Abstract:BACKGROUND Desquamative gingivitis (DG) is a clinical manifestation common to several diseases. It is known that most cases of DG are caused by mucous membrane pemphigoid (MMP), oral lichen planus (OLP), or pemphigus vulgaris (PV). Early recognition and treatment of these diseases can improve the prognosis, but diagnostic delays are common in patients with DG because obtaining a diagnostic biopsy is technically challenging. A biopsy technique designed to maintain the gingival epithelium for patients with DG was developed. The usefulness of this technique is discussed. METHODS This study is based on a retrospective review of 27 DG cases. A stab-and-roll technique was used to obtain gingival tissue. This technique is designed to reduce lateral forces on the epithelium during the procedure and to thereby prevent the inadvertent removal of the epithelium from the biopsy specimen. A total of 52 biopsies comprising 27 for hematoxylin and eosin (H&E)-stained samples and 25 for direct immunofluorescence (DIF) testing were reviewed. RESULTS Fifty-one of the 52 biopsies (98.1%) maintained the epithelium. Only one biopsy (1.9%) showed that the epithelium was totally absent. Therefore, H&E and DIF features of 51 biopsies were analyzed. Definitive diagnoses of the diseases causing DG included MMP (13 cases), PV (eight cases), and OLP (six cases). CONCLUSIONS A diagnostic biopsy was obtained from the gingiva of patients with DG using the stab-and-roll technique. The gingival epithelium was well maintained, and the relationship with the underlying connective tissue was diagnostic. In the future, this stab-and-roll biopsy technique may facilitate early diagnosis and treatment of diseases causing DG.

Abstract:Abstract This study explored relationships between sense of humor, stress, and coping strategies. Undergraduate students (N=258) from introductory psychology courses completed a perceived stress scale, an everyday problems scale, a state anxiety inventory, a sense of humor scale, and a scale assessing their preferred coping strategies. High and low sense of humor groups were determined by selecting participants with self-reported sense of humor at one standard deviation above and below the overall mean on the sense of humor scale. The high sense of humor group appraised less stress and reported less current anxiety than a low sense of humor group despite experiencing a similar number of everyday problems in the previous two months. The high humor group was more likely to use positive reappraisal and problem-solving coping strategies than the low humor group. A weaker relationship existed between appraisal of stress and number of problems in the low humor group because this group perceived greater stress at low and average number of everyday problems than the high humor group. The results were discussed as supporting the role of humor in restructuring a situation so it is less stressful, and the relationship of humor to both emotion-focused and problem-focused coping strategies.
Effective-Component Compatibility of Bufei Yishen Formula III Combined with Electroacupuncture Suppresses Inflammatory Response in Rats with Chronic Obstructive Pulmonary Disease via Regulating SIRT1/NF- κ B Signaling

Abstract:Objective: To explore more efficient treatments for chronic obstructive pulmonary disease (COPD), effective-component compatibility of Bufei Yishen formula III (ECC-BYF III) and electroacupuncture were tested on rats with COPD, and silent information regulator transcript-1 (SIRT1)/nuclear factor-kappaB (NF-κB) signaling was further investigated to interpret the therapy. Methods: In total, 70 rats were randomly divided into control (Control), model (Model), aminophylline (APL), ECC-BYF III, electroacupuncture (EA), ECC-BYF III+EA, and sham electroacupuncture (SA) groups. Cigarette smoke exposure combined with repeated bacterial infections was used to establish COPD models in 1-12 weeks. From 13 to 20 weeks, the ECC-BYF III and APL groups received corresponding drugs; the EA group received electroacupuncture therapy, wherein Dazhui (GV 14), Feishu (BL 13), and Shenshu (BL 23) points were selected; the ECC-BYF III+EA group received ECC-BYF III intragastrically combined with electroacupuncture; and the SA group received simulated electroacupuncture (nonacupoint). Pulmonary function, pulmonary histopathology, the expressions of SIRT1/NF-κB signaling, and inflammation-related mRNA and protein were detected. Results: Significant deterioration was observed in pulmonary function and pulmonary histopathology in rats with COPD (P < 0.01), and inflammatory state was illustrated by increased levels of interleukin- (IL-) 6 and tumor necrosis factor alpha (TNF-α) and decreased levels of IL-10 (P < 0.01). After the intervention of APL, ECC-BYF III, EA, and ECC-BYF III+EA, both pulmonary function and pulmonary histopathology were improved (P < 0.05 and P < 0.01), whereas the levels of IL-6 and TNF-α were decreased and IL-10 was increased (P < 0.05 and P < 0.01). Additionally, the mRNA expressions of IL-6, TNF-α, NF-κB, and acetylated NF-κBp65 (Ac-NF-κB) were noted to decrease, and SIRT1 and IL-10 were increased (P < 0.05 and P < 0.01); the protein expression of SIRT1 was upregulated, and NF-κBp65 and Ac-NF-κB were downregulated (P < 0.05 and P < 0.01). The effect of ECC-BYF III+EA was better in terms of improving pulmonary function and alleviating inflammation than that of the other treatment groups (P < 0.01 and P < 0.05). Conclusions: ECC-BYF III, electroacupuncture, and their combination can suppress inflammation, among which the combination therapy has been proven to be the most effective treatment, and the mechanism may be involved in activating SIRT1/NF-κB signaling.
Mitigation of renal microvascular injury in diabetic kidney disease model mice by electroacupuncture

Abstract:Objective Renal microvascular injury, as the result of diabetic toxicity, plays a vital role in the pathogenesis of diabetic kidney disease (DKD) during diabetes progression. Here, we investigated whether electroacupuncture (EA) could ameliorate renal microvascular impairment to prevent DKD and its underlying mechanism. Methods The male db/db mice with Leprdb mutation were used as the model of type 2 diabetes mellitus-induced DKD and treated with EA at “Zusanli (ST36)” and “Weiwanxiashu (EX-B3)” acupoints for 4 weeks. Age-matched wild-type mice were used as control group. Renal protection of EA was evaluated by mouse urine production, water consumption, renal index and tubules dilation. Two-photon microscope imaging was applied to visualize renal microvascular blood flow in vivo. Immunostaining and western blot analysis were used to detect the glomerular alternations and inflammatory signaling. Results EA significantly attenuated renal dysfunction in db/db mice. The protective effect of EA on renal microvascular recovery was observed both in function and structure analysis. Firstly, EA restored the renal microvascular blood flow in db/db mice. Then, glomerular hypertrophy and glomerular barrier destruction were suppressed after EA, as respectively demonstrated by the reduction of glomerular dilation, Collagen Ⅳ and Claudin-1 deposits. In mechanism, EA suppressed the diabetes-induced inflammatory response in renal microvessels, presenting as the downregulation of inflammatory cytokines interleukin-1β (IL-1β) and tumor necrosis factor (TNF-α), intercellular cell adhesion molecule-1 (ICAM-1) activation, and macrophage infiltration after EA treatment. Conclusion These findings indicated the benefits of EA against renal microvascular impairment and DKD progression, which was associated with the action of anti-inflammation, and supported EA as a promising modality for DKD management.

Abstract:A fourth ventricle lesion containing both areas of typical ependymoma and subependymoma has been studied with the electron microscope. Both foci show ependymal and astrocytic elements. In the subependymoma, a larger number of astrocytes, more frequent cytoplasmic degeneration, and increased glial filaments in the ependymal cells are found. These cellular changes in the subependymoma yield a distinctive light microscopic appearance. However, at the ultrastructural level, the similarity of cellular components, i.e. ependymal and astrocytic cells, makes us believe that subependymoma is a variant of ependymoma and not pure astrocytoma. Tissue culture study and further electron microscopic observation of these in vitro cells have also confirmed the presence of both ependymal and astrocytic cells. Like in vivo ependymal cells, these in vitro ependymal cells form not only glial filaments but also rosettes with specialized cell junctions, microvilli, and cilia. Perivascular pseudorosette formation is also described in detail.
Electroacupuncture improves peripheral neuropathy by up-regulating Sirt1/PGC-1α/TFAM pathway in type 2 diabetes rats with peripheral neuropathy

Abstract:Objectives: To observe the effect of electroacupuncture (EA) on activation of silent information regulator 1 (Sirt1)/peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α)/mitochondrial transcription factor A (TFAM) pathway in type 2 diabetes (T2DM) rats with peripheral neuropathy (DPN) , so as to explore its possible mechanisms underlying improvement of DPN. Methods: Thirty male SD rats were randomly divided into blank control group (n=8) and DPN model group (n=22) which were further divided into model group (n=8) and EA group (n=8) after successful modeling. The model of T2DM was established by high-fat diet and low-dose intraperitoneal injection of streptozocin (35 mg/kg). For rats of the EA group (anesthetized with isoflurane), EA stimulation (2 Hz/15 Hz, 2 mA) was applied to "Tianshu"(ST25) for 20 min, once daily, 6 times a week for 6 weeks. The blood glucose level, body weight, area under curve (AUC) of glucose tolerance test, and hind-paw mechanical pain threshold and thermal pain threshold were observed. The intra-epidermal nerve fiber density (IENFD) of the hind-foot pad was observed by immunofluorescence staining. The motor nerve conduction velocity (MNCV) of the sciatic nerve was measured by using electrophysiological method. H.E. staining was used to observe the histopathological changes of the sciatic nerve after modeling. Transmission electron microscopy (TEM) was used to observe the ultrastructural changes of the sciatic nerve. The protein expressions of energy-related Sirt1, PGC-1α and TFAM in the sciatic nerve was detected by Western blot. Results: Compared with the blank control group, the model group had a higher blood glucose contents and AUC (P<0.001), a slower MNCV (P<0.01), and a decrease in the body weight and in the mechanical and thermal pain thresholds (P<0.001) and IENFD (P<0.001), and in the expression levels of Sirt1, PGC-1α and TFAM (P<0.05, P<0.01). In contrast to the model group, the EA group had a decrease in the blood glucose contents and AUC (P<0.05, P<0.01), and an increase in mechanical and thermal pain thresholds, MNCV, IENFD, and expression levels of Sirt1, PGC-1α and TFAM proteins (P<0.01, P<0.05). In addition, results of histopathological and ultrastructural changes of the sciatic nerve showed more fragmented and disordered distribution of axons on the transverse section, and extensive separation of myelin and axons, uneven myelin thickness, axonal degeneration and irregular shape in the model group, whereas in the EA group, the axons on the transverse section were relatively more dense and more complete, the myelin sheath of the sciatic nerve was relatively uniform, and the axonal shape was relatively regular with relatively milder lesions. Conclusions: EA up-regulates the expressions of Sirt1, PGC-1α, TFAM in T2DM rats with DPN, which may be associated with its functions in improving and repairing the injured peripheral nerves in rats with DPN.
Effects of Electroacupuncture on Bladder Dysfunction and the Expression of PACAP38 in a Diabetic Rat Model.

Abstract:Objective: To explore the effects and the possible mechanism of electroacupuncture (EA) on diabetic bladder dysfunction (DBD) in streptozotocin-high fat diet (STZ-HFD) induced type 2 diabetes mellitus (T2DM) rats.Methods: The experiment was divided into Control, diabetic bladder dysfunction, electroacupuncture, and Sham electroacupuncture group. After 8 weeks of electroacupuncture intervention, the body mass, 24 h urine volume, intraperitoneal glucose tolerance test (IPGTT), and urodynamics were detected. After the wet weight of the bladder was detected, the hematoxylin-eosin (HE), Masson’s trichrome, and TUNEL were used to analyze histological changes. The PACAP38 expressions in the bladder were detected by Real-time PCR and Western blot.Results: Compared to the Control group, the bladder wet weight, 24 h urine volume, blood glucose, maximum bladder capacity, bladder compliance, bladder wall thickness, the smooth muscle/collagen ratio, and apoptosis rate of the diabetic bladder dysfunction group were significantly increased. Moreover, the body mass and leak point pressure were significantly reduced. Compared with the Sham electroacupuncture group, the bladder wet weight, maximum bladder capacity, bladder compliance, bladder wall thickness, and apoptosis rate of the electroacupuncture group were significantly reduced. In contrast, the leak point pressure was increased. The PACAP38 mRNA and PACAP38 protein expression of the diabetic bladder dysfunction group were significantly lower than the Control group, while electroacupuncture treatment could upregulate PACAP38 mRNA levels and PACAP38 protein expression of diabetic bladder dysfunction model rats.Conclusion: electroacupuncture could ameliorate bladder dysfunction in the diabetic bladder dysfunction model rats by reversing bladder remodeling, which might be mainly mediated by regulating the PACAP38 level.

Abstract:It is generally accepted that the media exerts a powerful influence over what the general public feels and believes. This study examined references to epilepsy and the law found in general media communications from January 1993 to December 1994 in the UK. These articles were extracted from general audience material mainly newspapers and magazines. Any article which referred to the legal aspects of epilepsy were then selected. These included those relating to legislation e.g. driving, state benefits and those relating to criminal law e.g. convictions. The reports were analyzed in terms of type of communication, space allotted, circulation and terminology used both in the title and the general text. An attempt was also made to classify the general image which the article portrayed. It is accepted that content analysis itself provides no direct data about the nature of the communicator, the audience or its effects and this method has cautiously been used to classify and describe the manifest content of the method of communication. The study concludes that although it is encouraging that legal aspects of epilepsy are receiving media attention, this attention could be used in a more positive manner in promoting awareness, rather than many of the cases examined which appeared to use negative images in order to pander to popular audience appeal.

Abstract:BACKGROUND:High blood sugar caused by diabetic encephalopathy(DE) can lead to excessive accumulation of reactive oxygen species in the brain, induce oxidative stress, and subsequently cause neuronal degeneration and apoptosis. The Nrf2/HO-1 signaling pathway is one of the most important pathways in oxidative stress response, but the precise mechanism of EA treatment for DE and its specific mechanism of action on the Nfr2/HO-1 pathway remain unclear. METHODS:Male Wistar rats were randomly assigned to four groups: normal, solvent, model, and EA, with 10 rats per group. A DE rat model was induced by intraperitoneal injection of streptozotocin. EA was applied to stimulate the "Zusanli" (ST36) and "Weiwanxiashu" (EXB3) points bilaterally, alternately for 30 min each, once a day for 4 weeks in the EA group. The rats' fasting blood glucose(FBG) levels were measured with a glucometer. The Morris water maze was used to evaluate their learning and memory abilities. The morphology of neurons in the CA1 area of the hippocampus was observed by Nissl staining. Detection of protein expression of Nrf2 and HO-1 in the CA1 area of the hippocampus was performed by immunohistochemistry and immunoblotting. RESULTS:EA treatment reduced blood glucose levels, improved learning and memory abilities, increased the number of neurons in the hippocampal CA1 area, and upregulated the expression of Nrf2 and HO-1 in rats with DE. EA treatment may inhibit oxidative stress by modulating the Nrf2/HO-1 pathway in the hippocampal CA1 area, exerting a protective effect on neuronal cells in the hippocampal area in DE. CONCLUSION:EA enhances the learning and memory abilities of rats with DE by regulating the Nrf2/HO-1 pathway in the CA1 area of the hippocampus. This indicates that EA has the potential to protect neurons by reducing oxidative stress.
Modelling of the inventory replenishment problem with a two-segment piecewise linear demand

Abstract:The classical inventory replenishment problem with a linear function in demand uses a ‘single-segment’ linear function as its demand and can be modelled by a simple algorithm. Moreover, this article extends the algorithm to provide a heuristic solution for the inventory replenishment model with a two-segment linear function in demand called the ‘two-segment piecewise linear demand model’. In addition, this article proposes a general procedure for solving both models. Meanwhile, several examples taken from the literature illustrate our algorithm for these two models with convincing results. Furthermore, this study shows that when the demand is a two-segment piecewise linear function over time, it is better to use the proposed algorithm rather than devising a decoupled solution approach by treating segments separately. Finally, a sensitivity analysis of two factors, demand and cost, is performed. The model is highly extensible and applicable, so it can serve as an inventory planning tool to solve the replenishment problem.
Electroacupuncture Attenuates Cerebral Ischemia-Reperfusion Injury in Diabetic Mice Through Adiponectin Receptor 1-Mediated Phosphorylation of GSK-3β.

Abstract:Diabetes mellitus substantially increases the risk of stroke and enhances brain's vulnerability to ischemia insult. Electroacupuncture (EA) pretreatment was proved to induce cerebral ischemic tolerance in normal stroke models. Whether EA could attenuate cerebral ischemia injury in diabetic mice and the possible underlying mechanism are still unrevealed. Male C57BL/6 mice were subjected to streptozotocin (STZ) for diabetic models. After inducing focal cerebral ischemia model, the levels of plasma and cerebral adiponectin (APN) were measured as well as the expression of cerebral adiponectin receptor 1 (AdipoR1) and 2 (AdipoR2). The neurobehavioral score, infarction volume, and cellular apoptosis were evaluated with or without AdipoR1 short interfering RNA (siRNA). The role of phosphorylation of glycogen synthesis kinase 3 beta (GSK-3β) at Ser-9 in the EA pretreatment was also assessed. EA pretreatment increased both plasma and cerebral APN levels and enhanced neuronal AdipoR1 in diabetic mice. In addition, EA reduced infarct size, improved neurological outcomes, and inhibited cell apoptosis after reperfusion. These beneficial effects were reversed by AdipoR1 knockdown. Furthermore, EA increased GSK-3β phosphorylation (p-GSK-3β) in the ipsilateral penumbra. Augmented p-GSK-3β induced neuroprotective effects similar to those of EA pretreatment. In contrast, dampened p-GSK-3β could reverse the neuroprotective effects of EA. In addition, the increase in p-GSK-3β by EA was abolished by AdipoR1 knockdown. We conclude that EA pretreatment increases the production of APN, which induce protective effects against cerebral ischemia-reperfusion injury through neuronal AdipoR1-mediated phosphorylation of GSK-3β in diabetic mice.
Cortical Spreading Depression Is Neuroprotective: The Challenge of Basic Sciences

Abstract:Hall et al (Headache. 2004;44:204-208) recorded cortical activity during migraine visual aura using magnetoencephalography. These authors conclude that their results are consistent with the theory of cortical spreading depression (CSD). If CSD is the neuro-physiological equivalent—and basis—of the migraine aura, it cannot be the attack-initiating event because the migraine prodrome—that precedes both the headache as well as the aura—can last several hours or a few days.1 About 20 years ago, Pearce underscored that CSD is not the inaugural event at the level of the brain.2 The advent of sophisticated neuroimaging does not diminish the importance of such fundamental pathophysiological considerations. Any theoretical model must correspond with clinical reality, by which benchmark the theory of CSD has serious drawbacks.3,4 Pharmacological agents that do not cross the blood-brain barrier (BBB) instantaneously abort migraine aura.4 In cats, CSD is not associated with release of calcitonin gene-related peptide.5 Dihydroergotamine, acetylsalicylic acid, metoprolol, or valproate do not affect CSD in the cat.6 Spreading depression cannot be elicited in the brain stem of the adult rat, even after conditioning.7 The brain stem is the focus of several pathogenetic concepts for migraine. Premature philosophic commitment encourages scientists to ignore contradictory evidences. A Medline review reveals a number of studies that show the neuroprotective effect of CSD following brain injury or ischemia.8,9 CSD is a powerful yet largely benign stimulus that acutely is capable of providing long-lasting ischemic tolerance probably through ischemic preconditioning; prolonged increase in atrial natriuretic peptide after an acute episode of CSD possibly contributes to CSD-induced neuroprotection.10 The neuroprotective effect of CSD has been known since about two decades; needle puncture of the brain improves survival following forebrain ischemia in mice.11 There is little evidence to support the popular belief that CSD is an important brain pathophysiological mechanism. Insofar as migraine pathophysiology is concerned, the ability of atenolol—that does not cross BBB—to prevent migraine with or without aura heralds the demise of a popular neurological theoretical concept.12 The scientific imperative to explain occurrence of migraine aura must not dull our ability to think logically.
[Effects of antianginal drugs on left ventricular diastolic filling in patients with ischemic heart disease].

Abstract:To study the acute effects of isosorbide dinitrate (ISDN), nifedipine, propranolol and placebo on left ventricular diastolic filling in patients with myocardial ischemia, we repetitively performed exercise radionuclide angiography (RNA) following random assignment of each drug in 12 patients with effort angina pectoris, and 5 normal controls. Left ventricular time activity curves were constructed using computer technique. We obtained changes of hemodynamic parameters such as endodiastolic volume, systemic vascular resistance and ejection fraction. The peak filling rate (PFR), and filling rate in the first third of the diastolic period (1/3 FR) were used as the indexes of LV diastolic filling. 1/3 FR remained unchanged during peak exercise by the patients, while it increased significantly in normal controls. After administration of ISDN and nifedipine, 1/3 FR increased significantly during peak exercise by the patients. Especially after nifedipine, 1/3 FR improved significantly compared with the improvement after ISDN, although these two drugs had the same effects on systolic function. These findings showed that 1/3 FR was reliable as an indicator of LV diastolic filling in ischemic heart disease, and nifedipine improved LV diastolic filling not only by improving ischemic changes in myocardium but also by it's direct effect on left ventricular relaxation.
Electro-acupuncture preconditioning abrogates the elevation of c-Fos and c-Jun expression in neonatal hypoxic-ischemic rat brains induced by glibenclamide, an ATP-sensitive potassium channel blocker

Abstract:This study aimed to clarify the neuroprotective mechanism of electro-acupuncture (EA) preconditioning on hypoxic-ischemic brain injury (HIBI). Using Western blot, the expression of c-fos protein (c-Fos) and c-jun protein (c-Jun) induced by glibenclamide, an ATP-sensitive potassium (KATP) channel blocker was examined from cerebral cortical and hippocampal samples in neonatal hypoxic-ischemic rats, with or without EA preconditioning. EA was performed on Hegu (LI4), a well-known acupoint commonly used in Oriental medicine for the treatment of neuronal injury resulting from hypoxia–ischemia (HI). Preconditioned rats were treated with either diazoxide, a KATP channel opener, glibenclamide, or sterile saline injected into the left lateral ventricle (i.c.v.), with or without EA administration before HI insult. Interestingly, low c-Fos and c-Jun expressions were found both in diazoxide and EA groups, 24 h after HI. Furthermore, significant differences in relative optical density (ROD) were found between glibenclamide and HI control groups (P≤0.05), as well as between the group administered glibenclamide after EA and the HI control group (P≤0.05). However, the level of c-Fos and c-Jun expression in the group administered glibenclamide after EA was significantly lower than in the glibenclamide group (P≤0.05). The present findings indicate that the effectiveness of EA preconditioning against HIBI may be mediated via the opening of KATP channels.
Effects of electroacupuncture stimulation at different spinal segmental levels in a rat model of diabetes mellitus

Abstract:Background: Diabetes mellitus (DM) is associated with high morbidity, mortality and economic cost. Studies have shown that acupuncture can improve many symptoms of DM. Objectives: To examine for differences in effects of electroacupuncture (EA) stimulation at Weiwanxiashu, BL15 and BL23 in the streptozotocin (STZ)-induced DM rat model, to help guide clinical selection of acupuncture points. Methods: 90 male rats weighing 160Â±5 g were used. 12 rats were control fed (Normal group) and 78 were fed a high-fat high-sugar diet for 8 weeks and underwent intraperitoneal STZ injection to model DM. 60 animals that met modelling criteria were randomly divided into an untreated DM group and four groups receiving EA at Weiwanxiashu (DM+WWX group), BL15 (DM+BL15 group), BL23 (DM+BL23 group) or a non-traditional acupuncture point on the tail (DM+Tail group). Fasting blood glucose, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and insulin levels were determined and an oral glucose tolerance test (OGTT) performed. Results: EA at Weiwanxiashu had a glucose-lowering effect on the 21st and 28th days, decreased TC, TG and LDL-C levels, increase insulin levels and improved glucose tolerance. EA at BL15 had a glucose-lowering effect on the7th, 14th and 21st days of intervention but did not impact lipids, insulin or OGTT parameters. EA at BL23 or on the tail had no significant effects. Conclusion: EA at Weiwanxiashu and BL15 had differential effects on metabolic markers in the STZ-induced rat model of DM. These effects may be explained neuroanatomically by variations in the segmental innervation of the tissues at these locations.
Acupuncture improve proteinuria in diabetic kidney disease rats by inhibiting ferroptosis and epithelial-mesenchymal transition

Abstract:Objective: To explore the mechanism of acupuncture to relieve diabetic proteinuria in Diabetic Kidney Disease (DKD). Methods: A total of 10 male Sprague-Dawley rats were randomly selected as the negative control group (NC), and a further 30 rats were fed a high-fat diet (HFD) and intraperitoneal streptozotocin (STZ). The DKD model rats in the model group (DKD) and the acupuncture group (DKD + Acu) were randomly assigned. After 4 weeks of intervention, collected urine, peripheral blood, and renal tissues from all rats, and assessed blood urea nitrogen, serum creatinine, triglyceride, 24-h urine protein quantification, and blood glucose, left kidney weight, kidney body ratio index, then observe changes in renal histology in the rats. The renal cortex tissues of three rats from each group were sent for transcriptomic analysis. According to the results of transcriptomic analysis, various kits were used to detect SOD, MDA, GSH, GSH-px, and iron concentration. The expression levels of GPX4 and System Xc-, members of the antioxidative stress pathway, and TfR 1, SLC39A14, FTH 1, and SLC40A1, involved in iron metabolism, in the kidney tissues were measured by western blotting and reverse transcription-quantitative PCR. The expression of mesenchymal phenotype markers and podocyte-specific markers were evaluated by immunofluorescence. Results: Acupuncture promoted the levels of GSH, GSH-px, and SOD, decreased the level of MDA (P < 0.05), promoted the expression of GPX4 and System Xc- (P < 0.05), decreased the expression of TfR1 and SLC39A14 (P < 0.01), and increased the expression of FTH 1 and SLC40A1 (P < 0.05), inhibited the expression of TGF-β1, desmin, FSP-1, and α-SMA (P < 0.05), promoted the expression of Nephrin, Podocin, and CD2AP (P < 0.05). Conclusion: Improving the ability of podocytes to prevent oxidative stress and restoring iron ion homeostasis, can improve Ferroptosis and block epithelial-mesenchymal transition, improve podocyte injury, restore filtration function, and reduce proteinuria in DKD rats.

Abstract:Objective: To observe the effects of electroacupuncture (EA) on renal fibrosis, the expression of transforming growth factor-β1 (TGF-β1) and epithelial mesenchymal transition (EMT) marker proteins in renal tissue of spontaneously hypertensive rats (SHR), so as to explore the underlying mechanism on EA alleviating hypertensive renal impairment. Methods: Twenty-four male SHR were randomly divided into model group, losartan group and EA group, with 8 rats in each group, and eight male Wistar-Kyoto rats were taken as the normal group. Rats in the losartan group received gavage of losartan potassium solution (3 mg/mL, 30 mg·kg-1·d-1),once every other day for 12 weeks. Rats in the EA group received EA stimulation at bilateral "Shenshu" (BL23) and "Geshu" (BL17) (2 Hz/15 Hz, 1.0 mA), 15 min each time, once every other day for 12 weeks. The systolic blood pressure of caudal artery was measured before, and 4, 8 and 12 weeks after the intervention. The 24-hour urinary protein was measured before, and 6 and 12 weeks after the intervention. Histopathological changes of the left renal tissue were observed under light mircoscope after H.E. stain. Extracellular matrix (ECM) in renal tissues was observed after periodate Schiff staining. Basement membrane and collagen fibers were observed after Masson staining with collagen volume fraction (CVF) evaluated. The expression of TGF-β1 mRNA in the right renal was detected by real-time fluorescence quantitative PCR. The expression of TGF-β1 and EMT marker E-cadherin, α-SMA and fibronectin (FN) proteins in the left renal tissue was measured by immunohistochemistry. Results: In the model group, irregular arrangement of nephrocytes, renal tubule atrophy, lumen stenosis, ECM hyperplasia and deposition, scar and sclerosis were observed, which were relatively milder in the EA and losartan groups. Compared with the normal group, tubulointerstitium CVF, systolic blood pressure of caudal artery before, and at 4, 8 and 12 weeks after the intervention, 24-hour urinary protein before, and at 6 and 12 weeks after the intervention, the expression of TGF-β1 mRNA, area of TGF-β1, α-SMA and FN positive staining in renal tissues were significantly increased (P<0.01), while the area of E-cadherin positive staining was significantly decreased (P<0.01) in the model group. Compared with the model group, tubulointerstitium CVF, systolic blood pressure of caudal artery at 4, 8 and 12 weeks after the intervention, 24-hour urinary protein at 6 and 12 weeks after the intervention, the expression of TGF-β1 mRNA, area of TGF-β1, α-SMA and FN positive staining in renal tissues were significantly decreased (P<0.01，P<0.05), while area of E-cadherin positive staining was significantly increased (P<0.01) in the losartan and EA groups. Compared with the losartan group, the area of E-cadherin was conside-rately increased (P<0.01), while the area of α-SMA protein decreased (P<0.01) in the EA group. Conclusion: EA could effectively alleviate hypertension and renal interstitial fibrosis in SHR, the mechanism of which may be related to its function in reducing the expression of TGF-β1 and inhibiting EMT in renal tissue.

Abstract:Glycosaminoglycans (GAGs) have been implicated in many functions of human blood cells. Normal human monocytes grown on plastic culture dishes for five days display macrophage-like morphology and functional characteristics concurrent with the appearance of a 4,6-disulfated chondroitin. This process has been regarded as in vitro differentiation. HL-60 human promyelocytic leukemia cells produce chondroitin 4-sulfate, which is the major GAG constituent of normal granulocytes and monocytes. These cells can be induced by phorbol myristate acetate (PMA) to develop many macrophage-like characteristics. This study investigates GAG production by HL-60 cells after prolonged exposure to PMA. Disulfated chondroitin molecules were not detected. This biochemical abnormality may be an important factor in the functional deficiencies of these induced cells.
