Abstract:A versatile synthetic route for the preparation of 1,4-diketones from simple starting materials is described and applied to the preparation of a wide range of diketones. The two most important steps in the reaction are the fluoride ion-catalysed Michael addition of a nitroalkane to a vinyl ketone and the subsequent transformation of the nitro ketone to the diketone using supported permanganate as the oxidant. The interaction of F– with nitroethane has been studied and a number of sources of F– have been tested in the Michael addition reaction. Potassium fluoride supported on alumina and potassium permanganate supported on silica gel are remarkably efficient reagents for the Michael addition and Nef transformation stages, respectively, although it is important to determine the best reagent loadings and drying conditions for maximum reaction efficiency to be achieved.

Abstract:Michael addition reaction, known since the 19th century, appeared as a relevant method with a great potential to design new organofluorinated fine chemicals and enlarged their structural diversity. In the Michael addition reaction for the synthesis of organofluorine compounds, the fluorinated blocks can be both used as the Michael donors and acceptors, but the latter is relatively rare. Abstract:Reagent loading and drying conditions have been compared for alkali metal fluride (MF)–alumina supported reagents, by reaction studies and a series of analytical techniques. The reaction studies indicate that whereas a combination of low MF loadings and highly efficient drying procedures give the best reagents for catalytic reactions, somewhat higher loadings and more moderate drying conditions give the best reagents for non-catalytic reactions. The analytical studies enable some rationalisation of the reaction results as well as providing important clues concerning the nature of the active-basic species. The formation of OH– on the reagent surface cannot be entirely responsible for the high basicity of the reagents and it seems that co-ordinately unsaturated F– ions on the reagent surface play an important role in the behaviour of the reagents. High MF loadings and high drying temperatures result in significant structural damage to the reagents with resulting loss in activity.

Abstract:A series of molecular boron, aluminium, and gallium difluoride complexes [{(ArNCMe)2CH}MF2] (M = B, Al, Ga) are reported as catalysts for the defluorofunctionalisation of electron-deficient arenes. Thiodefluorination reactions between TMS–SPh and poly(fluorinated aromatics) proceed with group 13 catalysis under forcing conditions. Evidence is presented for the fluoride entering the catalytic cyclic through a metathesis reaction with TMS–SPh to form metal thiolate intermediates, e.g. [{(ArNCMe)2CH}MF(SPh)], that are then competent nucleophiles for addition to the aromatic substrate, likely through a cSNAr mechanism. Attempts to expand the scope of reactivity to include the hydrodefluorination of electron-deficient arenes with the same group 13 difluoride catalysts met with limited success. Activity could, however, be recovered through addition of NaBArF4 as a catalytic additive (ArF = 3,5-C6H3(CF3)2). NMR titrations suggest that NaBArF4 is capable of coordinating to aluminium and gallium fluoride complexes, most likely through weak M–F---Na interactions (M = Al, Ga) and can play a role in lowering the barrier of metathesis between [{(ArNCMe)2CH}MF2] and Et3SiH to form the group 13 hydrido fluoride [{(ArNCMe)2CH}M(H)F], facilitating catalytic turnover. DFT calculations indicate this weak interaction, leads to a polarization of the M–F bond. The discovery of this additive effect has potentially broad implications in developing new reactivity and applications of thermodynamically stable metal–fluorides.

Abstract:The development of an efficient alternative to the widely employed Balz‐Schiemann deaminative fluorination method (i.e. without using aryl diazonium tetrafluoroborate salt) is a challenging task. Herein, we report a convenient one‐pot method for the FeCl3‐nanocellullose mediated mechanochemical synthesis of fluoroarenes through the selective substitution of an aromatic amino group by fluorine group using pyrylium tetrafluoroborate (Pyry‐BF4) and sodium fluoride (NaF) via in situ formation of pyridinium salt intermediate. The scope of the present protocol includes synthesis of twenty‐eight organofluorine compounds with excellent yields via a selective substitution (SNAr) of an amino group by fluorine. The presented concise methodology opens a pathway to access new chemical spaces for the late‐stage functionalization in pharmaceutical industries.

Abstract:A new organocatalytic asymmetric Michael addition reaction by cleavage of carbon-carbon bonds through a mild release of trifluoroacetate has been developed. The reported method generates the decarboxylated γ-nitro-α-fluorocarbonyl products with excellent enantioselectivies (up to 98% ee) and good diastereoselectivies (up to 20:1 dr).

Abstract:Fluorine's Smooth Introduction Carbon-fluorine bonds are emerging as increasingly versatile constituents of drugs, agrochemicals, and positron emission tomography tracers. Elemental F 2 gas is in principle an efficient reagent for their preparation, but its extreme reactivity requires special handling precautions. Substantial research has therefore focused on promoting selective reactivity of more conveniently handled fluoride ion salts. Liu et al. (p. 1322 ) present a manganese catalyst that transfers fluoride to a range of hydrocarbons in conjunction with a hypervalent iodine-based oxidant. Mechanistic studies implicate a manganese difluoride intermediate that reacts with alkyl radicals generated by a preceding manganese oxo.

Recent advances in the activation of CaF2 via mechanochemical methods have inspired investigation of the fundamental properties of Ca–F bonds in molecular complexes. However, the paucity of well-defined molecular Ca–F-containing complexes undermines systematic understanding of the factors governing nucleophilic fluoride delivery. Here we report the use of a multidentate bis(phenoxide) ligand based on a 1,4,7-triazacyclononane (TACN) scaffold for the formation of well-defined Ca–F complexes, and demonstrate their capabilities in fluoride delivery. Key to this synthesis is an unprecedented desymmetrization step carried out on the TACN ligand within the Ca coordination sphere. A series of stable anionic dinuclear calcium fluoride complexes has then been accessed and characterized by NMR spectroscopy (critically by 19F NMR) and X-ray crystallography. Nucleophilic fluoride delivery can be used to generate C–F, Si–F and S–F bonds, with a notable advance over amide-derived ligand scaffolds being the reduced extent of side reactions derived from competing attack on the electrophile by the less nucleophilic O-donor anionic ligand set.

Abstract:Abstract Diazocarbonyl compounds are useful substrates in di- and trifunctionalization reactions based on F/CF3/SCF3 introduction. In the presented reactions, various electrophilic F/CF3/SCF3-transfer reagents were used. The majority of the reactions were based on rhodium catalysis and the application of various oxygen nucleophiles, such as alcohols, cyclic/acyclic ethers, and carboxylic acids. The oxyfluorination reactions were further developed to provide a new fluorine-18 labeling method. Density functional theory (DFT) modeling studies were performed to get a deeper mechanistic understanding of these reactions. These DFT modeling studies indicated that the catalytic reactions proceed through formation of rhodium carbene and oxonium ylide intermediates. The oxonium ylides undergo tautomerization to enol ether type species that subsequently react with the electrophilic F/CF3/SCF3-transfer reagents. We also present an arylation–trifluoromethylthiolation reaction for simultaneous introduction of C–SCF3 and C–C bonds into diazocarbonyl compounds. Abstract:The incorporation of fluorine atoms within the structure of organic compounds is known to exert a significant impact on their electronic properties, thereby modulating their reactivity in diverse chemical transformations. In the context of our investigation, we observed a striking illustration of this phenomenon. A Michael addition involving gem -difluorovinyl and trifluorovinyl acceptors was successfully achieved, demonstrating high stereoselectivity. This selectivity was further elucidated through theoretical calculations. Using this methodology, a series of new α,β-unsaturated amides, both fluorinated and nonfluorinated, were synthesized. Abstract:Since the days of the “Manhattan Project” our knowledge of the chemistry of fluorine‐con‐taining compounds has increased tremendously. At first only considered out of curiosity, this area of research bordering between inorganic and organic chemistry has developed from modest beginnings to such a complexity of interesting ramifications that it would be very difficult to review all aspects in detail. The motivating force behind the developments in this field were, above all, the unusual properties the numerous compounds acquire on introduction of fluorine substituents. The spectrum of unusual modes of behavior extend from extreme stabilization in fluorine‐containing polymers and blood‐substitutes to drastic increases in reactivity in pharmacologically and phytomedicinally active substances and in dyes. Moreover, one physical peculiarity of the fluorine atom, its magnetic moment, opens a new route to the decyphering of metabolic processes.—Parallel to the ever increasing number of fluorinated compounds there has been a concomitant increase in the variety of methods of synthesis and reagents, which is frequently difficult to overview in entirety, even for the specialists. Older, simple methods such as halogen metathesis with metal fluorides and electrofluorination are being refined as far as higher selectivity is concerned, and completely new, very reactive substances such as hypofluorites and noble‐gas fluorides enable fascinating reactions on complex substrates, e. g. steroids and nucleobases. The present review is an attempted documentation and classification of the methods as well as a critical appraisal, not least from the standpoint of efficiency and economic aspects. The spectrum of changes the fluorine substituents can bring about in organic molecules extends from extreme stabilization (e. g. in polymers and blood‐substitutes) to drastic increases in reactivity (e. g. in Pharmaceuticals).—A selection of fluorinating agents is given below.

Abstract:Activation of trifluoromethyltrimethylsilane by potassium fluoride in N,N-dimethylacetamide provides a powerful source of trifluoromethide which is capable of substituting aromatic nitro and cyano groups under nucleophilic conditions, albeit in low yield. The trifluoromethide generated in this system is also a potent base which leads to a number of interesting side reactions via deprotonation of the substrate.

Abstract:Nucleophilic routes to selectively fluorinated aromatic compounds starting from cheap and readily available nitroaromatics are reviewed. Me3SiCF3, in conjunction with a fluoride, can be used as either a fluorinating or trifluoromethylating agent, and is capable of both the trifluoromethyldenitration and trifluoromethyldecyanation of an aromatic molecule. Nucleophilic trifluoromethylthiolation can be achieved using thiophosgene and a fluoride.
