Abstract:The determination of biogenic amines (BAs) plays an important role in ensuring food safety. In this study, the occurrence of BAs in 98 wine samples readily available to Chinese consumers, as well as the dynamics of BAs during the spontaneous fermentation of Cabernet Sauvignon and Marselan wines were determined by an improved ultra-performance liquid chromatography (UPLC) method. It was found that this novel approach presented a reduced Dns-Cl amount (0.55 mL), lower limits of detection (1.5-4.2 mu g/L) and quantification (4.9-14.1 mu g/L), and a much shorter analysis time (6 min). The cumulative BA levels varied significantly, from 3.46 +/- 0.05 mg/L in Sauvignon Blanc to 59.12 +/- 0.09 mg/L in Cabernet Sauvignon. Putrescine was the most abundant amine in wine samples, with concentrations ranging from 0.32 +/- 0.12 mg/L in Cabernet Sauvignon to 31.17 +/- 0.27 mg/L in Pinot Noir, while spermine was not detected in 80% of the wine samples and had the lowest concentration (average 0.205 +/- 0.13 mg/L). None of the investigated samples presented relevant levels of histamine and tyramine above the limits specified by the European Food Safety Authority (EFSA). Thus, wines available in China are not a serious toxicological risk regarding the type and content of BAs. The malolactic fermentation (MLF) and the later stage of alcohol fermentation (AF) were found to be critical control points (CCPs) in the spontaneous fermentation of both Marselan and Cabernet Sauvignon wines. Moreover, the contents of putrescine, tyramine and tryptamine were significantly correlated with the contents of other amines, and, thus, could be used as important indicators of the quality and safety of wines. In general, these results are helpful to assess the risk of dietary intake of BAs and to evaluate the winemaking processes by food safety authorities.

Abstract:Biogenic amines produced during fermentation are harmful and deteriorate the sensory experience of food. However, methods for overcoming the impact of biogenic amines in Huangjiu, a traditional Chinese food, have not been considered for research. This review analyzes various studies that have evaluated the content of biogenic amine content in Huangjiu, the formation of biogenic amines, and associated factors. Furthermore, the study discusses the health hazards of biogenic amines, and summarizes the prevalent approaches to determine their content in Huangjiu. Finally, the strategies to reduce the content of biogenic amines in Huangjiu have been listed. Although the analysis of biogenic amines in food products is crucial, government legislation and stringent recommended limits for biogenic amines in Huangjiu are nonexistent. This necessitates advanced research on regulating biogenic amines in Huangjiu production, along with the development of real-time monitoring techniques. Further efforts are required to accelerate the establishment of relevant regulations and ensure the safety of fermented foods.

Abstract:Biogenic amines (BAs) are a class of bioactive organics produced during the fermentation of soy sauce. A high concentration of BAs may bring about serious physiological and toxicological effects on the human body. In this study, we reported an optimized process to produce soy sauce with lower BA concentration and found the contents of putrescine, cadaverine and histamine increased with the increase of fermentation temperature but decreased with the increase of NaCl concentration. The final content of total BAs with improved fermentation was 105.56 +/- 0.13 mg/L, which was reduced by 89.11% compared to traditional brewing. Besides, the pilot production test was performed to verify the optimized conditions and physicochemical indexes were measured to better understand the change principle of the chemical compounds. Taken together, we present an effective process to inhibit the formation of BAs while ensuring that characteristic nutrients are not lost.

Abstract:This study evaluated the levels of eight biogenic amines in 59 craft beers of five styles and monitored the changes during beer fermentation, showing that putrescine and tryptamine were the most abundant at maximum values of 46.14 mg/L and 89.97 mg/L, respectively. This research indicated for the first time that dark beer, such as Stout/Porter, displayed the highest total biogenic amine content due to considerable tryptamine accumulation, with a maximum value of 116.95 mg/L. The total biogenic amine level increased gradually during the segmental saccharification and main fermentation stages, representing the two critical control points for their formation during beer fermentation. This study provides a theoretical basis and technical guidance for the safe and stan-dardized production of craft beer and the formulation of biogenic amines limit standards, which is highly sig-nificant for protecting the health of consumers.

Abstract:Biogenic amines (BAs) are natural contaminants of wine that originate from decarboxylase microorganisms involved in fermentation processes. The primary relevance of biogenic amines in food could have both toxic effects on consumers’ health (i.e., allergic reactions, nausea, tremors, etc.), if present at high concentrations, and concurrently it can be considered as a remarkable indicator of quality and/or freshness. Therefore, the presence of nine biogenic amines [Tryptamine (TRP), ß-phenylethylamine (ß-PEA), putrescine (PUT), cadaverine (CAD), histamine (HIS), serotonin (SER), tyramine (TYR), spermidine (SPD), and spermine (SPM)] was investigated in red and white wine samples, which differed in the winemaking processes. The qualitative-quantitative determination of BAs was carried out by chromatographic methods (HPLC-UV/Vis and LC-ESI-MS). The analysis showed that both winemaking processes had all the nine BAs considered in the study at different amounts. Data showed that red wines had a higher concentration of PUT (10.52 mg L−1), TYR (7.57 mg L−1), and HIS (6.5 mg L−1), the BAs most involved in food poisoning, compared to white wines, probably related to the different type of fermentation (alcoholic and malolactic).

Abstract:The decarboxylation of the corresponding amino acids by microorganisms leads to the formation of biogenic amines (BAs). From a toxicological point of view, BAs can cause undesirable physiological effects in sensitive individuals, particularly if their metabolism is blocked or genetically altered. The current study aimed to monitor and evaluate the content of eight biogenic amines (BAs) in 232 samples of wines (white, rosé, red) produced in the Central European region (Zone B). White wines (180 samples), rosé wines (17 samples), and red wines (35 samples) were analyzed. High-performance liquid chromatography equipped with a ultraviolet–visible diode array detector (UV/VIS DAD) was applied to identify and quantify the BAs present in wines. In general, histamine (HIS), tyramine (TYM), putrescine (PUT), cadaverine (CAD), phenylethylamine (PEA), spermine (SPN) and spermidine (SPD) were detected in all tested wine samples. Tryptamine (TRM) was not present in any of the samples examined. In white and red wines, SPD, TYM, and PUT were most often detected. Regarding rosé wines, the three major BAs were SPN, TYM, and CAD. The BA content in red wines was generally higher than in rosé and white wines. However, HIS concentrations above the recommended limit of 10 mg/L were detected in 9% of the red wine samples. In addition, alarming levels of PUT, HIS, TYM, and PEA, with serious potential impact on consumer health, were recorded in two red wine samples. On the whole, the presence and concentrations of BAs in wine should be constantly evaluated, primarily because alcohol intensifies the hazardous effects of BAs.

Effects of the microbial community on the formation of volatile compounds and biogenic amines during the traditional brewing of Hongqu rice wine

Abstract:As a typical representative of Chinese rice wine (Huangjiu), Hongqu rice wine is famous for its red color, mellow taste and strong fragrance. However, due to the open brewing environment and traditional fermentation technology, there are some safety risks in traditional brewed Hongqu rice wine, such as a certain amount of biogenic amines. In this study, the dynamic changes and the differences of microbial communities and volatile flavor components between two types of Hongqu rice wine with high and low biogenic amine contents (LBAW and HBAW) during the traditional brewing were systematically investigated. The results showed that the total biogenic amine contents in LBAW and HBAW were 20.91 and 69.06 mg/L, respectively. The contents of putrescine, cadaverine, spermine and spermidine in HBAW were significantly higher than those in LBAW, and it was noteworthy that spermine content in HBAW was 17.62 mg/L, which was not detected in LBAW. In addition, the volatile flavor characteristics of the two kinds of Hongqu rice wine were obviously different. The contents of acetophenone, n-butyl butanoate and benzothiazole were obviously higher in HBAW, while the contents of isoamyl acetate, ethyl lactate, ethyl caprate and phenylethyl alcohol were significantly higher in LBAW. High-throughput sequencing of 16S/ITS amplicon revealed that Weissella, Kosakonia, Pantoea, Monascus, Saccharomyces and Millerozyma were the predominant microbial genera during the traditional brewing of HBAW, while Weissella, Kosakonia, Monascus, Saccharomyces and Issatchenkia were the predominant microbial genera during the traditional brewing of LBAW. Correlation analysis revealed that biogenic amines were significantly negatively correlated with unclassified_o_Saccharomycetales, Cyberlindnera, Zygoascus, Aspergillus and Acinetobacter, but positively correlated with Lactobacillus, Pediococcus, Millerozyma and Apiotrichum. In addition, we also found that Lactobacillus, Pediococcus and Saccharomyces were significantly positively correlated with most of the volatile flavor components, while Candida, Trichosporon and Monascus were significantly negatively correlated with most of the volatile flavor components. In addition, bioinformatical analysis based on PICRUSt demonstrated that the key enzymes for biogenic amine biosynthesis were more abundant in the microbial community of HBAW than LBAW. These findings demonstrate that the formations of volatile flavor and biogenic amines in Hongqu rice wine are influenced by microbial community during the fermentation. This work facilitates scientific understanding of the formation mechanism of biogenic amines, and may be useful to develop effective strategies to improve the quality of Hongqu rice wine.

Abstract:This study examined the biogenic amine content of the brewing raw material (wheat Qu, seed starter, mash or liquid) during fermentation and during the maturation and storage process. Pre-column derivatization and high-performance liquid chromatography were employed to analyse the dynamic changes in biogenic amines during the Chinese rice wine brewing process. Some aliphatic amines (putrescine, cadaverine, spermine) were already found to be present in the glutinous rice and wheat Qu, but the content was below 2.88 mg/kg. None of the amines were detected in the brewing water. The biogenic amine content in the seed starter exhibited a large variation range (16.43-87.72 mg/L), which could influence the total content of biogenic amines prior to fermentation. A much more obvious increase in biogenic amines stemmed from the fermentation process, with the presence of a biogenic amine-producing bacteria and precursor amino amines. In the first 2 days, the biogenic amine content increased sharply, followed by a slow increase over the next 4 days, and the content reached a maximum value on day 6; subsequently there was a decrease or fluctuation in the concentration. After clarification and cooking, the biogenic amine content increased significantly; however, a slight decrease was observed during storage, especially for putrescine and histamine. Putrescine and tyramine were the most prevalent amines in all of the samples. Overall, the content of biogenic amines was found to be within the safe level for human health, ranging from 54.52 to 146. 35 mg/L. Copyright (C) 2013 The Institute of Brewing & Distilling

Abstract:A new HPLC method for determining biogenic amines in wines is developed. This method is based on pre-column amine derivatization, further separation of derivatives and on-line hyphenation of HPLC to atmospheric pressure chemical ionization mass spectrometry (APCI-MS). Biogenic amines have been derivatized with 1,2-naphthoquinone-4-sulfonate at 65 degrees C and pH 9.2 for 5 min. The separation of derivatives has been accomplished in a C(18) analytical column using an elution gradient based on increasing the percentage of methanol. Derivatives have been ionized in positive mode and detected by selected ion monitoring. The operating conditions of the APCI-MS system (voltages, temperatures and gases) have been thoroughly optimized to obtain the maximum sensitivity for all analytes. In the selected conditions, APCI-MS spectra display little fragmentation and good signal-to-noise ratio. Depending on the amine characteristics, the main spectral peaks are due to mono- and di-derivative products. Figures of merit of the method have been established under the selected conditions using red wine samples. Recoveries ranging from 94% to 106% have been obtained which prove excellent accuracy of the method in the determination of histamine, putrescine, cadaverine, tryptamine, phenylethylamine, tyramine and serotonin in red wines. The proposed method has been applied to the analysis of commercial wines from different Spanish regions.
Determination of Biogenic Amines in Semi-Dry and Semi-Sweet Chinese Rice Wines from the Shaoxing Region

Abstract:The aim of this study was to investigate the levels of nine biogenic amines in two types of Chinese rice wine (semi-dry and semi-sweet) from the Shaoxing region. Thirty-nine samples from different manufacturers were analyzed by reversed-phase high-performance liquid chromatography and ultraviolet detection after pre-column derivatization with benzoyl chloride. Serotonin and tyramine were the most prevalent amines (100%) followed by histamine, cadaverine and putrescine (95.2-88.1%). Spermine, spermidine, 2-phenylethylamine and tryptamine were not detected in any sample. The most prominent biogenic amine was serotonin followed by putrescine, tyramine, cadaverine and histamine, their mean contents being 53.3, 24.3, 19.2, 9.08 and 8.83 mg/L, respectively. The total biogenic amine contents were variable, ranging from 29.3 to 260 mg/L, with 115 mg/L on average. Significantly higher (P < 0.05) serotonin and total biogenic amine levels were detected in semi-sweet rice wines. Significant and strong correlations (r >= 0.75, P < 0.01) were observed between some individual biogenic amine contents and between some individual and total biogenic amine concentrations. Three clusters comprising rice wines of different biogenic amines content profile were identified using a combination of hierarchical and k-means clustering. (C) 2012 Elsevier Ltd. Abstract:Ethyl carbamate(EC),a known genotoxic compound,is mainly found in grape wine and other alcoholic beverages.It is mainly formed by the reaction between urea(produced by the breakdown of arginine by the yeast) and citrulline(produced by the degradation of arginine by malolactic bacteria and ethanol).In this paper,the carcinogenic features and the formation pathway of EC in grape wine and the factors influenc-ing EC formation were introduced.The measures to reduce EC in grape wine were discussed,which provided theoretical evidences for reducing EC content in grape wine.
Determination of 10 Mycotoxins in Wine, Baijiu, and Huangjiu of the Chinese Market by Liquid Chromatography Tandem Mass Spectrometry and Exposure Estimation

Abstract:In this study, liquid chromatography tandem mass spectrometry (LC–MS/MS) was employed to analyze the prevalence of 10 mycotoxins in 140 samples from the Chinese market, aiming to assess the exposure of Chinese individuals to these mycotoxins through the consumption of wine, baijiu, and huangjiu. Mycotoxins were detected in 98% of the samples, with fumonisins (FBs), deoxynivalenol (DON), and zearalenone (ZEN) exhibiting positive rates exceeding 50%. Regarding the exposure of the Chinese population to mycotoxins resulting from alcoholic beverage consumption, fruit wine intake made a relatively significant contribution to aflatoxin exposure, while baijiu showed a relatively significant contribution to ZEN exposure (1.84%). The analysis of the correlation between grape variety, wine region, and mycotoxin content demonstrated that FBs, ZEN, and DON were significantly influenced by grape variety and wine region. This research holds great significance in protecting human life and health, as well as in the production of safer alcoholic beverages.

Abstract:As one of the typical representatives of Chinese rice wine (Huangjiu), Hongqu rice wine is produced with glutinous rice as the main raw material and Hongqu as the fermentation starter. The complex microbial flora in the brewing process may have a great influence on the formation of the flavor quality and drinking safety of Hongqu rice wine. Previous studies have shown that high biogenic amine (BA) content in rice wine has potential physiological toxicity and has become a bottleneck problem restricting the development of the rice wine industry. This study aimed to evaluate the regulatory effects of the microbial community on the formation of BAs and volatile flavor components during the brewing of Hongqu rice wine. The results demonstrated that histamine, putrescine, cadaverine, tyramine, tryptamine, spermine, and spermidine were the main BAs in Hongqu rice wine. The contents of putrescine, cadaverine, histamine, tyramine, and spermidine in Hongqu rice wine of HBAs (with higher BAs content) were significantly higher than those in LBAs (with lower BAs content). GC-MS testing results showed that there were significant differences in the composition of volatile organic compounds (VOCs) between HBAs and LBAs. Among them, VOCs such as 2-methoxy-4-vinylphenol, ethyl caprate, phenethyl acetate, ethyl lactate, ethyl myristate, ethyl palmitate, ethyl n-octadecanoate, ethyl oleate, and ethyl linoleate were identified as the characteristic volatile components with significant differences between HBAs and LBAs. Microbiome analysis based on metagenomic sequencing revealed that unclassified_g_Pantoea, Klebsiella pneumoniae, Panobacter disperse, unclassified_f_Enterobacteriaceae, Leuconostoc mesenteroides, and Saccharomyces cerevisiae were the dominant microbial species in the HBA brewing process, while Weissella confuse, Pediococcus acidilactici, Saccharomyces cerevisiae, and Aspergillus niger were the dominant microbial species in the LBA brewing process. Furthermore, correlation heatmap analysis demonstrated that BAs were positively related to Lactobacillus curvatus, Lactococcus lactis, and Leuconostoc mesenteroides. Bioinformatical analysis based on the KEGG database revealed that the microbial genes encoding enzymes involved in BAs' synthesis were more abundant in HBAs, and the abundances of microbial genes encoding enzymes related to BAs' degradation and the metabolism of characteristic volatile components were higher in LBAs. Overall, this work provides important scientific data for enhancing the flavor quality of Hongqu rice wine and lays a solid foundation for the healthy development of the Hongqu rice wine industry.

Abstract:Safety and quality are significant challenges for food; namely, safety represents a big threat all over the world and is one of the most important goal to be achieved in both Western Society and Developing Countries. Wine safety mainly relies upon some metabolites and many of them are of microbial origin. The main goal of this review is a focus on two kinds of compounds (biogenic amines and mycotoxins, mainly Ochratoxin A) for their deleterious effects on health. For each class of compounds, we will focus on two different traits: (a) synthesis of the compounds in wine, with a brief description of the most important microorganisms and factors leading this phenomenon; (b) prevention and/or correction strategies and new trends. In addition, there is a focus on a recent predictive tool able to predict toxin contamination of grape, in order to perform some prevention approaches and achieve safe wine.

Abstract:Many amine compounds are exist in Beer.Whose taste and quality are closely related to the content and the kind of the amine compounds.A method for trace amine compounds analysis in beer was developed in this paper.The amine compounds with-NH2 and-NH-in beer were analyzed by HPLC separation and LIFD determination,using FITC as derivative regent.Then the nitrosamines in beer were quantitative analyzed by low-temperature vacuum distillation combined with GC-FID.Further more,various kinds of beer on sell were analyzed.Using the developed method,the result indicated that there existed some differences of amine compounds among different brands and different series of beer.Setting the integral threshold at 1mv,the largest number of HPLC peaks is 53 and the least number is 38.In the chromatograms of beer samples,Analyze the content of nitrosamines in three kinds of beer and trace nitrosamines have been detected in two kinds of them.The method has a high sensitivity,good stability and is easily operated.It can be used for detection of quality of beer and fake and shoddy products and is valuable significance to control the content of carcinogenic amine compounds.It is very important for the health of people.The method is also suitable for amine compounds analysis in other beverage and food.

Abstract:To determine the effect of cofermentation of Saccharomyces cerevisiae and different LABs on prune wine quality, this study compared phenolic compounds, organic acids, soluble sugars, biogenic amines and volatile flavor compounds among different treatments. The results showed that inoculation of LAB increased DPPH and total flavonoid content. Malic acid content was reduced in HS, HB and HF. Histamine content in S, F and B was lower than the limits in French and Australian wines. 15 phenolic compounds were identified. Yangmeilin and chlorogenic acid were detected only in HS, HF and HB. 51 volatile flavor compounds were identified, esters being the most diverse and abundant. 14 volatile flavor compounds with OAV > 1 contributed highly to the aroma of prune wine. 9 chemical markers including resveratrol, rutin, and catechin were screened to explain intergroup differences by OPLS-DA. This study provides new insights into the processing and quality analysis of prunes.
One-pot derivatization/magnetic solid-phase extraction combined with high-performance liquid chromatography-fluorescence detection for rapid analysis of biogenic amines in alcoholic beverages

Abstract:The determination of biogenic amines (BAs) in alcoholic beverages is crucial for assessing their health impact, ensuring beverage quality, and guaranteeing safety. Herein, a rapid one-pot derivatization/magnetic solid-phase extraction (OPD/MSPE) method was proposed using 6-aminoquinolinyl-N-hydroxysuccinimide carbamate as the derivatization reagent and magnetic hydroxyl-functionalized multi-walled carbon nanotubes as the extraction material. Integration of derivatization and extraction steps simplifies the sample preparation process, taking only three minutes and eliminating the need for centrifugation by utilizing magnetic sorbent. The resulting desorption solution was directly analyzed by high-performance liquid chromatography-fluorescence detection (HPLC-FLD) without any evaporation or reconstitution steps. The integrated OPD/MSPE-HPLC-FLD method demonstrates excellent linearity (R2 > 0.992), accuracy (relative recoveries: 85.1-109.2%), precision (RSDs≤9.7%) and detection limits (limits of detection: 0.3-2 ng/mL). It has been successfully applied to determine free BAs in various alcoholic beverages, including red wine, Baijiu, Huangjiu, and beer. This method enables rapid, sensitive and precise analysis of BAs in alcoholic beverages.
Detection and analysis of electrochemical signals in wine fermentation process

Abstract:This study presents an in-depth analysis of the electrochemical fingerprint spectra of different yeast strains (BV818, L2323, and ExcellenceXR) during the fermentation process of red grape wine. It was found that the electrochemical fingerprint spectra significantly change as fermentation progresses, indicative of molecular breakdown and the generation of electrochemically active small molecules. For instance, the sugar content exhibited a decline over time with BV818 showing the fastest consumption rate. Similarly, CO 2 emissions and alcohol concentration showed an increase over time, with BV818 yielding the highest results. The total acidity initially increased and then decreased, while the total phenolic content displayed a trend of initial increase followed by a decrease. Anthocyanin levels dropped rapidly within the first 48 h of fermentation and then stabilized. A significant correlation was observed between the changes in electrochemical fingerprint spectra and the total acid and total phenol content. These findings highlight the potential application of electrochemical fingerprinting for monitoring the wine fermentation process and evaluating the impact of different yeast strains on wine quality and composition. Further research will be focused on a more detailed analysis of changes in related substances and broader applications of this technique in the wine industry.

Abstract:Abstract Biogenic amines in gastrointestinal tract are the important metabolites of dietary protein and amino acids under the help of gut digestive enzymes and microbes, which play a crucial role in the regulation of intestinal functions, including digestion, absorption, and local immunity. However, high concentrations of biogenic amines can induce adverse reactions and are harmful to animal health. Therefore, it is crucial to have a clear understanding of how different biogenic amines interact with body intestinal function signaling pathway and to monitor the content of biogenic amines in the gastrointestinal tract. And in turn, the proper concentration of dietary protein and balanced amino acids for humans and livestock could be given. Though numerous methods have been developed and improved for the detection of biogenic amines in foods or wines，much less attention was paid straightly on the determination of amines level in gastrointestinal tract. In this article, we mainly focus on the interaction of amines with intestinal function signaling pathway and the broad impacts on animal physiology, and our modified method to accurately and quickly detect the biogenic amines in the digests of animal intestine.
