Molecular detection of lymph-node micrometastases in cervical cancer

Abstract:Objective:To investigate expression and significance of telomerase activity and human papillomavirus(HPV) in pelvic lymph nodes of human cervical carcinoma.Methods:HPV and telomerase in sixty-one cervical carcinomatous lymph nodes were examined by FQ-PCR and PCR-ELISA,respectively.Benign lymph nodes from fifteen patients with benign disease were utilized as negative control.Relation of lymph node micrometastasis and cliniopathological factors was analyzed by statistical method.Results:Positive rate of HPV16/18 and telomerase in lymph nodes of cervical carcinoma were all more than routine H-E staining(P<0.01),while these molecules were negative in the control group.The difference was statistically significant.Copies of HPV and telomerase activity in lymph nodes involved were higher than that in lymph nodes without nonmetastasis,while less than that in primary tumor.There was direct correlation of telomerase and HPV in nodes of cervical cancer(r=0.60,P<0.01).Lymph nodes micrometastasis was correlated with clinical stage and depth of muscular invasion.Conclusions:Compared with traditionary pathology method,detection of HPV and telomerase is available method for lymph node micrometastasis examination in cervical carcinoma.
