Abstract:Asymmetric cyanation of a class of easily available olefins with a favorable cyanide source ethyl cyanoformate (CNCOOEt) was realized by an interesting modular catalyst. High yields and ee values were obtained for a range of substrates under solvent-free and mild reaction conditions. The products obtained could be easily transformed to the enantioenriched useful intermediates 5, 6, and pharmaceutically important gamma-aminobutyric acid 7.

Abstract:An efficient process based on the gold-catalyzed redox reaction has been developed to oxidize 1,2-diarylacetylene or ynamide to 1,2-diaryldiketone or α-keto imide respectively. This process can tolerate a variety of functional groups and affords 1,2-dicarbonyl compounds in excellent yields under mild reaction conditions.

Abstract:Bioinspired manganese complexes of N-4 ligands and graphene oxide (GO) synergistically catalyze the highly enantioselective epoxidation of olefins (up to 99% ee), which is a rare example with GO as a co-catalyst in asymmetric catalysis. GO as a new and key additive not only successfully functions in catalytic amounts, but also has a positive effect for improving the enantioselectivity of the asymmetric epoxidation compared with the traditional stoichiometric organic carboxylic acid method [e.g., chalcone, 95% ee (3.5 mg GO) vs. 84% ee (5 equiv., 75 mg acetic acid), ethyl cinnamate, 84% ee (3.5 mg GO) vs. 19% ee (5 equiv., 75 mg acetic acid)]. The X-ray photoelectron spectroscopy (XPS) spectra of GO before and after the reaction indicate that the intensities of C-O become stronger after the reaction, which may have a certain relationship with hydrogen peroxide (H2O2) and gives a reasonable rationale for the large consumption of H2O2. Also, part of the hydrogen peroxide was used for the oxidation of GO.

Abstract:Optically active epoxides play important roles in pharmaceutical, agricultural and fine chemical syntheses. There are many chiral medications with pharmacodynamic activity in nature that can be synthesized by chiral epoxides. In recent years, researchers have developed a variety of biocatalysts for the asymmetric epoxidation of alkenes, which use oxygen or hydrogen peroxide as eco-friendly and low-cost oxidants, to provide better chemo-, regio- and stereoselectivity under moderate reaction conditions. In this paper, the advances, opportunities and challenges of the asymmetric epoxidation of unactive alkenes by biocatalyst are reviewed.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:Since the introduction of the Shi catalyst, the organocatalytic epoxidation of olefins has become an area of continued research interest. To explore the possibility to enhance the efficiency and stability of the catalyst, the synthesis of a galactose‐derived trifluoromethyl ketone, and its use for stereoselective epoxidation reaction that exploits a transitory dioxirane as active species are herein reported. The trifluoromethyl ketone was built on the C6 of a protected D‐galactopyranose. The organocatalyst was obtained smoothly in a few steps, and when utilized for stereoselective epoxidation exhibited excellent stability as no chemical alterations were observed during the reaction. The stereoselectivity, although sometimes moderate, appears surprisingly high considering the conformational freedom of the trifluoromethyl ketone moiety. DFT calculations were performed to investigate the interactions involved in regulating the observed selectivities. Moreover, a new mnemonic model has been developed to serve as a fast‐predicting tool for epoxidation of new substrates.

Control experiments and density functional theory (DFT) calculations revealed this successful stereocontrol might rely on hydrogen-bond-mediated dynamic kinetic resolution.

Abstract:A variety of organocatalysts for the asymmetric direct aldol reactions of ketones with alpha-keto acids were designed on the basis of molecular recognition and prepared from proline and aminopyridines. The organic molecule 8e, derived from proline and 6-methyl-2-amino pyridine, was the best catalyst, affording excellent enantioselectivities (up to 98% ee) for the direct aldol reactions of acetone or 2-butanone with a wide range of a-keto acids and for the reactions of various acyclic aliphatic ketones with 3-(2-nitrophenyl)2-oxopropanoic acid. The aldol adducts could be converted to 2-hydroxy-gamma-butyrolactones by reaction sequences of diastereoselective reduction and lactonization. Experimental and theoretical studies on the transition states revealed that the amide N - H and the pyridine N of the organocatalyst selectively form hydrogen bonds with the keto oxygen and the carboxylic acid hydroxy of the a-keto acid, respectively. These two hydrogen-bonding interactions are important for the reactivity and enantioselectivity of the direct asymmetric aldol condensation.

Abstract:Asymmetric epoxidation of olefins by using an α,α-dimethylmorpholinone-containing chiral ketone catalyst (4) has been investigated. This ketone, which has the combined features of several previously studied catalysts, is an effective catalyst for trans- and trisubstituted olefins, and up to 97% ee has been achieved. The α,α-dimethyl group has significant impact on spiro and planar transition states.

Abstract:Asymmetric epoxidation of alpha,beta-unsaturated ketones has been extensively studied and several important procedures have been developed in the last decade. This review addresses the most significant advances in asymmetric epoxidation of alpha,beta-unsaturated ketones using proline-derived alpha,alpha-diarylprolinols as catalysts. Special attention has been paid to the enantioselective epoxidation of chalcones, alpha,beta-unsaturated trifluoromethyl, trichloromethyl ketones and beta,gamma-unsaturated alpha-keto esters based on the reseach of our group.

Abstract:A new chrial organosulfide was synthesized through an unexpected Wagner-Meerwein rearrangement. This organosulfide could catalyze the epoxidation reaction of various aromatic aldehydes smoothly with benzyl bromide to give trans-diaryl epoxide in satisfactory yields (60-84%) with excellent diastereoselectivities (trans:cis = 95:5-100:0) and good to excellent enantioselectivities (86-96% ee).

Abstract:Herein we report an electrochemical strategy for the construction of polysubstituted oxazoles from easily available ketones and acetonitrile under room temperature. The method shows high efficiency, a broad substrate scope, and requires no external chemical oxidant. Mechanistic investigations suggest that the reaction proceeds through a Ritter-type reaction/oxidative cyclization using acetonitrile as the reactant and solvent. X-ray crystallographic analysis reveals that the reaction involves the attack of acetonitrile on the carbonyl carbon instead of the α-carbon.

Abstract:A general asymmetric deconstructive alkynylation of cyclic oximes with terminal alkynes was reported using copper/chiral cinchona alkaloid-based N,N,P-ligand catalysts and used to produce more than 60 examples of enantioenriched γ- and δ-alkynyl nitriles in a highly functional group-compatible manner. A wide variety of low-cost terminal alkynes and easily available cyclic oximes with substituent diversity were found to be well tolerated with the current asymmetric transformation with good functional group tolerance. Abstract:The method for converting epoxy carbamates to oxazolidinones is outlined in this study. This innovative approach combines intermolecular nucleophilic epoxide ring opening with intramolecular acyl substitution in a single step, thereby facilitating rapid conversion. Significantly, this method effectively addresses challenges associated with the use of expensive reagents, harsh reaction conditions, and prolonged reaction times, presenting a more efficient alternative. Demonstrating favorable reactivity across a diverse range of aryl groups, as well as benzyl or tert‐butyl carbamates, the method consistently yields satisfactory results, with oxazolidinone formation ranging from 55% to 99% in over 24 examples. The synthesized oxazolidinones, including 5v–5x, hold substantial promise as intermediates for the synthesis of crucial synthetic molecules. Furthermore, the versatility of this method is underscored by its successful application in the formation of the 6‐membered ring 1,3‐oxazinan‐2‐one. These findings not only contribute to the advancement of synthetic methodologies but also pave the way for a streamlined synthesis of important pharmaceutical compounds.
