Abstract:Understanding the effects of genotype, environment and their interactions on rice quality is of great importance for rice breeding and cultivation. In this study, six rice varieties with two indica, two japonica and two indica–japonica types of rice were selected and planted at ten locations in Zhejiang Province to investigate the genotype (G) × environment (E) on physicochemical and sensory properties and the differences of volatile organic compounds (VOCs) among the three types of rice. Analysis of variances showed that apparent amylose content (AC), total protein content (PC), alkali spreading value (ASV), RVA profiles, and appearance (ACR), palatability (PCR), and sensory evaluation value (SEV) of cooked rice and texture of cooled cooked rice (TCCR) were mainly affected by genotypic variation, whereas the smell of cooked rice (SCR) was mainly affected by environment (p < 0.05). The G × E effect was significant for most parameters. The weather in the middle and late periods of filling had important effects on the formation of rice quality, especially on setback (SB) and pasting temperature (PT) (p < 0.01). They were negatively correlated with the texture of cooked rice (TCR) and SEV (p < 0.05). Peak viscosity (PV) and breakdown (BD) were positively related to the sensory evaluation parameters (p < 0.01) and could be used to predict cooked rice quality. A total of 59 VOCs were detected, and indica, japonica and indica–japonica had 9, 6 and 19 characteristic compounds, respectively. The principal component analysis showed that the physicochemical and sensory properties and VOCs of indica–japonica rice were more stable than those of indica and japonica rice at ten locations in Zhejiang Province. It is helpful for rice breeders to understand how the environment affects the physicochemical, sensory properties and VOCs of the three rice types, and it is also important for food enterprises to provide rice products with stable quality.

Abstract:Background: Cereal food is widely consumed mainly as the staple food of people. As a decisive factor that directly affects the quality evaluation and consumer acceptance of cereals, aroma has been paid increasing attention. Currently, the research on the formation mechanism and regulation measures of cereal aroma is still weak. Strategies of aroma analysis are constantly updated. Thus, it is necessary to systematically review the research status in the field of cereal aroma. Scope and approach: The main volatile compounds, and their formation mechanisms, traditional and emerging analysis strategies, and regulation measures in cereals were reviewed. Key findings and conclusions: Significant variability of volatile compounds has been observed between different cereal varieties, and between raw cereals and their processed products. Overall, aldehydes, heterocyclic compounds and alcohols contributed more to their overall aroma. The formation of cereal aroma is a complex process, which mainly includes genetic factors, Maillard reaction (MR), caramelization reaction (CR), degradation of lipids, amino acids and proteins, microbial metabolism, and their interactions. Currently, accurate, fast, safe, real-time and non-destructive analysis technologies combined with machine learning (ML) has been used in the identification, evaluation and prediction of cereal aroma, and the outcome is better than the traditional methods. The adjustment of precursor substances and reaction conditions, addition of exogenous additives and adoption of emerging processing technologies are the commonly used aroma regulation measures. Apart from the evaluation and prediction of aroma, ML has also realized the direct and indirect synthesis of desirable aroma, which is a new trend of aroma regulation in cereals.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Rice aroma was a comprehensive result of numerous volatiles and human sense. More than 300 volatiles were reported in rice. Rice aroma was widely researched by using sensory evaluation, gas chromatography (GC) method, and electronic nose (E-nose). Genetic factor was the main cause for rice aroma. However, the same rice variety might result in different aroma quality due to different planting, processing and storage.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Scope and aproach</h3><p>The main purpose of this review is to elaborate the main volatiles contributing rice aroma, methods used for rice aroma evaluation and factors affecting rice aroma. Most volatiles with odor descriptions, odor thresholds and retention indices were summarized. Sensory evaluation, GC method, and E-nose were elaborated with respect to rice aroma analysis. The effects of gene, planting, processing and storage on rice sample were pointed out.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Key findings and conclusions</h3><p>2-Acetyl-1-pyrroline (2-AP), aldehydes, heterocyclics, alcohols play important roles in rice aroma quality. GC-O-MS which combines the advantages of sensory descriptive analysis and GC-MS, can not only realize detailed sensory analysis of odor quality but also realize quantitative and qualitative detection of volatiles, playing an important role in exploring key aroma-active compounds. Besides of traditional electronic sensors, mass spectrometry and colorimetric sensors were used for E-nose, making it more novel and applicable. Water washing, high hydrostatic pressure, roasting and parboiling were thought as effective processing to improve rice flavor. Degree of milling and storage significantly affected rice flavor.</p>

Abstract:Eating-quality is one of the most important qualities for rice. With the development and application of molecular biology techniques, studies of its genetic and molecular bases have made great progresses. This paper summarizes these progresses from the following aspects: the relation between starch component and structure and cooked rice texture, gene location and QTL analysis, the key enzymes involved in the starch synthesis and corresponding gene expression and regulation on starch quality. The possible approaches to further research are discussed.

Abstract:Aromatic rice is globally favored for its distinctive scent, not only increasing nutritional value but also enhancing economic importance. However, apart from 2-acetyl-1-pyrroline (2-AP), the metabolic basis of aroma remains elusive, and the genetic underlying of the accumulation of fragrance metabolites are largely unknown. Here, we revealed 2-AP and fatty acid-derived volatile metabolites (FAVs) as key contributors to rice aroma by combining aroma rating with molecular docking. Using volatilome-based GWAS, we identified two regulatory genes that determine the natural variation of these fragrance metabolites. We demonstrated that OsWRKY19 not only enhances fragrance by negatively regulating OsBADH2 but also promotes agricultural traits in rice. Additionally, we revealed OsNAC021 that negatively regulates FAVs through the LOX pathway, and the knockout of it resulted in the over-accumulation of grain FAVs without a yield penalty. Our findings provide a compelling example of deciphering the genetic regulatory mechanisms underlying rice fragrance and pave the way for the creation of aromatic rice varieties.
Mutual Relations between Texture and Aroma of Cooked Rice—A Pilot Study

Abstract:Texture and aroma are two important attributes for the eating quality of cooked rice, but their mutual relations are not clear. Cooked rice with a desirable texture might suffer from a deteriorated aroma property. To better understand the relations between texture and aroma, six different rice varieties with desirable eating qualities have been selected, with their texture and aroma profile characterized by a texture analyzer and gas chromatography-ion mobility spectrometry, respectively. A large variance of textural attributes and a total number of 39 major volatile organic components were observed for these cooked rice varieties. Pearson correlation showed that the hardness of cooked rice was positively correlated with the content of E-2-hexenal, 2-hexanol-monomer, 1-propanol, and E-2-pentenal, while stickiness was positively correlated with 5-methyl-2-furanmethanol and dimethyl trisulfide. Possible underneath mechanisms were discussed for these relations. These results could help the rice industry to develop rice products with both desirable texture and aroma property.

Abstract:The formation of rice aroma is a complex process that is influenced by genetic and environmental factors. More than 500 fragrance compounds have been documented in fragrant rice, among which 2-AP dominates the aroma of rice. This paper introduced the identification of OsBadh2 in the biosynthesis of 2-AP in rice. Then, non-enzymatic and enzymatic pathways of the 2-AP biosynthesis have been comprehensively investigated. In detail, 2-AP biosynthesis-associated enzyme, such as OsBADH2, OsP5CS, OsGAD, OsGAPDH, OsProDH, OsOAT, OsODC and OsDAO, have been summarized, while MG and fatty acids are also implicated in modulating the biosynthesis of 2-AP by providing the acetyl groups. Moreover, extensive collections of traditional fragrant rice varieties have been collated, together with the OsBadh2 haplotypes of 312 fragrant rice germplasm in China. And finally, genetic engineering of OsBadh2 and other genes in the 2-AP biosynthesis to develop fragrant rice are discussed.

Abstract:The changes of the reducing sugars,amino acids,oils,fatty acids,and the effect of the oxygen on the formation of the flavor were investigated. The results showed that the reducing sugars, fatty acids and some amino acids contents decreased,which showed that the reducing sugars,fatty acids and some amino acids took part in the Maillard reaction that formed peanut fragrance after roasting peanut. Due to different contents and different structures of the amino acids in peanut,they had different effects on the peanut aroma formation. Oxygen was destructive in the formation of the flavor,so the process of the flavor formation needed to be operated under oxygen free conditions.
Flavor Formation and Characteristics of Fermented Rice Cake

Abstract:Flavor compounds analysis was carried out at different stages of fermented rice cake making from Simiao rice by yeast inoculation to study the flavor characteristics of fermented rice cake and their formation mechanism.The formation of characteristic taste and volatile compounds was found during both fermentation and steaming.Flavor free amino acids were formed mainly as a result of the thermal decomposition of proteins during steaming,of which bitter amino acids were the most prominent,followed by sweet and tasty ones.Moreover,a number of volatile compounds were formed during fermentation,and alkanes were mostly formed during steaming along with other volatile substances such as alcohol,acids,phenols and ketones,thus forming the special aroma of fermented rice cake.In conclusion,the taste compounds of fermented rice cake are mainly formed by thermal degradation,while the formation of aroma compounds are associated with both microbial and thermal degradation.

Abstract:ABSTRACT Descriptive sensory analysis has identified over a dozen different aromas and flavors in rice. Instrumental analyses have found over 200 volatile compounds present in rice. However, after over 30 years of research, little is known about the relationships between the numerous volatile compounds and aroma/flavor. A number of oxidation products have been tagged as likely causing stale flavor. However, the amounts of oxidation products, singly or collectively, that need to be present for rice to have stale or rancid flavor have not been established. Only one compound, 2‐acetyl‐1‐pyrroline (2‐AP; popcorn aroma) has been confirmed to contribute a characteristic aroma. Furthermore, 2‐AP is the only volatile compound in which the relationship between its concentration in rice and sensory intensity has been established. This article discusses the challenges of measuring aroma and flavor instrumentally and by human sensory panels and reviews research examining the effects of genetic, preharvest, and postharvest factors on volatile compound profiles and the aroma and flavor of cooked rice.

Abstract:Molecular basis of rice aroma formation is sparsely known and developmental programs driving biochemical pathways towards aroma is in infancy. Here, discovery and targeted proteo-metabolome of non-aromatic and aromatic rice seeds across developmental stages identified a total of 442 aroma-responsive proteins (ARPs) and 824 aroma-responsive metabolites (ARMs) involved in metabolism, calcium and G-protein signaling. Biochemical examination revealed ARM/Ps were linked to 2-acetylpyrrolidine, γ-aminobutyrate, anthocyanin, tannins, flavonoids and related enzymes. Pairwise correlation and clustering showed positive correlation among ARM/Ps. Consistent with aroma-related QTLs, ARPs were mapped on chromosomes 3,4,5,8 and were mainly compartmentalized in cytoplasm and mitochondria. ARM/P-correlation network identified associations related to metabolism and signaling. Multiple reaction monitoring (MRM) confirmed role of catechins, quinic acid and quercetin in aroma formation. Pathway enrichment, multivariate analysis and qRT-PCR validated that calcium and G-protein signaling, aromatic/branched-chain aminoacid, 2-acetylpyrrolidine, oxylipin, melvonate and prenylpyrophosphate pathways, indole, phenylacetate, flavonoid, cinnamoic ester govern aroma formation in rice.

Abstract:Aroma, an important quality characteristic of plant fruits, is produced by volatile organic compounds (VOCs), mainly terpenes, aldehydes, alcohols, esters, ketones, and other secondary metabolites, in plant cells. There are significant differences in the VOC profile of various fruits. The main pathways involved in the synthesis of VOCs are the terpenoid, phenylalanine, and fatty acid biosynthesis pathways, which involve several key enzyme-encoding genes, transcription factors (TFs), and epigenetic factors. This paper reviews the main synthetic pathways of the main volatile components in fruit, summarizes studies on the regulation of aroma formation by key genes and TFs, summarizes the factors affecting the fruit aroma formation, describes relevant studies on the improvement of fruit flavor quality, and finally proposes potential challenges and prospects for future research directions. This study provides a theoretical basis for the further precise control of fruit aroma quality and variety improvement.

Metabolome and Transcriptome Unveil the Correlated Metabolites and Transcripts with 2-acetyl-1-pyrroline in Fragrant Rice

Abstract:Fragrance is a valuable trait in rice varieties, with its aroma significantly influencing consumer preference. In this study, we conducted comprehensive metabolome and transcriptome analyses to elucidate the genetic and biochemical basis of fragrance in the Shangsixiangnuo (SSXN) variety, a fragrant indica rice cultivated in Guangxi, China. Through sensory evaluation and genetic analysis, we confirmed SSXN as strongly fragrant, with an 806 bp deletion in the BADH2 gene associated with fragrance production. In the metabolome analysis, a total of 238, 233, 105 and 60 metabolic compounds exhibited significant changes at the seedling (S), reproductive (R), filling (F), and maturation (M) stages, respectively. We identified four compounds that exhibited significant changes in SSXN across all four development stages. Our analyses revealed a significant upregulation of 2-acetyl-1-pyrroline (2AP), the well-studied aromatic compound, in SSXN compared to the non-fragrant variety. Additionally, correlation analysis identified several metabolites strongly associated with 2AP, including ethanone, 1-(1H-pyrrol-2-yl)-, 1H-pyrrole, and pyrrole. Furthermore, Weighted Gene Co-expression Network Analysis (WGCNA) analysis highlighted the magenta and yellow modules as particularly enriched in aroma-related metabolites, providing insights into the complex aromatic compounds underlying the fragrance of rice. In the transcriptome analysis, a total of 5582, 5506, 4965, and 4599 differential expressed genes (DEGs) were identified across the four developmental stages, with a notable enrichment of the common pathway amino sugar and nucleotide sugar metabolism in all stages. In our correlation analysis between metabolome and transcriptome data, the top three connected metabolites, phenol-, 3-amino-, and 2AP, along with ethanone, 1-(1H-pyrrol-2-yl)-, exhibited strong associations with transcripts, highlighting their potential roles in fragrance biosynthesis. Additionally, the downregulated expression of the P4H4 gene, encoding a procollagen-proline dioxygenase that specifically targets proline, in SSXN suggests its involvement in proline metabolism and potentially in aroma formation pathways. Overall, our study provides comprehensive insights into the genetic and biochemical mechanisms underlying fragrance production in rice, laying the foundation for further research aimed at enhancing fragrance quality in rice breeding programs.

Abstract:Aglycone precursors and glycosidases are two important compounds in tea aroma formation. The present paper reviews constitutes of oolong tea floral aroma, involving its existence form in leaf tissue and release mechanism. β-primeverosidase and β-glucosidase are proved to be two of the most important glycosidases in tea aroma formation via physiology, biochemistry and molecular biology researches. These enzymes are induced by stresses during tea processing operations, resulting in the release of aromatic voltiles.

Abstract:By simulating the aroma changes during in vitro mastication, we can better understand the aroma changes during rice eating, which is helpful in studying people's sensory preferences. To investigate the rice aroma released during the in vitro mastication, the present study analyzed rice bolus's odor fingerprints in vitro mastication using electronic nose and gas chromatography-ion mobility spectrometry (GC-IMS). The electronic nose analysis results showed significant differences in the flavor of japonica rice in vitro mastication. In addition, GC-IMS determined 30 volatile organic compounds (VOCs) during rice in vitro mastication. Among these compounds, the most important content was aldehydes, followed by ketones and alcohols. Although the concentration of various chemicals was relatively high in cooked rice, most compounds decreased after mastication. The concentration of propan-2-ol, ethanol, and methanol increased after mastication. Multivariate data analysis showed that isoamyl sovalerate, pentanal, hexanal, acetone, hexanal, and limonene were the main VOCs of japonica rice during in vitro mastication. GC-IMS and e-nose analyses are complementary and recommended for using the two techniques to achieve the VOCs' rapid and comprehensive detection during in vitro mastication. Results from this study allowed us to understand rice flavor during oral processing.

Analysis of volatile compounds in rice porridge of different japonica rice varieties in Northeast China

Abstract:Japonica rice is a widely cultivated and consumed cereal globally. There are many studies on the aroma of japonica rice, but few are on japonica rice porridge. Electronic nose and headspace solid-phase microextractiongas chromatography-mass spectrometry (HS-SPME-GC-MS) techniques combined with sensory evaluation were used to analyze the volatile compounds of 21 kinds of japonica rice porridges from Northeast China. The results of electronic nose analysis showed that different varieties of rice porridges had distinct aromas, and principal component analysis effectively distinguished between them. A total of 69 volatile compounds were identified by GC-MS, including 19 hydrocarbons, 17 aldehydes, 9 alcohols, 3 ketones, 3 phenols, 5 esters, 4 ethers, 4 acids, and 5 other compounds. Then the odor activity value (OAV) was calculated to determine the contribution of each aroma compound to the overall aroma of rice porridge. Finally, it is found that the key aroma compounds of japonica rice porridge were hexanal, octanal, nonanal, decanal, (E)-2-nonenal, naphthalene, (E, E)-2,4decadienal, butylated hydroxytoluene, 1-dodecanol, and 2-pentadecanone. This study preliminarily determined the key aroma substances and aroma attributes in japonica rie porridge, which can provide a valuable theoretical basis for selecting rice raw materials in the japonica rice porridge processing industry.

Abstract:Controlling the Maillard reaction may affect the generation of 2-acetyl-1-pyrroline, the key aroma compound in rice. In this study, the kinetics of 2-acetyl-1-pyrroline accumulation in the glucose/proline model system was comprehensively investigated and extra methylglyoxal or glyoxal was added to enhance 2-acetyl-1-pyrroline concentrations during rice cooking. Using the multi-response kinetic modeling to derive kinetic parameters, the formation of glyoxal, as the reactive intermediate, was rate-determining for the overall generation rate of 2-acetyl-1-pyrroline. Besides, although 2-acetyl-1-pyrroline generation was easier to occur with lower activation energy, much higher depletion rates of 2-acetyl-1-pyrrroline at 120 degrees C and 140 degrees C led to maximal 2-acetyl-1-pyrroline accumulation at the lower temperature of 100 degrees C. Furthermore, the inclusion of 0.05 mu mol/kg additional methylglyoxal in cooked rice significantly enhanced 2-acetyl-1-pyrroline generation. The work suggested that the development of rice products with superior flavor quality may be achieved by the slight accumulation of intermediates prior to thermal processing.
Characterization of volatile aroma compounds in cooked black rice

Abstract:Black rice ( Oryza sativa L.), an aromatic specialty rice popular in Asia, has a unique flavor, the volatile chemistry of which has not been reported. The objectives of this research were to study volatile profiles of cooked black rice and to characterize the odor-active compounds. Thirty-five volatile compounds were identified by gas chromatography-mass spectrometry using a dynamic headspace system with Tenax trapping. Aldehydes and aromatics were quantitatively in the greatest abundance, accounting for 80.1% of total relative concentration of volatiles. The concentration of 2-acetyl-1-pyrroline (2-AP) was high, exceeded only by hexanal, nonanal, and 2-pentylfuran. A total of 25 odor-active compounds, determined by gas chromatography-olfactometry, were applied to principal component analysis, demonstrating significant differences between a black and a traditional white rice cultivar in terms of aroma and explaining 93.0% of the total variation. 2-AP, guaiacol, indole, and p-xylene largely influenced the difference between the aroma in cooked black and white rice. 2-AP and guaiacol were major contributors to the unique character of black rice based on odor thresholds, relative concentrations, and olfactometry.
