Abstract:MicroRNA*s are about 22nt noncoding RNAs, which are processed from precursors with a characteristic hairpin secondary structure in the biogenesis of microRNAs. Recently, miRNA* strands were shown to mediate post-transcriptional regulatory networks, rather than serve merely as non-functional by-product in general view. Unlike miRNAs bound to AGO1, miRNA* strands are bound to AGO2 to form RISC duplex to mediate RNAi, which is similar to siRNA. This paper mainly reviewed the recent research progresses on miRNA*, such as the biosynthesis, biological characteristics, and functions.

Abstract:Since the beginning of this century, microRNAs (miRNAs), which are tiny RNA molecules, have become one of the major research topics on gene expression regulation in both animals and plants. The major task of miRNA study is to elucidate how the miRNAs are expressed in vivo, how they exert regulatory effects on their targets, and how they can be qualitatively or quantitatively cloned. For these purposes, the methodology of miRNA study has been developed and significantly improved in recent years. The focus here is on a number of powerful methods for plant miRNA research including bioinformatics tools and experimental approaches being used for upstream or downstream analysis of miRNAs or miRNA cloning. Some discrepancies exist in the miRNA research methodology between plants and animals, for example, 5' modified RACE (Rapid Amplification of cDNA Ends) can be used for cleavage target validation only in plants. However, numerous common methods are shared by these two miRNA research areas. Thus, this review will enhance our understanding of miRNA research methodology in organisms.

Abstract:MicroRNAs(miRNAs) are a group of endogenous non-coding single-stranded RNAs which extensively exist in eukaryotic cells and viruses. They are about 21–24 nucleotides long and highly conserved in phylogeny. For their important roles in the developmental timing control and disease occurrence, miRNAs have become one of the hotspots in biological research. Here we briefly review the research advances in knowledge of the plant miRNAs on their synthesis, structural characteristics, mechanism of action and especially the functions.

Abstract:MicroRNAs (miRNAs) are a recently discovered family of endogenous, noncoding RNA molecules approximately 22 nt in length. miRNAs modulate gene expression post-transcriptionally by binding to complementary sequences in the coding or 3′ untranslated region of target messenger RNAs (mRNAs). It is now clear that the biogenesis and function of miRNAs are related to the molecular mechanisms of various clinical diseases, and that they can potentially regulate every aspect of cellular activity, including differentiation and development, metabolism, proliferation, apoptotic cell death, viral infection and tumorgenesis. Here, we review recent advances in miRNA research, and discuss the diverse roles of miRNAs in disease.

Abstract:Approximately 22 nucleotide-long non-coding small RNAs (ncRNAs) play crucial roles in physiological and pathological activities, including microRNAs (miRNAs). Long ncRNAs often stay in the cytoplasm, modulating post-transcriptional gene expression. Briefly, miRNA binds with the target mRNA and builds a miRNA-induced silencing complex to silence the transcripts or prevent their translation. Interestingly, data from recent animal and plant studies suggested that mature miRNAs are present in the nucleus, where they regulate transcriptionally whether genes are activated or silenced. This significantly broadens the functional range of miRNAs. Here, we reviewed and summarized studies on the functions of nuclear miRNAs to better understand the modulatory networks associated with nuclear miRNAs.

Abstract:MicroRNA(miRNA) is a kind of small non-protein-coding RNA about 22 nucleotides in length.MicroRNA can regulate organism growth and development by mediating the degradation of complementary mRNA or inhibiting translation.MicroRNA play important roles in homeostatic processes such as development,cel1 proliferation,cell apoptosis.It was certified that miRNA participate the processes of tumorigenesis and development and its expression profiles can be used as the indicator of tumor diagnosis and prognosis.Mutation,deletion or high expression of miRNA was found to correlate with various human cancer.miRNA is involved in tumor cell proliferation,differentiation and apoptosis processes as function as oncogenes or tumor supressors.In this review,we have summarized the research of miRNA in tumorigenesis,development,diagnosis and therapy.

Abstract:MicroRNA(miRNA)是一类长约21～25nt的内源性单链小分子RNA(small RNA),可调节与其序列互补mRNA的表达,普遍存在于高等生物界,在不同物种间具有高度的保守性,表达具有细胞特异性或组织特异性.通过与mRNAs特异性的碱基配对,引导靶序列进入降解或(和)翻译抑制,以此参与个体发育、细胞分化、细胞增殖、物种进化以及疾病发生过程中的基因表达调控.该文简述了近年对miRNA的形成、功能,作用机制等研究的新进展,并对其在基因沉默中的作用作了重点概述。

Abstract:Ever since microRNAs (miRNAs) were first recognized as an extensive gene family >20 years ago, a broad community of researchers was drawn to investigate the universe of small regulatory RNAs. Although core features of miRNA biogenesis and function were revealed early on, recent years continue to uncover fundamental information on the structural and molecular dynamics of core miRNA machinery, how miRNA substrates and targets are selected from the transcriptome, new avenues for multilevel regulation of miRNA biogenesis and mechanisms for miRNA turnover. Many of these latest insights were enabled by recent technological advances, including massively parallel assays, cryogenic electron microscopy, single-molecule imaging and CRISPR–Cas9 screening. Here, we summarize the current understanding of miRNA biogenesis, function and regulation, and outline challenges to address in the future.

Abstract:microRNAs is a group of small non-coding RNAs that play a negative regulation role in expression of target genes at post-transcriptional level by inhibition or degradation of target mRNAs after combination of the seed sequence (SS) in microRNAs with the SS-binding sequences usually located at 5'ends of target mRNAs.microRNAs was firstly found in Caenorhabditis elegans.Subsequently,many different microRNAs in eukaryocytes were revealed.In eukaryocytes,microRNA precursors are transcribed at first and then become functional microRNAs with 21-23 nt in size after splice.Most of eukaryocytic microRNAs combime with the sequences at 3'end of target mRNAs that cause the translation inhibition or degradation of the mRNAs.In the recent years,many different prokaryocytes,such as bacteria,have been confirmed to possess microRNAs.However,the microRNAs in prokaryotes such as bacteria are 50-400 nt in size and have the biological activity without splice.Moreover,the characteristics,action sites and mechanisms of the prokaryotic microRNAs have some certain diversity compared to the eukaryotic microRNAs.Our review briefly introduce the major regulation mechanisms of gene expression as well as the general characteristics of microRNAs and their regulation mechanisms of gene expression in prokaryocytes and eukaryocytes,which will provide a basis for further and profound study on the gene expression regulation and pathogenic mechanisms of prokaryotic microbial pathogens.

Abstract:MicroRNAs (miRNAs) are a family of endogenous, small (approximately 22 nucleotides in length), noncoding, functional RNAs. With the development of molecular biology, the research of miRNA biological function has attracted significant interest, as abnormal miRNA expression is identified to contribute to serious human diseases such as cancers. Traditional methods for miRNA detection do not meet current demands. In particular, nanomaterial-based methods, nucleic acid amplification-based methods such as rolling circle amplification (RCA), loop-mediated isothermal amplification (LAMP), strand-displacement amplification (SDA) and some enzyme-free amplifications have been employed widely for the highly sensitive detection of miRNA. MiRNA functional research and clinical diagnostics have been accelerated by these new techniques. Herein, we summarize and discuss the recent progress in the development of miRNA detection methods and new applications. This review will provide guidelines for the development of follow-up miRNA detection methods with high sensitivity and specificity, and applicability to disease diagnosis and therapy.

Abstract:MicroRNAs (miRNAs), which are evolutionarily well-conserved, 21- to 23-nucleotide-long, small non-coding RNAs, are widely involved in the regulation of multiple biological processes, such as development, organogenesis, cell proliferation, cell differentiation, and apoptosis. Abstract:microRNAs (miRNAs) are short single-stranded non-coding RNAs between 21 and 25 nt in length in eukaryotic organisms, which control post-transcriptional gene expression. Through complementary base pairing, miRNAs generally bind to their target messenger RNAs and repress protein production by destabilizing the messenger RNA and translational silencing. They regulate almost all life activities, such as cell proliferation, differentiation, apoptosis, tumorigenesis, and host-pathogen interactions. Methylation modification is the most common RNA modification in eukaryotes. miRNA methylation exists in different types, mainly N 6 -methyladenosine, 5-methylcytosine, and 7-methylguanine, which can change the expression level and biological mode of action of miRNAs and improve the activity of regulating gene expression in a very fine-tuned way with flexibility. In this review, we will summarize the recent findings concerning methylation modifications of miRNA, focusing on their biogenesis and the potential role of miRNA fate and functions.
