Abstract:Objective To estimate the bioequivalence and pharmacokinetics of two estradiol and norethisterone acetate-containing tablets in healthy post menopausal female volunteers.Methods A single oral dose of test(domestic) and control(imported) estradiol and norethisterone acetate-containing tablets was given 2 mg estradiol and 1 mg norethisterone to 20 volunteers in an open randomized crossover design.The serum concentration of estradoil,estone and norethisterone was determined by chemiluminescence immunoassay method,radioimmunoassay method and HPLC-MS/MS method,respectively.The pharmacokinetic parameters and relative bioavalibility were calculated to evaluate the bioequivalence of two preparations.Results The principle pharmacokinetic parameters of test and control were as follows:①estradoil,Cmax were(102.92±26.68)and(93.52±18.91)pg·mL-1,tmax were(1.05±0.78)and(2.32±1.54)h,t1/2 were(18.97±11.94)and(26.74±16.83)h,AUC0–t were(1621.53±737.76)and(1620.47±735.12)pg·h·mL-1,AUC0–∞ were(1980.43±821.71)and(2187.88±955.43)pg·h·mL-1,the relative bioavalibility F0–t and F0–∞ were(99.84±17.78)% and(94.41±24.98)%,respectively.②estone,Cmax were(313.72±93.89)and(313.05±103.76)pg·mL-1,tmax were(3.93±1.32)and(4.16±1.21)h,t1/2 were(11.86±3.76)and(12.04±5.10)h,AUC0–t were(4480.56±1751.37)and(4622.88±1726.57)pg·h·mL-1,AUC0–∞ were(4665.80±1765.01) and(4878.20±1855.23)pg·h·mL-1,the relative bioavalibility F0–t and F0–∞ were(97.47±15.18)% and(96.71±15.11)%,respectively.③norethisterone,Cmax were(12.73±6.53)and(12.62±6.32)pg·mL-1,tmax were(1.39±0.69)and(1.15±0.57)h,t1/2 were(7.04±2.66)and(7.98±2.66)h,AUC0–t were(46.92±28.16)and(49.28±31.46)ng·h·mL-1,AUC0–∞ were(52.99±32.96)and(56.81±37.07)ng·h·mL-1,the relative bioavalibility F0–t and F0–∞ were(99.74±16.19)% and(97.15±16.04)%,respectively.The main pharmacokinetic parameters showed no statistically significant difference between two preparations.Conclusion The two preparations of estradiol and norethisterone acetate-containing tablets were bioequivalent.
