Abstract:OBJECTIVE:To evaluate the correlation of CD44v6 and sCD44v6 expression with the biological activity of gastric carcinoma. METHODS:Mucosa samples from 103 gastric carcinoma patients and 10 healthy persons (control) were examined using immunohistochemical SP method; sCD44v6 level in peripheral blood samples was detected with ELISA in 86 gastric cancer patients, 30 gastric ulcer patients and 30 healthy controls. The follow-up period was 3-91 months for 88 patients in CD44v6 group and 1-91 months for 55 patients in sCD44v6 group. RESULTS:(1) Positive expression of CD44v6 was found in 60.2% of gastric carcinoma, where as 0% in the normal gastric mucosa. CD44v6 protein expression was positively correlated with TNM stage, perigastric lymph node metastasis, presence of cancerous embolic angiopathy and Borrmann classification (P <0.05), but was not correlated with invasion depth, differentiation, metastasis and survival (P > 0.05); 77.8% of the patient with liver metastasis had a strong CD44v6 protein expression. (2) sCD44v6 level in the peripheral blood of gastric carcinoma patient was significantly higher than that in gastric ulcer patient or healthy control; Compared with palliative gastrectomy, sCD44v6 level significantly went down after radical operation. No relationship between sCD44v6 level and the pathological features was found, which included invasion depth, perigastric lymph node metastasis, presence of cancerous embolic angiopathy, differentiation and Borrmann classification; The survival in the high sCD44v6 level group was longer than that in low sCD44v6 level group ( P = 0.0281), but no significant difference was observed by Cox Regression analysis (P = 0.415). (3) No apparent correlation was observed between CD44v6 expression in gastric cancer and sCD44v6 concentration in peripheral blood (P>0.05). CONCLUSION:Detection of CD44v6 expression in the gastric cancer may be helpful in evaluating the biological features and the survival in gastric carcinoma. The level of sCD44v6 in the blood may be also helpful in differential diagnosis, evaluation of surgical treatment and biological activity for gastric cancer. No apparent correlation is observed between CD44v6 expression in the cancer and that of sCD44v6 in the blood.
[Nuclear Expression of S100A4 is Associated with Lymph Node Metastasis in Gastric Carcinoma].

Abstract:OBJECTIVE:To investigate the intracellular localization of S100A4 in gastric carcinoma cells and the relationship between S100A4 expression status and lymph node metastasis of gastric carcinoma.METHODS:Western blotting analysis was performed to locate the expression of S100A4 protein in sub-fraction components of frozen tissues. S100A4 protein expression was also determined by immunohistochemical method in 131 samples of gastric cancer and 20 samples of matched metastatic lymph nodes.RESULTS:Thirty-two of 131 (24.4%) gastric carcinoma showed positive S100A4 nuclear expression and 50/131 (38.2%) carcinoma showed positive cytoplasmic expression. In 32 samples with positive S100A4 nuclear expression, 30 (93.8%) carcinomas had positive lymph node metastases. S100A4 nuclear expression level was higher in gastric carcinoma with lymph node metastasis (29.1%) than that without lymph node metastasis (7.1%) (P=0.016).CONCLUSION:Nuclear expression of S100A4 is associated with lymph node metastasis of gastric carcinoma.
Changes of Histology and Expression of Mmp-2 and Nm23-H1 in Primary and Metastatic Gastric Cancer

Abstract:AIM: To investigate the changes of histology and expression of MMP-2 and nm23-H1 in primary and metastatic gastric cancer.METHODS: One hundred and seventy-seven gastric cancer patients with lymph node and/or distal metastasis between 1997 and 2001 were reviewed. Differences in histology of the primary and metastatic gastric cancer were assessed. MMP-2 and nm23-H1 immunoreactivity was compared in 44 patients with tumor infiltration to the serosa layer.RESULTS: Poorly and moderately differentiated metastatic gastric cancer was found in 88.7% (157/177) and primary gastric cancer in 75.7% (134/177) of the patients. The histological type of metastatic gastric cancer that was not completely in accordance with the preponderant histology of primary gastric cancer was observed in 25 patients (14.1%). MMP-2 immunoreactivity in metastatic gastric cancer was significantly stronger than that in primary gastric cancer, while nm23-H1 immunoreactivity showed no difference in primary and metastatic gastric cancer.CONCLUSION: Metastatic gastric cancer presents more aggressive histological morphology and higher MMP-2 immunoreactivity than primary gastric cancer. This heterogeneity may elicit a possible mechanism of gastric cancer metastasis. (C) 2008 WJG. All rights reserved.
[Regulation Mechanism Study of S100A6 on Invasion and Metastasis in Gastric Cancer].

Abstract:OBJECTIVE:To detect the expression of S100A6 in gastric cancer, and to investigate the regulation mechanism of S100A6 in invasion and metastasis of gastric cancer.METHODS:Expression of S100A6 protein in gastric cancer specimens, tissue adjacent to cancer, liver and lymph node metastasis tissue specimens was detected by immunohistochemical staining in 166 patients with gastric cancer from January 1995 to December 2001. Their association with clinicopathological factors was analyzed. Chromatin Immunoprecipitation-chip was used to detect the downstream factors potentially regulated by S100A6 in gastric cancer cell lines KATO3. S100A6 gene was transfected into gastric cancer cell line AGS, and cell invasion experiment and real time Q-polymerase chain reaction(RT Q-PCR) were used to detect the cell invasive ability and the mRNA expression of invasion-related factors (CDK5 and FLJ12438) in transfection group, negative control group and blank control group, respectively.RESULTS:Low expression of S100A6 protein was found in cytoplasm of peritumoral tissues. In gastric cancer, liver and lymph node metastasis tissues, S100A6 protein expression was up-regulated in cytoplasm and (or) nuclei, especially in the tumor cells of invasive edge. The expression rates of gastric cancer, liver and lymph node metastasis tissues were 67.5%(112/166), 92.9%(26/28) and 100% (30/30) respectively. The high expression of S100A6 was associated with tumor local invasion, lymph node metastasis, cancer embolus, distant metastasis and TNM stages(all P<0.05). The transmembrane cell number was 31.3±5.5 in the S100A6 transfection group, significantly higher than that in negative control group (7.7±1.5) and blank control group (9.3±2.1)(both P<0.05), indicating an increase of cell invasion after S100A6 transfection. In transfection group, CDK5 mRNA expression was significantly higher than that in negative control group and blank control group(P<0.05). While FLJ1243 mRNA expression was similar among the three groups(P<0.05).CONCLUSION:S100A6 may affect the malignant biological behavior of gastric cancer cells by regulating the expressions of down-stream invasion-associated factors, such as CDK5.
Relationship between the CXC Chemokine Receptor-4 /CXCL12 Signal Transduction Pathway and Lymph Node Metastasis of Gastric Carcinoma

Abstract:Objective:To investigate the role of CXC chemokine receptor-4 (CXCR4) and CXC chemokine ligand-12 (CXCL12) in tumorigenesis of gastric carcinoma. Methods:The expression levels of various clinicopathological markers, such as CXCR4, CXCL12, cyclin D1, and Bcl-xL, were obtained from histological specimens of 40 patients with gastric carcinoma by Western blot analysis. Results:The expression of CXCR4 and CXCL12 was significantly higher in gastric cancer than in matched normal mucosa (P<0.05). The expression of CXCR4 and CXCL12 was higher in lymph node metastases that were present in 22 of the 32 cases compared to the primary tumor. The expression level of CXCR4 and CXCL12 was related to lymph node metastasis (P<0.05). Conclusion:Overexpression of CXCR4 and CXCL12 is found in primary gastric cancer and lymph node metastases. The CXCR4 signal transduction pathway may play an important role in lymph node metastasis of gastric cancer.
Expression and corelation of activated c-Src protein tyrosine kinase, HER2 and Ki-67 in gastric cancer

Abstract:Objective To study the expression of p-Src(Y419),the activated form of c-Src kinase,HER2,Ki-67 in gastric cancer,and explore their correlation.Methods Immunohistochemical investigations were carried out to study the expression of p-Src(Y419),HER2 and Ki-67 and their correlation in 123 gastric cancer specimens and 56 corresponding adjacent specimens.Results The median of staining score of p-Src(Y419) in gastric cancer tissues and adjacent nontumorous tissues was 80 and 30,respectively,with a significant difference.Activated c-Src correlated with tumor size,differentiation,depth of wall invasion,and pTNM classification.The high expression rate of HER2 was 39% in gastric cancer,while there was no expression of HER2 in adjacent nontumorous tissues,and the difference was significant(P<0.05).High expression of HER2 correlated with the well differentiated or intestinal type,and pure adenocarcinoma.The range of Ki-67 index in the two kinds of tissues was 0-98% and 0-20%,and the median Ki-67 index was 70% and 3.5%,respectively,which was significant.The high expression of Ki-67 significantly correlated with pure adenocarcinoma,intestinal type,and cancer embolus,and with well differentiated cancer tissues in a significant tendency.High expression of HER2 correlated with high levels of activated c-Src and Ki-67 expression in gastric cancer tissues.There was no such relationship in adjacent nontumorous tissues.High levels of activated c-Src correlated with high levels of Ki-67 expression in the two kinds of tissues.Conclusion Activation of c-Src and expression of HER2 and Ki-67 are of significance in gastric cancer.They should be considered as markers of the biological behavior of gastric caner.Furthermore,activation of c-Src may be associated with HER2 and Ki-67 expression.

Abstract:Objective To study the relationship between the expression of somatostatinand p21~(WAF1/CIP1)protein and biological behavior of gastrointestinal carcinoid.Methods The immunohistochemistry S-P method was adopted to examine the expression of somatostatinand p21~(WAF1/CIP1) protein in thirty-six cases of gastrointestinal carcinoid.Results In thirty-six cases of gastrointestinal carcinoid,somatostatin and p21~(WAF1/CIP1)protein were mostly expressed in typical carcinoid(P0.05).The positive rates of somatostatindecreased significantly(P0.01) and the expression of p21~(WAF1/CIP1)was correlated with the invasion and metastasis of carcinoid(P0.05).Conclusion The low expression of somatostatinand p21~(WAF1/CIP1) protein might play a great role in the differentiation and development of gastrointestinal carcinoid,and can be used for prognostic judgement.
A S100A14-CCL2/CXCL5 signaling axis drives breast cancer metastasis

Abstract:Rationale: Chemokines contribute to cancer metastasis and have long been regarded as attractive therapeutic targets for cancer. However, controversy exists about whether neutralizing chemokines by antibodies promotes or inhibits tumor metastasis, suggesting that the approach to directly target chemokines needs to be scrutinized.Methods: Transwell assay, mouse metastasis experiments and survival analysis were performed to determine the functional role of S100A14 in breast cancer. RNA-Seq, secreted proteomics, ChIP, Western blot, ELISA, transwell assay and neutralizing antibody experiments were employed to investigate the underlying mechanism of S100A14 in breast cancer metastasis. Immunohistochemistry and ELISA were performed to examine the expression and serum levels of S100A14, CCL2 and CXCL5, respectively.Results: Overexpression of S100A14 significantly enhanced migration, invasion and metastasis of breast cancer cells. In contrast, knockout of S100A14 exhibited the opposite effects. Mechanistic studies demonstrated that S100A14 promotes breast cancer metastasis by upregulating the expression and secretion of CCL2 and CXCL5 via NF-κB mediated transcription. The clinical sample analyses showed that S100A14 expression is strongly associated with CCL2/CXCL5 expression and high expression of these three proteins is correlated with worse clinical outcomes. Notably, the serum levels of S100A14, CCL2/CXCL5 have significant diagnostic value for discerning breast cancer patients from healthy individuals.Conclusions: S100A14 is significantly upregulated in breast cancer, it can promote breast cancer metastasis by increasing the expression and secretion of CCL2/CXCL5 via RAGE-NF-κB pathway. And S100A14 has the potential to serve as a serological marker for diagnosis of breast cancer. Collectively, we identify S100A14 as an upstream regulator of CCL2/CXCL5 signaling and a metastatic driver of breast cancer.

[Clinicopathological and Biological Features of Micrometastasis in Early Gastric Cancers].

Abstract:OBJECTIVE:To investigate the clinicopathological and biological features of micrometastasis in early gastric cancers.METHODS:Eleven cases of early gastric cancer with micrometastasis (micrometastatic, MM group) and 46 cases of early gastric cancer with lymph node metastasis (control group) were included in the study. Immunochemical staining of ssDNA, bcl-2, p53, E-cadherin, Ki-67, CD34 was performed. The superficial lesions, invasive fronts and lymph nodes were examined in both groups.RESULTS:Positive rate of ssDNA at the superficial lesions in MM group was higher than that in control group. In MM group the positive rate of ssDNA in micrometastasis was higher than that at invasive fronts and in lymph nodes. Positive rate of bcl-2 at the superficial lesions in micrometastasis was higher than that at invasive fronts and lymph nodes. Positive rate of c-myc at the superficial lesions in MM group was higher than that in control group. Positive rate of E-cadherin and the percentage of microvascular areas at the lymph nodes in MM group was lower than those in control group. Proliferative ability of cancer cells at superficial lesions and lymph nodes in MM group was lower than those in control group. Lymph nodes <3 mm in micrometastasis accounted for 27.3%.CONCLUSION:The pathological and biological features of micrometastasis in early gastric cancer show low positive rate of ssDNA, E-cadherin, Ki-67 and low percentage of microvascular areas at the lymph nodes.
Expression and Significance of Angiopoietin-2 in Gastric Cancer.

Abstract:AIM: To investigate the expression and pathological factors of Angiopoietin-2 (Ang-2) in primary gastric cancers and adjacent normal tissues.METHODS: The expression of Angiopoietin-2 and VEGF were studied in 72 primary gastric cancers and adjacent normal tissues from the same patients by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and immumohistochemistry.RESULTS: Ang-2 was mainly expression in tumor cells. There were significantly difference between expression of Ang-2 in primary gastric cancer and in adjacent normal tissue samples (P=0.003). It was statistically correlation between Ang-2 and VEGF expression in tumors (P=0.0055). With regard to Ang-2 expression in tumors, there were significant difference between early stage and advanced stage (P=0.017), and significant difference between positive vascular involvement and negative vascular involvement (P=0.032). However, there was no significant difference between moderate-poor differential type and high differential type (P=0.908), between positive lymph node metastasis and negative lymph node metastasis (P=0.752), between positive serosal invasion and negative serosal invasion (P=0.764). The cases with expression of Ang-2 were increasing with advanced stage and vascular involvement.CONCLUSION: The results manifested that Angiopoietin-2, coordinated with VEGF, play role in regulating tumor angiogenesis of gastric cancer.
Expression and Significance of VEGF and MMP-9 in Gastric Carcinoma

Abstract:Objective To investigate the expression of vascular endothelial growth factor C(VEGF-C),vascular endothelial growth factor D(VEGF-D) and matrix metalloproteinase-9(MMP-9) in gastric cancer and their correlation with lymph node metastasis.Methods 108 cases of gastric carcinoma tissues were detected for the expression of MMP-9,VEGF-C and D by immunohistochemical staining,and normal stomach tissues were employed as a control.Results The positive rates of VEGF-C,D and MMP-9 in gastric cancer were significantly higher when compared with the control(65.7%,77.8%,75.0% vs 15%,20%,10% respectively;both P0.05).Furthermore,in gastric cancer with lymph node metastasis,the expression of VEGF-D,MMP-9 were more evident than that of the cases without lymph node metastasis(87.0%,83.3%vs31.5%,33.3%),whereas VEGF-C expression did not show eminent difference(66.7%,64.8%)(P0.05).Conclusion The expression of VEGF-D,MMP-9 is closely related to lymph node metastasis and staging of gastric cancer,and they are promising targets for clinical therapies.

Abstract:To investigate whether S100A14 and S100A4 expression correlates with metastatic potential and prognosis in colorectal cancer (CRC), we firstly used RT-PCR analysis to detect mRNA expression of S100A14 and S100A4 in 40 pairs of fresh tumor samples matched with adjacent normal tissues. We then evaluated the clinical significance of our findings with immunohistochemistry on 115 samples of formalin-fixed and paraffin-embedded tumors on tissue microarrays. Typically, we identified decreased S100A14 mRNA levels (52.5%, 21/40), and increased S100A4 mRNA levels (70.0%, 28/40) in primary CRC samples. In addition, down-regulated or absent S100A14 expression was detected in 56.5% of samples (65/115) and was correlated with poor differentiation (P=0.010). In contrast, overexpressed S100A4 was detected in 57.4% of samples (66/115) and was associated with lymph node metastasis (P=0.001). Simultaneous S100A14 low-expression and S100A4 high-expression was correlated with high CRC metastatic potential (P<0.001). Taken together, the signature derived from the combined expression status of S100A14 and S100A4 could be a valuable prognostic indicator in CRC.
Overexpression and Clinicopathological Significance of NET-1,Ki67 and CD34 in Gastric Carcinoma

Abstract:Objective To investigate the overexpression of NET-1、Ki67 and CD34 and clinicopathological significance in gastric carcinoma.Methods The overexpression of NET-1、Ki67 and CD34 was detected in 86 cases of gastric carcinoma paraffin-embeded sections by immunohistochemical method. Immunohistochemical trimarkers double staining method was used to detect all positive cases with the three markers and show the adjacent relationship and accompanying conditions between NET-1、Ki67 and CD34 on the one slide.All cases were followed up over 60 months.Results The overexpression rates of NET-1,Ki67 and CD34 in gastric carcinoma were 56.98%、74.42% and 62.79%,respectively. There was significant positive correlation between these three markers (P <0.05). The overexpression rate of these markers were remarkably positive correlated with tumor clinical stages(P<0.05)and negatively correlated with tumor survival rate(P<0.05)and metastasis (P<0.05). The survival rate within 3 and 5 years were dramatically shorter in the overexpression grouTP of these three markers than that in low expression grouTP and the negative grouTP(P<0.05,P<0.01). The overexpression of NET-1 and Ki67 were negatively correlated with carcinoma differentiation (P<0.01). The overexpression of NET-1 and CD34 was positively correlated with infiltration and lymph node status of tumor (P<0.05),respectively.Conclusion The overexpression of NET-1,Ki67 and CD34 proteins was related with the proliferation,angiogenesis and poor prognosis in gastric carcinoma.
S100A4 Over-Expression Underlies Lymph Node Metastasis and Poor Prognosis in Colorectal Cancer

Abstract:AIM:To develop lymph node metastasis (LNM)-associated biomarkers for colorectal cancer (CRC) using quantitative proteome analysis. METHODS:Differences in protein expression between primary CRC with LNM (LNM CRC) and without LNM (non-LNM CRC) were assessed using methyl esterification stable isotope labeling coupled with 2D liquid chromatography followed by tandem mass spectrometry (2D-LC-MS/MS).The relationship to clinicopatholog-ical parameters and prognosis of candidate biomarkers was examined using an independent sample set. RESULTS:Forty-three proteins were found to be differentially expressed by at least 2.5-fold in two types of CRC.S100A4 was significantly upregulated in LNM CRC compared with non-LNM CRC, which was confirmed by Western blotting, immunohistochemistry and real-time quantitative polymerase chain reaction.Further immunohistochemistry on another 112 CRC cases showed that overexpression of S100A4 frequently existed in LNM CRC compared with non-LNM CRC (P < 0.001).Overexpression of S100A4 was significantly associated with LNM (P < 0.001), advanced TNM stage (P < 0.001), increased 5-year recurrence rate (P < 0.001) and decreased 5-year overall survival rate (P < 0.001).Univariate and multivariate analyses indicated that S100A4 expression was an independent prognostic factor for recurrence and survival of CRC patients (P < 0.05).CONCLUSION: S100A4 might serve as a powerful biomarker for LNM and a prognostic factor in CRC.
Identification of metastasis candidate proteins among HCC cell lines by comparative proteome and biological function analysis of S100A4 in metastasis in vitro

Abstract:Widespread metastasis of hepatocellular carcinoma (HCC) was a complex cascade of events, which is still beyond full appreciation. Screening key proteins, which play a critical role in metastasis, using high-throughput proteomics approach help discover valuable biomarkers and elucidate the mechanism of metastasis. This study was to find out some metastasis candidate proteins among HCC cell lines with various metastatic potential by comparative proteomics, and then further validate the biological function of these proteins in metastasis in vitro. The protein profiles of metastatic HCC cell lines (MHCC97H and MHCC97L) displayed obvious differences compared with nonmetastatic ones (Hep3B). Twenty-six metastasis candidate proteins, which were identified by on-line LC-ESI-MS/MS, such as S100 calcium-binding protein A4 (S100A4), annexin 1, etc., might have much application in diagnostic procedures and prognosis evaluation. S100A4, as a leading different metastasis candidate protein, which overexpressed only in the metastatic cells, was selected for further investigation. A series of assays related to invasion and metastasis in vitro, including cell motility, invasion, and matrix metalloproteinases (MMPs) secretion, were performed in MHCC97H/antisense recombinant plasmid to S100A4 (pcDNA3.1(+) AS S100A4) and the mock controls. All the data in the present study suggested that S100A4 might contribute to HCC invasion and metastasis through two paths of matrix metalloproteinase (MMP9) secretion regulation and strengthened motility and invasion properties.
Expressions of SIRT 1, E-cadherin and MLH 1 in Gastric Cardia Carcinoma and Their Relationship

Abstract:Objective SIRT 1 is a NAD+(nicotinmide nicotinamide adenine dinucleotide)-dependent deacetylase,which may be involved in the formation and progression of many types of carcinoma,and plays an important role in regulating protein function and gene silencing of certain tumor-related genes.The objective of the research is to investigate the clinicopathological significance of the expressions of SIRT 1,MLH 1 and E-cadherin in gastric cardia carcinoma and their relationship.Methods We detected the expressiond of SIRT 1,MLH 1 and E-cadherin proteins in the gastric cardia carcinoma tissues of 176 patients using the tissue microarray(TMA) technique and immunohistochemistry staining.Results The expression of SIRT 1 was positively correlated with lymph node metastasis(P=0.032) and TNM stages(P = 0.034),that of E-cadherin negatively with the histological differentiation(P=0.029),lymph node metastasis(P=0.000) and TNM stages(P=0.003),and that of MLH 1 showed no correlation with the clinicopathological features of gastric cardiac carcinoma,lymph node metastasis and TNM stages(P0.05).The complete follow-up data of 90 cases exhibited that the three-year survival rate and and mean survival time were significantly lower and shorter in the SIRT 1-positive than in the SIRT 1-negative patients(P=0.024),higher and longer in the patients with a strong positive E-cadherin expression than in those with a weak one(P=0.001),and higher and longer,too,in those with a positive MLH 1expression than in those with a negative one(P = 0.045).Conclusion The expression of SIRT 1 in gastric cardia carcinoma is associated with lymph node metastasis,TNM stages and survival time,but it is not an independent prognostic factor,and that of E-cadherin is negatively correlated with the degree of malignancy.SIRT 1 may promote the formation of gastric cardia carcinoma and influence its biological behavior by silencing tumor suppressor genes,such as E-cadherin and MLH 1.
Expression of S100A4 and matrix metalloproteinase 2 and their correlations with clinicopathological features in esophageal squamous cell carcinoma

Abstract:AIM:To investigate the expression of S100A4 and matrix metalloproteinase 2(MMP-2)in esophageal squamous cell carcinoma and adjacent normal mucosa,and their correlation with clinicopathological features. METHODS:SP immunohistochemistry was used to detect the expression of S100A4 and MMP2 proteins in 100 cases of esophageal squamous cell carcinoma and adjacent normal mucosa. RESULTS:The positive rates of S100A4 and MMP2 protein expression were 52.00% and 67.00%,respectively,in esophageal squamous cell carcinoma,while only 26.00% and 31.00% in normal mucosa.The positive rates of S100A4 and MMP2 protein expression in esophageal squamous cell carcinoma were significantly higher than those in normal mucosa(P<0.01). The expression of S100A4 and MMP2 protein was significantly associated with differentia- tion,depth of tumor invasion and lymph node metastasis(P<0.05).A positive correlation was observed between S100A4 and MMP2 protein in esophageal squamous cell carcinoma. CONCLUSION:The overexpression of S100A4 protein may produce a marked effect in the gen- esis,development and progression of esophageal squamous cell carcinoma through regulation of MMP2 protein overexpression.S100A4 may be a potential marker for evaluating the prognosis of esophageal carcinoma.
Expression of S100A6 and S100A4 in matched samples of human colorectal mucosa, primary colorectal adenocarcinomas and liver metastases

Abstract:Objective: S100A6 and S100A4, two of S100 protein family, have been suggested to be associated with cancer tumorigenesis and metastasis. The aim of this study was to evaluate the expression levels of S100A6 and S100A4 in matched samples of primary human colorectal adenocarcinomas (T), adjacent normal colorectal mucosa (N) and liver metastases (M). This gave us the advantage of directly comparing levels of S100A6 and S100A4 expression within the same genetic background. Methods: In matched samples of N, T and M from 10 colorectal adenocarcinoma patients, expressions of S100A6 and S100A4 were studied by Western blot and immunohistochemical analyses using specific antibodies against each protein. Results: The expression levels of S100A6 were significantly higher in T than in N (p < 0.05), while those of S100A4 showed no difference between T and N. There were no significant differences in the expression levels of S100A6 or S100A4 between M and T. Similar results were obtained by immunohistochemical analysis. Moreover, S100A6 was stained more intensely in invading fronts than in central portions of both T and M. Conclusions: The observed differential expression of S100A6 and S100A4 suggests that S100A6, rather than S100A4, is associated with human colorectal adenocarcinoma tumorigenesis and invasion/metastasis.
