Abstract:Obesity and its major co-morbidity, type 2 diabetes, have been an alarming epidemic prevalence without an effective treatment available. Sterol regulatory element-binding proteins (SREBPs) are major transcription factors regulating the expression of genes involved in biosynthesis of cholesterol, fatty acid and triglyceride. Therefore, inhibition of SREBP pathway may be a useful strategy to treat obesity with type 2 diabetes. Here, we identify a small molecule, Ganoderic Acid A (GAA), inhibits the SREBP expression and decreases the cellular levels of cholesterol and fatty acid in vitro. GAA also ameliorates body weight gain and fat accumulation in liver or adipose tissues, and improves serum lipid levels and insulin sensitivity in high fat diet (HFD)-induced obese mice. Consistently, GAA regulates SREBPs target genes and metabolism associated genes in liver or adipose tissues, which may directly contribute to the lower lipid level and improvement of insulin resistance. Taken together, GAA could be a potential leading compound for development of drugs for the prevention of obesity and insulin resistance.
Ximenynic Acid Regulation of N-3 PUFA Content in Liver and Brain

Abstract:Background/Aims: Ximenynic acid is a rare conjugated enyne fatty acid found primarily in plants in the Santalaceae family. It has been reported that sandalwood seed oil (SWSO) affects fatty acid metabolism in animal studies; however, the effects of pure ximenynic acid remain unclear. The present study aimed to study the impact of SWSO and ximenynic acid on n-3 fatty acid metabolism in the liver and brain. Methods: Thirty C57BL/6 male mice aged 4 weeks were fed SWSO (1.0 mL/20 g bodyweight), olive oil (OO), or a combination of SWSO and OO (n = 10/group) for 8 weeks. Liver and brain fatty acid compositions were determined using gas chromatography. HepG2 cells were treated with up to 150 μM ximenynic acid and oleic acid for 48–72 h. The expression and abundance of genes and proteins relevant to n-3 fatty acid metabolism pathways were investigated. Results: The intake of SWSO in mice elevated the levels of total n-3 fatty acids and decreased total n-9 fatty acids in the liver (p < 0.05) compared with the OO group. In contrast, total n-3 fatty acids were significantly decreased in the brain (p < 0.05). HepG2 cells treated with ximenynic acid for 48 h showed significant reductions in n-9 fatty acids and docosahexaenoic acid (C22:6n-3) (p < 0.05) compared with HepG2 cells treated with oleic acid. In HepG2 cells, stearoyl-CoA desaturase (SCD) and fatty acid desaturase 2 (FADS2) gene expression, as well as FADS2 protein expression, were significantly down-regulated after a 72-h incubation with 150 μM of ximenynic acid compared with the vehicle (p < 0.05). Conclusion: Ximenynic acid may regulate fatty acid metabolism by suppressing the expression of key enzymes of lipid metabolism. In contrast, SWSO, which has a high level of C18:3n-3, positively affected n-3 fatty acid synthesis in mouse liver compared to pure ximenynic acid. We hypothesize that a high level of precursor C18:3n-3 in SWSO promotes the endogenous synthesis of C22:6n-3 despite the presence of ximenynic acid.

Abstract:Hormone sensitive lipase is a key enzyme in fat metabolism.It catalyzes the release of fatty acids and glycerin from trigly ceride in order to meet the need of animal.Its structure consists of three regions:regulatory,catalytic and lipid binding.The regulatory site can regulate the hormone sensitive lipase activity.The small variation on the activity of hormone sensitive lipase can influence the lean percentage and backfat traits.The concentration of dietary energy,ratio of protein to amino acids and dietary lipids are all influence the hormone sensitive lipase activity.
Leptin limits hepatic lipid accumulation and inflammation via vagal activation of the JAK2-STAT3/AMPK pathway

Abstract:Background: Non-alcoholic fatty liver disease (NAFLD) begins with hepatic lipid accumulation, and leptin has anti-steatosis properties. In this study, we investigated the effects of leptin on hepatic steatosis and inflammation through the vagal pathway independently of the inhibitory effect of food intake. Methods: Male Sprague-Dawley rats were matched for food intake after the high-fat diet (HFD)-induced obesity model and were injected intraperitoneally with leptin or leptin + lidocaine for 6 weeks. Control rats received equal volumes of saline. Adipose tissue mass, NAFLD activity scores (NAS), hepatic inflammatory factors, hepatic triglyceride content and hepatic lipid metabolism-related protein levels were evaluated. Results: Leptin ameliorated HFD-induced hepatic lipid accumulation, improved NAS, and decreased tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and monocyte chemotactic protein-1 (MCP-1) levels in the presence of matched intake. Lidocaine decreased the phosphorylation of signal transducer and activator of transcription 3 (p-STAT3) expression in the nucleus tractus solitarius (NTS) and abrogated the leptin-mediated improvement. Leptin increased hypothalamic phosphorylated Janus kinase 2 (p-JAK2) and p-STAT3 expression, as well as the expression of mitochondrial respiratory chain-related genes. Leptin also increased hepatic phosphorylated adenosine 5'-monophosphate-activated protein kinase (p-AMPK) expression and phosphorylation of its downstream target acetyl Co A carboxylase 1 (ACC1), reducing de novo lipogenesis. Conclusions: Our results suggest that leptin ameliorated hepatic lipid accumulation and inflammation by activating the JAK2-STAT3/AMPK pathway through the vagal pathway independently of the inhibitory effect of ingestion. Leptin has the potential to be a drug for early NAFLD treatment.
Effects and Mechanisms of GLP-1 on Fatty Acid Metabolism in Insulin-Resistant 3T3-L1 Adipocytes

Abstract:AIM To explore the effects and mechanisms of glucagon like peptide-1(GLP-1) on fatty acid metabolism.METHODS Through ELISA and Western blot,the present study was to investigate the effects and mechanisms of GLP-1 on fatty acid metabolism in insulin-resistant 3T3-L1 adipocytes which were induced by high glucose and insulin.RESULTS ELISA showed that GLP-1 regulated medium free fatty acid(FFA) concentrations in a insulin dependent way: FFA content rised when GLP-1 combined with insulin;GLP-1 decreased FFA content without insulin.Similarly,GLP-1 increased fatty acid synthase(FAS) content with insulin;when there was no insulin in medium,FAS could not be affected by GLP-1.Western blot revealed that GLP-1 could enhance protein kinase B(PKB) phosphorylation.When treated with PKB phosphorylation inhibitor LY294002 or Wortmannin with insulin could inhibit the increased effect of GLP-1 on FFA content.GLP-1 had no effect on PKB phosphorylation when there was no insulin existence,but could diminish hormone-sensitive lipase(HSL) concentration in this case.CONCLUSION GLP-1 can enhance insulin sensitivity and decrease HSL concentration in insulin resistant 3T3-L1 adipocytes.Insulin affects GLP-1 regulation of fatty acid metabolism in resistance 3T3-L1 adipocytes.
Synthesis and biological activity of novel barbituric and thiobarbituric acid derivatives against non-alcoholic fatty liver disease

Abstract:Forty-four barbituric acid or thiobarbituric acid derivatives were synthesized and evaluated for their effects on adipogenesis of 3T3-L1 adipocytes by measuring the expression of adiponectin in vitro. Four compounds (3a, 3o, 3s, 4t) were found to increase the expression of adiponectin and lower the leptin level in 3T3-L1 adipocytes at respective concentration of 10 μM. Among them, 3s showed the most efficacious. Oral administration of 3s effectively reduced body weight, liver weight, and visceral fat and regulated serum levels of biochemical markers in the high-fat/diet-induced Wistar rats. Histopathological evaluation of liver sections by Oil Red O and H&E staining confirmed 3s as a potent, orally active molecule for reducing fat deposition against non-alcoholic fatty liver disease.

Abstract:Along with the increasing prevalence of obesity worldwide, the deleterious effects of high-calorie diet are gradually recognized through more and more epidemiological studies. However, the concealed and chronic causality whitewashes its unhealthy character. Given an ingenious mechanism orchestrates the metabolic adaptation to high-fat high-fructose (HFF) diet and connive its lipotoxicity, in this study, an experimental rat/mouse model of obesity was induced and a comparative transcriptomic analysis was performed to probe the mystery. Our results demonstrated that HFF diet consumption altered the transcriptomic pattern as well as different high-calorie diet fed rat/mouse manifested distinct hepatic transcriptome. Validation with RT-qPCR and Western blotting confirmed that SREBP1-FASN involved in de novo lipogenesis partly mediated metabolic self-adaption. Moreover, hepatic ACSL1-CPT1A-CPT2 pathway involved in fatty acids β-oxidation, played a key role in the metabolic adaption to HFF. Collectively, our findings enrich the knowledge of the chronic adaptation mechanisms and also shed light on future investigations. Meanwhile, our results also suggest that efforts to restore the fatty acids metabolic fate could be a promising avenue to fight against obesity and associated steatosis and insulin resistance challenged by HFF diet.

Abstract:Objective: To investigate the role of mitogen activated protein kinase phosphatase-1 (MKP-1) in regula-tion of transcription activity of farnesoid X receptor. Methods: Expression levels of FXR and MKP-1 mRNA and protein in liver specimens of obese mice were measured by real-time quantitative PCR and Western blot, respectively. The effect of MKP-1 on regulating transcription activity of FXR was determined by co-immunoprecipitaion and luciferase reporter as-says. Results: FXR expression was down-regulated and MKP-1 expression was up-regulated in liver of obese mice (P<0.05). The protein-protein interaction of MKP-1 and FXR was confirmed by co-immunoprecipitation and luciferase reporter assays. Overexpression of MKP-1 could inhibit FXR transcription activity, while MKP-1 silencing could promote tran-scription activity of FXR. Conclusions: MKP-1 might be a co-repressor for FXR and play a role in the regulation of hep-atic glucose and lipid metabolism.
Antiobesity Effect of Flaxseed Polysaccharide via Inducing Satiety due to Leptin Resistance Removal and Promoting Lipid Metabolism through the AMP-Activated Protein Kinase (AMPK) Signaling Pathway

Abstract:Obesity is a metabolic syndrome worldwide that causes many chronic diseases. Recently, we found an antiobesity effect of flaxseed polysaccharide (FP), but the mechanism remains to be elucidated. In this study, rats were first induced to develop obesity by being fed a high-fat diet. The obese rats were then fed a control diet, AIN-93M (group HFD), or a 10% FP diet (group FPD). The body weight, body fat, adipose tissue and liver sections, serous total triglycerides, levels of fasting blood glucose in serum, serous insulin, inflammatory cytokines in serum, and serous proteins within the leptin-neuropeptide Y (NPY) and AMP-activated protein kinase (AMPK) signaling pathway were determined and analyzed. FP intervention significantly reduced body weight and abdominal fat from 530 ± 16 g and 2.15% ± 0.30% in group HFD to 478 ± 10 g and 1.38% ± 0.48% in group FPD, respectively. This effect was achieved by removing leptin resistance possibly by inhibiting inflammation and recovering satiety through the significant downregulation of NPY and the upregulation of glucagon-like peptide 1. Adiponectin was then significantly upregulated probably via the gut-brain axis and further activated the AMPK signaling pathway to improve lipid metabolism including the improvement of lipolysis and fatty acid oxidation and the suppression of lipogenesis. This is the first report of the proposed antiobesity mechanism of FP, thereby providing a comprehensive understanding of nonstarch polysaccharides and obesity.

Abstract:Introduction: High concentrations of nonesterified fatty acids (NEFA) is the key of characteristic of fatty liver in dairy cows. Therefore, the aim of this study was to investigate the effect of high concentration of NEFA on lipid metabolism in hepatocytes through the lipidomic approach and molecular biology techniques. Methods: Stimulate AML-12 cells with different concentrations of NEFA, observe the cellular lipid accumulation, and select 0.6 mM NEFA stimulation concentration for subsequent experiments. Collect cells for lipidomics analysis. Results: High concentration of NEFA (0.6–2.4 mM) significantly reduced the cell viability in a concentration-dependent manner, indicating that high concentrations of NEFA have lipotoxicity on hepatocytes. In addition, NEFA promoted triglycerides (TAG) accumulation, increased the mRNA expression of the lipogenic molecules SREBP1c and FASN, and decreased the mRNA expression of lipolytic molecules CPT1A and HSL in hepatocytes. Mechanistically, high concentration of NEFA induced lipid metabolism disorders in hepatocytes by regulating metabolic pathways such as glycerol phospholipid metabolism, glycosyl phosphatidylinositol anchored biosynthesis, triglyceride metabolism, sphingolipid metabolism, and inositol phosphate metabolism. Discussion: High concentration of NEFA is lipotoxic to cells, promoting lipid accumulation. LPE (18:2), LPE (18:3), LPE (18:1) via glycerophospholipid metabolism, glycosylphosphatidylinositol (GPI)-anchor biosynthesis, glycerolipid metabolism, sphingolipid metabolism, and inositol phosphate metabolism, indicating their potential regulation role in the pathogenesis of fatty liver.

Abstract:Despite its wide distribution in the central nervous system, the presence of Neuropeptide Y (NPY) in peripheral tissues has been detected. White adipose tissue (WAT) is a new site of NPY synthesis and secretion. The development of brown-like adipocytes in WAT is controlled by hypothalamic NPY neurons through interaction with sympathetic nervous system (SNS). However, whether peripheral NPY has a direct effect on induction of the Uncoupling protein1 (UCP1)-positive adipocytes is unknown. We have used adipocytes derived from C3H10T1/2 stem cells as a model of brown-like adipocyte, and investigated the role of NPY in their differentiation and activation. In general, NPY had no effect on brown adipogenesis of C3H10T1/2 stem cell, but suppressed db-cAMP activation of brown-like adipocytes, which was due to blunting brown fat-relevant gene expression and mitochondrial function. NPY showed suppression in a receptor-dependent manner, inhibition of endogenous cAMP production and cAMP-PKA-dependent pathways p38 MAPK and CREB phosphorylation were involved in the downstream mechanisms. A novel role of NPY in the peripheral is presented, which may help decrease energy expenditure in WAT of obese subjects.
Effect of GLP-1 receptor agonist on lipolysis in adipose tissue of obese mice and its underlying mechanism

Abstract:AIM:To investigate the effects of glucagon-like peptide-1 (GLP-1) receptor agonist exendin-4 on white adipose tissue (WAT) and the underlying mechanisms.METHODS:Male C57BL/6J mice (8 weeks) were chal-lenged by high-fat diet for 12 weeks, and were randomly divided into saline group and exendin-4 group.The mRNA expres-sion of sirtuin 1 (SIRT1), adipose triglyceride lipase (ATGL), TNF-αand adiponectin of WAT was detected by real-time PCR.3T3-L1 adipocytes or mouse embryonic fibroblasts cells were treated with exendin-4 for 24 h.The protein levels of SIRT1, ATGL and hormone-sensitive lipase (HSL) were determined by Western blot.RESULTS:Exendin-4 significantly decreased epididymal fat weight, fasting blood glucose and serum triglyceride levels ( P<0.05) , and reduced body weight and serum TNF-αlevel.The mRNA expression of SIRT1, ATGL and adiponectin in WAT was all significantly up-regulated by exendin-4, which were contrary to the down-regulation of TNF-αmRNA expression (P<0.05).Exendin-4 promoted the protein expression of SIRT1, ATGL, and HSL in 3T3-L1 adipocytes in a dose-dependent manner.Less lipid droplets with up-regulation of lipolytic protein expression were observed when combined with SIRT1 agonist treatment, which were suppressed by SIRT1 inhibitor.Deletion of SIRT1 led to larger adipocytes with more lipid droplets, and the effect of ex-endin-4 on the lipolysis disappeared when SIRT1 was deficient.CONCLUSION:Exendin-4 promotes lipolysis in WAT of obese mice via activation of SIRT1.
[Effects of Medium-Chain Fatty Acid on Weight Gain and Lipids Metabolism in Obese Rats].

Abstract:OBJECTIVE:Some researchers considered medium chain fatty acids a group of potential agents in the prevention of obesity for their particular metabolism characters.METHODS:In this study, the authors investigated the effects of medium chain fatty acids on body weight gains, food intake, adipose depots and serum lipid profiles. Healthy male Sprague-Dawley rats were fed for 38 days with over-fat diets and oils containing MCFA at different level.RESULTS:Significant decreases of body weight [( 295.2 +/- 29.7) g vs. (324.6 +/- 9.1) g] (P < 0.05), the weight of adipose pads [(5.30 +/- 1.57) g vs. (7.20 +/- 1.74) g](P < 0.05), the level of leptin in serum[(1.61 +/- 0.39) ng/ml vs. (2.04 +/- 0.46) ng/ml] (P < 0.05), the sizes of fat cells but increase of HDL-C [(0.73 +/- 0.12) mmol/L vs. (0.60 +/- 0.06)mmol/L] (P < 0.05) were noted in the group fed MCFA at 0.975 g/kg bw per day as compared with those of the group fed soybean oil.CONCLUSION:The results suggested the role of MCFA in prevention of obesity and there is no adverse effect on lipid metabolism if the proportion of MCFA to total fat scouce is mild.
Apigenin, a Modulator of PPARγ, Attenuates HFD-induced NAFLD by Regulating Hepatocyte Lipid Metabolism and Oxidative Stress Via Nrf2 Activation

Abstract:Lipid metabolic disorders and oxidative stress in the liver are key steps in the progression of nonalcoholic fatty liver disease (NAFLD), which is a major risk factor for the development of metabolic syndrome. To date, no pharmacological treatment for this condition has been approved. Our previous study has found that the food-derived compound apigenin (Api) significantly attenuates obesity-induced metabolic syndrome by acting as a peroxisome proliferator-activated receptor gamma modulator (PPARM). Herein, a high fat diet (HFD) induced NAFLD model was used to dig out whether Api had the effect on NAFLD. The results showed that Api had obvious effect in restraining NAFLD progression, including attenuating HFD induced lipid accumulation and oxidative stress in vivo. As a PPARM, although Api did significantly inhibit the expression of PPARγ target genes encoding the protein associated with lipid metabolism, it had no obvious activating effect on PPARγ. Interestingly, we found that Api promoted Nrf2 into the nucleus, thereby markedly activating Nrf2 to inhibit the lipid metabolism related genes and increase the oxidative stress related genes. Further Nrf2 knockdown/knockout and overexpression experiments showed that Api regulating PPARγ target genes was dependent on Nrf2 activation and the activation of Nrf2 counteracted the activation effect of PPARγ by Api. Importantly, we also found that Api might bind with Nrf2 via auto dock and ITC assay. Therefore, our results indicate that Api ameliorates NAFLD by a novel regulating mode of Nrf2 and PPARγ in inhibiting lipid metabolism and oxidative stress abnormity.

Abstract:Objective To investigate the effects of fish oil diet on the expression of leptin in fat tissue,and the levels of serum leptin and insulin in high-fat diet induced obese rats.Methods After fed a high-fat diet(lard,20/100 WT) for 8 w,the obese rats,30g heavier than controls,were separated and divided into two groups: lard group and fish oil group.The lard group was continuously fed high fat diet,while the fish oil group fed the same diet except that fish oil substituted for lard.After feeding for another 4 w,the blood samples were drawn for analysis.After the rats were killed,the retroperitoneal and epididymal fat masses were carefully collected and weighed.The levels of serum insulin and leptin were measured by RIA.The adipose tissue samples were fixed in 10% buffered formalin and embedded in paraffin.The tissue was sectioned(10μm) and stained with hemotoxylin and eosin,and adipocyte diameter was determined using a microscope equipped with a graduated ocular micrometer.Total RNA in retroperitoneal fat tissue was extracted and the level of leptin mRNA was evaluated by RT-PCR.Results(1) Compared to lard diet,fish diet significantly reduced weight gain,retroperitoneal and epididymal fat masses.A declining trend was also shown in the levels of leptin mRNA,serum leptin and serum insulin,but without significant difference.(2) The level of serum leptin in lard group and fish oil group was significantly related to the level of serum insulin.Conclusion Fish oil feeding for 4 w ameliorates obesity,but it does not decrease the levels of leptin expression,serum leptin and insulin significantly.[ACTA NUTRIMENTA SINICA,2013,35(2):146-149]
Different Expression of Fatty Acid Synthase (FAS) in Adipose Tissue from High-Fat Diet Induced Obesity-Prone and Obesity-Resistant Rats

Abstract:Objectives To explore the different expression of fatty acid synthase (FAS) in white adipose tissue (WAT) of obesity- prone (OP) and obesity- resistant (OR) rats on a high- fat diet. Methods After one week of free access to a standard diet,12 rats were selected randomly as standard control group and were fed on standard diet,the 60 remaining rats were separated on the basis of one week body weight percentage gained to OP or OR group,and were being fed on high- fat diet for another 6 weeks. The body weight, Lee's index,the wet weight of epitesticle,retroperitoneal and subcutaneous fat pad were measured after sacrificing the rat. The expression of FAS mRNA in WAT was detected by RT- PCR technique and the expression of FAS protein was determined by Western blotting. Results In OP rats,the speed of body weight gain,the Lee's index and the wet weight of fat pad at epitesticle,retroperitoneal and subcutaneous sites were higher than those in OR rats (P 0.05=. There were higher levels of FAS mRNA expression in OP rats than in OR rats. The OP rats showed higher expression of FAS protein in WAT than OR rats. Conclusions Different expression of FAS in WAT existing between high- fat induced obesity- prone and obesity- resistant rats suggest that FAS may be involved in the susceptibility to obesity.

Abstract:Abstract Hyperlipidemia poses a serious threat to human health and evaluating the ability of natural active substances to regulate disorders of lipid metabolism is the focus of food functionality research in recent years. Bioactive peptides are distinguished by their broad range of sources, high nutritional content, ease of absorption and use by the body, and ease of determining their sequences. Bioactive peptides have a wide range of potential applications in the area of medicines and food. The regulation of lipid metabolism disorder caused by bioactive peptides from different sources provides a reference for the development and research of bioactive peptides for lipid reduction. Abstract:Histologic spectrum studies in patients revealed fatty acid binding proteins 1 (FABP1) as a potential new target for the treatment of metabolic associated fatty liver disease. However, there is no FABP1 inhibitor has been reported except the first-in-class FABP1 inhibitor bearing acid moiety reported by our laboratory. Herein, we firstly report the structure-activity relationship of novel non-carboxylic acid FABP1 inhibitors, which resulted in the identification of the potent and selective FABP1 inhibitor 30. The IC50 value of compound 30 for subtype FABP4 in the same family was greater than 80 μM. Moreover, compound 30 significantly alleviated the hepatic steatosis in DIO mice, which is equivalent to that of clinical drug obeticholic acid. This study might be provided a promising probe for the development of FABP1 inhibitors and thus can help to further elucidate the pharmacology of FABP1.
