Abstract:Four new compounds ( 1 – 4 ) were isolated from the whole plants of two species of Delphinium , including two C 20 -diterpenoid alkaloids, umbrodines A and B ( 1 and 2 ), and a dibenzoxazepinone, umbrolide A ( 3 ) from Delphinium umbrosum Hand.-Mazz. and a C 20 -diterpenoid alkaloid, kingiadine ( 4 ) from Delphinium kingianum Bruhl. ex Huth. Ten known diterpenoid alkaloids were also isolated. Their structures were elucidated via HR-ESIMS, IR, and NMR data. Lycoctonine ( 11 ) and delectinine ( 12 ) exhibited appreciable cardiac activity. Furthermore, 11 and 12 showed cardioprotective effects against doxorubicin-induced toxicity in H9c2 cells, with the maximum protection rates of 61.63% and 51.18%, respectively. Graphical

Abstract:Three new diterpenoid alkaloids ( 1 , 2 , 3 ) and seventeen known ( 4–20 ) compounds were isolated from the whole plant of Delphinium sherriffii Munz (Ranunculaceae). Their structures were elucidated by various spectroscopic analyses, including IR, HR-ESI-MS, 1D and 2D NMR spectra. All compounds were evaluated for the inhibitory activity of Sf9 cells and compound 5 exhibited the strongest cytotoxicity (IC 50 = 8.97 μM) against Sf9 cell line.

Abstract:Six diterpenoid alkaloids, namely, pachycentine ( 1 ), deacetylswinanine A ( 2 ), siwanine A ( 3 ), tatsiensine ( 4 ), deacetyltatsiensine ( 5 ), and 6-deoxydeltamine ( 6 ), were isolated from a China-specific Delphinium plant (family Ranunculaceae), Delphinium pachycentrum Hemsl. Their structures were established via detailed spectroscopic analyses, including IR, HR-ESI-MS, 1D and 2D NMR techniques. Pachycentine ( 1 ) is a previously undescribed hetisine-type C 20 -diterpenoid alkaloid, and compounds 5 and 6 were synthetic intermediates newly identified as natural products. In addition, compounds 2 – 4 were isolated from this species for the first time. The chemotaxonomic significance of all the isolates was summarized. Abstract:Thirteen new benzamide alkaloids, delphiniumines A–M (1–13), together with one known analogue (14), were isolated from Delphinium anthriscifolium Hance. All of the structures were determined by spectroscopic and spectrometric analyses. Absolute configuration for 1 was established using experimental and calculated ECD data, as well as by X-ray crystallography analysis. Compound 1 possesses a previously undescribed polysubstituted cyclopentene carbon framework. Compound 2 was isolated as an artifact from 1 during the extraction process. Compound 7 is glycosylated with a β-D-glucose unit. Compound 13 bears a chlorine substituent. Abstract:The genus Delphinium is a rich source of diterpene alkaloids. Chemical investigation on an alkaloid rich extract of the whole parts of Delphinium turkmenum resulted in the isolation of three C 19 -diterpene alkaloids ( 1-3 ) and a palmitic acid derivative ( 4 ). The chemical structures were elucidated by analysis of 1D and 2D-NMR and comparison the data with those reported in the literature. Notably, all isolated compounds were reported for the first time from D. turkmenum.

Abstract:Delphinium grandiflorum , one of the well-known Delphinium species, contains diverse bioactive diterpenoid alkaloids (DAs), which promoted a systematic phytochemical study on this plant. As a result, two new DAs, namely 16-demethyl tuguaconitine ( 1 ) and 6- O -acetyl-16-demethyldelsoline ( 2 ), together with one known analogue pseudophnine A ( 3 ) were obtained from the whole herbs of D. grandiflorum . Their structures were elucidated by use of extensive spectroscopic analysis (HR-ESIMS, IR, 1D and 2D NMR). Additionally, the inhibitory effects of these obtained individual compounds were also evaluated in vitro for lung cancer cell line (A549), liver cancer cell line (SMMC-7721), and breast cancer cell line (MCF-7). The results of cytotoxic activities showed that only compound 2 exhibit weak inhibitory effect on SMMC-7721 and MCF-7, and their IC 50 values were 37.4 and 33.1 μM, respectively. However, compound 1 and 3 had no obvious cytotoxic activities (IC 50 >80 μM).

Abstract:Two new C19‐diterpenoid alkaloids of the lycoctonine‐type (liangshanine A and liangshanine B) and nineteen known compounds (3‐21) were isolated from the whole plant of Delphinium liangshanense W. T. Wang, and all the compounds were identified by different spectroscopic analyses, such as IR, HR‐ESI‐MS and NMR. All the compounds were isolated from this plant for the first time and tested for the anti‐proliferation effects on MH7A and SF9 cells to figure their anti‐rheumatoid arthritis and anti‐insect activity, but none of them showed remarkable activity.

Abstract:The chemical constituents have been isolated from Delphinium tatsienense Franch Var Chinghaiense by column chromatography and preparative thin layer chromatography. The structures of the diterpenoid alkaloids were elucidated by IR, EA, MS, H-1 NMR, C-13 NMR as well as DEPT. Six compounds were obtained, among which one is a new ditepenoid alkaloid, four axe known diterpenoid alkaloids and the other is P-sitosterol.

Abstract:Delphinium grandiflorum L. (family Ranunculaceae), one of the most important and widely distributed Delphinium species, has received considerable interest due to its extremely high medicinal value. The discovery of novel metabolites from D. grandiflorum supported and broadened its application as an herbal medicine. In this study, the whole herb of D. grandiflorum was phytochemically investigated to obtain fourteen C 19 -lycaconitine-type diterpenoid alkaloids ( 1 – 14 ), including six undescribed alkaloids, grandiflolines A–F ( 1 – 6 ). The structural elucidation of them was accomplished by detailed spectroscopic analyses, mainly including HR-MS, 1D and 2D NMR ( 1 H– 1 H COSY, NOESY, HMBC and HSQC), and IR spectra. New alkaloids 1 – 3 and 5 possess a characteristic △ 2,3 functional group in the A ring, while compounds 5 and 6 feature a rare OH-16 substituent. In addition, known compounds 7 – 12 were isolated from D. grandiflorum for the first time. Moreover, according to its medicinal use, new alkaloids 1 – 6 were estimated for their potential in vitro anti-inflammatory effects, and some of them exhibited inhibitory effects on NO production in LPS-activated RAW 264.7 macrophages. Our work enriched the chemical diversity of D. grandiflorum and the genus Delphinium and presented beneficial information for further investigations.

Abstract:Cyano tends to have better biological activity, but it is rarely reported in natural products, especially in the C 20 -diterpene alkaloids. Herein, three unprecedented C 20 -diterpenoid alkaloids, brunonianines A-C ( 1 - 3 ), possessing rare cyano functional group as well as an atisine backbone constructed from a phenethyl substituent and a tetrahydropyran ring, along with four C 19 -alkaloids ( 4 – 7 ) and one amide alkaloids ( 8 ), were isolated from the whole plant of Delphinium brunonianum Royle. Compounds 1 – 3 are also the first atisine type diterpenoid alkaloids with cyano group obtained from nature. The structures of the previously undescribed compounds were elucidated by HR-ESI-MS, 1D/2D NMR spectroscopic data and electronic circular dichroism calculations and single-crystal X-ray diffraction. Reasonable speculations have also been made regarding the biogenic synthetic pathways of compounds 1 – 3 . In addition, the inhibitory activity of all compounds was also tested against four tumor lines: A549, Caco-2, H460 and Skov-3, where compound 2 (IC 50 2.20 ± 0.21 μM) showed better inhibitory activity against Skov-3 cells than the hydroxycamptothecin. Using flow cytometry, cell staining, migration and invasion analysis, and Western blot, compound 2 was found to arrest cells in the G2/M phase and was able to effectively inhibit cell motility to achieve potent anti-tumor effects. In addition, compound 2 can effectively induce apoptosis by activating the Bax/Bcl-2/Caspase-3 signaling pathway.

Abstract:Two new isoquinoline alkaloids, decumbensine (1) and decumbendine (2), together with nine known compounds including tetrahydropalmatine (3), bicuculline (4), allocryptopine (5), bulbocapine (6), protopine (7), kikemanine (8), decumbenine B (9), decumbenine C (10), and muramine (11) were isolated from the seedling of Corydalis decumbens (Thunb.) Pers. (Papaveraceae). Their structures were decisively elucidated by spectroscopic analysis including 1D and 2D NMR techniques. Furthermore, the biogenetic pathways of two new compounds were proposed, which would provide valuable information for further understanding of the isoquinoline alkaloids of C. decumbens.

Abstract:This research aimed to excavate compounds with activity reducing hepatocytes lipid accumulation from Delphinium brunonianum. Four novel diterpenoid alkaloids, brunodelphinine B–E, were isolated from D. brunonianum together with eleven known diterpenoid alkaloids through a phytochemical investigation. Their structures were elucidated by comprehensive spectroscopy methods including HR-ESI-MS, NMR, IR, UV, CD, and single-crystal X-ray diffraction analysis. The inhibitory effects of a total of 15 diterpenoid alkaloids on hepatocytes lipid accumulation were evaluated using 0.5 mM FFA (oleate/palmitate 2:1 ratio) to induce buffalo rat liver (BRL) cells by measuring the levels of triglyceride (TG), total cholesterol (TC), alanine transaminase (ALT), aspartate transaminase (AST), and the staining of oil red O. The results show that five diterpenoid alkaloids—brunodelphinine E (4), delbruline (5), lycoctonine (7), delbrunine (8), and sharwuphinine A (12)—exhibited significant inhibitory effects on lipid accumulation in a dose-dependent manner and without cytotoxicity. Among them, sharwuphinine A (12) displayed the strongest inhibition of hepatocytes lipid accumulation in vitro. Our research increased the understanding on the chemical composition of D. brunonianum and provided experimental and theoretical evidence for the active ingredients screened from this herbal medicine in the treatment of the diseases related to lipid accumulation, such as non-alcoholic fatty liver disease and hyperlipidemia.

Abstract:Caesaldecins A and B (1a/1b), a pair of epimeric cassane-type diterpenoids containing an ether linkage between C-6 and C-20 and an alpha,beta-butenolide ring in the northern hemisphere, were isolated from the leaves of the medicinal plant Caesalpinia decapetala (Roth) Alston, along with a new labdane-type diterpenoid, termed as 8(17),11(Z),13(E)-trien-15,18-dioic acid (2). Structures of 1 and 2 were established by extensive spectroscopic analyses, including HRESIMS, 1D and 2D NMR, and electronic circular dichroism (ECD) calculations. The in vitro antibacterial and inhibitory activities against a panel of bacteria and cancer cell lines were evaluated, respectively.
