Abstract:A mild method for the preparation of tryptanthrin and its derivatives by copper(I) catalysis with two identical isatins in the presence of KHCO 3 as a base is described. Furthermore, more complicated tryptanthrin derivatives are obtained in two steps with two different isatins in one pot. This work features readily available starting materials, mild reaction conditions, simple operation, good functional group tolerance, high yields and selectivity. This transformation not only provided a new access to tryptanthrin and its derivatives, but also enriched the chemistry of isatins and copper catalysis.

Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:Enantioselective synthesis of tetrahydroisoquinolines bearing an all-carbon quaternary stereogenic center, was achieved via asymmetric C-H activation with high enantioselectivities (up to 93% ee). Fair substrate tolerance was indicated throughout the scope investigation and no evident loss of enantioselectivity was exhibited in late-stage derivatization. This study provides incentives for the construction of diverse chiral isoquinoline derivatives, which are prevalent among pharmaceuticals, natural products, etc.

Abstract:Transition-metal-catalyzed C-N bond formation reactions have been widely studied in recent decades, but site-selective intermolecular C-N cross-coupling between the carbon and nitrogen sites of two identical molecules is less explored. Herein, we report the highly selective copper-catalyzed oxidative cyclization/C-N cross-coupling of two tryptamines with O2 as the terminal oxidant, which provides practical and direct access to a new class of indole-linked-pyrroloindoline compounds in moderate to good yields. Mechanistic studies suggested that the coordination between the copper catalyst and the tryptamine substrate is key for chemoselectivity control.

Abstract:The total syntheses of aspidospermidine, N-methylaspidospermidine, N-acetylaspidospermidine, and aspidospermine were achieved from a common pentacyclic indoline intermediate. The common pentacyclic indoline intermediate was synthesized on a gram scale through a Stork-enamine alkylation of 1H-pyrrolo[2,3-d]carbazole derivatives, which were prepared through a Brønsted acid-catalyzed tandem cyclization of tryptamine-ynamide. Abstract:The first example of cyclization cross-coupling of tryptophols and tryptamines has been realized by copper catalysis with air or oxone as the terminal oxidant, resulting in the direct construction of a new class of heterocyclic 3a,3a′-bisindolines in moderate to good yields with high chemoselectivities. A series of mechanistic control experiments were also conducted, indicating that the copper catalyst selectively coordinates with the nitrogen moiety of the tryptamine to initiate the oxidation, and a nucleophilic-alkylation process is proposed for the carbon–carbon bond-forming in the reaction. Abstract:Dearomatization of indoles through a charge transfer complex constitutes a powerful tool for synthesizing three-dimensional constrained structures. However, the implementation of this strategy for the dearomatization of tryptamine-derived isocyanides to generate spirocyclic scaffolds remains underdeveloped. In this work, we have demonstrated the ability of tryptamine-derived isocyanides to form aggregates at higher concentration, enabling a single electron transfer step to generate carbon-based-radical intermediates. Optical, HRMS and computational studies have elucidated key aspects associated with the photophysical properties of tryptamine-derived isocyanides. The developed protocol is operationally simple, robust and demonstrates a novel approach to generate conformationally constrained spirocyclic scaffolds, compounds with high demand in various fields, including drug discovery.

Abstract:In this work, we successfully explored a one-pot strategy to synthesize valuable chiral 2-functionalized tetrahydroquinoline motifs through the precise modification of amides derived from commercially available natural amino acids and their derivatives via Pd/Cu−catalyzed cascade γ−C(sp3)−H arylation/C−N coupling with good maintenance of chirality. This methodology offers an alternative route for facile access to valuable chiral tetrahydroquinolines motifs with good regioselectivity and broad functional group compatibility for drug discovery. The synthetic applications of this methodology to the late−stage functionalization of natural products, drugs, and biomolecules have also been well−demonstrated.

Abstract:A highly enantioselective tandem Michael addition of tryptamine-derived oxindoles to alkynones was developed by taking advantage of a chiral N,N-dioxide Sc(OTf)(3) catalyst. The reaction enables the facile preparation of enantioenriched spiro[pyrrolidine-3,3-oxindole] compounds, which provides a novel strategy for the synthesis of monoterpenoid indole alkaloids. As a demonstration, the asymmetric synthesis of strychnos alkaloids [(-)-tubifoline, (-)-tubifolidine, (-)-dehydrotubifoline] was achieved in 10-11 steps.

Abstract:The development of transition-metal-catalyzed methods for the synthesis of P-chiral phosphine derivatives poses a considerable challenge. Herein, we present a direct Pd/Xiao-Phos-catalyzed cross-coupling reaction of easily accessible secondary phosphine oxides and aryl bromides, which provides rapid access to P-chiral phosphine oxides. The reaction proceeds efficiently with a wide array of reaction partners to deliver various tertiary phosphine oxides in up to 96% yield and 97% ee. Moreover, the synthesis of DiPAMP ligand and its analogues was also realized, which demonstrates a suitable pathway to switching the branched chain of DiPAMP.

Abstract:The highly efficient and stereo-specific synthesis of the enantioenriched versatile building blocks—namely, β-vinyltryptamines—by rhodium-catalyzed allylic substitutions of vinylaziridines and indoles is presented. Besides indoles, pyrroles can also serve as competent carbon nucleophiles in the current reaction, which is different from the previous works. To demonstrate the synthetic utility of our method, up to 11 natural product and pharmaceutically relevant chiral indole scaffolds were synthesized in highly efficient reaction sequences. Notably, asymmetric formal synthesis of a potent constrained analogue of MS-245 and a nNOS and 5-HT1B/1D receptor inhibitor are also reported.

Abstract:Acyloxymethyl (ACOM) ethers of hydroxytryptamines such as psilocin are potential prodrugs for the psychedelic treatment of mental disorders. Previous synthetic approaches suffer from insufficient selectivity and very low yields. We report a novel synthetic route towards ACOM derivatives of tryptamines, including the chemoselective installation of a carbamate protecting group at the indole nitrogen by means of a Heller-Sarpong reagent and final deprotection under extremely mild conditions. This enables delicate transformations such as the O acyloxymethylation of hydroxytryptamines or the N2-acyloxymethylation of sumatriptan. Several O-ACOM ethers of hydroxytryptamines have thus been obtained and their membrane permeability and stability in various media including human saliva and plasma were studied. The pharmacokinetic profile of the ACOM ethers is governed by the steric bulk of the acyl moiety. Short half-lives in human saliva will likely preclude the sublingual or buccal application of ACOM ethers of hydroxytryptamines, while other routes of administration may be pursued.
