Abstract:Water-soluble, regioregular polythiophene (PT) derivatives are conformational sensitive to external stimuli, and their spectral responses are determined by conformation changes, π-π interactions and scattering of visible lights originated from PT aggregates. In this paper, we review the synthesis of water-soluble PTs and their photophysical properties, and then highlight our efforts toward the development of PT-based optical sensors for small molecules. Mechanism studies indicate that the interplay between side-chain steric/electrostatic repulsive and interchain attractive interactions is responsible for the assembly and disassembly of water-soluble PTs in aqueous media. Consequently, the planarization/deplanarization of PT backbones provides the possibility for colorimetric and fluorescent dual detection of small bioanalytes and chemical species.

Abstract:We investigated a water-soluble conjugated polymer (WSCP) with pendant disulfide linkages to poly(ethylene glycol) (PEG) chains, which is a highly efficient ratiometric probe with solubility-induced fluorescence conversion for thiol detection. This WSCP was doped with a low-bandgap fluorophore, 1,4-dithienyl benzothiadiazole (DBT), and was modified with PEGs by disulfide linkages to increase its water solubility. The free probe exhibited good solubility in aqueous solution (28 mg/mL) and showed purple fluorescence because of the low doping ratio of DBT. The separation of water-soluble PEG chains from the conjugated backbone induced by the cleavage of the disulfide linkages would lead to a significant decrease of the water solubility of the probe. The combined utilization of scanning electron microscopy, dynamic light scattering, and fluorescence spectrophotometer further confirmed that decreased solubility produced an aggregation of the hydrophobic conjugated backbone and subsequently increased fluorescence resonance energy transfer efficiency from the conjugated backbone to DBT which manifested as fluorescence conversion from purple to red. The fluorescence ratiometry (I-628/I-420) of the probe varied from the lowest value of 0.095 to 1.15 (12-fold maximum enhancement). The detection limit was 2.56 mu g/mL (0.021 mM). The WSCP probe was confirmed to be a good sensing material with high selectivity for thiols by examining various biological molecules. We also successfully achieved the imaging of intracellular thiols in HeLa cell. Considering that the disulfide could be replaced by other cleavable linkages, such a fluorescence ratiometry induced by decreased solubility could be utilized for detecting other chain-cleavable biomolecules, which would contribute to the development of new probes based on conjugated polymers.

Abstract:We investigated a novel water-soluble conjugated polymer (WSCP) for thiol detection based on "turn-off" effect. This WSCP was modified with poly(ethylene glycol) (PEG) by disulfide linkages to achieve good solubility in aqueous solution (34 mg/mL) and high quantum yield (0.47). The separation of water-soluble PEG chains from the conjugated backbone induced by the cleavage of the disulfide linkages would lead to a significant decrease of the water solubility and a dramatical fluorescence quenching of the probe. The combined intuitive images and fluorescence spectrophotometer further confirmed that decreased solubility produced an aggregation of the hydrophobic conjugated backbone. The fluorescence intensity of the probe showed a good linear relationship with glutathione (GSH) (1-200 nmol.L-1), and the detection limit was 16 nmol.L-1. This WSCP probe was confirmed to be a good sensing material with high selectivity to thiols by testing various biological molecules. And this WSCP probe exhibited good detection effect to intracellular thiols by testing Hela cells. Considering the good sensitivity and selectivity, the probe could be further used in vivo. In conclusion, this conjugated polymer probe made up for the drawbacks of the micromolecue probes and contributed to the development of new probes based on conjugated polymers.
Investigation of Conformation Transition of Poly(n-Isopropylacrylamide) by Surface Stress Detection Using Micro-Cantilever

Abstract:A new method based on micro-cantilever sensors was presented and used to investigate conformation transition of macromolecules. Poly(N-isopropylacrylamide)(PNIPAM)were grafted onto one surface of a micro-cantilever by self assemble monolayer method. Then the micro-cantilever was immersed into distilled water in which the temperature can be adjusted in the range of 20—40℃. The deflection of the micro-cantilever induced by conformation transition of PNIPAM chains was measured using optical lever technique. The results show that the micro-cantilever deflects upon heating and opposite deflection occurs upon cooling, which indicates that the surface stress of the micro-cantilever changes when the conformation of PNIPAM changes upon heating and cooling. The surface stress changes continuously over the range of 20—40℃. However, a sharp change appears around the low critical solution temperature (～32℃), at which a coil-globule transition occurs in free aqueous solution of PNIPAM. The whole process is irreversible and shows a clear hysteresis, which can be attributed to hydrogen bonding and the possible chain entanglement formed during the collapse process.

Abstract:Integrating electron-accepting dicyanovinyl (DCV) moieties with aggregation-induced emission (AIE)-active 1,1-dimethyl-2,3,4,5-tetraphenylsilole (DMTPS) skeleton on the meta-positions of the 2,5-phenyl groups via simple Knoevenagel condensation derived the novel polar silole derivative, i.e. DMTPS-m-DCV. Systematic and comparative studies on the structures, properties and application in the thiol detection of DMTPS-m-DCV and its para-isomer DMTPS-p-DCV shed light on their structure-property relationships. The multiple rotors and non-planar 3D configurations bestow aggregation-enhanced emission (AEE) or AIE feature on them and the A-D-A type electronic structures impart intramolecular charge transfer (ICT) effect to them. The reactive DCV units endow these two isomers with the capability of specifically recognizing thiol species. The subtle variation in the substitution position of DCV units leads to a number of differences in the crystal and electronic structures, thermal stability, fluorescence properties, solvatochromism, ICT effect, thiol-reactivity, and thus detection performances. Benefiting from their AIE/AEE plus ICT attributes, both DMTPS-p-DCV and DMTPS-m-DCV perform well as ratiometric probes for thiols with minimal background noise, remarkable blue shift in fluorescence maxima, large enhancement in fluorescence intensity as well as high contrast in the biocompatible aqueous solutions or on the solid matrices. Intriguingly, DMTPS-p-DCV could differentiate cysteine (Cys) and homocysteine (Hcy) from glutathione (GSH) relying on the distinctive differences in kinetics. The fluorescence turn-on ratiometric probe DMTPS-m-DCV holds a superb sensitivity to Cys with a detection limit lower than 0.5 μM. Fast and sensitive responses of TLC plates with DMTPS-m-DCV spots to Cys in water demonstrated the practicability of these simple and handy test strips.

Abstract:The conformation transition of macromolecules is a very important problem,because it is the groundwork of understanding life phenomena at all levels.It is also a gundamental problem in life sciences.In this study, thermally sensitive poly(N-isopropylacrylamide)(PNIPAM) brushes were grafted on top surface of a micro-cantilever.Using the optical lever amplification technique to monitor the bending of the micro-cantilever,we investigated the thermally induced conformation transition of PNIPAM chains.The results indicate that the conformation transition occurs over a broad range of temperatures(20℃~40℃) and the collapse-swelling process has a clearly hysteresis effect.On the other hand,the surface stress changes at the grafted surface of the micro-cantilever reflect the micro-mechanism in the conformation transition process.It should be noted that the present method is based on measuring the mechanical signal during the conformation transition process.It has a higher sensitivity and wider application fields.

Abstract:A conjugated alternative copolymer (P0), containing tetraphenylethylene (TPE) and phenylene-ethynylene (PE) units, was prepared by the Sonogashira coupling polymerization with high yield (96.8%). With the aid of activated ester on the PE unit, differently functionalized copolymers (P1 and P2) were derived from P0, and their structures were well characterized using multiple spectroscopic methods. Due to the incorporation of TPE units into the polymer structure, all of these copolymers (P0, P1 and P2) showed aggregation enhanced emission (AEE) properties as revealed by the fluorescence behaviors observed in THF/water and THF/hexane mixtures. As AEE-active polymers, they were investigated for use as fluorescent probes for explosive detection. Using picric acid (PA) as a model explosive, the Stern-Volmer curves showed a super-amplification effect for all these copolymers, and the largest fluorescence quenching constant (k(SV)) was calculated to be 56 100 M-1 for P1 in THF/water mixture with 90% water fraction. This value is comparable to that reported for TPE-containing hyperbranched polymers. By the reaction of the chiral primary amine with P0, the chiral dichroism (CD) signal was recorded for the derived copolymer P1 and the evident Cotton effect shifted to around 380 nm, indicating that the chirality was transferred from the side chain to the main chain. By the substitution of the hydrophobic activated ester with the highly hydrophilic poly(ethylene glycol) (PEG(1000)), the obtained copolymer P2 could be molecularly dispersed in water and gave a clear aqueous solution. The film casting from the P2 THF solution displayed a moderate hydrophilic surface static contact angle of 66.8 degrees, which varied to 33.6 degrees after immersion in THF/water mixture (1 : 1 by volume) for hours, and the microstructural morphology underwent a change from coarse spheres to microfibers, as suggested by scanning electron microscope (SEM) images.

Abstract:Using a water-soluble conjugated polymer (CP) as a sensing probe, we developed a rapid colorimetric detection strategy for pH-driven conformational conversion of DNA i-motif structure. Two sensing configurations were designed: one used CP only to detect the conversion between i-motif and random-coiled state of a C-rich single-strand DNA, the other used CP and a complementary single-strand DNA to investigate the conversion of duplex to i-motif equilibrium. All the conversions would lead to color change observed directly with naked eyes within a few minutes. The limitation of detection (LOD) is as low as 40nM. More importantly, reversible conformational conversions by adjusting the pH of the system could also be detected.

Abstract:Two-dimensional spectral analysis based on two-dimensional correlation spectroscopy has been proved to be a very powerful tool to study the structural changes of various physical-chemical systems at the molecular level. This review summarize the recent research progress of two-dimensional correlation spectroscopy and its derivative technique, perturbation correlation moving window, in the investigation of thermoresponsive water-soluble polymers, especially LCST (lower critical solution temperature)-type polymers. Around LCST, LCST-type polymers in aqueous solutions undergo coil-to-globule conformational changes while in gel, volume phase transitions LCST-type polymers exhibit collapsing and swelling, which can be well traced by in-situ IR spectroscopy. Based on dynamic IR spectra, the combined one- and two-dimensional correlation spectroscopy analysis can be used to easily determine the transition temperature, transition temperature range, temperature-responsiveness, and the sequence order of different species, which are very helpful to illustrate the whole transition process. In this review, we will mainly emphasize on the typical applications of two-dimensional correlation spectroscopy analysis in the systems of LCST-type homopolymers, copolymers and mixtures, gels, and polymer brushes.

Abstract:There exists a critical need in biomedical molecular imaging and diagnostics for molecular sensors that report on slight changes to their local microenvironment with high spatial fidelity. Herein, a modular fluorescent probe, termed StyPy, is rationally designed which features i) an enormous and tunable Stokes shift based on twisted intramolecular charge transfer (TICT) processes with no overlap, a broad emission in the far-red/near-infrared (NIR) region of light and extraordinary quantum yields of fluorescence, ii) a modular applicability via facile para-fluoro-thiol reaction (PFTR), and iii) a polarity- and viscosity-dependent emission. This renders StyPy as a particularly promising molecular sensor. Based on the thorough characterization on the molecular level, StyPy reports on the viscosity change in all-DNA microspheres and indicates the hydrophilic and hydrophobic compartments of hybrid DNA-based mesostructures consisting of latex beads embedded in DNA microspheres. Moreover, the enormous Stokes shift of StyPy enables one to detect multiple fluorophores, while using only a single laser line for excitation in DNA protocells. The authors anticipate that the presented results for multiplexing information are of direct importance for advanced imaging in complex soft matter and biological systems.

Abstract:Conjugated polymers (CPs) with large, delocalised molecular structures exhibit unique optical and electrochemical characteristics that can be used as excellent sensing elements. Recently, research on chemical and biological sensors that use water-soluble CPs as transducers has generated intense interest. Two main sensing mechanisms are used for the detection of DNA-related events, such as hybridisation, mismatch, single nucleotide polymorphism (SNP), SNP genotyping, conformational changes, and cleavage of the nucleic acids. One mechanism takes advantage of the fluorescence resonance energy transfer (FRET) between CPs and a chromophore label on the nucleic acid probes in which a series of cationic polyfluorene, polythiophene and polyarylene derivatives are frequently used. The other mechanism relies on the conformational effects of CPs, which is induced by combination of the specific targets in which cationic polythiophene derivatives are often used. The electron transfer property of CPs are always used to design high sensitive electrochemical DNA biosensors. Here we review recent progress in the development of optical and electrochemical DNA biosensors based on water-soluble CPs.

Abstract:Water-soluble conjugated polymers (WSCPs) have been applied to a wide range of fields such as chemical sensing, bio-sensing and bio-imaging. The advantages of WSCPs for these applications include their high brightness, good photo-stability, structure variety and sufficient solubility in aqueous solution. WSCPs are usually decorated with water-soluble ionic groups or polymers on their sides or end groups. Sugars are a kind of biomolecules existing naturally and most of them have the features of water-solubility, therefore in recent years researchers have introduced sugar compounds into conjugated polymers leading to water-soluble conjugated glycopolymers with the biological properties of carbonhydrates. In this review, we summarize the recent development in the synthesis of water-soluble conjugated glycopolymers and their applications in terms of lectin-sensing, bacteria-sensing and live-cell imaging. Finally, we point out the peculiarity, developing trend and the challenges of water-soluble conjugated glycopolymers.

Abstract:Thiophenol (PhSH) is an important sulfhydryl compound in organic synthesis, but it is also a volatile environmental pollutant with high toxicity to organisms. Herein, we reported a novel near-infrared (NIR) probe (1) for turn-on fluorescence detection of PhSH. The probe was prepared by coupling 2,4-dinitrophenyl (DNP) to a dicyanoisophorone-based fluorophore (2). PhSH can specifically perform a nucleophilic aromatic substitution on probe 1 and result in the release of fluorophore 2, thus achieving a turn-on fluorescence response (λem = 693 nm). A dramatic color change from red (λabs = 525 nm) to blue (λabs = 668 nm) was also observed. This fluorescent assay displayed a large Stokes shift (∼133 nm) and a high sensitivity for PhSH, as well as a low detection limit (34 nM). Moreover, probe 1 was successfully applied to monitor PhSH in real water samples and image PhSH in living cells.

Abstract:A new kind of hydrophilic copolymer poly(HEMA-co-VNP) was designed and synthesized by the radical copolymerization of 2-hydroxyethylmethacrylate (HEMA) and 2-{[2-(4-vinylbenzyloxy) naphthalen-1-yl] methyleneamino}-(S)-2-phenylethanol (VNP). The desired copolymer showed a highly selective red-shifted emission and a unique chiroptical response upon HSO4- binding in organic solution. UV-vis and H-1 NMR spectroscopic studies revealed that the hydrogen bonding between the imine moiety in the VNP repeating units and HSO4- is crucial for the high selectivity of the receptor to this anion. As expected, the incorporation of HEMA into the polymer matrix endowed the copolymer excellent hydrophilicity, flexibility and good film-forming properties. Thus, high-quality film sensors could be easily fabricated on quartz plates through spin-casting techniques. The resultant polymeric films can recognize HSO4- ions among a series of common anions in aqueous solution with high selectivity and sensitivity. The promising new film probe for HSO4- has distinct characteristics such as a rapid response, enough stability in an aqueous media and practicality.

Abstract:Functionalization of π-conjugated polymers is dispensable for solubilization of the rigid and hydrophobic backbones in water. However, polymer aggregation is always present and leads to issues with complication and reproducibility in spectral properties. Herein, we reported a simple and robust method to make a series of conjugated polymer nanostructures by a crosslinking strategy. In favor of multivalency effect, polythionenes with various chain lengths were functionalized with branched polyethylenimine (PEI) via ester aminolysis reaction under mild conditions. Photophysical studies revealed the conjugated backbones could be well stabilized and dispersed in water. By taking advantage of intermolecular recognition interaction between copper ions and cationic PEI, we applied crosslinked polythiophenes as a nano probe at very low concentration (0.01 mg/mL) to fluorescently detect copper ions with high sensitivity up to 10 nM and selectivity over other metal ions in aqueous solutions, without occurrence of detectable aggregates. The overall performance of our nano probes outperforms reported water-soluble polymers-based probes, particularly in probe availability and manipulation as well as selective copper detection capability.

It is found that when the pH varies, the microcantilever deflects because of the changing surface stress. By analysing the electrostatic repulsive effect, the surface stress change is related to the conformation transition of PAA from a collapse state to a swelling state. This method offers the interaction information among the polymer chains during the conformational transition and affords an alternative way to study conformational change of polymers.
