Effects of Variations in Design of Experiment on the Result of Diagnosis of Pancreatic Cancer with Serum Tumor Marker (CA19-9)

Abstract:Objective To explore how sample size and control selection might influence the result in the analysis of serum tumor marker CA19-9 diagnostic parameters for pancreatic cancer. Method The diagnostic parameters of CA19-9 (determined with CLIA) were analyzed in the following individuals: ① Patients with pancreatic cancer (n=307) ② patients with other benign pancreato-billiary diseases (n=173) ③ subjects asking for routine physical check-ups (n=200) ④ healthy controls (n=500). Results The sensitivity and area under the ROC of CA19-9 was very unevenly distributed when the involved specimen size was less than 90. The specificity in controls with different kind of members also differed significantly, with the highest ones in healthy controls (100%), which was significantly higher than that in patients with benign pancreato-billiary diseases (P<0.05). Conclusion Sample size of more than 90 was recommended for serum CA-199 diagnostic parameters analysis in the diagnosis of pancreatic carcinoma. Specificity in controls with different kinds of members varied significantly.
