Abstract:Dendrimer is one type of macromolecules with well-defined, highly-branched and nano-scaled architectures, composed of three distinct domains: core, branches and terminal groups. Its potential functions and applications are explored based on the unique physical and chemical features due to special molecular architecture. Using polyhedral oligomeric silsesquioxanes (POSS) as the core of dendrimers is an effective and facile way to simplify the tedious repetitive steps of preparation and separation. Since POSS allows eight branches to radiate from a silicon-oxygen right cubic core, the dendrimers can be constructed in a well-controlled globular, three-dimensional framework, and large numbers of peripheral groups can be obtained at relatively low generation numbers. As a kind of novel nano-hybrid supermolecules, POSS-based dendrimers have attracted considerable interest in materials science. In this review, we briefly introduce the synthetic approaches of some typical POSS-based dendrimers categorized by the different chemical composition of branching units, and then focus on the potential functions and applications of this nano-hybrid material in the field of catalysis, gene and drug delivery, liquid crystals, light harvesting and energy transfer. The specific and excellent properties imparted by the incorporation of POSS into dendrimers are also presented. Finally, the advances of POSS-based dendrimers are prospected.

Abstract:The synthesis and characterization of a series of peptide dendrons with glycin (Gly) and glutamate (Glu) as building blocks were described. The dendrons were prepared with convergent method by protection, deprotection and DCC coupling reactions. With the characterization of elemental analysis, H-1-NMR, C-13-NMR, FTIR, EI-MS and MAILD-TOF MS, the designed molecular structure was proved. It could be predicted that the obtained compounds will exhibit unique properties and potential applications in the fields of biology, medicine and so on. Further studies on the properties of these compounds are in progress.

Abstract:Fluorophore-labeled glycodendrimers have potential use in the study of carbohydrate-protein interactions by fluorescence spectroscopy and imaging methods. The current solution-phase methods for preparation of such glycoconjugates are labour intensive. On the other hand, the intrinsically more efficient solid-phase methods have been explored only at low generations. Herein we disclose a direct, expedient glycodendrimer synthesis from commercially available or easily prepared building blocks by machine-assisted solid-phase peptide synthesis (SPPS). Large, monodisperse 4th- and 5th-generation polylysine dendrons are prepared and capped with 16 and 32 mannose residues, respectively, in a single synthetic operation. Incorporation of a C-terminal lysine residue in the 4th-generation dendron allows fluorescence labelling with a number of common labels on resin, in organic solvent or in aqueous buffer, as required. A single HPLC purification is sufficient in all cases to obtain a homogeneous sample. The monodispersity of the glycodendrons is confirmed by MALDI-TOF. FITC-labeled 4th-generation glycodendron is an excellent probe for the imaging studies of mannose-receptor-mediated entry of into dendritic cells by confocal fluorescence microscopy.

Abstract:A series of amphiphilic dendrons (G1-G4) have been designed and synthesized, which have a highly branched aliphatic hydrocarbon skeleton and a hydrophilic hydroxyl functionality to enable conjugation with other substrates. The higher generation dendrons (G3 and G4) were synthesized by a double-stage convergent method, which shortened the synthetic route significantly and provided the products in an efficient manner. The key branching step involved a double alkyl-metal addition to an ester functionality followed by deoxygenation of a resulting tertiary alcohol by triethylsilane.

Abstract:A series of poly(ether-amide) dendrimers with amino acids and peptides as the peripheral functional groups was synthesized, and their structures were confirmed by nuclear magnetic resonance (NMR) and electrospray ionization-mass spectrometry (ESI-MS) spectrometry. Molecular dynamics simulation of the peptide dendrimers in solution was performed, indicating that, the prior conformations of the dendrimers were atom number dependent, i.e., with the increases of the atom number, the prior conformations were more spherical. Also, the amino acid α-C atom radial distribution indicated that, with larger peripheral groups, more back-folding of the dendrimers occurred.

Abstract:[GRAPHICS] A novel family of giant pi-conjugated dendrimers (G0, G1, and G2) solely constructed by 5,5,10,10,15,15-hexahexyltruxene units has been developed in a convergent manner through a Suzuki cross-coupling reaction. The overall yields to such large rigid conjugated dendrimers are quite satisfying. The structures and purity of these nanosize rigid dendrimers are verified by H-1 and C-13 NMR, MALDI-TOF MS, and elemental analysis.
Access to Diverse Glutamic Acid Dendrons and a Janus Peptide Dendrimer Using an Atypical Solid Phase Synthesis

Abstract:The negligible cytotoxicity of anion surface-linked dendrons makes glutamic acid-based dendrons a potential candidate for materials and biological applications. Despite the inherent drawbacks of the conventional solution phase synthesis of glutamic acid-based dendrons, there have been no advancements in these protocols. Herein, we demonstrate the first-ever convergent solid phase synthesis of dendrons, up to fourth generation, having glutamic acid branching points produced by preactivation of dicarboxylic acid groups with N-hydroxysuccinimide and simultaneous coupling with amine groups of two growing peptide chains, with excellent yields (30-70%). In addition to the general advantages, such as the easy workup, a final single purification step, and an overall short synthesis duration, the convergent solid phase synthesis allowed us to chemically synthesize glutamic acid branching-based dendrons that cannot be accessed by standard divergent solid phase synthesis. This method has also been validated for its application in synthesizing hard-to-achieve Janus peptide dendrimers in a single stretch on a solid support. Our work corroborates the efficacy of controlled -COOH activation to accomplish an atypical solid phase synthesis of diverse glutamic acid dendrons in a convergent fashion. This is the first example of a Janus peptide dendrimer being synthesized on a solid support, utilizing both convergent and divergent approaches simultaneously.

Abstract:Novel dendrimers consisting of a triphenylamine core and 1st to 3rd generations of 9-phenylcarbazole-based dendrons were synthesized by Suzuki coupling reaction through convergent approach. Their structures were confirmed by two-dimensional correlated H-H COSYand C-H HSQC NMR spectra, MALDI-TOF MS and elemental analysis. The dendrimers exhibit excellent thermal stability with 5% weight loss temperatures over 540 degrees C. The computer modeling reveals that the dendrons in dendrimers greatly twisted with the generation, leading to the dendrimers decreased crystalline ability. Of interest is the observation that, for an identical dendrimer, the solid film displays the similar UV absorption and luminescence emission profiles to the solution sample, indicating that, after evaporation of solvent, the rigid dendrimer can well maintain its conformational morphology and the aggregation or stacking of the chromophoric groups is significantly inhibited. All the dendrimers can emit intense fluorescence with narrow full width at half maximum (FWHM) around 46-50 nm. Moreover, with the incremental generation, the quantum efficiencies remarkably increase from 64 to 95%, suggesting that the highly contorted and bulky dendrons effectively decrease energy wastage and non-radiative decay. The synergistic effect of electron-donating triphenylamine core and 9-phenylcarbazole-based dendrons results in the HOMO energy level of -5.36 eV for the 3rd-generation dendrimer, very close to the work function of the ITO/PEDOT electrode (-5.2 eV), which characteristic is very advantageous for the hole injection and transport materials.

Abstract:The design and synthesis of the first examples of AB4 and AB5 dendritic building blocks with complex architecture are reported. Structural and retrostructural analysis of supramolecular dendrimers self-assembled from hybrid dendrons based on different combinations of AB4 and AB5 building blocks with AB2 and AB3 benzyl ether dendrons demonstrated that none of these new hybrid dendrons exhibit the previously encountered conformations of libraries of benzyl ether dendrons. These hybrid dendrons enabled the discovery of some highly unusual tapered and conical dendrons generated by the intramolecular back-folding of their repeat units and of their apex. The new back-folded tapered dendrons have double thickness and self-assemble into pine-tree-like columns exhibiting a long-range 7/2 helical order. The back-folded conical dendrons self-assemble into spherical dendrimers. Non-back-folded truncated conical dendrons were also discovered. They self-assemble into spherical dendrimers with a less densely packed center. The discovery of dendrons displaying a novel crown-like conformation is also reported. Crown-like dendrons self-assemble into long-range 5/1 helical pyramidal columns. The long-range 7/2 and 5/1 helical structures were established by applying, for the first time, the helical diffraction theory to the analysis of X-ray patterns obtained from oriented fibers of supramolecular dendrimers.

Synthesis of Dendrimer-Like Molecules with Partial Carbon Chain via Iterative Single Unit Monomer Insertions

Abstract:Carbon-chain dendritic polymers hold unique properties and promising applications. However, synthesizing carbon-chain dendrimers, beyond conjugated ones, remains a challenge. Here, the use of the iterative single unit monomer insertion technique for synthesizing 2.5 generation partial-carbon-chain dendrimers (G2.5) is described, utilizing bismaleimide as the core, a maleimide-trithiocarbonate conjugate as the branching unit, and indene as the spacer unit, following a divergent growth strategy. The optimized conditions for synthesizing the maleimide-trithiocarbonate branching unit are a bismaleimide to trithiocarbonate ratio of 5:1 and a reaction time of 30 min. The structures are verified using 1H nuclear magnetic resonance, gel permeation chromatography, and matrix-assisted laser desorption/ionization-time of flight mass spectra. A four-arm star polymer is then synthesized using the G2.5 as the core. This synthesis of a partial-carbon-chain dendrimer establishes a foundational step toward creating all-carbon-chain ones and may open new application avenues in material science.

Abstract:Dendritic compounds (so-called dendrimers) with nano-scale represent unique symmetric and spherical structures. They are usually prepared through two fundamental methods based on molecular level which involved the convergent method and divergent method, although the synthetic methods have a tendency towards the diversity and functionalization. The outershell of dendrimers with rich peripheral reactive sites are easily modified by small functional molecules. Moreover, the higher generation dendrimers also possess more exposed functional groups on the surface prior to the previous one and they are much easily customized for much more applications. Consequently, based on previous researches, this review summarized wide applications of dendritic compounds in many fields which were investigated in detail, including gene vector, drug carrier, catalysis, sensor industry, photoelectric material, etc. Dendrimers provide promising tools for the cellular delivery of molecular cargos ranging in size from small molecules and peptides to proteins and DNA. More importantly, it is necessary to explore new synthetic methods and undiscovered applications of new dendrimers.

Abstract:Heterocyclic unit 2 containing complementary donor-donor-acceptor (DDA) and acceptor-acceptor-donor (AAD) hydrogen bonding arrays at an angle of about 601 Was designed to self-assemble into a hexamer. To investigate whether this unit could self-assemble dendrimers, the 2,8-diamino-2-N-ethylpyrimido-(4,5-b)(1, 8)naphthyridine-3H-4-one subunit was synthesized with a first (2a), second (2b), and third generation (2c) Frechet-type dendron attached to the 8-amino group. The synthesis of 2a-c was accomplished in 11 steps beginning with 2,6-diaminopyridine and the corresponding dendron bromide. Studies using H-1 NMR, size exclusion chromatography (SEC), and dynamic light scattering (DLS) support the cooperative formation of cyclic hexamers in apolar solvents. The stability of the self-assembled dendrimers is dependent on the size of the attached dendron, and mixing studies with 2a-c indicate their usefulness in constructing dynamic combinatorial libraries.

Abstract:The synthesis of a glycodendrimer by incorporating repetitive mannoside units onto a glucoside core was carried out to multivalently probe fundamental carbohydrate protein interactions. The dendritic structure was constructed by a modified procedure that utilized multiple glycosylations between a thioether glycosyl donor and five elongated spacer arms of a glycosyl acceptor. The completed dendrimer bears a full carbohydrate structure, and thus should find its potential application in the study of mannose lectin interactions.
