Abstract:The synthesis of tetraarylmethanes has long been a challenge in the field of synthetic chemistry. In this study, a series of tetraarylmethanes were successfully synthesized through the formal nucleophilic substitution reaction of indol-2-yl diaryl methanol catalyzed by Brønsted acid. The key success of this study lies in suppressing the influence of water molecules by forming hydrogen bonds with the TFE solvent. This process leads to the formation of active 2-indole imine methide (2-IIM) intermediates, ensuring the successful synthesis of tetraarylmethanes. Furthermore, some of the products also exhibited potential anticancer activity.

Abstract:We describe an alkylation reaction of indoles with sodium alkoxides for the synthesis of bis(indolyl)methanes. 1-Tetralone proved to be an efficient hydrogen acceptor, thus avoiding the use of precious transition metals such as Ru and Ir. This reaction features transition-metal-free conditions, uses readily available starting materials, and can be conducted on a gram scale.

Abstract:A new efficient strategy for the synthesis of 3-trifluoromethyl-indoles was developed through tandem cyclization/trifluoromethylation of 2-alkynylanilines and Umemoto's reagent under effect of CuBr. The reaction features the use of cheap copper salt, mild reaction conditions and high yield of products. The preliminary mechanism study indicates the solvent dimethylacetamide acts as the role of dealkylation.

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:Addition of interelement compounds across alkynes is a straightforward strategy for the synthesis of densely functionalized alkenes, which are versatile units in numerous biologically active compounds. Of these processes the bis-silylation of alkynes is of particular interest, due to the synthetic use of the alkenyl silane reaction products. While cis -bis-silylation of alkynes is well known, trans -bis-silyation of alkynes is relatively underdeveloped. Here, we report the intermolecular trans -bis-silylation of terminal alkynes, using Pd-catalysis and disilane reagent 8-(2-substituted-1,1,2,2-tetramethyldisilanyl)quinoline (TMDQ), to selectively form trans -bis-silylated alkenes. The reaction process was found to be compatible with aryl and alkyl alkynes as well as those bearing electron-withdrawing groups and other alkenyl or alkynyl functionalities. The use of the reaction was displayed through late-stage functionalization of terminal alkynes bearing natural product or drug motifs and onward synthetic transformations of the trans -bis-silylated alkenes reaction products. Experimental and computational mechanistic studies reveal that the reaction probably proceeds through a combined cis -bis-silylation and Z / E isomerization process and that the use of TMDQ as the limiting reagent is key to the desired reactivity.

Abstract:A method for the synthesis of unsymmetrical bis(indolyl)methanes (BIMs) using simple indoles and readily available alcohols as the coupling partners under both heat and 390–400 nm light conditions was developed. Detailed research of the mechanism demonstrated that the diaryliodonium salt‐mediated transformation undergoes a nucleophilic hydroxymethyl radical formation process. Moreover, the diaryliodonium salt‐mediated method is also applicable for synthesizing symmetrical BIMs.

Abstract:A metal‐free and novel thiolation of indole with sulfinyl amides in the presence of tetrabutylammonium iodide (TBAI), affording 3‐indole thioethers in good to excellent yields. A mild, efficient and highly regioselective thiolation of indoles by sulfinyl amides via direct C−H functionalization is described. Sulfinyl amides could be reduced to thioethers and submitted as the thiolation reagents were developed. This reaction provides a useful method for the synthesis of 3‐indole thioethers and 2,3‐dithioindole derivatives. This mix‐and‐go approach also tolerates broad substrate scope and outstanding functional group compatibility. In addition, this preparation could be easily conducted on a gram scale with high efficiency.

Abstract:A method for the synthesis of symmetrical and unsymmetrical 3,3'‐bis(indolyl)methanes (3,3'‐BIMs) has been developed through the direct heteroarylation of indolyl‐containing di(hetero)aryl phosphonium salts with indole derivatives in aqueous media. This metal‐free reaction proceeds smoothly under mild conditions, eliminating the need for hazardous solvents and expensive additives/catalysts typically used in conventional synthesis routes, thereby highlighting the synthetic practicality and efficiency of the process. The success of this approach is largely attributed to the role of water as a promoter, facilitating the C‐P bond functionalization of the phosphonium salts. Furthermore, the reaction demonstrates a broad substrate scope, good functional group tolerance, and excellent scalability. Moreover, the protocol can also be extended to the synthesis of triindolylmethane compound and diarylmethylated indole.

Abstract:Herein, we describe an efficient transition-metal-free regioselective C(3)alkylation of indoles for the synthesis of bis(indolyl)methanes and 3-styryl indoles. Nitrobenzene is employed as the oxidant to oxidize the alcohols in the presence of a strong base and the reaction avoids the use of transition metals such as Ru and Mn. The protocol provides a favorable route to access biologically active compounds such as arundine, vibrindole A, and turbomycin B.

Abstract:We describe an efficient catalyst- and metal-free aminoboration of alkynes to 3-borylated indoles using one of the least expensive boron sources, BCl3. The major dichloro(indolyl)borane products can be used for the in situ construction of useful 3-Bpin indoles. This simple and mild method provides a novel and efficient approach for the synthesis of various 3-borylated indoles with high regioselectivity and a wide substrate scope.

Abstract:Oxone (3.5mol%) was found to be an effective catalyst for the condensation reactions of indoles with aldehydes to afford bis-indolylmethanes in good to excellent yields within 10–30min. The method is simple and economic. The catalyst is found to be recyclable.

Abstract:An efficient method for the highly regio‐ and stereoselective synthesis of 1,2,3‐trisubstituted indanes from diarylmethanols and allylamides through iron(III) chloride hexahydrate‐catalyzed intermolecular [3+2] cycloaddition reaction is reported. A range of 1,2,3‐trisubstituted indanes were prepared in good to excellent yields with excellent stereoselectivities for various combinations of diarylmethanols and allylamides. The choice of solvent is critical to the catalysis. Mechanistic studies indicated that the generation of a benzylic cation is involved in this catalytic process, and the reaction proceeds via the initial formation of diarylmethyl ethers. Abstract:An efficient palladium-catalyzed intramolecular deacetylative dearomatization reaction of 3-acetoxyindoles has been developed. A range of tetracyclic indolin-3-ones bearing C2-quaternary stereocenters are achieved in good yields, showing a wide substrate scope for this reaction. A preliminary enantioselective reaction is established to furnish the product in 63% ee by using (R,R,R)-phosphoramide-PE as a chiral ligand.
