Abstract:For constructing a bifunctional antioxidative enzyme with both superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities, a supramolecular artificial enzyme was successfully constructed by the self-assembly of the Mn(III)meso-tetra[1-(1-adamantyl methyl ketone)-4-pyridyl] porphyrin (MnTPyP-M-Ad) and cyclodextrin-based telluronic acid (2-CD-TeO3H) through host–guest interaction in aqueous solution. The self-assembly of the adamantyl moieties of Mn(III) porphyrin and the β-CD cavities of 2-CD-TeO3H was demonstrated by the NMR spectra. In this supramolecular enzyme model, the Mn(III) porphyrin center acted as an efficient active site of SOD and tellurol moiety endowed GPx activity. The SOD-like activity (IC50) of the new catalyst was found to be 0.116μM and equals to 2.56% of the activity of the native SOD. Besides this, supramolecular enzyme model also showed a high GPx activity, and a remarkable rate enhancement of 27-fold compared to the well-known GPx mimic ebselen was observed. More importantly, the supramolecular artificial enzyme showed good thermal stability.

Abstract:A manganese(II) complex has been prepared with a proline-derived pentadentate ligand (Pro3Py), and it can be converted to a peroxo-manganese(III) complex in the presence of H2O2 and triethylamine. The resulting peroxomanganese(III) complex was well characterised by UV-vis, EPR and ESI-MS techniques, and the geometric structure was discussed based on DFT calculations.

Abstract:An artificial smart bifunctional enzyme with both superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities was successfully constructed by the self-assembly of a porphyrin core with four suspensory adamantyl moieties and beta-cyclodextrin-terminated temperature-sensitive copolymer through host-guest interaction in aqueous solution. The Mn(III) porphyrin, Mn(III) meso-tetra[1-(1-adamantyl methyl ketone)-4-pyridyl] porphyrin (MnTPyP-M-Ad), was designed as both a "supramolecular linker" and an efficient active site of SOD and the temperature-responsive block copolymer (beta-CD-PEG-b-PNIPAAm-Te) with incorporated tellurium moieties in the PNIPAAm chain as GPx active sites was synthesized by ATRP. As a new type of artificial bifunctional enzymes, it exhibited stable SOD-like activity of 0.137 mu M (IC50) at 37 degrees C, and high GPx catalytic efficiency with temperature-responsive dependence characteristic, and the new artificial enzyme was found to exhibit the highest GPx catalytic efficiency (v(0) 8.11 mu M min(-1)) in close to physical temperature.

Abstract:Superoxide dismutase (SOD), glutathione peroxidase (GPX), and catalase (CAT) play crucial roles in balancing the production and decomposition of reactive oxygen species (ROS) in living organisms. These enzymes act cooperatively and synergistically to scavenge ROS. In order to imitate the synergism of these enzymes, we designed and synthesized a novel 32-mer peptide (32P) on the basis of the previous 15-mer peptide with GPX activity and a 17-mer peptide with SOD activity. Upon the selenation and chelation of copper, the 32-mer peptide was converted to a new Se- and Cu-containing 32-mer peptide (Se-Cu-32P) that displayed both SOD and GPX activities, and its kinetics was studied. Moreover, the novel peptide was demonstrated to be able to better protect vero cells from the injury induced by the xanthine oxidase (XOD)/xanthine/Fe2+ damage system than its parents. Thus, this bifunctional enzyme imitated the synergism of SOD and GPX and could be a better candidate of therapeutic medicine.
Tin Porphyrin-Based Nanozymes with Unprecedented Superoxide Dismutase-Mimicking Activities

Abstract:As the oxidative stress is related to human aging and many diseases, a diversity of antioxidant biomimetic enzymes to eliminate reactive oxygen species in vivo and maintain the redox balance has attracted intensive attention. Of particular interest are superoxide dismutase (SOD)-mimicking artificial enzymes that bear inherent characteristics of natural counterparts but overcome their deficiencies in thermal and acidic stability. Inspired by the metallized active center of natural SODs, here, we engineered different groups of metalloporphyrins and found that Sn-metallized porphyrins can act as novel SOD mimics, in which Sn-metallized meso-tetra(4-carboxyphenyl) porphine (Sn-TCPP) can more effectively catalyze the disproportionation of superoxide radical anions (•O<sub>2</sub><sup>-</sup>) into hydrogen peroxide and oxygen. Especially, Sn-TCPP-based metal-organic frame nanozyme (Sn-PCN222) displays an unusually high catalytic activity that remarkably exceeds those of commonly used counterparts. Such unprecedented catalytic behaviors are proposed to depend on the Sn(IV)/Sn(II) transition at the center of Sn-TCPP. In addition, the metal-organic framework (MOF) nanozymes also display higher thermal and acidic stability than natural SODs. Interestingly, we find that Sn-complexed methylated tetra-(4-aminophenyl) porphyrin shows an aggregation-induced SOD activity in an acidic environment, whereas conventional SOD mimics do not function well in this case. Given these unique features, our reported Sn-porphyrin-based nanozymes would be potent alternatives for natural SODs to be widely used in clinical treatments of oxidative stress-related diseases.

Abstract:Superoxide dismutase (SOD), glutathione peroxidase (GPX), glutathione S-transferase (GST) and glutathione reductase (GR) play crucial roles in balancing the production and decomposition of reactive oxygen species (ROS) in living organisms. In order to imitate the synergy of the enzymes, we designed and generated a recombinant protein, which comprises of a Schistosoma japonicum GST (SjGST) and a bifunctional 35-mer peptide with SOD and GPX activities. The engineered protein demonstrated SOD, GPX and GST activities simultaneously. This trifunctional enzyme with SOD, GPX and GST activities is expected to be the best ROS scavenger.

Abstract:The antioxidant enzymes, such as superoxide dismutase, catalase, glutathione peroxidase, and glutathione S-transferase contribute dominatingly to enhance cellular antioxidant defense against oxidative stress. They act cooperatively to scavenge reactive oxygen species, and not one of them can singlehandedly clear all forms of reactive oxygen species. On the basis of the structural understanding for these natural enzymes, many mimics with multifunctional activities had been obtained by chemical synthesis, biosynthesis, and protein fusion techniques. Some of them display remarkable antioxidant cooperative effect in living model which possess potential application in medicine as potent antioxidants. This review summarizes aspects of some multifunctional mimics which have been reported so far. Abstract:A novel hepta-coordinated mononuclear manganese(II) complex, [Mn(L)(2)(H2O)](ClO4)(2)(.)2.5H(2)O [L = 1,2-bis(2'-pyridylmethyleneamino)propane], was obtained by assembly reaction of a di-Schiff base ligand L with manganese(II) perchlorate. The crystal structure shows that one of the L ligand acts as a quadridentate ligand, while the other one serves as a bidentate. The Mn(II) complex was found to have activity of dismutation of superoxide (O-2(-)) in the riboflavin-methioninenitro blue tetrazolium assay.
ZIF-8-derived Hollow Carbon Polyhedra with Highly Accessible Single Mn-N6 Sites As Peroxymonosulfate Activators for Efficient Sulfamethoxazole Degradation

Abstract:The adsorption and protective calcination methods were combined to synthesize single Mn atoms catalyst with more Mn-Nx exposure for peroxymonosulfate (PMS) activation. During the carbonization process, the surface adsorbed Mn species in zeolitic imidazolate framework-8 (ZIF-8) and the surrounding graphitic carbon nitride (gC3N4) jointly maintained the morphology of precursor and enriched Mn-N6 sites near the catalyst surface. The hollow and hierarchically porous structures induced by the erosive function of g-C3N4 further facilitated the exposure of Mn-N6 sites and mass transfer processes. Herein, the derived hollow carbon polyhedron catalyst (CZ8-Mn@g-C3N4) possessed highly accessible edge-host Mn-N6 sites and ultra-high specific surface area (1846.91 m2 g- 1). C-Z8-Mn@g-C3N4 exhibited remarkable catalysis for sulfamethoxazole (SMX) degradation at an extremely low dosage (0.01 g L-1), particularly, up to 3.8 times of promotion was obtained compared to traditional single Mn atoms catalyst (66.9 vs 17.8 L g- 1 min-1). Besides, the catalytic performance of C-Z8Mn@g-C3N4 was not obviously affected by initial pH (3.14-11.00), inorganic anions and natural organic matters (NOMs). Experiments and Density functional theory (DFT) calculations revealed that Mn-N6 sites not only had stronger PMS adsorption capacity than graphitic N, but also enhanced the electron transfer between catalyst and PMS, thus promoting the redox and self-decomposition of PMS to produce SO4 & BULL;-, & BULL;OH and singlet oxygen (1O2) (as major active oxygen species). This work provides new insights into the regulation of active sites exposure in single-atom catalysts for enhanced catalytic performance.
Selenium-mediated Micellar Catalyst: an Efficient Enzyme Model for Glutathione Peroxidase-Like Catalysis.

Abstract:Mimicking the properties of the selenoenzyme glutathione peroxidase (GPx) has inspired great interest. In this report, a selenium-containing micellar catalyst was successfully constructed by the self-assembly of the cationic surfactant hexadecyltrimethylammonium bromide (CTAB) with benzeneseleninic acid (PhSeO2H) through hydrophobic and electrostatic interaction in water. The selenium-containing micellar catalyst demonstrated substrate specificity for both 3-carboxy-4-nitrobenzenethiol (ArSH, 2) and cumene hydroperoxide (CUOOH), and their complexation was confirmed by UV and fluorescence spectra. More importantly, it demonstrated high GPx activity in two assay systems. It is about 126 times more effective than the well-known GPx mimic ebselen in the classical coupled reductase assay system; however, by using hydrophobic substrate ArSH (2) as an alternative of glutathione (GSH, 1), the micellar catalyst exhibited remarkable 500-fold and 94 500-fold rate enhancements compared with that of PhSeO2H and PhSeSePh.
Gene Manipulation Based Selenium-Containing Peptide Exhibiting Synergism of Sod and Gpx

Abstract:In this paper, we constructed a novel bifunctional superoxide dismutase(SOD)/glutathione peroxidase(GPx) mimic, a selenium-, copper-containing 35-mer peptide conjugate(Se-Cu-35P) in which a three-amino acid linker(Gly-Asn-Gly) connects the C-terminus of 17-mer polypeptide SOD mimic with the N-terminus of 15-mer polypeptide GPx mimic. The SOD and GPx activities of Se-Cu-35P are two orders of magnitude lower than those of natural SOD and GPx, respectively. It provides a GPx activity 56-fold higher than Ebselen(a well-known GPx mimic). The glutathione(GSH) binding constant is 5.6×102 L·mol−1. Se-Cu-35P synergistically resists against the inactivation by H2O2 and protects the mitochondria from oxidative damage in a dose dependent manner. These results highlight the challenge of generating an efficient SOD/GPx synergism mimic. It could facilitate the studies of the cooperation of GPx and SOD and could be a potential therapeutic agent for the treatment of ROS-mediated diseases.

Abstract:[N,N′-butylenebis(2-acetylpyridineiminato)] copper(II) diperchlorate, a new model compound of CuZn-SOD (Cu(II)Zn(II)-superoxide dismutase) was synthesized and studied. Its spectral properties and crystal structure showed that it has planar square configuration distorted toward tetrahedron, being very similar to the active center of Cu(II)Zn(II)-SOD. Its activity is 10% of CuZn-SOD and twenty-eight times that of Na2 [Cu(salicylate)2].

Abstract:The enzymes manganese superoxide dismutase and manganese lipoxygenase use MnIII-hydroxo centres to mediate proton-coupled electron transfer (PCET) reactions with substrate. As manganese is earth-abundant and inexpensive, manganese catalysts are of interest for synthetic applications. Recent years have seen exciting reports of enantioselective C-H bond oxidation by Mn catalysts supported by aminopyridyl ligands. Such catalysts offer economic and environmentally-friendly alternatives to conventional reagents and catalysts. Mechanistic studies of synthetic catalysts highlight the role of Mn-oxo motifs in attacking substrate C-H bonds, presumably by a concerted proton-electron transfer (CPET) step. (CPET is a sub-class of PCET, where the proton and electron are transferred in the same step.) Knowledge of geometric and electronic influences for CPET reactions of Mn-hydroxo and Mn-oxo adducts enhances our understanding of biological and synthetic manganese centers and informs the design of new catalysts. In this Feature article, we describe kinetic, spectroscopic, and computational studies of MnIII-hydroxo and MnIV-oxo complexes that provide insight into the basis for the CPET reactivity of these species. Systematic perturbations of the ligand environment around MnIII-hydroxo and MnIV-oxo motifs permit elucidation of structure-activity relationships. For MnIII-hydroxo centers, electron-deficient ligands enhance oxidative reactivity. However, ligand perturbations have competing consequences, as changes in the MnIII/II potential, which represents the electron-transfer component for CPET, is offset by compensating changes in the pKa of the MnII-aqua product, which represents the proton-transfer component for CPET. For MnIV-oxo systems, a multi-state reactivity model inspired the development of significantly more reactive complexes. Weakened equatorial donation to the MnIV-oxo unit results in large rate enhancements for C-H bond oxidation and oxygen-atom transfer reactions. These results demonstrate that the local coordination environment can be rationally changed to enhance reactivity of MnIII-hydroxo and MnIV-oxo adducts.

Abstract:Based on a biomimetic point of view, a new artificial enzyme model was designed to imitate the biocatalytic behavior of native-chloroperoxidase hemoenzyme. Transition metals catalyzed C–H chlorination and represented the C–H activation process. Cysteine–metal complexes of Mn(II), Fe(III), Co(II), Ni(II), Cu(II) and Zn(II) were prepared and encapsulated in a sodium dodecyl sulfate (SDS)/dodecyl trimethyl ammonium bromide (DTAB) vesicular mixture to imitate a native-chloroperoxidase model at pH 3. Current biocatalysts also behaved as a catalase at pH 3 and as a peroxidase at pH 1. Surprisingly, "Cu(II)–cysteine/SDS/DTAB" nanozyme indicated the maximum chloroperoxidase, catalase and peroxidase activity or efficacy among other mentioned intermediate metal complexes including manganese (2+), cobalt (2+), zinc (2+), iron (3+) and nickel (2+). The performed structural analysis by transmission electron microscopy and dynamic light scattering illustrated different nanozyme shapes and surface charges at distinct pHs. Zeta potential of colloids indicated the presence of Cu(II)–cysteine active site inside the nanozyme. Differential scanning calorimetry shows different stabilities and fluidities of nanozyme at different pHs. FTIR spectroscopy was utilized to investigate the structural complexation of Cu(II)–cysteine active essence. The present study demonstrated that "the sulfur" moiety of l-cysteine is ligated to copper ion at pH 1 and 3 in order to form Cu(II)–cysteine center as a multi-functional active site. Accordingly, the current study presents the preparation of an efficient pseudo-vesicular Cu(II)–cysteine supramolecular system with pH-switchable multi-functional enzymatic ability.Graphic abstract

Abstract:A five-co-ordinate manganese(II) complex [Mn(ntb)(Hsal)][ClO4], where ntb is tris(benzimidazol-2-ylmethyl)-amine and Hsal(-) is the anion of salicyclic acid (H(2)sal), has been prepared and characterized by X-ray crystallography. The manganese adopts a distorted trigonal-bipyramidal co-ordination geometry with a N4O ligand donor set and bears structural similarities to the active site of manganese superoxide dismutase. The electrochemical properties of the complex were studied in MeCN by cyclic voltammetry. The complex can catalyse the dismutation of superoxide (O-2(-)) effectively in the riboflavin-methionine-nitro blue tetrazolium assay.

Abstract:Readily exchangeable water molecules are commonly found in the active sites of oxidoreductases, yet the overwhelming majority of studies on small-molecule mimics of these enzymes entirely ignores the contribution of water to the reactivity. Studies of how these enzymes can continue to function in spite of the presence of highly oxidizing species are likewise limited. The mononuclear MnII complex with the potentially hexadentate ligand N-(2-hydroxy-5-methylbenzyl)-N,N',N'-tris(2-pyridinylmethyl)-1,2-ethanediamine (LOH) was previously found to act as both a H2O2-responsive MRI contrast agent and a mimic of superoxide dismutase (SOD). Here, we studied this complex in aqueous solutions at different pH values in order to determine its (i) acid-base equilibria, (ii) coordination equilibria, (iii) substitution lability and operative mechanisms for water exchange, (iv) redox behavior and ability to participate in proton-coupled electron transfer (PCET) reactions, (v) SOD activity and reductive activity toward both oxygen and superoxide, and (vi) mechanism for its transformation into the binuclear MnII complex with (H)OL-LOH and its hydroxylated derivatives. The conclusions drawn from potentiometric titrations, low-temperature mass spectrometry, temperature- and pressure-dependent 17O NMR spectroscopy, electrochemistry, stopped-flow kinetic analyses, and EPR measurements were supported by the structural characterization and quantum chemical analysis of proposed intermediate species. These comprehensive studies enabled us to determine how transiently bound water molecules impact the rate and mechanism of SOD catalysis. Metal-bound water molecules facilitate the PCET necessary for outer-sphere SOD activity. The absence of the water ligand, conversely, enables the inner-sphere reduction of both superoxide and dioxygen. The LOH complex maintains its SOD activity in the presence of •OH and MnIV-oxo species by channeling these oxidants toward the synthesis of a functionally equivalent binuclear MnII species.

Abstract:To strengthen the enzyme-mimic catalytic activity of classical transition metal copper and iron oxide, bimetal Cu-Fe oxide mesoporous nanoparticles (Cu-Fe MNPs) were prepared in one pot under the microwave-assisted conditions. The proposed Cu-Fe MNPs possessed a well-distributed mesoporous structure with an average aperture of 3.91 nm and a high BET specific surface area of 197.93 m2g−1. Its peroxidase-like kinetic parameters (Km/Vmax) to catalyze the redox of TMB and H2O2 were 0.011 mM/3.01×10−8 M·s−1 and 57.44 mM/4.12×10−8 M·s−1, respectively. In particular, either toxic o-aminophenol (o-AP) or bioactive glutathione (GSH) could deteriorate its peroxidase-like property with obvious color changing from blue to colorless for the proposed Cu-Fe MNPs-TMB-H2O2 system. Under the optimized conditions, Cu-Fe MNPs-TMB-H2O2 could selectively response o-AP and GSH with quite low detection limits of 1.57×10−8 mol/L (S/N=3) for o-AP and 3.35×10−8 mol/L (S/N=3) for GSH respectively. When applied for visual monitoring of o-AP and GSH in real aqueous, fruit juice and medicinal samples, the detection recovery (%) is between 96.8~101.4 % for o-AP and 97.1~105.8 % for GSH. The mechanism for synergic-enhancement peroxidase-like activity of Cu-Fe MNP and these for selective recognition of o-AP and GSH were investigated in detail.

Abstract:Many diseases are associated with the overproduction of hydroperoxides that inflict cell damage. A novel cyclodextrin derivative, 6A,6B-diseleninic acid-6A',6B'-selenium bridged beta-cyclodextrin (6-diSeCD), was synthesized to be a functional mimic of glutathione peroxidase (GPX) that normally removes these hydroperoxides. The mimic had high catalytic GPX activity of 13.5 U/micromol, which is 13.6-fold higher than ebselen (PZ51), and was chemically and biologically stable in vitro. Antioxidant activity was studied by ferrous sulfate/ascorbate-induced mitochondria damage model system. These data show that the mimic has great antioxidant activity. Such mimics may result in better clinical therapies for diseases mediated by hydroperoxides.

Abstract:Replacement of the native heme cofactor by manganese protoporphyrin IX (MnPPIX) to reconstitute manganese myoglobin (MnIIIMb) is an important approach to investigate the reactivity of the Mn center inside protein scaffolds. However, unlike the Mn porphyrin synthetic model compounds, MnPPIX reconstituted myoglobins (MnIIIMb) have no reactivity in the epoxidation of styrene using H2O2, which was attributed to the low reactivity of the MnIVO intermediate after homocleavage of the O–O bond in manganese peroxide. To address this issue, we herein chose Oxone® (2KHSO5·KHSO4·K2SO4), a well-known oxidant undergoing O–O bond heterocleavage. After screening 7 mutants and wild-type MnIIIMb, we found that the L29H/F43H mutant could generate a new species ([MnIVO]+˙), tentatively assigned by using UV-vis and EPR spectra, through heterocleavage of the O–O bond. Computational docking showed hydrogen bonds between three distal histidines (H64, L29H and F43H) and anions, which increase the binding affinity to persulfate. With Oxone® as the oxidant, MnIIIMb (L29H/F43H) showed the highest reactivity toward the epoxidation of styrene, different from that with the H2O2 oxidant. This work demonstrates the first example of MnPPIX reconstituted Mb which could catalyze styrene epoxidation and provides new insights to further explore the reactivity of the Mn center in protein scaffolds.
