Detection of replicative state of HBV in peripheral blood mononuclear cells by nuclease digestion and nested PCR

Abstract:AIM:To investigate the existence and replicative ability of HBV DNA in peripheral blood mononuclear cells (PBMCs) of chronic HBV-infected patients. METHODS:HBV DNA in PBMCs was detected by nested PCR; covalently closed circular DNA (cccDNA) in PBMCs was detected by Hirt method, mung-bean nuclease digestion and nested primers which crossed single chain and three chains for cleaning rcDNA as possible as we could. RESULTS:cccDNA in PBMCs could be detected after nuclease digestion and nested PCR, while rcDNA wasn't detected; total positive rates of HBV DNA and cccDNA in PBMCs were 62.5% and 45.8%, respectively; HBV DNA positive rate in PBMCs were significantly higher than cccDNA positive rate (P<0.01).HBV DNA positive rates (80.0%) and cccDNA positive rates(45.0%) in PBMCs were significantly higher in HBeAg positive group than in HBeAg negative group(61.7%, 30.0%, P<0.01). CONCLUSION: The methods of nuclease digestion and nested PCR can effectively avoid the influence of rcDNA on cccDNA amplification; HBV not only infects PBMCs but also participates in replication; the high positive rates of HBV DNA and cccDNA in PBMCs were detected in HBeAg positive patients.
