use. Similarly, this review makes it possible to appreciate that the preparation and separation of COS with well-defined structures could boost the discovery of the specific regulatory mechanisms that each oligomer species can activate (or repress), that is, defense mechanisms in plants.

Abstract:ABSTRACT Chitooligosaccharides (COS) are derived from chitosan, which can be used as nutraceuticals and functional foods. Because of their various biological activities, COS are widely used in the food, medicine, agriculture, and other fields. COS were prepared by chitosanase from Pseudoalteromonas sp. SY39 and their anti-obesity activity was researched in mice in this study. The effects of hydrolysis time, temperature, the ratio of enzyme to chitosan, and pH on the productivity of COS were discussed. Preparation process of COS was established in a 5-L fermenter. COS were characterized and their anti-obesity activity was studied in animal experiments. The results showed that COS could effectively reduce serum lipid levels and obesity in mice, and have a good anti-obesity activity. The preparation technology and remarkable anti-obesity activity of COS further expand their applications in the food and pharmaceutical industries. GRAPHICAL ABSTRACT Chitooligosaccharides (COS) were effectively prepared by using chitosanase under the optimum conditions, which had excellent anti-obesity activity in mice.

Abstract:Chitosan, which is derived from chitin, is the only known natural alkaline cationic polymer. Chitosan is a biological material that can significantly improve the living standard of the country. It has excellent properties such as good biodegradability, biocompatibility, and cell affinity, and has excellent biological activities such as antibacterial, antioxidant, and hemostasis. In recent years, the demand has increased significantly in many fields and has huge application potential. Due to the poor water solubility of chitosan, its wide application is limited. However, chemical modification of the chitosan matrix structure can improve its solubility and biological activity, thereby expanding its application range. The review covers the period from 1996 to 2022 and was elaborated by searching Google Scholar, PubMed, Elsevier, ACS publications, MDPI, Web of Science, Springer, and other databases. The various chemical modification methods of chitosan and its main activities and application research progress were reviewed. In general, the modification of chitosan and the application of its derivatives have had great progress, such as various reactions, optimization of conditions, new synthetic routes, and synthesis of various novel multifunctional chitosan derivatives. The chemical properties of modified chitosan are usually better than those of unmodified chitosan, so chitosan derivatives have been widely used and have more promising prospects. This paper aims to explore the latest progress in chitosan chemical modification technologies and analyze the application of chitosan and its derivatives in various fields, including pharmaceuticals and textiles, thus providing a basis for further development and utilization of chitosan.

Abstract:In view of rapidly growing interest in the amino polysaccharide chitosan/chitin and its oligosaccharides as functional biopolymer, a recent progress of basic and application studies in chitosan/chitin oligosaccharides chemistry is reviewed as well as some basic aspects of this specialty biomass resource. A special emphasis is placed on the controlled modification reactions to prepare chitin derivatives with well-defined structures and thereby to construct sophisticated molecular architecture having various advanced functions. The reactions discussed here include acylation, quaternary ammonium salt, carboxyalkylation, graft copolymerization, quaternary salt formation, Schiff base formation, reductive alkylation, microwave modification. For conducting modification reactions in a facile and controlled manner, some soluble chitosan/chitin oligosaccharides derivatives are convenient. To fully explore the high potential of these specialty biopolymers, basic and application researches are being made extensively.

Abstract:Chitosan is a product of the deacetylation of chitin, which is widely found in nature. Chitosan is insoluble in water and most organic solvents, which seriously limits both its application scope and applicable fields. However, chitosan contains active functional groups that are liable to chemical reactions; thus, chitosan derivatives can be obtained through the chemical modification of chitosan. The modification of chitosan has been an important aspect of chitosan research, showing a better solubility, pH-sensitive targeting, an increased number of delivery systems, etc. This review summarizes the modification of chitosan by acylation, carboxylation, alkylation, and quaternization in order to improve the water solubility, pH sensitivity, and the targeting of chitosan derivatives. The applications of chitosan derivatives in the antibacterial, sustained slowly release, targeting, and delivery system fields are also described. Chitosan derivatives will have a large impact and show potential in biomedicine for the development of drugs in future.

Abstract:Chitosan, an amino polysaccharide mostly derived from crustaceans, has been recently highlighted for its biological activities that depend on its molecular importantly, for some advanced biomaterials, the homogeneity of the chitosan structure is an important factor in determining its biological activity. Here we review emerging enzymes and cell factories, respectively, for in vitro and in vivo preparation of chitosan oligosaccharides (COSs), focusing on advances in the analysis of the AP and structural modification of chitosan to tune its functions. By 'mapping' current knowledge on chitosan's in vitro and in vivo activity with its MW and AP, this work could pave the way for future studies in the field.

Abstract:Chitin is an abundant polysaccharide primarily produced as an industrial waste stream during the processing of crustaceans. Despite the limited applications of chitin, there is interest from the medical, agrochemical, food and cosmetic industries because it can be converted into chitosan and partially acetylated chitosan oligomers (COS). These molecules have various useful properties, including antimicrobial and anti-inflammatory activities. The chemical production of COS is environmentally hazardous and it is difficult to control the degree of polymerization and acetylation. These issues can be addressed by using specific enzymes, particularly chitinases, chitosanases and chitin deacetylases, which yield better-defined chitosan and COS mixtures. In this review, we summarize recent chemical and enzymatic approaches for the production of chitosan and COS. We also discuss a design-of-experiments approach for process optimization that could help to enhance enzymatic processes in terms of product yield and product characteristics. This may allow the production of novel COS structures with unique functional properties to further expand the applications of these diverse bioactive molecules.

Abstract:Chitosan oligosaccharide (COS) is known for its unique biological activities such as anti-tumor, anti-inflammatory, anti-oxidant, anti-bacterial activity, biological recognition, and immune enhancing effects, and thus continuous attracting many research interests in drug, food, cosmetics, biomaterials and tissue engineering fields. In comparison to its corresponding polymer, COS has much higher absorption profiles at the intestinal level, which results in permitting its quick access to the blood flow and potential contacting with blood components. However, the effects of COS on blood components remain unclear to date. Herein, two COS with different molecular weight (MW) were characterized by FTIR and 1H NMR, and then their effects on human blood components, including red blood cells (RBCs) (hemolysis, deformability, and aggregation), coagulation system [activated partial thromboplastin time (APTT), prothrombin time (PT), thrombin time (TT), and the concentration of fibrinogen (Fib)], complement (C3a and C5a activation), and platelet (activation and aggregation), were comprehensively studied. In the case of RBCs, COS exhibited a low risk of hemolysis in a dose and molecular weight dependent manner and the irreversible aggregation was observed in their high concentration. For coagulation system, COS has a mild anticoagulation activity through blocking the intrinsic coagulation pathway. In addition, COS showed no effect on complement activation in C3a and C5a and on platelet activation while inhibition of platelet aggregation was evident. Finally, the mechanism that effects of COS on blood components was discussed and proposed.

Abstract:A series of phenolic acid chitooligosaccharide (COS) derivatives synthesized by two mild and green methods were illuminated in this paper. Seven phenolic acids were selected to combine two kinds of COS derivatives: the phenolic acid chitooligosaccharide salt derivatives and the phenolic-acid-acylated chitooligosaccharide derivatives. The structures of the derivatives were characterized by FT-IR and 1H NMR spectra. The antioxidant experiment results in vitro (including DPPH-radical scavenging activity, superoxide-radical scavenging activity, hydroxyl-radical scavenging ability, and reducing power) demonstrated that the derivatives exhibited significantly enhanced antioxidant activity compared to COS. Moreover, the study showed that the phenolic acid chitooligosaccharide salts had stronger antioxidant activity than phenolic-acid-acylated chitooligosaccharide. The cytotoxicity assay of L929 cells in vitro indicated that the derivatives had low cytotoxicity and good biocompatibility. In conclusion, this study provides a possible synthetic method for developing novel and nontoxic antioxidant agents which can be used in the food and cosmetics industry.

Abstract:The objectives of the present study are to synthesize a series of chitosan oligosaccharide-O-Terpenol (COS-O-Ter) derivatives and their implication to evaluate in vitro antibacterial activity. Herein, a general strategy is described for preparing COS-O-Ter derivatives, including substitution and deprotection reactions. The structures of COS-O-Ter derivatives were characterized by FT-IR, 1H NMR, XRD, TGA, and elemental analysis. COS-O-Ter derivatives revealed the excellent solubility and in vitro antibacterial activity. Moreover, their antibacterial activities were more sensitive to Staphylococcus aureus (S. aureus) than Escherichia coli (E. coli) indicating the effective potential application of COS-O-Ter derivatives as natural antibacterial agents. The aforementioned study opens a pave to expand the application scope of COS and its derivatives in the food and pharmaceutical industries.

Abstract:Chitosan, a natural and abundant polysaccharide, offers several advantageous properties, such as excellent biocompatibility, biodegradability, and antimicrobial activity, making it a highly versatile material. However, its extensive application is hindered by its insolubility in water and most organic solvents, due to numerous intramolecular/intermolecular hydrogen bonds. To overcome this limitation and optimize its benefits, chitosan is often chemically modified to enhances its utility in various industries. Biomedical materials, environmental management, biological papermaking, food preservation, and the daily chemical industry are a few examples. Modified chitosan retains the inherent properties of chitosan and acquires new physicochemical characteristics as a result of introduced functional groups. Hydrophilic modifications typically include carboxylation, phosphorylation, and quaternary ammonium modification reactions. The specific properties and applications of chitosan derivatives depend on the grafted groups. This review summarizes the reaction conditions, properties, and applications of hydrophilic chitosan derivatives in drug delivery and food preservation, serving as a reference for the development and use of chitosan-based biomaterials. Abstract:Chitosan derivatives, and more specifically, glycosylated derivatives, are nowadays attracting much attention within the scientific community due to the fact that this set of engineered polysaccharides finds application in different sectors, spanning from food to the biomedical field. Overcoming chitosan (physical) limitations or grafting biological relevant molecules, to mention a few, represent two cardinal strategies to modify parent biopolymer; thereby, synthetizing high added value polysaccharides. The present review is focused on the introduction of oligosaccharide side chains on the backbone of chitosan. The synthetic aspects and the effect on physical-chemical properties of such modifications are discussed. Finally, examples of potential applications in biomaterials design and drug delivery of these novel modified chitosans are disclosed.

Abstract:Background: Bio-based materials, as the plentiful and renewable resources for natural constituents which are essential for biomedical and pharmaceutical applications, have not been exploited adequately yet. Chitosan is a naturally occurring polysaccharide obtained from chitin, which has recently attracted widespread attention owing to its excellent activity. This review shows the methods of extraction and modification of chitosan and provides recent progress of synthesis and use of chitosan-based materials in biological applications. Methods: By consulting the research literature of the last decade, the recent progresses of functional chitosan-based materials for biological applications were summarized and divided into the methods of extraction chitosan, the chemical modification of chitosan, chitosan-based materials for biological applications were described and discussed. Results: Chemical modification of chitosan broadens its applications, leading to developing numerous forms of chitosan-based materials with excellent properties. The excellent bioactivity of chitosan-based material enables it serves potential applications in biomedical fields. Conclusion: Chitosan-based materials not only exhibit the excellent activities of chitosan but also show other appealing performance of combined materials, even give the good synergistic properties of chitosan and its composite materials. Further studies are needed to define the ideal physicochemical properties of chitosan for each type of biomedical applications. The development of various functional chitosan-based materials for biological applications will be an important field of research, and this kind of material has important commercial value.
