Abstract:AIM:To develop lymph node metastasis (LNM)-associated biomarkers for colorectal cancer (CRC) using quantitative proteome analysis.METHODS:Differences in protein expression between primary CRC with LNM (LNM CRC) and without LNM (non-LNM CRC) were assessed using methyl esterification stable isotope labeling coupled with 2D liquid chromatography followed by tandem mass spectrometry (2D-LC-MS/MS). The relationship to clinicopathological parameters and prognosis of candidate biomarkers was examined using an independent sample set.RESULTS:Forty-three proteins were found to be differentially expressed by at least 2.5-fold in two types of CRC. S100A4 was significantly upregulated in LNM CRC compared with non-LNM CRC, which was confirmed by Western blotting, immunohistochemistry and real-time quantitative polymerase chain reaction. Further immunohistochemistry on another 112 CRC cases showed that overexpression of S100A4 frequently existed in LNM CRC compared with non-LNM CRC (P<0.001). Overexpression of S100A4 was significantly associated with LNM (P<0.001), advanced TNM stage (P<0.001), increased 5-year recurrence rate (P<0.001) and decreased 5-year overall survival rate (P<0.001). Univariate and multivariate analyses indicated that S100A4 expression was an independent prognostic factor for recurrence and survival of CRC patients (P<0.05).CONCLUSION:S100A4 might serve as a powerful biomarker for LNM and a prognostic factor in CRC.
Expression Status of S100A14 and S100A4 Correlates with Metastatic Potential and Clinical Outcome in Colorectal Cancer after Surgery.

Abstract:To investigate whether S100A14 and S100A4 expression correlates with metastatic potential and prognosis in colorectal cancer (CRC), we firstly used RT-PCR analysis to detect mRNA expression of S100A14 and S100A4 in 40 pairs of fresh tumor samples matched with adjacent normal tissues. We then evaluated the clinical significance of our findings with immunohistochemistry on 115 samples of formalin-fixed and paraffin-embedded tumors on tissue microarrays. Typically, we identified decreased S100A14 mRNA levels (52.5%, 21/40), and increased S100A4 mRNA levels (70.0%, 28/40) in primary CRC samples. In addition, down-regulated or absent S100A14 expression was detected in 56.5% of samples (65/115) and was correlated with poor differentiation (P=0.010). In contrast, overexpressed S100A4 was detected in 57.4% of samples (66/115) and was associated with lymph node metastasis (P=0.001). Simultaneous S100A14 low-expression and S100A4 high-expression was correlated with high CRC metastatic potential (P<0.001). Taken together, the signature derived from the combined expression status of S100A14 and S100A4 could be a valuable prognostic indicator in CRC.

Abstract:Objective The development and progression of many cancers may be related to SALL4, the role and molecular mechanism of which are unclear in colon adenocarcinoma (COAD). Methods The SALL4 expression in adjacent normal mucosa tissues and carcinoma tissues of patients with COAD was detected through bioinformatic analysis based on TCGA database and immunohistochemistry. Single-cell analysis showed that the expression of SALL4 in normal tissue was noticeably low. GSEA analysis suggested that the SALL4 upregulated the GO and pathway of growth and cancer development and downregulated metabolization pathway. The relationship between lymph node metastasis, histological grading, clinical staging, and the expression of SALL4 in carcinoma tissues was analyzed. The upregulated or downregulated SALL4 expression of COAD cell lines was established. The influence of SALL4 on COAD cells invasion and proliferation was detected using plate cloning assay and Transwell. The expressions of EMT-related proteins E-cadherin, N-cadherin, vimentin, and Twist were detected by Western blot. The EMT phenotype was analyzed by immunofluorescence. Results The study confirmed that the expression of SALL4 was upregulated in COAD and positively correlated with the degree of tumor differentiation, tumor staging, and metastasis. The overexpression of SALL4 was related to a poor prognosis, promoted the invasion and proliferation of colorectal cancer cells, and accelerated the occurrence of EMT, which was characterized by upregulation of Twist, vimentin, and N-cadherin expressions and downregulation of E-cadherin. The immunofluorescence staining confirmed the EMT phenotype. On the contrary, knocking out SALL4 gene reversed EMT, weakened cell proliferation and invasion, inhibited upregulation of Twist, vimentin, and N-cadherin expressions and downregulation of E-cadherin. Conclusion To sum up, TNM grading, histological grading, and lymphatic metastasis were significantly correlated with SALL4 in tumor tissues. SALL4 played a vital role in tumor proliferation, invasion, and tumor EMT and may be a novel target for COAD.

Abstract:OBJECTIVE:To investigate the intracellular localization of S100A4 in gastric carcinoma cells and the relationship between S100A4 expression status and lymph node metastasis of gastric carcinoma.METHODS:Western blotting analysis was performed to locate the expression of S100A4 protein in sub-fraction components of frozen tissues. S100A4 protein expression was also determined by immunohistochemical method in 131 samples of gastric cancer and 20 samples of matched metastatic lymph nodes.RESULTS:Thirty-two of 131 (24.4%) gastric carcinoma showed positive S100A4 nuclear expression and 50/131 (38.2%) carcinoma showed positive cytoplasmic expression. In 32 samples with positive S100A4 nuclear expression, 30 (93.8%) carcinomas had positive lymph node metastases. S100A4 nuclear expression level was higher in gastric carcinoma with lymph node metastasis (29.1%) than that without lymph node metastasis (7.1%) (P=0.016).CONCLUSION:Nuclear expression of S100A4 is associated with lymph node metastasis of gastric carcinoma.
[Correlations of S100A4 Protein Expression to Invasion and Metastasis of Non-Small Cell Lung Cancer].

Abstract:BACKGROUND & OBJECTIVE:Metastasis-associated protein S100A4 is overexpressed in many malignant tumor cells; it may play a pivotal role in invasion and metastasis of malignant tumors. This study was to determine the expression of S100A4 in human non-small cell lung cancer (NSCLC), and to investigate its correlations to invasion and metastasis of NSCLC. METHODS:The expression of S100A4 in 41 specimens of NSCLC and 6 specimens of normal lung tissues was detected by SP immunohistochemistry. The correlations of S100A4 to clinicopathologic features of NSCLC were analyzed. RESULTS:The positive rate of S100A4 was significantly higher in NSCLC than in normal lung tissues (70.7% vs. 16.7%, P<0.05), and was significantly higher in adenocarcinoma than in squamous cell carcinoma (90.0% vs. 52.4%, P<0.01). The positive rate of S100A4 was significantly higher in stage III-IV than in stage II and stage I NSCLC (100.0% vs. 66.7% and 30.0%, P<0.01), while there was no obvious difference between the latter 2 groups (P>0.05). The positive rate of S100A4 was significantly higher in NSCLC with lymphatic metastasis than in NSCLC without lymphatic metastasis (90.0% vs. 52.4%, P<0.01), and significantly higher in NSCLC with tumor size of > or = 3 cm than in NSCLC with tumor size of < 3 cm (91.3% vs. 44.4%, P<0.001). The expression of S100A4 was closely related to lymphatic metastasis (r=0.480, P=0.001), and tumor size (r=0.288, P=0.017). No significant correlation was found between the expression of S100A4 and pathologic grade of NSCLC (P>0.05). CONCLUSION:S100A4 expression is up-regulated in NSCLC, and closely related to lymphatic metastasis, TNM stage and tumor size, which suggest an important role of S100A4 in the invasion and metastasis of NSCLC.

Abstract:S100A4 expression is associated with poor clinical outcomes of patients with pancreatic cancer. The effects of loss or gain of S100A4 were examined in pancreatic cancer cell lines. S100A4 downregulation remarkably reduces cell migration and invasion, inhibits proliferation and induces apoptosis in pancreatic tumor cells. S100A4 downregulation results in significant cell growth inhibition and apoptosis in response to TGF-β1, supporting a non-canonical role of S100A4 in pancreatic cancer. The role of S100A4 in tumor progression was studied by using an orthotopic human pancreatic cancer xenograft mouse model. Tumor mass is remarkably decreased in animals injected with S100A4-deficient pancreatic tumor cells. P27 Kip1 expression and cleaved caspase-3 are increased, while cyclin E expression is decreased, in S100A4-deficient pancreatic tumors in vivo . S100A4-deficient tumors have lower expression of vascular endothelial growth factor, suggesting reduced angiogenesis. Biochemical assays revealed that S100A4 activates Src and focal adhesion kinase (FAK) signaling events and inhibition of both kinases is required to maximally block the tumorigenic potential of pancreatic cancer cells. These findings support that S100A4 plays an important role in pancreatic cancer progression in vivo and S100A4 promotes tumorigenic phenotypes of pancreatic cancer cells through the Src-FAK mediated dual signaling pathway.
Expressions of S100A4 in lung cancer tissues and its clinical significance

Abstract:Objective To investigate the expression of S100A4 protein in lung cancer tissues and further to discuss its clinical significance in lung cancer. Methods The study selected 45 cases of lung squamous cell carcino-ma, 50 cases of adenocarcinoma and 23 cases of small cell lung carcinoma. The expression of S100A4 protein in vari-ous types of lung cancer and adjacent non-malignant tissues were detected by immunohistochemistry method respec-tively, and quantitative analysis of the difference among them were done by Image-Pro Plus 6. 0 software system. Re-sults The expression of S100A4 protein in all cases of lung squamous cell carcinomas and adenocarcinoma was sig-nificantly higher than that in the adjacent non-malignant tissues and bronchial mucosa inflammatory lesions (P<0. 01). The expression of S100A4 protein was not detected in small cell lung carcinoma, but it was detected in stromal vascular. The S100A4 protein expression of lung squamous cell carcinomas and adenocarcinoma was significantly higher in the group with lymphatic metastasis than in the group without lymphatic metastasis (P<0. 05). The S100A4 pro-tein expression of the lung squamous cell carcinomas and adenocarcinoma was significantly higher in the group with distant metastasis than in the group without distant metastasis (P<0. 05). The S100A4 protein expression of lung squamous cell carcinomas and adenocarcinoma was not related with gender, age, tumor size or pathological grade ( P>0. 05). Conclusion The S100A4 protein play an important role in the proliferation and metastasis of non small cell lung cancer, and the detection of S100A4 protein is helpful to judge the prognosis of non small cell lung cancer. At the same time, it may play a certain role in invasion and metastasis of small cell lung cancer.
The Expression of S100A4 Protein in Tumor-stroma of Human Non-small Cell Lung Cancer and Its Clinical Significance

Abstract:Objective To investigate the expression and clinical significance of S100A4 protein in tumor-stroma of non-small cell lung cancer(NSCLC) to study its correlation with invasion, metastasis and prognosis. Methods Immunohistochemical staining(S-P method)for S100A4 protein expression was performed in tissue sections from 130 patients with NSCLC operated and to analyze association of S100A4 protein with clinicopathological parameters in lung cancer and prognosis. Results The total positive expression rates of S100A4 protein in stroma of NSCLC was 72.3%. The positive expression rates of S100A4 protein in stroma of squamous cell carcinoma, adenocarcinoma, adenosquamous carcinoma and large cell lung cancer were 84.3%,59.6%,70.0% and 75% respectively.The expression of S100A4 protein was significantly associated with lymph node metastasis (χ2=18.91, P=0.000), distant metastasis(χ2=5.51, P=0.019) and TNM stage (χ2=21.54, P=0.000). The 3 years survival rates of patients whose tumour-stroma stained positive for S100A4 was lower than that of patients whose tumour-stroma stained negative (36.2% vs. 63.9%, P=0.003). Cox' risk ratio model analysis indicated that age ≤50 years (OR=1.866), lymph node metastasis(OR=1.826), distant metastasis(OR=6.224), lower histology differentiation and undifferentiation (OR=1.793), TNM stage Ⅲ-Ⅳ (OR=2.573) and positive expression of S100A4 protein in stroma of NSCLC(OR=1.776) were significantly independent prognostic factors which affected survival. Conclusion Expression of S100A4 protein in stroma of NSCLC is significantly associated with invasion, metastasis, TNM stage and prognosis. S100A4 protein might become a marker for prediction of tumor progression of disease and clinical therapy.

Abstract:Introduction Hypopharyngeal squamous cell carcinoma (HSCC) is among the most lethal tumors encountered in the head and neck and frequently involves regional metastasis. However, the mechanism underlying the aggressiveness of HSCC remains elusive. S100A4 is a well-established metastasis-promoting regulator in a variety of malignancies, but its role in HSCC has not yet been identified. Objectives Our objectives were to explore the expression levels of S100A4 in HSCC tumors and its association with clinicopathological parameters and the clinical prognosis of HSCC and to confirm its role in the metastatic process of the HSCC FaDu cell line in vitro. Methods We assessed the expression levels of S100A4 with immunohistochemistry (IHC) in HSCC tumors ( n = 71) and adjacent normal tissues ( n = 44). In vitro experiments were performed to explore the impact of S100A4 knockdown on biological phenotypes of human HSCC FaDu cell line, including migration, invasion, proliferation, apoptosis, and cell cycle. Results The expression of S100A4 was elevated in HSCC tumors compared with adjacent normal tissues and positively correlated with cervical lymph node metastasis in this HSCC patient cohort. In vitro experiments showed that S100A4 knockdown significantly impaired migration and invasion and increased the proportion of cells in G0/G1 phase with no change in proliferation or apoptosis in FaDu cells. Additionally, nuclear S100A4 expression proved to be an independent prognostic indicator in patients with HSCC. Conclusion This study demonstrated for the first time that S100A4 expression is upregulated in HSCC tumors and that this upregulation is positively correlated with cervical lymph node metastasis of this malignancy. The metastasis-promoting role of S100A4 was further validated in the HSCC FaDu cell line, indicating that S100A4 is a potential therapeutic target for HSCC. Furthermore, this study suggests that nuclear S100A4 expression could be considered a prognostic biomarker for HSCC.
Overexpression of S100A4 is Closely Associated with the Progression and Prognosis of Gastric Cancer in Young Patients.

Abstract:The aim of this study was to determine the correlation of S100A4 expression with the progression, prognosis and clinical pathology of gastric cancer (GC) in young pateints. A total of 85 tumor tissues with corresponding adjacent normal tissues and 62 non-metastatic lymph nodes (LNs) with corresponding metastatic LNs were obtained from young GC patients (<40 years old) who underwent surgery between January 2001 and December 2006. The expression of S100A4 was detected by RT-PCR and immunohistochemistry. Differences in the expression of S100A4 mRNA or protein were observed among the GC tissues, matched normal gastric mucosa, non-metastatic LNs and metastatic LNs. The expression of S100A4 mRNA and protein in GC tissues and metastatic LNs was significantly higher compared with that in the matched normal gastric mucosa and non-metastatic LNs, respectively (P<0.05). The overexpression of S100A4 was significantly associated with parameters involved in tumor progression and poor prognosis, including tumor size (P=0.017), Lauren classification (P=0.002), histological classification (P= 0.010), histological differentiation (P= 0.000), Borrmann classification (P=0.020), tumor-node-metastasis (TNM) stage (P=0.000), LN metastasis (P=0.000) and distant metastasis (P=0.024). Multivariate analysis suggested that patient age (P=0.035), tumor size (P=0.002), TNM stage (P=0.001) and S100A4 upregulation (P=0.000) were independent prognostic indicators for the disease. The overexpression of S100A4 in young GC patients is significantly associated with the clinicopathological characteristics. S100A4 may be used as a biomarker to predict the progression and poor prognosis of GC in young patients.

Abstract:Aim: Accumulated evidence has suggested a relationship between S100A4 protein expression and the development and progression of pancreatic cancer (PC) while its role in diagnosis and prognosis of PC still keeps inconsistent. To obtain definitive associations between S100A4 and PC, a meta-analysis was conducted. Materials and Methods: The PubMed and Chinese National Knowledge Infrastructure databases were electronically searched to identify studies reporting an association between S100A4 protein and PC. Statistical analyses were undergone with the utilization of STATA version 12.0 software. Results: Nine clinical studies with a total of 545 tumor samples were included in the meta-analysis. Results revealed that increased S100A4 expression were associated with the tumor-node-metastasis stages of PC (III-IV vs. I-II: odds ratio [OR] = 5.50, 95% confidence interval [95% CI] = 3.13-9.67, P < 0.001). Also, compared with 1-2 histologic grade of PC samples, S100A4 protein was expressed more frequently in samples with 3-4 histologic grade (grades 1-2 vs. grades 3-4: OR = 2.57, 95% CI = 1.05-6.24, P = 0.038). Conclusion: This meta-analysis showed that overexpression of S100A4 seems to be associated with tumor progression and poor prognosis of PC patients.
Downregulation of S100A4 Expression by RNA Interference Suppresses Cell Growth and Invasion in Human Colorectal Cancer Cells.

Abstract:S100A4 protein, a member of the S100 superfamily of calcium-binding proteins, is frequently observed in various types of human cancers, including colorectal cancer (CRC). Our previous investigations have demonstrated that the overexpression of S100A4 is associated with lymph node metastasis and poor prognosis in CRC; however, its biological roles in CRC remain unclear. In the present study, we compared the expression of S100A4 at the mRNA and protein levels in six CRC cell lines, and found that the expression levels roughly coincided with their invasiveness. Using RNA interference, we suppressed S100A4 expression in SW620 CRC cells with highly invasive potential and S100A4 high expression. The specific knockdown of S100A4 strongly suppressed cell growth, migration and invasion activities. Furthermore, employing metastasis-related gene mRNA microarrays, we found four genes to be significantly dysregulated (more than 2-fold) after downregulation of S100A4, including three downregulated genes (MMP9, MMP10 and CDH11) and one upregulated gene (TIMP4). Our present results indicate that S100A4 may positively regulate tumor cell proliferation, invasion and metastasis associated with multiple molecules. Thus, the inhibition of S100A4 might be a potentially novel approach to treatment for CRC.
Restoration of S100A4 Expression Enhances Invasive and Metastatic Potentials of COLO16 Cutaneous Squamous Cancer Cells.

Abstract:BACKGROUND: S100A4 promotes cancer metastasis but is frequently silenced in human cutaneous squamous cell carcinomas/c-SCCs due to DNA methylation, which may explain the less metastasized property of c-SCCs.OBJECTIVE: This study aims to check 1) whether the metastatic potential of S100A4-negative human c-SCC cells could be enhanced when S100A4 expression is restored in COLO16 c-SCC cells with S100A4 methylation and 2) the correlation of S100A4 expression and the differentiation grades and invasiveness of human c-SCC tumors.METHODS: The motility and invasion of parent and transfected COLO16 cells are examined by the use of 24-well modified Boyden chambers, scratched wound healing assay and nude mouse transplantation tumor model. Meanwhile, the correlation of S100A4 expression with growth patterns and grade of differentiation of c-SCC surgical specimens are analyzed.RESULTS: S100A4 expression is successfully restored in COLO16 cells after plasmid lipofectamine transfection. Transwell and scratched wound healing assays shows that the invasion and migration activities of S100A4-expressing transfectants are higher than that of parent COLO16 cells. Subcutaneous and foot pad c-SCC models are established by injecting 5 x 10(6)/100 l parental and S100A4-expressing COLO16 cells to BALB/c-nu/nude mice, respectively. Histological examination confirms the differences of invasiveness between the parent cells and the transfectants. Regional lymph node metastases are found only in the mice bearing S100A4-expressing tumors. S100A4 expression levels and frequencies are significantly different (P < 0.001) between the well and the poorly differentiated c-SCCs and closely correlated with tumor invasion (P < 0.05).CONCLUSIONS: S100A4 confers invasive and metastatic potentials on human c-SCCs. The low incidence of metastasis of c-SCCs, especially the well differentiated ones, might be due to the infrequent S100A4 expression. S100A4 can be regarded as a negative prognostic biomarker or a metastasis-risk factor of human c-SCCs.

Abstract:Intercellular cross-talk plays important roles in cancer progression and metastasis. Yet how these cancer cells interact with each other is still largely unknown. Exosomes released by tumor cells have been proved to be effective cell-to-cell signal mediators. We explored the functional roles of exosomes in metastasis and the potential prognostic values for hepatocellular carcinoma (HCC). Exosomes were extracted from HCC cells of different metastatic potentials. The metastatic effects of exosomes derived from highly metastatic HCC cells (HMH) were evaluated both in vitro and in vivo. Exosomal proteins were identified with iTRAQ mass spectrum and verified in cell lines, xenograft tumor samples, and functional analyses. Exosomes released by HMH significantly enhanced the in vitro invasion and in vivo metastasis of low metastatic HCC cells (LMH). S100 calcium-binding protein A4 (S100A4) was identified as a functional factor in exosomes derived from HMH. S100A4rich exosomes significantly promoted tumor metastasis both in vitro and in vivo compared with S100A4low exosomes or controls. Moreover, exosomal S100A4 could induce expression of osteopontin (OPN), along with other tumor metastasis/stemness-related genes. Exosomal S100A4 activated OPN transcription via STAT3 phosphorylation. HCC patients with high exosomal S100A4 in plasma also had a poorer prognosis. In conclusion, exosomes from HMH could promote the metastatic potential of LMH, and exosomal S100A4 is a key enhancer for HCC metastasis, activating STAT3 phosphorylation and up-regulating OPN expression. This suggested exosomal S100A4 to be a novel prognostic marker and therapeutic target for HCC metastasis.

Abstract:Rationale: Chemokines contribute to cancer metastasis and have long been regarded as attractive therapeutic targets for cancer. However, controversy exists about whether neutralizing chemokines by antibodies promotes or inhibits tumor metastasis, suggesting that the approach to directly target chemokines needs to be scrutinized.Methods: Transwell assay, mouse metastasis experiments and survival analysis were performed to determine the functional role of S100A14 in breast cancer. RNA-Seq, secreted proteomics, ChIP, Western blot, ELISA, transwell assay and neutralizing antibody experiments were employed to investigate the underlying mechanism of S100A14 in breast cancer metastasis. Immunohistochemistry and ELISA were performed to examine the expression and serum levels of S100A14, CCL2 and CXCL5, respectively.Results: Overexpression of S100A14 significantly enhanced migration, invasion and metastasis of breast cancer cells. In contrast, knockout of S100A14 exhibited the opposite effects. Mechanistic studies demonstrated that S100A14 promotes breast cancer metastasis by upregulating the expression and secretion of CCL2 and CXCL5 via NF-κB mediated transcription. The clinical sample analyses showed that S100A14 expression is strongly associated with CCL2/CXCL5 expression and high expression of these three proteins is correlated with worse clinical outcomes. Notably, the serum levels of S100A14, CCL2/CXCL5 have significant diagnostic value for discerning breast cancer patients from healthy individuals.Conclusions: S100A14 is significantly upregulated in breast cancer, it can promote breast cancer metastasis by increasing the expression and secretion of CCL2/CXCL5 via RAGE-NF-κB pathway. And S100A14 has the potential to serve as a serological marker for diagnosis of breast cancer. Collectively, we identify S100A14 as an upstream regulator of CCL2/CXCL5 signaling and a metastatic driver of breast cancer.

Abstract:This study was designed to explore the correlation between expressions of SATB1 and S100A4 and their relationships to the clinicopathologic parameters of colorectal carcinoma (CRC). Expressions of SATB1 and S100A4 were evaluated by immunohistochemistry in a cohort of 131 primary CRC patients undergone surgical resection from 2005 to 2007. SATB1 and S100A4 were positively expressed in 48.9% and 54.2% of CRC cases, respectively. SATB1 and S100A4 expressions in tumor tissues were significantly higher than those in the corresponding normal tissues. A positive correlation was observed between SATB1 and S100A4. Moreover, the levels of SATB1 and S100A4 were both significantly associated with invasion, lymph node status, and TNM stage of CRC, whereas S100A4 expression was also correlated with distant metastasis. Multivariate analysis revealed that SATB1 expression was an independent prognostic indicator for poor survival of CRC. Further survival analysis indicated that co-expression of SATB1 and S100A4 suggested a worse 5-year overall survival rate in CRC patients. Thus, combined analysis of SATB1 and S100A4 expressions may be valuable in determining the development and progression of CRC. Co-expression of SATB1 and S100A4 is an unfavorable prognostic indicator and may be useful in the follow-up of patients with CRC.

Abstract:Patients diagnosed with lymph node (LN) metastatic liver cancer face an exceedingly grim prognosis. In-depth analysis of LN metastatic patients' characteristics and tumor cells' interactions with human lymphatic endothelial cells (HLECs), can provide important biological and therapeutic insights. Here we identify at the single-cell level that S100A6 expression differs between primary tumor and their LN metastasis. Of particular significance, we uncovered the disparity in S100A6 expression between tumors and normal tissues is greater in intrahepatic cholangiocarcinoma (ICC) patients, frequently accompanied by LN metastases, than that in hepatocellular carcinoma (HCC), with rare occurrence of LN metastasis. Furthermore, in the infrequent instances of LN metastasis in HCC, heightened S100A6 expression was observed, suggesting a critical role of S100A6 in the process of LN metastasis. Subsequent experiments further uncovered that S100A6 secreted from tumor cells promotes lymphangiogenesis by upregulating the expression and secretion of vascular endothelial growth factor-D (VEGF-D) in HLECs through the RAGE/NF-kB/VEGF-D pathway while overexpression of S100A6 in tumor cells also augmented their migration and invasion. Taken together, these data reveal the dual effects of S100A6 in promoting LN metastasis in liver cancer, thus highlighting its potential as a promising therapeutic target.

Abstract:Invasion and metastasis are hallmarks of lung cancer and other malignant tumors. The activation of many genes has been shown to be important in tumor progression. We studied the corelationship of S100A4 gene (mts1) expression with the prognosis evaluation of lung cancer. Total RNAs were extracted from 46 cases of patient with lung cancer, and 4 lung cancer cell lines (A549, GLC-82, NCI-H446, NCI-H460), S100A4 gene expressions were detected at mRNA level by reverse transcription-polymerase chain reaction (RT-PCR). Up-regulation of S100A4 gene was shown in 29 of 46 (63.0%) lung cancer patients and all of the lung cancer cell lines, especially in younger patients (89.5% vs 44.4%, p <0.01), poor differentiated lung cancer patients (81.0% vs 48.0%, p <0.05), mediastinum lymphonodus metastasis (77.3% vs 20%, p <0.05) and the IIIa stage patients (73.1% vs 0, p <0.05). S100A4 gene played an important role in lung cancer progression, correlated well with the condition of the lung cancer differrentiation, mediastinum lymphonodus metastasis, and the prognosis for the patients of lung cancer. It was likely that S100A4 gene as a prognostic, predictive, or even diagnostic factor in clinical practice.
