Abstract:Dendrimer is one type of macromolecules with well-defined, highly-branched and nano-scaled architectures, composed of three distinct domains: core, branches and terminal groups. Its potential functions and applications are explored based on the unique physical and chemical features due to special molecular architecture. Using polyhedral oligomeric silsesquioxanes (POSS) as the core of dendrimers is an effective and facile way to simplify the tedious repetitive steps of preparation and separation. Since POSS allows eight branches to radiate from a silicon-oxygen right cubic core, the dendrimers can be constructed in a well-controlled globular, three-dimensional framework, and large numbers of peripheral groups can be obtained at relatively low generation numbers. As a kind of novel nano-hybrid supermolecules, POSS-based dendrimers have attracted considerable interest in materials science. In this review, we briefly introduce the synthetic approaches of some typical POSS-based dendrimers categorized by the different chemical composition of branching units, and then focus on the potential functions and applications of this nano-hybrid material in the field of catalysis, gene and drug delivery, liquid crystals, light harvesting and energy transfer. The specific and excellent properties imparted by the incorporation of POSS into dendrimers are also presented. Finally, the advances of POSS-based dendrimers are prospected.

Abstract:A series of amphiphilic dendrons (G1-G4) have been designed and synthesized, which have a highly branched aliphatic hydrocarbon skeleton and a hydrophilic hydroxyl functionality to enable conjugation with other substrates. The higher generation dendrons (G3 and G4) were synthesized by a double-stage convergent method, which shortened the synthetic route significantly and provided the products in an efficient manner. The key branching step involved a double alkyl-metal addition to an ester functionality followed by deoxygenation of a resulting tertiary alcohol by triethylsilane.

Abstract:Fluorinated polyether dendritic sectors 4, 7 and 10 from the first to the third generations, respectively, were prepared by a convergent approach from readily available pentafluorophenol, phloroglucinol and 1,3-dibromopropane. These dendritic sectors have good solubility properties and are stable in both acidic and basic media. Hence they are useful dendritic building blocks for the preparation of functional dendrimers.

Abstract:Dendritic compounds (so-called dendrimers) with nano-scale represent unique symmetric and spherical structures. They are usually prepared through two fundamental methods based on molecular level which involved the convergent method and divergent method, although the synthetic methods have a tendency towards the diversity and functionalization. The outershell of dendrimers with rich peripheral reactive sites are easily modified by small functional molecules. Moreover, the higher generation dendrimers also possess more exposed functional groups on the surface prior to the previous one and they are much easily customized for much more applications. Consequently, based on previous researches, this review summarized wide applications of dendritic compounds in many fields which were investigated in detail, including gene vector, drug carrier, catalysis, sensor industry, photoelectric material, etc. Dendrimers provide promising tools for the cellular delivery of molecular cargos ranging in size from small molecules and peptides to proteins and DNA. More importantly, it is necessary to explore new synthetic methods and undiscovered applications of new dendrimers.

Abstract:Chemoselective reactions allow near-precision control over the polymer composition and topology to create sequence-controlled polymers with similar secondary and tertiary structures to those found in proteins. Dendrimers are recognized as well-defined macromolecules with the potential to mimic protein surface functionality due to the large number of functional groups available at its periphery with the internal structure acting as the support scaffold. Transitioning from using small-molecule dendrimers to dendritic macromolecules will not only allow retention of the high peripheral functionality but also provide an internal scaffold with a desired polymer composition within each generational layer. Here, we exemplify a systematic approach to creating a dendritic macromolecule with the placement of different polymer building blocks in precise locations within the internal structure and the placement of three different amino acid moieties clustered at the periphery. The synthesis of this ABC dendritic macromolecule was accomplished through iterative chemoselective reactions.

Abstract:We hereby report the synthesis of a novel type of dendritic structure, a hierarchically branched dendritic polymer, in which discrete dendrons are built at the branch points of a dendrimer-like skeleton. The synthesis consists of two stages, i.e., the preparation of dendrimer-like polystyrene through anionic polymerization and a chlorosilane coupling reaction as well as the formation of carbonate-containing dendrons from the primary hydroxy groups at the branch points of dendrimer-like polystyrene through highly selective reactions between imidazole carboxylic esters and primary hydroxyls. The hydroxy group is introduced by monoaddition of a 1,1-diphenylethylene derivative possessing-OSiBuMe2 during anionic polymerization followed by deprotection. A hierarchically branched dendritic polymer, G3-g3, containing 21 carbonate-derived dendrons and 167 (theoretically 232) hydroxyl groups (subsequently transformed into acetoxy groups for better solubility), in a single dendrimer-like polystyrene, is prepared and characterized by NMR monitoring the synthetic process. The ultimate product shows higher multiplicity of functional groups owing to the high-level dendritic or multiple dendrons on dendrimer-like structure.
Easy synthesis of dendrimer-like polymers through a divergent iterative end-grafting method

Abstract:We report here an easy method for the synthesis of dendrimer-like polymers with high branching functionality (1 -> 8). The synthetic process involves iterative grafting reactions in a divergent way. A multi-functional core containing short segments of polyisoprene (PI), either as a star-like block copolymer of isoprene and styrene or as a linear triblock copolymer of isoprene, styrene and isoprene (coded G1), is epoxidized on the double bonds and grafted with a living block copolymer, polyisoprene-b-polystyrenyllithium (PI-b-PSLi), again with a short PI segment, through the ring-opening reaction of oxirane by polymeric anions. The resulting graft polymer, G2, possesses a definite number of PI segments at the periphery. These PI segments are further epoxidized followed by the ring-opening addition of PI-b-PSLi, affording G3. Repeating the process leads to the synthesis of a dendrimer-like polystyrene up to 5th generation with a polydispersity lower than 1.21, as measured by SEC. A feature of the process is the easily accessible high chain density in the final product, although defects exist due to steric hindrance in the reactions of high generations. The solution properties of the dendritic products are investigated using viscometry and dynamic and static laser light scattering on the molecular conformation. The results support a compact globular conformation model for the dendrimer-like products. In addition, the chain density of the products from the star-like core is higher than that of products from a linear triblock core. AFM results show that the dendritic products adopt flattened conformations and tend to form lateral sphere-like aggregates on mica substrate.

Abstract:Dendrigraft polymers are a class of highly branched macromolecules belonging to the dendritic polymer family.Steady and growing interest is quite noticeable for research directed toward the synthesis、architecture and application of the dendrigraft.The highlight of this review is synthetic methodologies.Three distinct synthetic methodologies are considered for the preparation of these materials: divergent 'grafting-onto'、 divergent 'grafting-from' and convergent 'grafting-through'.Comparison of the other members of the dendritic polymer family: unlike the other dendritic counterparts,dendrigraft are not composed of repeatable monomeric units throughout the whole macromolecule,but only the side chains;dendrigraft have the unique core-shell structures just like the others;the size of dendrigraft are typically 1~2 orders of magnitude larger than their comparators.Interesting properties reported for dendrigraft are also discussed,as well as some applications.

Abstract:Synthesis of dendrimers has been directed toward process efficiency and structural diversity. We report a divergent approach to the preparation of dendrimers with both ABC and ABB branching structures from nonbranching monomers by combination of efficient orthogonal ABC Passerini multicomponent reaction (MCR) and ABB thiol-yne MCR. Two kinds of dendrimers were synthesized efficiently: (1) dendrimers with two generations in three steps and (2) dendrimers with two generations containing one kind of internal functional group and two kinds of surface functional groups in five steps. This new synthetic method offers an efficient access to dendrimers with structural diversity.
Access to Diverse Glutamic Acid Dendrons and a Janus Peptide Dendrimer Using an Atypical Solid Phase Synthesis

Abstract:The negligible cytotoxicity of anion surface-linked dendrons makes glutamic acid-based dendrons a potential candidate for materials and biological applications. Despite the inherent drawbacks of the conventional solution phase synthesis of glutamic acid-based dendrons, there have been no advancements in these protocols. Herein, we demonstrate the first-ever convergent solid phase synthesis of dendrons, up to fourth generation, having glutamic acid branching points produced by preactivation of dicarboxylic acid groups with N-hydroxysuccinimide and simultaneous coupling with amine groups of two growing peptide chains, with excellent yields (30-70%). In addition to the general advantages, such as the easy workup, a final single purification step, and an overall short synthesis duration, the convergent solid phase synthesis allowed us to chemically synthesize glutamic acid branching-based dendrons that cannot be accessed by standard divergent solid phase synthesis. This method has also been validated for its application in synthesizing hard-to-achieve Janus peptide dendrimers in a single stretch on a solid support. Our work corroborates the efficacy of controlled -COOH activation to accomplish an atypical solid phase synthesis of diverse glutamic acid dendrons in a convergent fashion. This is the first example of a Janus peptide dendrimer being synthesized on a solid support, utilizing both convergent and divergent approaches simultaneously.
Synthesis of Novel Poly(ester Amine) Dendrimers by Michael Addition and Acrylate Esterification

Abstract:Dendrimers are a new kind of polymers. They have gained widespread attentions due to their remarkable properties in recent years. In the present work, a facile and efficient synthetic approach to prepare poly(ester amine) dendrimers is presented. Three different generations of poly(ester amine) dendrimers are synthesized starting from using pentaerythritol tetraacrylate as a core, followed by Michael addition and acrylate esterification reactions. This synthetic protocol is highly efficient, high yielding, and environmentally benign. To the best of our knowledge, this is the first time that these novel poly(ester amine) dendrimers are synthesized by using Michael addition reaction as the key step. The chemical structures of these poly(ester amine) dendrimers were measured with Fourier transform infrared (FT-IR) spectroscopy, NMR, elemental analysis, gas permeation chromatography, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF). In addition, the hydroxyl value and solubilities of the poly(ester amine) dendrimers were also examined. These synthesized poly(ester amine) dendrimers, which have well-defined structures with very narrow polydispersities, can be used in many fields such as chemical engineering, medicine, and nanoengineering.
Synthesis of Dendronized Polymers by a “n + 2” Approach

Abstract:A series of dendronized polymers (DP) were synthesized by attaching second-generation dendron onto the preformed DP core of generation 1–3 (n + 2 approach). The obtained DP were compared to the reference DP prepared by the conventional graft-from (n + 1) approach using GPC, UV–vis, 1H NMR, and AFM analyses. The former two analyses showed that the newly prepared DP tend to have a more significant decrease in dendron integrity as the generation increases; however, the striking similarities from the latter two analyses suggest that the properties of the new samples do not much deviate from the reference DP by the graft-from approach. The advantages of the “n + 2” approach over the “n + 1” approach include shorter reaction time, higher yield, and easier purification. On top of these, it has the power of surpassing some selected postpolymerization steps, which are required for the conventional graft-from approach and may be detrimental in some occasions.
Divergent growth of poly(amidoamine) dendrimer-like branched polymers at the reducing end of cellulose nanocrystals

Abstract:This paper presents the growth of dendritic polymers at the reducing ends of cellulose nanocrystals by the "grafting from" approach. We took advantage of the chemically differentiated ends of cellulose nanocrystals to specifically synthesize dendrimers at their reducing end by the divergent approach. We used acid-amine coupling reactions in aqueous media to synthesize the carboxylic acid- or amine-terminated poly(amidoamine) dendrimers. The growth of dendrimer generations was monitored by UV and FTIR spectroscopies, and we successfully introduced up to 4 generations. The dendrimer growth at reducing ends was demonstrated by the nanocrystal adsorption driven by the peripheral amino groups onto gold surfaces. Hence, the results from quartz crystal microbalance with dissipation (QCM-D) pointed to a rather upright orientation of the dendrimer-modified cellulose nanocrystals. As the generation increased, the adsorbed layers appeared to be more flexible, which demonstrated that the functionality at the reducing end can successfully tune the properties of cellulose nanocrystals.

Abstract:Two DendriMac polymers (Dendri-hydr and Dendri-click) are efficiently and conveniently synthesized via the combination of living anionic polymerization (LAP) and hydrosilylation/click chemistry. Based on the end-capping of DPE derivatives (DPE-SiH and DPE-DA) toward polymeric anions, the polymeric core and arms are effectively synthesized, and the base polymers can be regarded as polymeric bricks. Hydrosilylation and click chemistry are used as coupling reactions to construct the DendriMac polymers with high efficiency and convenience. The numbers of branched arms are calculated by SEC as 5.84 and 6.08 for Dendri-hydr and Dendri-click, respectively, which indicate that the DendriMac architectures exhibit high structural integrity. Because of its independence, high efficiency, and convenience, the whole construction can be regarded as the "building of polymeric bricks."
