Abstract:Methylhexamethylenetetramine fluoride dihydrate has been prepared by conventional methods. It is moderately soluble in polar aprotic solvents, and shows good thermal stability, enabling it to be dried. The reagent is capable of converting activated nitroaromatics to the corresponding fluoroaromatics, with a high selectivity to monofluorodenitration being observed with some polysubstituted aromatics.

Abstract:The fluorodenitration of several nitroaromatics with tetramethylammonium salts has been found to be dependent on the basicity of the fluorinating species. Tetramethylammonium fluoride is highly basic and is capable of inducing H-D exchange in 1,3-dinitrobenzene, as well as deprotonating N,N-dimethylacetamide. Reaction of the fluoride with phthalic anhydride forms the bifluoride in situ. Tetramethylammonium bifluoride gives higher yields of the required fluoroaromatic, but slower reaction rates than the corresponding fluoride.

Abstract:Direct nucleophilic radiosynthesis of di-tert-butyl-(4-[18F]fluoro-1,2-phenylene)-dicarbonate was conducted through no-carrier-added electrochemical reactions. 10 mM of triethylamine acetic acid (Et3N·3HAc) in acetonitrile was used to release [18F]fluoride ion trapped in the quaternary ammonium anion exchange resin. The relationship between radiofluorination efficiency, concentration of the Et3N·3HAc, and the working potential were studied with a di-tert-butyl-(4-(tert-butyl)-1,2-phenylene)-dicarbonate substrate in nonaqueous acetonitrile solution. Radiochemical yields of 6.8 ± 1.3 % with specific activity of up to 13 GBq/µmol were achieved with a precursor concentration of 50 mM after a reaction time of 57 min at 0 °C.

Abstract:Activation of trifluoromethyltrimethylsilane by potassium fluoride in N,N-dimethylacetamide provides a powerful source of trifluoromethide which is capable of substituting aromatic nitro and cyano groups under nucleophilic conditions, albeit in low yield. The trifluoromethide generated in this system is also a potent base which leads to a number of interesting side reactions via deprotonation of the substrate.

Abstract:The excellent solubility of quaternary ammonium fluorides makes them attractive alternatives to traditional fluoride ion sources such as potassium fluoride. Unfortunately the high hydroscopicity and low thermal stability of most of these reagents severely restricts their synthetic value. Abstract:Nucleophilic routes to selectively fluorinated aromatic compounds starting from cheap and readily available nitroaromatics are reviewed. Me3SiCF3, in conjunction with a fluoride, can be used as either a fluorinating or trifluoromethylating agent, and is capable of both the trifluoromethyldenitration and trifluoromethyldecyanation of an aromatic molecule. Nucleophilic trifluoromethylthiolation can be achieved using thiophosgene and a fluoride.

Abstract:The development of an efficient alternative to the widely employed Balz‐Schiemann deaminative fluorination method (i.e. without using aryl diazonium tetrafluoroborate salt) is a challenging task. Herein, we report a convenient one‐pot method for the FeCl3‐nanocellullose mediated mechanochemical synthesis of fluoroarenes through the selective substitution of an aromatic amino group by fluorine group using pyrylium tetrafluoroborate (Pyry‐BF4) and sodium fluoride (NaF) via in situ formation of pyridinium salt intermediate. The scope of the present protocol includes synthesis of twenty‐eight organofluorine compounds with excellent yields via a selective substitution (SNAr) of an amino group by fluorine. The presented concise methodology opens a pathway to access new chemical spaces for the late‐stage functionalization in pharmaceutical industries.

Abstract:Abstract Among the many organic fluorine compounds containing nitrogen and sulfur, dialkylaminotrifluorosulfuranes and bis(dialkylamino)difluorosulfuranes have become very useful fluorinating agents. The first representative of this group, dimethylaminosulfur trifluoride was prepared in 1964, followed in 1970 by diethylaminosulfur trifluoride, which became popular under the acronym DAST. Both compounds were synthesized by treatment of the appropriate dialkylaminotrimethylsilane with sulfur tetrafluoride. Subsequently, bis(dialkylamino)sulfur difluorides were prepared by the reaction of dialkylaminosulfur trifluorides with dialkylaminotrimethylsilanes. Both classes of compounds were found to convert alcohols into fluorides, and aldehydes and ketones into geminal difluorides: Further uses include the preparation of acyl and sulfonyl fluorides from carboxylic and sulfonic acids and the conversion of sulfoxides into α‐fluoroalkyl sulfides. Later, the reaction of bis(dialkylamino)difluorosulfuranes with dialkylaminotrimethylsilanes was shown to give tris(dialkylamino)sulfonium difluorotrimethylsilicates (TASF). These salts no longer react with alcohols or carbonyl compounds, but they convert even relatively unreactive halides, such as primary chlorides, into fluorides under very mild conditions. Furthermore, they are readily soluble in organic solvents and are useful sources of “naked,” unsolvated fluoride ion. This chapter deals with the preparation of all three classes of reagents and their use as fluorinating agents. The literature is covered exhaustively until the end of 1984; some papers beyond that date are included.

Abstract:Herein, we disclose that pyridinium salts derived from abundant (hetero)anilines represent a novel precursor class for nucleophilic aromatic substitution reactions with [18F]fluoride. The value of this new 18F-fluorodeamination is demonstrated with the synthesis of over 30 structurally diverse and complex heteroaryl 18F-fluorides, several derived from scaffolds that were yet to be labelled with fluorine-18. The protocol tolerates heteroarenes and functionalities commonly found in drug discovery libraries, and is amenable to scale-up and automation on a commercial radiosynthesiser.

Abstract:Herein, by exploiting different activation modes of fluoroamides, we achieved α- and δ-C(sp3)-H alkylation of nitroalkanes with switchable regioselectivity. Cu catalysis enabled the interception of a distal C-centered radical by a N-centered radical to couple nitroalkanes and unactivated δ-C-H bonds. Abstract:With Et3N·3HF as the fluorinating reagent, a copper(I)- or silver(I)-catalyzed β/α-site-regiocontrolled trans-hydrofluorination of alkynamides has been achieved, affording the corresponding fluoro enamides in moderate to nearly quantitative yields with high regio- and stereoselectivity, respectively. The reaction proceeds under mild reaction conditions and exhibits good functional group tolerance. Further, the deuterium-labeling experiment confirmed the existence of the alkenylcopper intermediate.

Abstract:Amines are a fundamentally important class of biologically active compounds and the ability to manipulate their physicochemical properties through the introduction of fluorine is of paramount importance in medicinal chemistry. Current synthesis methods for the construction of fluorinated amines rely on air and moisture sensitive reagents that require special handling or harsh reductants that limit functionality. Here we report practical, catalyst-free, reductive trifluoroethylation reactions of free amines exhibiting remarkable functional group tolerance. The reactions proceed in conventional glassware without rigorous exclusion of either moisture or oxygen, and use trifluoroacetic acid as a stable and inexpensive fluorine source. The new methods provide access to a wide range of medicinally relevant functionalized tertiary β-fluoroalkylamine cores, either through direct trifluoroethylation of secondary amines or via a three-component coupling of primary amines, aldehydes and trifluoroacetic acid. A reduction of in situ-generated silyl ester species is proposed to account for the reductive selectivity observed.
