Abstract:Native chemical ligation (NCL) is a milestone reaction in protein chemical synthesis. The NCL involves a reaction between an unprotected peptide with C-terminal thioester and a second unprotected peptide with N-terminal cysteine, which could give a new peptide in a neutral aqueous buffer solution. The cysteine abundance of natural proteins is only 1.7%, which makes it difficult to find suitable cysteine reaction sites and hinders the widespread application of NCL. The "NCL-desulfurization" strategy extended the ligation site to alanine, inspiring chemists to mediate NCL reactions through different types of beta-thiolated amino acids. After the NCL is completed, native peptide can be obtained through desulfurization reactions. The "NCL-desulfurization" provides more options for protein synthesis. In recent years, several research groups have completed the synthesis of beta-thiolated phenylalanine, leucine, arginine, aspartic acid, asparagine, lysine, methionine, tryptophan, glutamic acid and glutamine, and applied NCL-desulfurization strategy to synthesize proteins. In this article, the synthesis and application of the above beta-thiolated amino acids are reviewed.

Abstract:Phosphonopeptides are important phosphorus analogs of natural peptides and have been widely applied as enzyme inhibitors and antibacterial agents. A series of phosphonodipeptides containing C-terminal α-aminoalkylphosphonic acids was synthesized in satisfactory to good yields conveniently from 2-(N-benzyloxycarbonylamino)alkanamides, aldehydes, and phosphorus trichloride via Mannich-type reaction and subsequent sequential hydrolysis. The reaction mechanism was proposed and verified by (31)P NMR tracing experiments. The current method is an efficient and convenient method for preparation of phosphonopeptides containing C-terminal α-aminoalkylphosphonic acids.

Abstract:N-thiophosphonylamino groups have been greatly overlooked as potentially potent, transition-state analogues, or tetrahedral--intermediate inhibitors of metalloproteases. Alkylthiophosphoramidates derivatives were synthesized y reaction of O-isopropyl phosphorodichloridothioate with an amino acid este followed by hydrolysis in 2M NH 4 OH. The reaction was monitored by 31 P NMR spectroscopy.

Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Abstract:Both sulfonopeptides and phosphonopeptides are important analogs of naturally occurring peptides and have been widely used as enzyme inhibitors and haptens for producing catalytic antibodies due to their tetrahedrally structural features. A series of hybrid sulfonophosphinodipeptides composing of taurines and 1-aminoalkylphosphinic acids were first and conveniently synthesized in satisfactory to good yields via a Mannich-type reaction of N -benzyloxycarbonylaminoalkanesulfonamides, aldehydes, and aryldichlorophosphines, and subsequent hydrolysis. The current method provides an efficient and direct synthesis of hybrid sulfonophosphinodipeptides.

Abstract:Thioamide peptides were synthesized in a straightforward one-pot process via the linkage of diverse natural amino acids in the presence of thiolphosphonate and trichlorosilane, wherein carbonyl groups were replaced with thiono compounds with minimal racemization. Experimental and computational mechanistic studies demonstrated that the trichlorosilane enables the activation of carboxylic acids via intense interactions with the Si-O bond, followed by coupling of the carboxylic acids with thiolphosphonate to obtain the key intermediate S-acyl dithiophosphate. Silyl-activated quadrangular metathesis transition states afforded the thioamide peptides. The potential applications of these thioamide peptides were further highlighted via late-stage linkages of diverse natural products and pharmaceutical drugs and the thioamide moiety.

Abstract:Aryl thiolates have unique reactive, redox, electronic, and spectroscopic properties. A practical approach to synthesize peptides containing thiophenylalanine has been developed via a novel Cu(I)-mediated cross-coupling reaction between thiolacetic acid and iodophenylalanine-containing peptides in the solid phase. This approach is compatible with all canonical proteinogenic functional groups, providing general access to aryl thiolates in peptides. Peptides containing thiophenylalanine (pK(a) 6.4) were readily elaborated to contain methyl, allyl, and nitrobenzyl thioethers, disulfides, sulfoxides, sulfones, or sulfonates.

Abstract:Fully unprotected peptide o-aminoanilides can be efficiently activated by NaNO2 in aqueous solution to furnish peptide thioesters for use in native chemical ligation. This finding enables the convergent synthesis of proteins from readily synthesizable peptide o-aminoanilides as a new type of crypto-thioesters. The practicality of this approach is shown by the synthesis of histone H2B from five peptide segments. Purification or solubilization tags, which are sometimes needed to improve the efficiency of protein chemical synthesis, can be incorporated into the o-aminoanilide moiety, as demonstrated in the preparation of the cyclic protein lactocyclicin Q.

Abstract:Thioamides are an important, but a largely underexplored class of amide bioisostere in peptides. Replacement of oxoamide units with thioamides in peptide therapeutics is a valuable tactic to improve biological activity and resistance to enzymatic hydrolysis. This tactic, however, has been hampered by insufficient methods to introduce thioamide bonds into peptide or protein backbones in a site-specific and stereo-retentive fashion. In this work, we developed an efficient and mild thioacylation method to react nitroalkanes with amines directly in the presence of elemental sulfur and sodium sulfide to form a diverse range of thioamides in high yields. Notably, this convenient method can be employed for the controlled thioamide coupling of multifunctionalized peptides without epimerization of stereocenters, including the late stage thioacylation of advanced compounds of biological and medicinal interest. Experimental interrogation of postulated mechanisms currently supports the intermediacy of thioacyl species.

Abstract:An efficient thiol-assisted one-pot synthesis of peptide/protein C-terminal thioacids was achieved by using peptide/protein hydrazides precursors at neutral pH and room temperature (about 20 degrees C). The transformation from hydrazides to thioacids was shown to be efficient for different C-terminal amino acids and was racemization-free. The in situ formed peptide-thioacids were further used for protein chemical synthesis and site-specific labelling successfully.

Abstract:We developed the first nickel-catalyzed direct ortho-thiolation of N-benzoyl α-amino acid derivatives. This novel strategy showed wide generality, functional tolerance, and high regioselectivity. In addition, dipeptide derivatives were also compatible with this transformation system, providing a potential protocol for the direct modification of peptide derivatives.

This conversion involves activating one of the ester moieties, resulting in a high yield. Its straightforward and comprehensive approach distinguishes the synthesis methodology, exhibiting a significant advancement in peptide analog chemistry. Phosphoryl peptidomimetics represent a novel class of peptide analogs that have garnered attention due to their unique synthetic pathways. Employing a combination of Tf2O and pyridine, the amino acid phosphoryl diester transforms into a phosphorodiamidate compound. Side chain-functionalized depsiphosphonopeptides were also prepared in satisfactory yields directly from one-pot reactions of benzyl carbamate, aldehydes, and diethyl (R,R)-2-chloro-1,3,2-dioxaphospholane-4,5-dicarboxylate.

Abstract:A general and practical cross-dehydrogenative coupling protocol between readily available trisubstituted α,β-dehydro α-amino carboxylic esters and H-phosphites is described. This C(sp<sup>2</sup>)–H phosphorylation reaction proceeds with absolute <i>Z</i>-selectivity promoted by silver salt in a radical relay manner. A Study of Tyrosine O-Phosphorylation via Atherton-Todd Reaction

Abstract:The tyrosine O-phosphorylation via Atherton-Todd reaction, which was a classical reaction for phosphorylation amines, was first studied in this paper. Reaction of Boc-Tyr-OCH3, which was selected as model substrate, with three dialkyl H-phosphonate diester in the presence of CCl4 and triethylamine produced the corresponding tyrosine O-phosphorylation products in a high yield (over 80%). The reaction was conducted in anhydrous THF solution and monitored by P-31 NMR and TLC. Then reaction of Boc-Tyr-OCH3 with a complicated H-phosphonate diester, O-3'-acetylthymidin-5'-yl O-isopropyl H-phosphonate diester, also gave a good result and this would be an alternative approach for the synthesis of nucleotide O-tyrosine (or tyrosine containing peptide) conjugate. Further, the successful O-phosphoration of three tyrosine containing protected dipeptides, Boc-Tyr-Val-OCH3, Boc-Ala-Tyr-OCH3, Boc-Tyr-Tyr-OCH3, with diisopropyl H-phosphonate diester demonstrated the effectiveness of the method.

Abstract:We report a facile synthetic strategy toward CH2-substituted phosphothreonine mimetics. Herein, inexpensive valine with a directing group was converted into homothreonine via palladium-catalyzed gamma-methyl C(sp(3))-H bond activation, followed by construction of a phosphorus-carbon bond via the well-developed Appel reaction and Michaelis-Becker reaction with a total yield of 30%. Furthermore, the derived mimetic was applied for solid-phase synthesis of two phosphopeptide inhibitors. This efficient synthesis provides a chance to prepare not only phosphopeptides but also phosphoproteins resistant to phosphatases.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:A model study of DNA-directed peptide ligation has been developed by transferring fluorescent reporting group from small molecule thioester to a DNA strand (template DNA) in the presence of a thiol-functionalized DNA strand (auxiliary DNA), mimicking the Native Chemical Ligation (NCL) reaction. This DNA-directed transfer shows dependence on the sequence complementarity of the two DNA strands, with in situ generated 4-thiolphenylmethyl functionalized oligonucleotide as the auxiliary DNA strand, under mild basic condition (pH=8.5), and with tris(2-carboxyethyl) phosphine hydrochloride (TCEP) as a reducing agent. Reactions with different amino acid α-thioesters resulted in varied transfer efficiencies from glycine to α-substituted amino acids. This study has provided the basic foundation to use DNA-programmed chemistry toward the chemical synthesis or unnatural modification of protein molecules.
