Abstract:Understanding the effects of genotype, environment and their interactions on rice quality is of great importance for rice breeding and cultivation. In this study, six rice varieties with two indica, two japonica and two indica–japonica types of rice were selected and planted at ten locations in Zhejiang Province to investigate the genotype (G) × environment (E) on physicochemical and sensory properties and the differences of volatile organic compounds (VOCs) among the three types of rice. Analysis of variances showed that apparent amylose content (AC), total protein content (PC), alkali spreading value (ASV), RVA profiles, and appearance (ACR), palatability (PCR), and sensory evaluation value (SEV) of cooked rice and texture of cooled cooked rice (TCCR) were mainly affected by genotypic variation, whereas the smell of cooked rice (SCR) was mainly affected by environment (p < 0.05). The G × E effect was significant for most parameters. The weather in the middle and late periods of filling had important effects on the formation of rice quality, especially on setback (SB) and pasting temperature (PT) (p < 0.01). They were negatively correlated with the texture of cooked rice (TCR) and SEV (p < 0.05). Peak viscosity (PV) and breakdown (BD) were positively related to the sensory evaluation parameters (p < 0.01) and could be used to predict cooked rice quality. A total of 59 VOCs were detected, and indica, japonica and indica–japonica had 9, 6 and 19 characteristic compounds, respectively. The principal component analysis showed that the physicochemical and sensory properties and VOCs of indica–japonica rice were more stable than those of indica and japonica rice at ten locations in Zhejiang Province. It is helpful for rice breeders to understand how the environment affects the physicochemical, sensory properties and VOCs of the three rice types, and it is also important for food enterprises to provide rice products with stable quality.

Abstract:High-quality rice industry plays an important role in the sustainable development of local economy and the poverty alleviation of border ethnic groups in Yunnan.Aromatic rice'Diantun 502'is the main variety of high-quality fragrant and soft rice production, and it is also the main aroma rice resource to improve the variety of fragrant and soft rice.However, the inheritance of'Diantun 502'aromatic trait has been still unclear so far.In this study, the fragrant gene badh2 specific primer and the grain chewing method were used to analyze the genetic regularity of fragrant trait in'Diantun 502'and the phenotypic differences of its hybrid offspring.The results indicated that the fragrant trait of'Diantun 502'was controlled by the recessive nuclear badh2 gene, and its genetic separation ratio was not affected by difference of either cytoplasmic background or planting environment.In the analysis ofplant andalthough there were no obvious agronomic traits related to aroma traits, the number of spikes, total spikes, effective spikes and seed setting rates of single plant with aroma traits were lower than those without fragrance.There was no significant correlation between aroma and others agronomic traits, but fertile grains per panicle, total grains per panicle, effective panicles per plant and seed-setting rate were lower than those no-fragrance plants inIn particular, there were lower fertile per panicle, seed-setting rate and low yield for fragrant strains inThe results would provide theoretical basis for the breed improvement and purification and rejuvenation of'Diantun 502'variety.

Abstract:Fragrance is a valuable trait in rice varieties, with its aroma significantly influencing consumer preference. In this study, we conducted comprehensive metabolome and transcriptome analyses to elucidate the genetic and biochemical basis of fragrance in the Shangsixiangnuo (SSXN) variety, a fragrant indica rice cultivated in Guangxi, China. Through sensory evaluation and genetic analysis, we confirmed SSXN as strongly fragrant, with an 806 bp deletion in the BADH2 gene associated with fragrance production. In the metabolome analysis, a total of 238, 233, 105 and 60 metabolic compounds exhibited significant changes at the seedling (S), reproductive (R), filling (F), and maturation (M) stages, respectively. We identified four compounds that exhibited significant changes in SSXN across all four development stages. Our analyses revealed a significant upregulation of 2-acetyl-1-pyrroline (2AP), the well-studied aromatic compound, in SSXN compared to the non-fragrant variety. Additionally, correlation analysis identified several metabolites strongly associated with 2AP, including ethanone, 1-(1H-pyrrol-2-yl)-, 1H-pyrrole, and pyrrole. Furthermore, Weighted Gene Co-expression Network Analysis (WGCNA) analysis highlighted the magenta and yellow modules as particularly enriched in aroma-related metabolites, providing insights into the complex aromatic compounds underlying the fragrance of rice. In the transcriptome analysis, a total of 5582, 5506, 4965, and 4599 differential expressed genes (DEGs) were identified across the four developmental stages, with a notable enrichment of the common pathway amino sugar and nucleotide sugar metabolism in all stages. In our correlation analysis between metabolome and transcriptome data, the top three connected metabolites, phenol-, 3-amino-, and 2AP, along with ethanone, 1-(1H-pyrrol-2-yl)-, exhibited strong associations with transcripts, highlighting their potential roles in fragrance biosynthesis. Additionally, the downregulated expression of the P4H4 gene, encoding a procollagen-proline dioxygenase that specifically targets proline, in SSXN suggests its involvement in proline metabolism and potentially in aroma formation pathways. Overall, our study provides comprehensive insights into the genetic and biochemical mechanisms underlying fragrance production in rice, laying the foundation for further research aimed at enhancing fragrance quality in rice breeding programs.

Abstract:Eating-quality is one of the most important qualities for rice. With the development and application of molecular biology techniques, studies of its genetic and molecular bases have made great progresses. This paper summarizes these progresses from the following aspects: the relation between starch component and structure and cooked rice texture, gene location and QTL analysis, the key enzymes involved in the starch synthesis and corresponding gene expression and regulation on starch quality. The possible approaches to further research are discussed.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Rice aroma was a comprehensive result of numerous volatiles and human sense. More than 300 volatiles were reported in rice. Rice aroma was widely researched by using sensory evaluation, gas chromatography (GC) method, and electronic nose (E-nose). Genetic factor was the main cause for rice aroma. However, the same rice variety might result in different aroma quality due to different planting, processing and storage.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Scope and aproach</h3><p>The main purpose of this review is to elaborate the main volatiles contributing rice aroma, methods used for rice aroma evaluation and factors affecting rice aroma. Most volatiles with odor descriptions, odor thresholds and retention indices were summarized. Sensory evaluation, GC method, and E-nose were elaborated with respect to rice aroma analysis. The effects of gene, planting, processing and storage on rice sample were pointed out.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Key findings and conclusions</h3><p>2-Acetyl-1-pyrroline (2-AP), aldehydes, heterocyclics, alcohols play important roles in rice aroma quality. GC-O-MS which combines the advantages of sensory descriptive analysis and GC-MS, can not only realize detailed sensory analysis of odor quality but also realize quantitative and qualitative detection of volatiles, playing an important role in exploring key aroma-active compounds. Besides of traditional electronic sensors, mass spectrometry and colorimetric sensors were used for E-nose, making it more novel and applicable. Water washing, high hydrostatic pressure, roasting and parboiling were thought as effective processing to improve rice flavor. Degree of milling and storage significantly affected rice flavor.</p>

The comparative test between the research results in China and abroad show the major flavor DNA is the same in the Chinese and overseas, yet some special flavors in China probably contains different DNA. Many Chinese researchers believe that there are some minor genes and it is hard to account for the flavor diversity with one coupe of recessive genes.

Abstract:The improvement of the grain quality of the hybrid rice is one of the difficult and hotspot problems in rice breeding for a long time.The theory and technology of aromatic hybrid rice was proposed by Hunan Hybrid Rice Research Center in 1989,and the first male sterile lines Xiangxiang 2 A and combination Xiangyou 63 of aromatic hybrid rice with good quality were bred.For twenty years,more than10 aromatic male sterile lines and 40 aromatic rice hybrids have been released in China.Some foreign research institutions especially the International Rice Research Institute and Indian Academy of Agricultural Sciences also bred a series of aromatic hybrid rice combinations,and they have achieved important accomplishment on the genetic expression and molecular biotechnological breeding of aroma characters.The extension and application of aromatic hybrid rice enriched the theory and practice of hybrid rice.In the present study,the accomplishments on aromatic hybrid rice breeding,genetic expression and biotechnological improvement of aroma characters,etc.were summarized to provide reference for further research in future.

Abstract:A total of 144 accessions of rice germplasm were screened for aromatic rice germplasm by using KOH immersion method and fragrance function markers in this experiment.Two F2 populations derived from Pingjing 11/ Heixiangdao、Jilinriluo/Heixiangdao were used to study the inheritance of fragrance.The results showed that 20 rice varieties produced fragrance when screening by KOH immersion method,while only 17 rice varieties with fragrance gene were screened by fragrance function markers GRFM04.The three rice germplasm only could be selected as aromatic rice by KOH immersion method were Heixiangdao,SY-xiang-7 and Qingxiangnuo.The fragrance of Heixiangdao was controlled by a pair of recessive gene.The fragrance of grains was consistent with the leaves in homozygote plants,but it was not completely consistent in heterozygote plants.

Abstract:With the development of modern biotechnology, the flavour or fragrance of aromatic rice is associated with the presence of 2-acetyl-1-pyrroline. A recessive gene (fgr) on chromosome 8 of rice has been linked to this important trait. In the present study, a F2 segregation population derived from a cross between Jingxiangmi with jasmine fragrant and MR63 without fragrance were used to study the inheritance of fragrance and map the fragrance gene in rice chromosome 8. Genetic mapping of molecular markers and the fragrance phenotype demonstrated that the InDel markers AP005537-17 and AP004463-13 flanked fgr, The physical distance between AP005537-17 and AP004463-13 is about 252 kb. The data suggested that fgr was closer to AP004463-13 (0.4 cM) than to AP005537- 17 (1.6 cM). Data analysis suggested that one Bacterial Artificial Chromosomes (BAC) (clone AP004463) was most likely to contain the gene. Ultimate forecast in the region may have 15 genes with the genes related to fragrance.

Abstract:Plants produce and emit a wide array of aroma compounds. These aroma compounds are extensively utilized in cosmetics, healthcare, and the food industry. In the last few years, the research on aroma compounds has made great progress. For some valuable economic crops including gain crops, fruits, vegetables and flowers, the main fragrance compounds have been identified. In this review, we summarized the important role and great potential of aroma compounds for crops and humans. Aroma compounds mainly originate from four major biosynthetic pathways in plants, including fatty acid, amino acid, terpenoid and carotenoid pathways, producing all kinds of substances including esters, alcohols, aldehydes, ketones, terpene, and sulfur compounds, etc. Importantly, we discussed the development of genetic engineering and its application potential in enhancing plant fragrance, especially the CRISPR/Cas9 system. We hope this review will provide insights into the fragrance improvement of the economic crops.

Abstract:The formation of rice aroma is a complex process that is influenced by genetic and environmental factors. More than 500 fragrance compounds have been documented in fragrant rice, among which 2-AP dominates the aroma of rice. This paper introduced the identification of OsBadh2 in the biosynthesis of 2-AP in rice. Then, non-enzymatic and enzymatic pathways of the 2-AP biosynthesis have been comprehensively investigated. In detail, 2-AP biosynthesis-associated enzyme, such as OsBADH2, OsP5CS, OsGAD, OsGAPDH, OsProDH, OsOAT, OsODC and OsDAO, have been summarized, while MG and fatty acids are also implicated in modulating the biosynthesis of 2-AP by providing the acetyl groups. Moreover, extensive collections of traditional fragrant rice varieties have been collated, together with the OsBadh2 haplotypes of 312 fragrant rice germplasm in China. And finally, genetic engineering of OsBadh2 and other genes in the 2-AP biosynthesis to develop fragrant rice are discussed.
Volatilome-based GWAS Identifies OsWRKY19 and OsNAC021 As Key Regulators of Rice Aroma

Abstract:Aromatic rice is globally favored for its distinctive scent, not only increasing nutritional value but also enhancing economic importance. However, apart from 2-acetyl-1-pyrroline (2-AP), the metabolic basis of aroma remains elusive, and the genetic underlying of the accumulation of fragrance metabolites are largely unknown. Here, we revealed 2-AP and fatty acid-derived volatile metabolites (FAVs) as key contributors to rice aroma by combining aroma rating with molecular docking. Using volatilome-based GWAS, we identified two regulatory genes that determine the natural variation of these fragrance metabolites. We demonstrated that OsWRKY19 not only enhances fragrance by negatively regulating OsBADH2 but also promotes agricultural traits in rice. Additionally, we revealed OsNAC021 that negatively regulates FAVs through the LOX pathway, and the knockout of it resulted in the over-accumulation of grain FAVs without a yield penalty. Our findings provide a compelling example of deciphering the genetic regulatory mechanisms underlying rice fragrance and pave the way for the creation of aromatic rice varieties.
Some Factors Affecting the Concentration of the Aroma Compound 2-Acetyl-1-pyrroline in Two Fragrant Rice Cultivars Grown in South China

Abstract:The fragrance potential of two fragrant rice (Oryza sativa L.) cultivars grown in South China was investigated in this study using headspace SPME and static headspace in conjunction with GC-MS. About a five-fold difference of 2-acetyl-1-pyrroline (2-AP) levels were observed among the two fragrant rice cultivars, with Guixiangzhan having the highest content (3.86 μg·g−1) comparable to that obtained with Thai KDML 105 rice. Other compounds instead of 2-AP were assumed to contribute to the characteristic aroma of Peizaruanxiang. The two cultivars were subjected to two preharvest treatments (planting density and harvest date) and different storage conditions (3 to 6 months at −4, 8, 20, and 30°C). Results were discussed in terms of grain yield, milling quality, grain appearance, and amylose and protein contents of rice samples associated with differing treatments. Highest 2-AP concentrations were obtained for Guixiangzhan and Peizaruanxiang with lower planting densities, the earliest harvesting time of 10 days after heading, the shortest storage time of 3 months, and the lowest storage temperature of −4°C. These findings indicate that manipulating pre- and postharvest treatments can greatly improve the specific attributes of the domestically produced cultivars. With that in mind, China could effectively increase its share of the domestic market of fragrant rice and even tap into the international market.

Abstract:Grain filling is a key step in determining the yield potential and grain quality of rice, which is directly related to the seed setting rate and 1000-grain weights. Rice grain filling is influenced by many factors, including the inherent characteristics of rice cultivar, plant hormones and environmental factors. At the molecular level, grain filling is also regulated by the related enzymes and genes during starch synthesis and transportation. Research progress on influence factors and related proteins and genes of rice grain filling were reviewed in the paper.

Abstract:Fragrant rice has various flavor types, mainly the popcorn flavor, corn flavor and lotus root flavor. Chinese fragrant rice from China and Thai fragrant rice from Thailand were analyzed. GC-MS was used to determine the volatile compounds of fragrant rice. It was found that there were 28 identical volatile compounds between Chinese and Thai fragrant rice. The key compounds of different flavor types of fragrant rice were obtained by comparing the common volatile compounds. The key compounds of the popcorn flavor were 2-butyl-2-octenal, 4-methylbenzaldehyde, ethyl 4-(ethyloxy)-2-oxobut-3-enoate and methoxy-phenyl-oxime. The key compounds of the corn flavor were 2,2′,5,5′-tetramethyl-1,1′-biphenyl, 1-hexadecanol, 5-ethylcyclopent-1-enecarboxaldehyde and cis-muurola-4(14), 5-diene. By using a combination of GC-MS and GC-O, the flavor spectrogram of fragrant rice was constructed, and the characteristic flavor compounds of each flavor type were identified. It was found that the characteristic flavor compounds of the popcorn flavor were 2-butyl-2-octenal, 2-pentadecanone, 2-acetyl-1-pyrroline, 4-methylbenzaldehyde, 6,10,14-trimethyl-2-pentadecanone, phenol and methoxy-phenyl-oxime. The characteristic flavor compounds of the corn flavor were 1-octen-3-ol, 2-acetyl-1-pyrroline, 3-methylbutyl 2-ethylhexanoate, methylcarbamate, phenol, nonanal and cis-muurola-4(14), 5-diene. The characteristic flavor compounds of the lotus root flavor were 2-acetyl-1-pyrroline, 10-undecenal, 1-nonanol, 1-undecanol, phytol and 6,10,14-trimethyl-2-pentadecanone. The resistant starch content of lotus root flavor rice was relatively high (0.8%). The correlation between flavor volatiles and functional components was analyzed. It was found that the fat acidity of fragrant rice was highly correlated (R = 0.86) with the characteristic flavor compounds, such as 1-octen-3-ol, 2-butyl-2-octenal and 3-methylbutyl-2-ethylhexanoate. The characteristic flavor compounds had an interactive contribution to the production of the different flavor types of fragrant rice.
