Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:Pyrone moieties are present in natural products and can be synthesized by a diverse range of synthetic methods, resulting in the formation of various derivatives through chemical modifications. Many pyrone-based derivatives are commercially available and are biocompatible. They are building blocks of various intermediates in organic synthesis. They possess remarkable biological properties including antimicrobial, antiviral, cytotoxic, and antitumor activity. These characteristics have made them valuable for the development of drugs. We have summarized recent developments in the synthesis of 2-pyrone and its derivatives and their potential applications. With regard to synthetic approaches, the focus has been on metal-free and transition metal-catalyzed reactions.

Abstract:Pyrroloquinoxalines have proved to be a very attractive scaffold for medicinal chemist in the recent past. These compounds were extensively studied as bioactive compounds and many of them are known to be biologically and medicinally useful molecules, such as anti-HIV agents, antimalarial agents, antagonist agents, anticancer agents, and PARP-1 inhibitors. Additionally, pyrroloquinoxalines are also important intermediates for the construction of 5-HT3 receptor agonists. In this review the developments in biological activities and synthetic methodologies of pyrroloquinoxalines are discussed.

Abstract:The pyrrole moiety is an important structural motif in functional materials, natural products, and pharmaceuticals. More and more synthetic strategies toward pyrroles have emerged, where various efficient building blocks are developed and these synthons enable the syntheses of pyrroles with different numbers of components. However, no review specifically summarizes the syntheses of pyrroles according to the type and number of employed building blocks. To aid researchers to design appropriate substrates for pyrrole synthesis, herein we summarized the advances in pyrrole syntheses and classified these reactions into four categories according to the number of employed components, which may shed light on developing more efficient synthetic methods toward substituted pyrroles.

Abstract:Thieno[2,3-b]pyridine derivatives are of great importance because of their remarkable biological properties.They are of chemical and pharmacological interests due to their isosterism with quinolines and isoquinolines,two important heterocycles involved in many alkaloids.Some of them have been used as herbicides,fungicides,pesticides,medicines and dyes.Three general routes have been described so far for the preparation of thieno[2,3-b]pyridine derivatives of this ring system:a)formation of the thiophene ring by cyclization of suitable substitutents of a pyridine;b)formation of the pyridine ring by cyclization of a suitable thiophene;c)condensation of unsaturated thionone derivatives.This paper reviews the advances in synthesis and applications of thieno[2,3-b]pyridine derivatives.

Abstract:The construction of N-aryl sulfonamides is usually an important reaction in organic synthesis because these compounds are privileged in the structure of many natural products and pharmaceutical molecules. Also, N-aryl sulfonamides are well-known due to their antibacterial, antitumor, and anti-HIV biological activities. Therefore, research on the laboratory synthesis of N-aryl sulfonamides is an important challenge in organic synthesis. In this review, we discussed on methods reported up to now for the synthesis of N-aryl sulfonamides with the goal of stimulating further progresses in this field. We believe that this review will open up a new road for the preparation of pharmaceutically and biologically active sulfonamides.

Abstract:Pyrazolones and pyrazoles, featuring nitrogen-nitrogen bonds, are two of the most important classes of heterocycles, owing to their widespread occurrence in medicinal chemistry and functional materials. The last decade has witnessed a rapid increase in the construction of chiral pyrazolone and pyrazole derivatives, with the application of pyrazolone derivatives as powerful synthons. Since our last review in 2018, a large number of new achievements has emerged in this area, requiring a timely update. Thus, this review summarizes these elegant achievements based on the multiple reactive sites of different pyrazolone synthons. In addition, important mechanisms and interesting biological investigations relating to the corresponding products are also discussed.

Abstract:To investigate the alkyl analog of insecticide chlorfenapyr, two series of 2‐alkyl‐4‐bromo‐5‐(trifluoromethyl)pyrrole‐3‐carbonitriles were synthesized with a cycloaddition as the key step. The target products were characterized by 1H‐NMR spectroscopy, elemental analysis, or HRMS. The insecticidal, herbicidal, and antifungal activities of the target compounds were evaluated and found that these compounds did not show much insecticidal activity, but compounds 4, 10, and 11 had very good fungicidal activities against Alternaria solani and Fusarium oxysporum. Moreover, compound 4 had an outstanding inhibition effect against pigweed.

Abstract:Imidazopyridine derivatives are of great importance because of their remarkable biological properties. They are of chemical and pharmacological interest due to their isosterism with indoles and aza-indoles, two important heterocycles involved in many alkaloids. Some of them have been used as herbicides, fungicides, pesticides, medicines and dyes. The imidazopyridines comprise four isomers: imidazo[1,2-a]pyridine, imidazo[1,5-a]pyridine, imidazo[4,5-b]pyridine, and imidazo[4,5-c]pyridine. This paper reviews the advances in the synthesis of imidazopyridine derivatives during the past ten years.

Abstract:The pyrrolo[2,3-b] pyridine core structure, a bioisostere of quinolones, is found in several molecules that possess important biological activity. We describe here a new, concise, three-step synthesis of pyrrolo[2,3-b]pyridines starting from L-alanine. A series of 4,7-dihydro-4-oxo-1H-pyrrolo[2,3-b]pyridine-5-carboxylic acid derivatives, which have not been previously reported, were synthesized using this approach.

Abstract:It has been reported that pyrazole and pyrimidine heterocyclic compounds maybe exhibit high pharmaceutical functions and pesticidal activity. A series of novel pyrimidylarnino-pyrazole derivatives were synthesized and their biological activities were studied. All of the products were confirmed by H-1 NMR and elemental analysis, and some of them were characterized by IR and MS. The bioassy results indicated that some of the title compounds have a high fungicidal activity or herbicidal activity. In addition, the structure-activity relationship was discusses.

Abstract:While remarkable progress has recently been made for the direct C‐H‐functionalization of azines, its application is still limited by a lack of accessible functional groups (primarily carbon‐based) and poor regioselectivity. In contrast, C2‐functionalized pyridines and quinolines can be easily synthesized by treating readily available N‐oxides with various reagents under appropriate activation conditions. This review seeks to comprehensively document the available synthetic methods for introducing functional groups at the C2 position of pyridines and quinolines. In this work, we highlight recent developments in the C2‐functionalization of pyridine and quinoline N‐oxides and address both the mechanisms and regioselectivity of the reactions. We also describe the pathways and reactive species involved in these processes and highlight a number of medically relevant nitrogen heteroaromatics.

Abstract:An atom-economic reaction sequence on a multigram scale for synthesizing 2-pyrone was developed starting from furfuryl alcohol, a renewable resource made from bran or bagasse, utilizing a large-scale thermal rearrangement of cyclopentadienone epoxide as the key step. Additionally, 6-substituted 2-pyrone natural product derivatives are readily accessible by this approach. Abstract:Drug derived from simplest and commercially available moieties such as ethylacetoacetate, hydrazine, malonitrile, aldehyde derivatives is advantageous to achieve the mono‐ and bis‐pyranopyrazole derivatives with the good yield and purity. Potent drugs can be chemically prepared by the condensation and Micheal addition reaction under green, catalyst free, solvent free conditions to depict an excellent inhibition potention against diseases. Structure‐activity relationship of the target molecules also showed significance of active substituents such as methoxy, halogen and nitro groups in drug moieties for the further improvement of medicinal drugs. Recent Synthesis of Mono‐ & Bis‐Pyranopyrazole Derivatives. Pyranopyrazole is an important structural unit in heterocyclic chemistry and is useful in pharmaceutical and agrochemical industries. The pyranopyrazole‐based compounds are known in the agricultural industry as herbicides, fungicides, and insecticide agents and in medicinal fields as antimicrobial, anticancer, and antiHIV agents. Despite its wide range of applications, the synthesis of pyranopyrazole has several limitations such as the cost of reagents, potential environmental risks, harsh reaction conditions, longer reaction times, laborious workup procedures, and unsatisfactory yields. However, many studies have been recently reported on the synthesis of pyranopyrazoles using mild, green, and environmentally friendly methods that also give higher yields. Different catalysts and conditions have been reported for the synthesis of pyranopyrazole using four, three, and two components. Among these, four‐component synthesis is commonly reported by using benzaldehyde, hydrazine derivatives, ethyl acetoacetate and malononitrile due to the commercial availability and stability of the reagents. This review summarized the recent synthesis of pyranopyrazole i. e., mono‐ and bis‐pyranopyrazole derivatives via four‐, three‐ and two‐component‐based reactions. Mono‐ and bis‐pyranopyrazole derivatives varied due to the number of pyranopyrazole units that contributed to play an important role in increasing the binding interaction and biological activities beneficial for pharmaceutical industries.
