Effects of Qiweikeli on Oxidative Stress and Pathologic Changes of the Kidneys in Type 2 Diabetic Rats

Abstract:Objective:To investigate the effects of on oxidative stress and pathologic changes of the kidney in type 2 diabetic rats.Methods:Type 2 Diabetes mellitus was induced in male SD rats by feeding with high fat diet combined with intraperitoneal injection of low-dose streptozotocin(30 mg/kg).Thirty-two diabetic rats were randomly assigned to the model,Qiweikeli,alpha lipoic acid and normal control groups.Rats in the group were performed intragastric administration of 20 g·kg-1·d-1 crude drug of and rats in the alpha lipoic acid group were administrated with 20 mg·kg-1·d-1 alpha lipoic acid,other two groups were administrated with distilled water by equal volume.Blood glucose,glycolated hemoglobin,total cholesterol,triglyceride,urine protein,renal function of diabetic rats were detected after 12 weeks.And meanwhile the renal cortex superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malonaldehyde(MDA)were determined.Pathologic changes of the kidney were evaluated.Results:Compared with the normal control group,the levels of 24 hour-urinary protein excretion of rats in DM groups were all higher(P0.01),while the creatinine clearance rate were lower(P0.05);GSH-Px and SOD activities were lower and MDA levels were higher in renal cortex of rats DM groups(P0.01).Pathological observation of rats in DM groups showed that the glomeruli were hypertrophic and glomerular basement membranes(GBM)became unevenly thicker,that the mesangial cells proliferated and mesenteric matrix increased,that the renal tubular epithelial cells were swollen and renal tubulointerstitial fibrosis appeared.Compared with the model group,treatments with Qiweikeli、alpha lipoic acid alleviated above changes(P0.01 or 0.05),but there are not significant differences between group and alpha lipoic acid gruop(P0.05).Conclusion:Oxidative stress may contribute to the development of type 2 diabetic nephropathy.Chinese Medicine Qiweikeli can effectively inhibit oxidative stress of the kidneys in type 2 diabetic rats and improve renal function and micromorphological structures.
