Abstract:Poultry industry has been recognized as a fast-developing sector aiming to produce low-cost and high-nutrient foods for human consumption. This review article aimed to focus on the significant roles of dietary betaine supplementation in the poultry industry. In this respect, different effects of betaine on performance and carcass traits, as well as its osmoregulatory, anticoccidial, immune-modulatory, and heat-stress alleviation activities, were discussed. Different concentrations of betaine supplementation can improve the feed conversion ratio, final body weight, breast muscle yield, egg production ratio, and reduce body fat contents in broiler chicken, turkey, duck, geese, and quail diets. Betaine supplemented with methyl groups can eliminate the need to have some methyl-group donors, including choline and methionine, therefore having positive effects on feed conversion ratio in poultry diets. The osmolytic character of betaine can alleviate heat stress and have a positive impact on tonic immobility, which consequently reduces stress in poultry. By inhibiting distinct developmental stages of Eimeria species, betaine reduces the damaging effects of coccidiosis on broiler chickens and improves intestinal structure and function. The immunological, cardiovascular, neurological, renal, and hepatic metabolic systems benefit from betaine's osmo-protective properties. Therefore, betaine has the potential to be considered as an alternative to feed additives and enhances the health status and productive performance of poultry.

Abstract:Betaine is widely used in animal nutrition to promote growth, development and methyl donor during methionine metabolism through nutritional reprogramming via regulation of gene expression. Prenatal betaine exposure is reported to modulate hypothalamic cholesterol metabolism in chickens, yet it remains unknown whether feeding hens with betaine-supplemented diet may affect hypothalamic cholesterol metabolism in F1 offspring. In this study, hens were fed with basal or betaine-supplemented (0.5%) for 30days, and the eggs were collected for incubation. The hatchlings were raised under the same condition up to 56days of age. Betaine-treated group showed significantly (P<0.05) higher plasma concentration of total cholesterol and HDL-cholesterol, together with increased hypothalamic content of total cholesterol and cholesterol ester. Concordantly, hypothalamic gene expression of SREBP2, HMGCR, and LDLR was significantly up regulated (P<0.05). Also, mRNA abundances of SREBP1, ACAT1 and APO-A1 were up-regulated, while that of CYP46A1 was significantly down-regulated (P<0.05). These changes coincided with a significant down-regulation of BDNF and CRH, and a significant up-regulation of NPY mRNA expression. Moreover, genes involved in methyl transfer cycle were also modulated. DNMT1 and BHMT were up-regulated (P<0.05) at both mRNA and protein levels, which was associated with significant modifications of CpG methylation on the promoter of SREBP-1, SREBP-2 and APO-A1 genes as detected by bisulfate sequencing. These results indicate that feeding betaine to hens modulates hypothalamic expression of genes involved in cholesterol metabolism and brain functions in F1 cockerels with modification of promoter DNA methylation.

Abstract:Eight hundred ISA laying hens of 20 weeks of age were tested to investigate the effects of betaine on contents of serum yolk precursors VLDL(very low-density lipoprotein) and VTG(vitellogenin) for 350 days.Laying hens were randomly allotted to four groups with four replicates each.The groups were fed a corn-soybean basal diet,supplemented with 0,400,600,800 mg·kg~(-1) betaine.It indicated that addition of 600 mg·kg~(-1) betaine significantly improved egg production by 8.67%(P0.05) compared with control.Supplementation of 600 mg·kg~(1) betaine increased concentration of VLDL by 34.61%(P0.05) and 53.41%(P0.03);VTG by 33.74%(P0.01) and 39.14%(P0.01),decreased activity of LPL in abdominal fat by 31.0%(P0.01) and 44.83%(P0.01) in 50 and 70-week old laying hens respectively.It indicates that the contents of serum yolk precursors VLDL and VTG were significantly affected by betaine in laying hen.

Abstract:This review focuses on the osmoregulatory function of betaine and its effect in terms of alleviating heat stress in poultry. Poultry appear to be particularly sensitive to temperature-associated environmental challenges, especially heat stress. High ambient temperatures are deleterious to productive performance in poultry, including broilers, laying hens, quails and turkeys, resulting in considerable economic losses. Heat stress impairs overall poultry production by decreasing feed intake and negatively affecting intestinal development, leading to reduced nutrient digestibility. Apart from inducing a high mortality rate, heat stress is known to depress growth rate and reduce meat yield in broilers. In layers, lower feed intake impairs ovarian function, leading to decreased feed efficiency, egg production and egg quality. In addition, reduced immune functions, such as thyroid activity and antibody production, are evident in poultry exposed to heat stress. Heat stress increases the production of oxidants, causing oxidative stress and lipid peroxidation of cell membranes. Poultry respond physiologically and behaviourally when encountering the negative effects of heat stress, attempting to return the body to homeostasis. This requires energy at the expense of weight gain or egg production. Due to its zwitterionic structure, betaine has osmoprotective properties that aid in protecting intestinal cell proteins and enzymes from environmental stress, including high ambient temperature, thereby counteracting performance losses. Betaine also exerts an osmoregulatory role in cells, regulating water balance, and this results in more stable tissue metabolism. Inclusion of betaine in the diet may be beneficial for alleviating physical reactions to heat stress, as indicated by increases in nutrient digestibility. In broilers, betaine supplementation increases weight gain and breast muscle yield, while improving feed conversion. In layers, betaine supplementation improves egg production, egg quality traits and immune indices. In conclusion, due to its osmoregulatory functions, betaine plays an important role in alleviating heat stress in poultry.

Abstract:Betaine as a feed additive, it has three active methyl groups. As a methyl donor, it regulates the expression of key genes of energy metabolism through epigenetic regulation mechanism, so as to regulate the energy metabolism of the body. On the one hand, it inhibits the aerobic oxidation of glu?cose, promotes gluconeogenesis, and improves the utilization of glucose. At the same time, it is in?volved in the synthesis of carnitine to promote the oxidation of fatty acid and reduce the deposition of abdominal lipid. On the other hand, betaine enhance protein anabolism and improve carcass quality by improving animal energy distribution. In this paper, we summarized that betaine as feed additive, the mechanism of energy metabolism and application in monogastric animals.

Abstract:Betaine is found ubiquitously in plants, animals, microorganisms, and rich dietary sources including seafood, spinach, and wheat bran. The chief physiological role of betaine is to function as a methyl donor and an osmolyte. Betaine also acts as an osmolyte, to maintain the avian’s cellular water and ion balance to improve the avian’s capacity against heat stress via preventing dehydration and osmotic inactivation. It helps in maintaining the protective osmolytic activity, especially in heat-stressed birds. Betaine may promote various intestinal microbes against osmotic variations and thus improve microbial fermentation activity. Previous studies showed that dietary supplementation of betaine in poultry diets could positively affect nutrients’ digestibility, reduce abdominal fat weight, and increase breast meat yield. In addition, betaine has been reported to protect internal organs and boost their performance. Its inclusion in poultry diet is sparing essential amino acids like choline and methionine. In addition, it may play an important role in lean meat production by positively affecting the lipid metabolism with increased fatty acids catabolism and thus reducing carcass fat deposition. The aim of this review article was to broaden the knowledge regarding betaine and its importance in the poultry industry to cope with the heat stress problem. Moreover, it should be added to the diet as a natural anti-stressor through different routes (water/feed) to overcome the heat stress problem. However, further studies need to be conducted at the genetic and molecular basis to elucidate the mechanism behind the betaine as a natural anti-heat agent to decrease the heat stress problem in the poultry industry.

Abstract:1. This study was conducted to investigate the effects of methionine and betaine supplementation on growth performance, carcase composition and lipid metabolism in growing broilers. 2. A total of 450 commercial broilers, 22 d of age, were randomly allocated to three groups, each of which included three replicates (50 birds per replicate). The groups received the same methionine-deficient diet supplemented with 0 or 1 g/kg methionine, or 0.5 g/kg betaine, respectively. 3. Methionine and betaine supplementation significantly improved weight gain and feed conversion. Supplemental methionine and betaine also significantly increased breast muscle yield and decreased abdominal fat content. Meanwhile, addition of methionine and betaine significantly increased the contents of creatine and free carnitine in liver, the activity of hormone-sensitive lipase in abdominal fat and the concentration of free fatty acid in serum, whereas uric acid concentration in serum was significantly decreased. 4. The results of this study suggest that betaine can spare methionine in its function as an essential amino acid and is as effective as methionine in improving performance and carcase quality of growing broilers if the diet is moderately deficient in methionine. The decrease in abdominal fat may be due to the increased carnitine synthesis in liver and hormone-sensitive lipase activity in abdominal fat.
Effect of Betaine on the Regulation of the Lipid Metabolism in Laying Hen

Abstract:The experiment was conducted to investigate the effects of betaine on lipid metabolism of laying hens. 1 600 ISA brown laying hens of 20 weeks of age were alloted randomly into 4 groups each with 4 replicates of 100 layers, the layers were given a corn-soybean basal diet with 0,400,600 and 800 mg/kg betaine respectirely. Results indicated that supplementation of 600 mg/kg betaine significantly improved lay performance, and increased egg production 8.67% (P＜0.04), decreased feed/egg ratio by 9.02% (P＜0.03) compared with control, but there were no significant differences in egg weight and feed intake. It was found that supplementation of 600 mg/kg betaine decreased weights of abdominal fat by 19.63% (P＜0.02) and 22.35% (P＜0.02), and decreased liver fat by 8.52% (P＜0.05) and 16.28% (P＜0.01) in 50 and 70 weeks of age respectively. Compared with control, in 70 weeks of age, activity of LPL in abdominal fat was decreased by 44.83% (P＜0.01), HSL activity in abdominal fat increased by 50.0% (P＜0.02), and MDH activity in liver was decreased by 28.74% (P＜0.03). Supplementation of 600 betaine increased levels of T3 by 43.29% (P＜0.05) and 43.95% (P＜0.02) in 50 and 70 weeks of age respectively, and increased the concentration of cAMP in hypothalamus and anterior pituitary by 59.89% (P＜0.01) and 24.03% (P＜0.05) respectively in 70 weeks of age. The concentration of serum gluclose (GLU), FFA and phospholipid were increased by 44.50% (P＜0.03), 19.66% (P＜0.03) and 7l.66% (P＜0.01) in50weeksofage. The concentration of serum gluclose (GLU), FFA and phospholipid were increased by 16.25% (P＜0.05), 22.51% (P＜0.02) and 22.42% (P＜0.01) in 70 weeks of age.

Abstract:Betaine has been found to alleviate oxidative stress, inflammation, and apoptosis. However, whether dietary betaine can protect late-laying hens against these adverse effects is unknown. Here, 270 65-week-old Jinghong-1 laying hens were randomly divided into the Control, 0.1% Betaine, and 0.5% Betaine groups and fed a basal diet, 0.1%, and 0.5% betaine supplemented diet, respectively. The trial lasted for seven weeks. Birds that consumed 0.5% betaine laid more eggs with thicker eggshells. Accordingly, uterine reduced glutathione (GSH), glutathione peroxidase (GSH-PX), and ovarian superoxide dismutase (SOD) contents were increased. The uterine calcium ion content and the mRNA expression of ovalbumin, ovotransferrin, and carbonic anhydrase two were increased. Moreover, ovarian IL-1β, Caspase-1, Caspase-8, and Caspase-9 mRNA expressions were decreased; luteinising hormone receptor (LHR) and follicle-stimulating hormone receptor mRNA expressions were increased. Furthermore, dietary betaine decreased the ovaries' mRNA expression of DNA methyltransferase 1 (DNMT)1, DNMT3a, and DNMT3b. The methylation level at the promoter region of ovarian LHR decreased. These results indicated that dietary betaine consumption with a concentration of 0.5% could increase the laying rate and the eggshell thickness during the late-laying period. The underlying mechanism may include antioxidative, anti-apoptosis, and hormone-sensitivity-enhancing properties.

Abstract:To investigate the effect of betaine supplementation on growth performance, muscle protein deposition, muscle nucleic acid and amino acid contents, and muscle proteome of broilers, 160 one-day-old male partridge shank broiler chickens were randomly divided into 2 groups with 8 replicates of 10 broilers each. Broilers were fed a basal diet alone, or a basal diet supplemented with 1000 mg/kg betaine. Compared with the control group, the betaine group significantly increased (p < 0.05) the broilers average daily gain, the levels of serum insulin-like growth factor-1 (IGF-1), growth hormone (GH), total protein (TP), the contents of muscle absolute protein deposition, RNA, Ser, Glu, Met, and Phe, and the ratio of RNA/DNA, and decreased (p < 0.05) the feed conversion ratio and serum blood urea nitrogen content. Moreover, proteomic analysis revealed 35 differentially abundant proteins (DAPs) in the betaine group compared with the control group, including 27 upregulated proteins and 8 downregulated proteins (p < 0.05). These DAPs were mainly related to cell differentiation, small molecule metabolic process, and tissue development. In conclusion, diets supplemented with 1000 mg/kg betaine improved growth performance and muscle protein deposition of broilers. Increased serum GH, IGF-1, and TP contents, and alterations in muscle nucleic acids, amino acids, and protein abundance levels were involved in this process.

Abstract:Managing and controlling environmental temperature conditions using practical strategies is crucial to avoid the negative impacts of high environmental temperature, improving poultry production and welfare. High environmental temperature is one of the significant factors challenging poultry production during hot seasons or in tropical areas causing heat stress (HS). The detrimental effects of HS on broilers range from reduced growth performance to impaired poultry meat quality. HS impairs physiological responses caused by alteration in blood parameters, which could lead to impaired product quality by reducing moisture content and altering the production of antioxidant enzymes resulting in increased oxidative stress. There has been a focus on the use of nutritional supplements as a cost effective HS amelioration strategy, such as betaine and polyphenols. Supplementing broiler chicken’s diets with polyphenols aims to enhance growth performance via reduced levels of oxidative stress in tissues under HS conditions. Furthermore, using betaine as an osmolyte aims to protect tissues during osmotic stress conditions. The current review reveals that betaine and polyphenols are essential under crucial conditions such as HS to protect tissues from oxidative damage.
Effects of Graded Levels of Betaine Supplementation on Growth Performance and Intestinal Morphology in Indigenous Young Yellow Feather Broilers

Abstract:This 28-d feeding trial was conducted to evaluate the effects of dietary supplementation with different levels of betaine on growth performance and intestinal morphology in Chinese indigenous young yellow feather broilers. A total of 360 male, one-d-old Huaixiang chickens with an average initial BW of 44.52 +/- 1.07 g were randomly allotted to 4 treatments. Each dietary treatment consisted of 6 replicate cages, with 15 birds per replicate. The 4 dietary treatments were corn-soybean meal-based diets and supplemented with 0, 500, 1000 and 2000 mg/kg betaine, respectively. During d 1-14, with increasing the dietary betaine levels from 0 to 2000 mg/kg, the BWG was improved (quadratic, P<0.05), whereas the FCR was decreased (linear, P<0.01). During d 14-28, dietary supplementation of graded level of betaine had no significant effects on BWG, feed intake and FCR (P>0.05). Overall (d 1-28), there was a quadratic increase in BWG (P<0.05) and a linear decrease in FCR (P<0.01) with increasing betaine supplementation. In addition, dietary supplementation with different levels of betaine increased the villus width (linear, P<0.05), serosal layer thickness, muscular thickness, and wall thickness of duodenum (quadratic, P<0.05), and the serosal layer thickness and wall thickness of jejunum were also improved by inclusion of different level of betaine (cubic, P<0.05). Moreover, dietary supplementation of betaine increased the length of ileum (quadratic, P<0.05). In conclusion, the betaine could be used as growth promoter for Chinese indigenous young yellow feather broilers (Huaixiang chickens), and the betaine also exerted positive effects on intestinal morphology. The suitable recommended supplemental amount is 500 mg/kg.

Abstract:Heat stress has a profound effect on poultry health and productivity. The present study evaluated whether feeding betaine could ameliorate long-term heat stress-induced impairment of productive performance in indigenous yellow-feathered broilers. A total of 240 five-week-old male broilers were randomly allocated to five treatments with six replicates of eight broilers each. The five treatments included a thermoneutral zone control group (TN, fed basal diet), a heat stress control group (HS, fed basal diet), and an HS control group supplemented 500, 1000, 2000 mg/kg betaine, respectively. The TN group was raised at 26 ± 1 °C during the whole study, HS groups exposed to 32 ± 1 °C for 8 h/day from 9:00 am to 17:00 pm. The results showed that heat stress decreased the body weight gain (BWG) and feed intake of broilers during 1–5, 6–10, and 1–10 weeks (p < 0.05). Dietary betaine tended to improve the BWG and feed intake of broilers under 5 weeks of heat stress (linear, p < 0.10), and betaine supplementation linearly increased the BWG and feed intake during 6–10 and 1–10 weeks (p < 0.05). Additionally, nitrogen retention was reduced by 5 weeks and 10 weeks of heat stress (p < 0.05), whereas dietary betaine could improve nitrogen retention in heat stressed broilers after both 5 and 10 weeks of heat stress (linear, p < 0.05). Moreover, this study observed that the trypsin activity of jejunum was decreased by 5 weeks of heat stress (p < 0.05), whereas betaine supplementation had quadratic effects on trypsin activity of jejunum in heat stressed broilers (p < 0.05). Furthermore, 10 weeks of heat stress induced a reduction of villus height of the duodenum and jejunum (p < 0.05), and decreased the villus height to crypt depth ratio of the jejunum (p < 0.05). Supplementation with betaine ameliorated the adverse effects of heat stress on these parameters (p < 0.05). Compared with the TN group, 10 weeks of heat stress reduced carcass and breast yield (p < 0.05) and betaine supplementation improved carcass and breast yield of heat stressed broilers (linear, p < 0.05). In conclusion, dietary supplementation of betaine could reduce the detrimental effects of long-term heat stress on growth performance, digestive function, and carcass traits in indigenous yellow-feathered broilers.
