Abstract:A new chiral manganese complex (C1) bearing a tetradentate nitrogen ligand containing chiral bipyrrolidine and benzimidazole moieties was prepared. The structure of C1 was confirmed by ESI-MS and crystallography. This manganese complex is an active catalyst for the asymmetric epoxidation of various olefins with excellent conversion (up to 99%) and high enantiomeric excess (up to 96% ee) with hydrogen peroxide as the oxidant in the presence of 2-ethylhexanoic acid or acetic acid. Compared with previous structurally similar manganese complexes with different diamine backbones (C2, cyclohexanediamine; C3, diamine from L-proline), C1 showed improved asymmetric induction, especially for simple olefins such as styrene derivatives and substituted chromene. The possible reasons for the improvement of the ee values are discussed in the text on the basis of the crystal structures of the manganese complexes.

Abstract:A series of Mn-complexes with tetradentate N4 ligands, introducing aromatic groups into 2-pyridylmethyl positions of N,N′-dimethyl-N,N′-bis(2-pyridylmethyl)ethane-1,2-diamine (mep), N,N′-dimethyl-N,N′-bis(2-pyridylmethyl)cyclohexane-trans-diamine (mcp), have been synthesized and applied for epoxidation of olefins using H2O2 as the oxidant. The Mn-complexes still possessed an octahedral mononuclear structure in a cis-α topology. These complexes showed good regioselectivity, high yields and turnover frequency (even up to 228,000h−1) with low catalyst loading (0.1–0.01mol%) for epoxidation of a family of olefins (including internal aromatic olefins, internal and terminal aliphatic olefins and diolefins).
Highly Enantioselective Oxidation of Spirocyclic Hydrocarbons by Bioinspired Manganese Catalysts and Hydrogen Peroxide

Abstract:Bioinspired manganese complexes have emerged as attractive catalysts for a number of selective oxidation reactions over the past several decades. In the present study, we report the enantioselective oxidation of spirocyclic compounds with manganese complexes bearing tetradentate N4 ligands as catalysts and aqueous H2O2 as a terminal oxidant under mild conditions; spirocyclic tetralone (1a) and its derivatives bearing electron-donating and -withdrawing substituents are converted to their corresponding chiral spirocyclic beta,beta'-diketones with high yields and enantioselectivities. Spirocyclic indanones are also converted to the beta,beta'-spirobiindanones with high enantioselectivities. Indeed, the reaction expands the diversity of chiral spirocyclic diketones via a late-stage oxidative process. In addition, it is of importance to note that the catalytic reaction can be easily scaled up and the chiral spirocyclic beta,beta'-diketones can be transformed into diol products. In mechanistic studies, we have shown that (1) ketones were yielded as products via the initial formation of alcohols, followed by the further oxidation of the alcohols to ketones, (2) hydrogen atom (H atom) abstraction from the methylene C-H bonds of 1a by a putative Mn(V)-oxo intermediate was proposed to be the rate-determining step, and (3) the C-H bond hydroxylation of 1a by the Mn(V)-oxo species was proposed to occur via oxygen rebound mechanism. On the basis of these results, we have proposed a plausible mechanism for the selective C-H bond oxidation of hydrocarbons by bioinspired manganese catalysts and hydrogen peroxide.
Combined experimental and theoretical study on the reactivity of compounds I and II in horseradish peroxidase biomimetics.

Abstract:For the exploration of the intrinsic reactivity of two key active species in the catalytic cycle of horseradish peroxidase (HRP), Compound I (HRP-I) and Compound II (HRP-II), we generated in situ [Fe(IV) O(TMP(+.) )(2-MeIm)](+) and [Fe(IV) O(TMP)(2-MeIm)](0) (TMP=5,10,15,20-tetramesitylporphyrin; 2-MeIm=2-methylimidazole) as biomimetics for HRP-I and HRP-II, respectively. Their catalytic activities in epoxidation, hydrogen abstraction, and heteroatom oxidation reactions were studied in acetonitrile at -15 °C by utilizing rapid-scan UV/Vis spectroscopy. Comparison of the second-order rate constants measured for the direct reactions of the HRP-I and HRP-II mimics with the selected substrates clearly confirmed the outstanding oxidizing capability of the HRP-I mimic, which is significantly higher than that of HRP-II. The experimental study was supported by computational modeling (DFT calculations) of the oxidation mechanism of the selected substrates with the involvement of quartet and doublet HRP-I mimics ((2,4) Cpd I) and the closed-shell triplet spin HRP-II model ((3) Cpd II) as oxidizing species. The significantly lower activation barriers calculated for the oxidation systems involving (2,4) Cpd I than those found for (3) Cpd II are in line with the much higher oxidizing efficiency of the HRP-I mimic proven in the experimental part of the study. In addition, the DFT calculations show that all three reaction types catalyzed by HRP-I occur on the doublet spin surface in an effectively concerted manner, whereas these reactions may proceed in a stepwise mechanism with the HRP-II mimic as oxidant. However, the high desaturation or oxygen rebound barriers during CH bond activation processes by the HRP-II mimic predict a sufficient lifetime for the substrate radical formed through hydrogen abstraction. Thus, the theoretical calculations suggest that the dissociation of the substrate radical may be a more favorable pathway than desaturation or oxygen rebound processes. Importantly, depending on the electronic nature of the oxidizing species, that is, (2,4) Cpd I or (3) Cpd II, an interesting region-selective conversion phenomenon between sulfoxidation and H-atom abstraction was revealed in the course of the oxidation reaction of dimethylsulfide. The combined experimental and theoretical study on the elucidation of the intrinsic reactivity patterns of the HRP-I and HRP-II mimics provides a valuable tool for evaluating the particular role of the HRP active species in biological systems.
A P450 Harboring Manganese Protoporphyrin IX Generates a Manganese Analogue of Compound I by Activating Dioxygen

Abstract:The self-sufficient cytochrome P450 102A1 (P450BM3), known for its highly efficient activation of molecular oxygen, was reconstituted with manganese protoporphyrin IX. This artificial enzyme (Mn-BM3) was found to catalyze the monooxygenation of various substrates by activating molecular oxygen. Investigation using mechanistic probes (i.e., radical clock substrates) revealed that hydroxylation by Mn-BM3 proceeds via a radical intermediate of the substrate, supporting the involvement of an “H atom abstraction/•OH recombination” mechanism, which is the commonly accepted mechanism for hydroxylation by P450s. A DFT study also indicated that the activation barrier of Mn-porphine in the rebound step is higher than that of Fe-porphine, consistent with the experimental results obtained in the radical clock experiment, wherein Mn-BM3 presents a slower rebound rate than that of heme-bound BM3.

Effect of distal histidines on hydrogen peroxide activation by manganese reconstituted myoglobin.

Abstract:Myoglobins provide an opportunity to investigate the effect of the secondary coordination sphere on the functionality and reactivity of non-native metal porphyrins inside well-defined protein scaffolds. In this work, we reconstituted myoglobin by the replacement of natural heme with manganese(III) protoporphyrin IX and firstly investigated the effect of distal histidine on the reaction of Mn-III porphyrin with H2O2 and one-electron oxidation of ABTS. We have prepared L29H, F43H, H64F, L29H/H64F, F43H/H64F, L29H/F43H and L29H/F43H/H64F mutants and reconstituted apo-myoglobins with manganese(III) protoporphyrin IX. Distal histidine at the 64 position plays an essential role in binding H2O2 through hydrogen bond formation, which facilitates the coordination of H2O2 to the Mn center. The second histidine at the 43 position is important in the cleavage of the O-O bond and to form the highly valent Mn(IV)-oxo intermediate. His29 has less efficiency to activate H2O2, because it is too far from the Mn center. The cooperative effect of dual distal histidines at positions 64 and 43 on the activation of H2O2 was observed and the F43H Mn(III)Mb mutant exhibited 5-fold and 10-fold reaction rate increases in the activation of H2O2 and one-electron oxidation of ABTS versus wild-type Mn(III)Mb. This is different from the distal histidine effect on the H2O2 activation by heme in Mb. This work will provide new insights to understand the fundamental chemistry of manganese in oxidation, and further construct biomimetic Mn models for peroxidase, inside or outside of protein scaffolds.
A Balancing Act: Stability versus Reactivity of Mn(O) Complexes

Abstract:A large class of heme and non-heme metalloenzymes utilize O2 or its derivatives (e.g., H2O2) to generate high-valent metal-oxo intermediates for performing challenging and selective oxidations. Due to their reactive nature, these intermediates are often short-lived and very difficult to characterize. Synthetic chemists have sought to prepare analogous metal-oxo complexes with ligands that impart enough stability to allow for their characterization and an examination of their inherent reactivity. The challenge in designing these molecules is to achieve a balance between their stability, which should allow for their in situ characterization or isolation, and their reactivity, in which they can still participate in interesting chemical transformations. This Account focuses on our recent efforts to generate and stabilize high-valent manganese-oxo porphyrinoid complexes and tune their reactivity in the oxidation of organic substrates. Dioxygen can be used to generate a high-valent Mn(V)(O) corrolazine (Mn(V)(O)(TBP8Cz)) by irradiation of Mn(III)(TBP8Cz) with visible light in the presence of a C-H substrate. Quantitative formation of the Mn(V)(O) complex occurs with concomitant selective hydroxylation of the benzylic substrate hexamethylbenzene. Addition of a strong H(+) donor converted this light/O2/substrate reaction from a stoichiometric to a catalytic process with modest turnovers. The addition of H(+) likely activates a transient Mn(V)(O) complex to achieve turnover, whereas in the absence of H(+), the Mn(V)(O) complex is an unreactive "dead-end" complex. Addition of anionic donors to the Mn(V)(O) complex also leads to enhanced reactivity, with a large increase in the rate of two-electron oxygen atom transfer (OAT) to thioether substrates. Spectroscopic characterization (Mn K-edge X-ray absorption and resonance Raman spectroscopies) revealed that the anionic donors (X(-)) bind to the Mn(V) ion to form six-coordinate [Mn(V)(O)(X)](-) complexes. An unusual "V-shaped" Hammett plot for the oxidation of para-substituted thioanisole derivatives suggested that six-coordinate [Mn(V)(O)(X)](-) complexes can act as both electrophiles and nucleophiles, depending on the nature of the substrate. Oxidation of the Mn(V)(O) corrolazine resulted in the in situ generation of a Mn(V)(O) π-radical cation complex, [Mn(V)(O)(TBP8Cz(•+))](+), which exhibited more than a 100-fold rate increase in the oxidation of thioethers. The addition of Lewis acids (LA; Zn(II), B(C6F5)3) to the closed-shell, diamagnetic Mn(V)(O)(TBP8Cz) stabilized a paramagnetic valence tautomer Mn(IV)(O)(TBP8Cz(•+))(LA), which was characterized as a second π-radical cation complex by NMR, EPR, UV-vis, and high resolution cold spray ionization MS. The Mn(IV)(O)(TBP8Cz(•+))(LA) complexes are able to abstract H(•) from phenols and exhibit a rate enhancement of up to ∼100-fold over the parent Mn(V)(O) valence tautomer. In contrast, a large decrease in rate is observed for OAT for the Mn(IV)(O)(TBP8Cz(•+))(LA) complexes. The rate enhancement for hydrogen atom transfer (HAT) may derive from the higher redox potential for the π-radical cation complex, while the large rate decrease seen for OAT may come from a decrease in electrophilicity for an Mn(IV)(O) versus Mn(V)(O) complex.

Abstract:A high-valent manganese(IV)-hydroxo porphyrin pi-cation radical complex, [Mn(IV)(OH)(Porp+center dot)(X)]+, was synthesized and characterized spectroscopically. The Mn porphyrin intermediate was highly reactive in alkane hydroxylation and oxygen atom transfer reactions. More importantly, the Mn porphyrin intermediate reacted with water at a fast rate, resulting in the dioxygen evolution. To the best of our knowledge, we report the first manganese Cpd I model compound bearing a porphyrin pi cation radical ligand with a high reactivity in oxidation reactions, including water oxidation.
Concerted proton-electron transfer reactions of manganese-hydroxo and manganese-oxo complexes

Abstract:The enzymes manganese superoxide dismutase and manganese lipoxygenase use MnIII-hydroxo centres to mediate proton-coupled electron transfer (PCET) reactions with substrate. As manganese is earth-abundant and inexpensive, manganese catalysts are of interest for synthetic applications. Recent years have seen exciting reports of enantioselective C-H bond oxidation by Mn catalysts supported by aminopyridyl ligands. Such catalysts offer economic and environmentally-friendly alternatives to conventional reagents and catalysts. Mechanistic studies of synthetic catalysts highlight the role of Mn-oxo motifs in attacking substrate C-H bonds, presumably by a concerted proton-electron transfer (CPET) step. (CPET is a sub-class of PCET, where the proton and electron are transferred in the same step.) Knowledge of geometric and electronic influences for CPET reactions of Mn-hydroxo and Mn-oxo adducts enhances our understanding of biological and synthetic manganese centers and informs the design of new catalysts. In this Feature article, we describe kinetic, spectroscopic, and computational studies of MnIII-hydroxo and MnIV-oxo complexes that provide insight into the basis for the CPET reactivity of these species. Systematic perturbations of the ligand environment around MnIII-hydroxo and MnIV-oxo motifs permit elucidation of structure-activity relationships. For MnIII-hydroxo centers, electron-deficient ligands enhance oxidative reactivity. However, ligand perturbations have competing consequences, as changes in the MnIII/II potential, which represents the electron-transfer component for CPET, is offset by compensating changes in the pKa of the MnII-aqua product, which represents the proton-transfer component for CPET. For MnIV-oxo systems, a multi-state reactivity model inspired the development of significantly more reactive complexes. Weakened equatorial donation to the MnIV-oxo unit results in large rate enhancements for C-H bond oxidation and oxygen-atom transfer reactions. These results demonstrate that the local coordination environment can be rationally changed to enhance reactivity of MnIII-hydroxo and MnIV-oxo adducts.
Asymmetric Epoxidation of Olefins Catalyzed by Substituted Aminobenzimidazole Manganese Complexes Derived from L ‐Proline

Abstract:A family of manganese complexes [Mn(Rpeb)(OTf)2] (peb=1‐(1‐ethyl‐1H‐benzo[d]imidazol‐2‐yl)‐N‐((1‐((1‐ethyl‐1H‐benzo[d]imidazol‐2‐yl)methyl) pyrrolidin‐2‐yl)methyl)‐N‐methylmethanamine)) derived from L‐proline has been synthesized and characterized, where R refers to the group at the diamine backbone. X‐ray crystallographic analyses indicate that all the manganese complexes [Mn(Rpeb)(OTf)2] exhibit cis‐α topology. These types of complexes are shown to catalyze the asymmetric epoxidation of olefins employing H2O2 as a terminal oxidant with up to 96% ee. Obviously, the R group of the diamine backbone can influence the catalytic activity and enantioselectivity in the asymmetric epoxidation of olefins. In particular, Mn(i‐Prpeb)(OTf)2 bearing an isopropyl arm, cannot catalyze the epoxidation reaction with H2O2 as the oxidant. However, when PhI(OAc)2 is used as the oxidant instead, all the manganese complexes including Mn(i‐Prpeb)(OTf)2 can promote the epoxidation reactions efficiently. Taken together, these results indicate that isopropyl substitution on the Rpeb ligand inhibits the formation of active Mn(V)‐oxo species in the H2O2/carboxylic acid system via an acid‐assisted pathway.

Abstract:The coexistence of aliovalent cations may facilitate the formation of electronic and ionic defects, altering the catalytic properties of manganese oxide. In this paper, a series of mixed-valent manganese oxides (MnOx) were prepared by a hydrothermal route and tested for heterogeneous activation of peroxydisulfate for Orange II degradation in aqueous solution. The morphology, crystal structure and surface chemistry of the MnOx were adjusted by the addition of ethanol in the hydrothermal process. The physicochemical properties were characterized and evidence from X-ray diffraction (XRD) and high-resolution transmission electron microscopy (HRTEM) indicated that MnOx exhibited a mixed crystal phase structure of Mn2O3, Mn3O4 and MnO2. The catalytic properties of the three samples prepared with 5%, 10%, and 20% volume fractions of ethanol (denoted as MnOx-5, MnOx-10, and MnOx-20) were found to be related to the valent states of Mn, and MnOx-10 exhibited a better performance with a mole ratio of Mn4+/ Mn2+ close to 1.0. The electrochemical characterization demonstrated that MnOx-10 performed a more effective redox process which may determine the catalytic activity of MnOx. Moreover, quenching tests, electron paramagnetic resonance (EPR) and gas chromatography-mass spectrometry (GC-MS) were used to evaluate the possible activation processes of peroxydisulfate (PDS) and the intermediates of Orange II degradation. This catalyst preparation strategy provides a new idea for the preparation of high-efficiency environmental catalysts.
Mn(II) Oxidation by the Multicopper Oxidase Complex Mnx: A Binuclear Activation Mechanism

Abstract:The bacterial protein complex Mnx contains a multicopper oxidase (MCO) MnxG that, unusually, catalyzes the two-electron oxidation of Mn(II) to MnO2 biomineral, via a Mn(III) intermediate. Although Mn(III)/Mn(II) and Mn(IV)/Mn(III) reduction potentials are expected to be high, we find a low reduction potential, 0.38 V (vs Normal Hydrogen Electrode, pH 7.8), for the MnxG type 1 Cu2+, the electron acceptor. Indeed the type 1 Cu2+ is not reduced by Mn(II) in the absence of molecular oxygen, indicating that substrate oxidation requires an activation step. We have investigated the enzyme mechanism via electronic absorption spectroscopy, using chemometric analysis to separate enzyme-catalyzed MnO2 formation from MnO2 nanoparticle aging. The nanoparticle aging time course is characteristic of nucleation and particle growth; rates for these processes followed expected dependencies on Mn(II) concentration and temperature, but exhibited different pH optima. The enzymatic time course is sigmoidal, signaling an activation step, prior to turnover. The Mn(II) concentration and pH dependence of a preceding lag phase indicates weak Mn(II) binding. The activation step is enabled by a pKa > 8.6 deprotonation, which is assigned to Mn(II)-bound H2O; it induces a conformation change (consistent with a high activation energy, 106 kJ/mol) that increases Mn(II) affinity. Mnx activation is proposed to decrease the Mn(III/II) reduction potential below that of type 1 Cu(II/I) by formation of a hydroxide-bridged binuclear complex, Mn(II)(μ-OH)Mn(II), at the substrate site. Turnover is found to depend cooperatively on two Mn(II) and is enabled by a pKa 7.6 double deprotonation. It is proposed that turnover produces a Mn(III)(μ-OH)2Mn(III) intermediate that proceeds to the enzyme product, likely Mn(IV)(μ-O)2Mn(IV) or an oligomer, which subsequently nucleates MnO2 nanoparticles. We conclude that Mnx exploits manganese polynuclear chemistry in order to facilitate an otherwise difficult oxidation reaction, as well as biomineralization. The mechanism of the Mn(III/IV) conversion step is elucidated in an accompanying paper .
Spectroscopic Observation of Iodosylarene Metalloporphyrin Adducts and Manganese(v)-Oxo Porphyrin Species in a Cytochrome P450 Analogue

Abstract:Different metalloporphyrin model compounds have been synthesized to study the mechanisms of cytochrome P450s with various terminal oxidants, and numerous intermediates have been reported. However, the detailed mechanism of the oxygen atom transfer from iodosylarene to the substrates remains unclear. Here we report the direct ultraviolet-visible spectroscopic observation of the soluble iodosylarene-manganese porphyrin adduct following catalytic oxidation using 2,4,6-tri-tert-butylphenol as the reductant. When the reductant is changed to cisstilbene, the rate-determining step also changes. Both the iodosylarene-manganese porphyrin adduct and [(porphyrin) Mn(V) = O] species may be simultaneously observed. In the absence of reductant, the adduct of iodosylarene with sterically hindered [Mn(meso-tetrakis(2,6-dichlorophenyl)porphinato)Cl] is immediately formed, and smoothly converted into a high-valent [(porpyrinato)Mn = O]. Electrospray ionization mass spectrometry analysis of the reaction further confirms the transformation between these species. This study provides an insight into the mechanism of oxygen transfer within the haem-containing enzymatic systems.

Abstract:Although manganese(II) is known to have no role in peroxymonosulfate (PMS) activation, through a series of sulfamethoxazole (SMX) oxidation experiments, we found that the addition of pyridine organic ligands can improve the catalytic activity and accelerate SMX oxidation. For the organic ligands to be effective: the stability constant of the Mn(III) complex should be higher than that of the Mn(II) complex. A positive correlation was observed between the SMX oxidation rate and Mn(II) concentration, and the maximum PMS utilization efficiency was achieved. Many shreds of evidence verified that neither center dot SO4- nor center dot OH was associated with SMX oxidation. The enhanced effect of phenanthroline on the Mn(II)/PMS system was attributed to the highly oxidative intermediate manganese species (Mn(V)), originating from the two-electron transfer reaction of complexed Mn(III) and PMS. Notably, the main oxidizing species did not change (eta-(PMSO2) similar to 100%) regardless of the initial PMSO concentration or pH value. Additionally, the analysis of SMX degradation products revealed that the oxygen transfer oxidation pathway was dominant in the Mn(II)/phenanthroline/PMS system, while the N radical coupling pathway also contributed significantly to SMX oxidation. This work offers new insights into the formation of high-valent manganese species and provides a potential strategy for applying low-concentration Mn(II) to wastewater treatment. Manganese porphyrin redox state in endothelial cells: Resonance Raman studies and implications for antioxidant protection towards peroxynitrite

Abstract:Cationic manganese(III) ortho N-substituted pyridylporphyrins (MnP) act as efficient antioxidants catalyzing superoxide dismutation and accelerating peroxynitrite reduction. Importantly, MnP can reach mitochondria offering protection against reactive species in different animal models of disease. Although an LC-MS/MS-based method for MnP quantitation and subcellular distribution has been reported, a direct method capable of evaluating both the uptake and the redox state of MnP in living cells has not yet been developed. In the present work we applied resonance Raman (RR) spectroscopy to analyze the intracellular accumulation of two potent MnP-based lipophilic SOD mimics, MnTnBuOE-2-PyP5+ and MnTnHex-2-PyP5+ within endothelial cells. RR experiments with isolated mitochondria revealed that the reduction of Mn(III)P was affected by inhibitors of the electron transport chain, supporting the action of MnP as efficient redox active compounds in mitochondria. Indeed, RR spectra confirmed that MnP added in the Mn(III) state can be incorporated into the cells, readily reduced by intracellular components to the Mn(II) state and oxidized by peroxynitrite. To assess the combined impact of reactivity and bioavailability, we studied the kinetics of Mn(III)TnBuOE-2-PyP5+ with peroxynitrite and evaluated the cytoprotective capacity of MnP by exposing the endothelial cells to nitro-oxidative stress induced by peroxynitrite. We observed a preservation of normal mitochondrial function, attenuation of cell damage and prevention of apoptotic cell death. These data introduce a novel application of RR spectroscopy for the direct detection of MnP and their redox states inside living cells, and helps to rationalize their antioxidant capacity in biological systems.
Enhancing Mn(Ii)-Binding and Manganese Peroxidase Activity in A Designed Cytochrome C Peroxidase Through Fine-Tuning Secondary-Sphere Interactions

Abstract:Noncovalent second-shell interactions are important in controlling metal-binding affinity and activity in metalloenzymes, but fine-tuning these interactions in designed metalloenzymes has not been fully explored. As a result, most designed metalloenzymes have low metal-binding affinity and activity. Here we identified three mutations in the second coordination shell of an engineered Mn(II)-binding site in cytochrome c peroxidase (called MnCcP.1, containing Glu45, Glu37, and Glu181 ligands) that mimics the native manganese peroxidase (MnP), and explored their effects on both Mn(II)-binding affinity and MnP activity. First, removing a hydrogen bond to Glu45 through Tyr36Phe mutation enhanced Mn(II)-binding affinity, as evidenced by a 2.8-fold decrease in the KM of Mn(II) oxidation. Second, introducing a salt bridge through Lys179Arg mutation improved Glu35 and Glu181 coordination to Mn(II), decreasing KM 2.6-fold. Third, eliminating a steric clash that prevented Glu37 from orienting toward Mn(II) resulted in an 8.6-fold increase in kcat/KM, arising primarily from a 3.6-fold decrease in KM, with a KM value comparable to that of the native enzyme (0.28 mM vs 0.19 mM for Pleurotus eryngii MnP PS3). We further demonstrated that while the effects of Tyr36Phe and Lys179Arg mutations are additive, because involved in secondary-shell interactions to different ligands, other combinations of mutations were antagonistic because they act on different aspects of the Mn(II) coordination at the same residues. Finally, we showed that these MnCcP variants are functional models of MnP that mimic its activity in both Mn(II) oxidation and degradation of a phenolic lignin model compound and kraft lignin. In addition to achieving KM in a designed protein that is similar to the that of native enzyme, our results offer molecular insight into the role of noncovalent interactions around metal-binding sites for improving metal binding and overall activity; such insight can be applied to rationally enhance these properties in other metalloenzymes and their models.

Abstract:The dual role of manganese oxides in peroxymonosulfate (PMS) activation for the degradation of organic contaminants makes the mechanism investigation complicated. Herein, a ( −1 1 1) crystal plane exposed δ-MnO 2 (SN-KMO) was synthesized to decipher the debate between direct oxidation process (DOP) and electron transfer process (ETP) mechanisms. Generally, 5 mg/L bisphenol A (BPA) could be efficiently removed in 15 min regardless of in the presence of high (0.5 mM) or low (0.05 mM) PMS concentration. Mechanisms analysis showed that nonradical pathways of DOP and ETP simultaneously contributed to the BPA degradation without the participation of radicals oxidation. Yet, contributions of DOP and ETP at certain PMS dosages differed significantly. Specifically, at high PMS concentration of 0.5 mM, DOP played the dominant role, and the overly protonated manganese active sites could bond with O-O bridged species derived from two PMS molecules to form outer-sphere complexes. Comparatively, at low PMS concentration of 0.05 mM, BPA oxidization was primarily dominated by ETP. The specific ETP was highly dependent on solution pH, and it only existed in the presence of high PMS concentration. The significance of this work lies in synthesizing a ( −1 1 1) crystal plane exposed δ-MnO 2 for efficient BPA degradation with low PMS dosage (0.05 mM) and discovering the unreported outer-sphere complexes for understanding the PMS activation mechanisms by manganese oxides.

Abstract:Bioinspired manganese complexes have emerged over recent decades as attractive catalysts for a number of selective oxidation reactions. However, these catalysts still suffer from oxidative degradation. In the present study, we prepared a series of porous Mn–N4 catalysts in which the catalytic units are embedded in the skeleton of porous organic polymers (POPs). These POP-based manganese catalysts demonstrated high reactivity in the oxidation of aliphatic C–H bonds and the asymmetric epoxidation of olefins. Furthermore, these catalysts could be readily recycled and reused due to their heterogeneous nature. Morphological characterization revealed that the Mn–N4 complex was individually distributed over a porous polymer network. Remarkably, the nature of the single-site catalyst prevented oxidative degradation during the reaction. Noncovalent Modulation Of Ph-Dependent Reactivity Of A Mn-Salen Cofactor In Myoglobin With Hydrogen Peroxide

Abstract:To demonstrate protein modulation of metal-cofactor reactivity through noncovalent interactions, pH-dependent sulfoxidation and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) oxidation reactivity of a designed myoglobin (Mb) containing non-native Mn-salen complex (1) was investigated using H2O2 as the oxidant. Incorporation of 1 inside the Mb resulted in an increase in the turnover numbers through exclusion of water from the metal complex and prevention of Mn-salen dimer formation. Interestingly, the presence of protein in itself is not enough to confer the increase activity as mutation of the distal His64 in Mb to Phe to remove hydrogen-bonding interactions resulted in no increase in the turnover numbers, while mutation His64 to Arg, another residue with ability to hydrogen-bond interactions, resulted in an increase in reactivity. These results strongly suggest that the distal ligand His64, through its hydrogen-bonding interaction, plays important roles in enhancing and fine-tuning reactivity of the Mn-salen complex. Nonlinear least-squares fitting of rate versus pH plots demonstrates that 1.Mb(H64X) (X = H, R and F) and the control Mn-salen 1 exhibit pK(a) values varying from pH 6.4 to 8.3. and that the lower pK(a) of the distal ligand in 1.Mb(H64X), the higher the reactivity it achieves. Moreover, in addition to the pK(a) at high pH, 1.Mb displays another pK(a) at low pH, with pK(a) of 5.0 +/- 0.08. A comparison of the effect of different pH on sulfoxidation and ABTS oxidation indicates that, while the intermediate produced at low pH conditions Could only perform sulfoxidation. the intermediate at high pH Could oxidize both sulfoxides and ABTS. Abstract:The presence of a nonheme metal, such as copper and iron, in the heme-copper oxidase (HCO) superfamily is critical to the enzymatic activity of reducing O-2 to H2O, but the exact mechanism the nonheme metal ion uses to confer and fine-tune the activity remains to be understood. We herein report that manganese and cobalt can bind to the same nonheme site and confer HCO activity in a heme nonheme biosynthetic model in myoglobin. While the initial rates of 02 reduction by the Mn, Fe, and Co derivatives are similar, the percentages of reactive oxygen species (ROS) formation are 7%, 4%, and 1% and the total turnovers are 5.1 +/- 1.1, 13.4 +/- 0.7, and 82.5 +/- 2.5, respectively. These results correlate with the trends of nonheme-metal-binding dissociation constants (35, 22, and 9 mu M) closely, suggesting that tighter metal binding can prevent ROS release from the active site, lessen damage to the protein, and produce higher total turnover numbers. Detailed spectroscopic, electrochemical, and computational studies found no evidence of redox cycling of manganese or cobalt in the enzymatic reactions and suggest that structural and electronic effects related to the presence of different nonheme metals lead to the observed differences in reactivity. This study of the roles of nonheme metal ions beyond the Cu and Fe found in native enzymes has provided deeper insights into nature's choice of metal ion and reaction mechanism and allows for finer control of the enzymatic activity, which is a basis for the design of efficient catalysts for the oxygen reduction reaction in fuel cells.
