Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:Pyridazine was prepared by hydrogenolysis of 3,6-dichloropyridazine obtained via chlorination of 3,6-pyridazine diol with phosphorus oxychloride.3,6-Pyridazine diol was prepared by cyclization of hydrazine dihydrochloride and maleic anhydride.The feasible reaction conditions were as follows: cyclization: the mole ratio of maleic anhydride to hydrazine dihydrochloride was 1∶1,100°C,3 h;chlorination: the mole ratio of 3,6-pyridazine diol to phosphorus oxychloride was 1∶12.5,100～105°C,5 h;catalytic hydrogenolysis: the mole ratio of 3,6-dichloropyridazine to triethylamine was 1∶2.6,50°C,H_2 0.5 MPa,6 h.The overall yield was 62.2%(mol).The product was identified by melting point test,()~1H NMR and elemental analysis.

Abstract:2-Amino-3-methyl pyridine was prepared with 2-cyano-methylpyridine as raw material via hydrolysis in dilute basic solution using H_2O_2 as oxidant and Hofmann degradation with NaBrO.The feasible reaction conditions were as follows:hydrolysis,(50°C),5% NaOH,3.5 h,10% of H_2O_2;Hofmann degradation,(60°C),12% NaOH,0.5 h.The overall yield was 74%(mol).The product obtained was identified by GC-MS、IR、()~1H NMR and elemental analysis.

Abstract:A general synthetic route of aminopyridines which are useful organic intermediates was investigated.The target compounds were synthesized starting from corresponding cyanopyridines,via incomplete hydrolysis in the presence of hydrogen peroxide using mixture of acetone-water and dilute base as solvent,and Hoffman degradation reaction with freshly made NaBrO.The feasible conditions of the above synthetic process were discussed taking the preparation of 4-aminopyridine as example.The optimum concentration of sodium hydroxide solution in the transformation of cyanopyridine to pyridinecarboxamide was 1.0 %.And the Hoffman degradation would be efficiently carried out at 75 ℃ in 12.5 % sodium hydroxide solution.

Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.
Synthesis of 3-Aminopyrrolidine and its N-alkylating Derivatives

Abstract:The target compounds——3-aminopyrrolidine dihydrochloride and its acetate are the key intermediates of tosufloxacin and other quinolone antibiotics. One of the synthetic routes of the target compounds is from benzylamine and ethylacrylate, which lead to 3-benzylaminopropionate by additional reaction, then affords 3-[N-(methoxycarbonylmethene)benzylamino]propionate through N-alkylation with methyl chloroacetate, further by Dieckmann cyclization, decarboxylation to obtain 1-benzyl-3-pyrrolidine, then oximate and hydrogenate it to gain the intermediate 3-amino-1-benzy1-pyrrolidone, which can be changed to target compounds easily. This route is much of industrial value because of the cheap and easily gained materials. But the original method presents many weakness such as low overall yield and troublesome operate process. We carried out our research work to improve the above method and got an obvious increase of N-alkylation yield from 62.4% to more than 95%, and also some improvement in some reaction process. Thus we succeed in getting an industrial route characterized by cheap and easily gained materials, convenient operate process and low cost.

Abstract:Pyrazole compounds are one kind of heterocyclic compounds with extensive bioactivity,and lots of pyrazole compounds have been commercialized both in the region of medicament and pesticide.In the process of finding out some novel pyrazole compounds,a~d four novel pyrazole intermediates were synthesized.Taking diethyl oxalate as starting substance,the novel compounds can be synthesized via condensation,cyclic condensation,alkylation,(bromination) and hydrolysis reactions.The yield of products was about 78%-90%.Their structures were confirmed by elementary analysis,1H-NMR,MS and IR.

Abstract:An efficient and practical method was developed for the synthesis of tetrahydro-1H-pyrrolo[1,2-a]imidazol-2-ones based on the decarboxylative cyclization reaction of alpha-ketoamides and proline. Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:Pyrimidine compounds are an important class of heterocyclic compounds. The excellent bioactivity of pyrimidine skeleton makes it of important research value in the treatment of diseases. Therefore, efficient synthesis of pyrimidine skeleton compounds is of great significance in the fields of synthetic chemistry and pharmaceutical chemistry. In the present research, a variety of methods for building pyrimidine rings have been reported. In this regard, we have reviewed the recent update on the synthesis methods of homo-trisubstituted pyrimidines compounds in details. Through the analysis of the reaction mechanism, the existing problems of the existing reactions are pointed out, and the new methods for the synthesis of pyrimidine rings are prospected.

Abstract:3-(3-Chloro-1,2,5-thiadiazol-4-yl)pyridine,which is a key intermediate of selective M1 receptor agonist xanomeline,was synthesized from 3-pyridine carboxaldehyde,sodium cyanide,and sulfur chloride.This procedure has advantage of convenient operations and mild reaction conditions.The yield is improved from 40% to 52% and the process is easy to be popularized in industry.
Synthesis, Antimicrobial and Antioxidant Activity of Some New Pyrazolines Containing Azo Linkages

Abstract:Background: Pyrazolines and azo-pyrazolines are influential groups of heterocyclic compounds with two nitrogen atoms inside the five-membered ring. They play an important role in a wide range of biological processes, such as antifungal, antioxidant, antimalarial and other antimicrobial activities. Objective: The main objective of this study is to synthesize some new heterocyclic compounds with antioxidant and antimicrobial activity Methods: One-pot three components and traditional synthesis of new azo-pyrazoline compounds were achieved in this work. The preparation process has been started by diazotizing 4-(6-methylbenzothiazol-2-yl) benzamine and its coupling reaction with 4-hydroxy acetophenone producing azo-acetophenone, followed by benzylation with benzyl chloride to form the starting material, azo-benzyloxy acetophenone. A series of substituted benzaldehydes were reacted with the latter compound via one pot and classical methods, forming new chalcones containing azo linkages and benzyloxy moieties, which were then converted into new target azo-pyrazoline derivatives. Results: The structures of the synthesized compounds were confirmed by spectroscopic techniques using FT-IR, 1H-NMR, 13C-NMR, and 13C- DEPT- 135 spectra. Finally, the synthesized compounds were screened for their antioxidant and antimicrobial activities against Staphylococcus aureus and Escherichia coli. Conclusion: Overall, the one-pot three-component synthesis of pyrazoline compounds generally provides advantages in terms of efficiency, simplicity, and time-consumption compared to classical synthesis methods. Hence, the study advocates the one-pot method because it eliminates the tedious process of making chalcones, which takes time, materials, and unnecessary effort. Therefore, this is the most convenient and effective approach to green chemistry.

Abstract:Drug derived from simplest and commercially available moieties such as ethylacetoacetate, hydrazine, malonitrile, aldehyde derivatives is advantageous to achieve the mono‐ and bis‐pyranopyrazole derivatives with the good yield and purity. Potent drugs can be chemically prepared by the condensation and Micheal addition reaction under green, catalyst free, solvent free conditions to depict an excellent inhibition potention against diseases. Structure‐activity relationship of the target molecules also showed significance of active substituents such as methoxy, halogen and nitro groups in drug moieties for the further improvement of medicinal drugs. Recent Synthesis of Mono‐ & Bis‐Pyranopyrazole Derivatives. Pyranopyrazole is an important structural unit in heterocyclic chemistry and is useful in pharmaceutical and agrochemical industries. The pyranopyrazole‐based compounds are known in the agricultural industry as herbicides, fungicides, and insecticide agents and in medicinal fields as antimicrobial, anticancer, and antiHIV agents. Despite its wide range of applications, the synthesis of pyranopyrazole has several limitations such as the cost of reagents, potential environmental risks, harsh reaction conditions, longer reaction times, laborious workup procedures, and unsatisfactory yields. However, many studies have been recently reported on the synthesis of pyranopyrazoles using mild, green, and environmentally friendly methods that also give higher yields. Different catalysts and conditions have been reported for the synthesis of pyranopyrazole using four, three, and two components. Among these, four‐component synthesis is commonly reported by using benzaldehyde, hydrazine derivatives, ethyl acetoacetate and malononitrile due to the commercial availability and stability of the reagents. This review summarized the recent synthesis of pyranopyrazole i. e., mono‐ and bis‐pyranopyrazole derivatives via four‐, three‐ and two‐component‐based reactions. Mono‐ and bis‐pyranopyrazole derivatives varied due to the number of pyranopyrazole units that contributed to play an important role in increasing the binding interaction and biological activities beneficial for pharmaceutical industries.

Synthesis of Some New 1, 3, 4-Oxadiazole, Pyrazole, and Pyrimidine Bearing Thienopyrazole Moieties

Abstract:Aim and objective: According to the literature survey, pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the present work, pyrazol- 3-one 1 was reacted with cyanoacetic acid hydrazide and elemental sulfur to afford the corresponding thieno[3,2-c]pyrazol-6-carbohydrazide 3 derivatives. The latter compound reacted with some electrophilic reagents such as DMF-DMA, triethylorthoformate, arylidenemalononitriles and chalcones under neat conditions to give substituted oxadiazole and pyrazole, respectively. The treatment of compound 3 with active methylene reagents such as acetylacetone, diethylmalonate, ethyl acetoacetate and ethyl cyanoacetate under suitable conditions afforded pyrazole derivatives 10, 11, 13, and 15, respectively. Novel pyrazolothienopyrimidine 27 and 30 were prepared from precursor 26 with carbon disulfide and triethylorthoformate, respectively. The chemical structures of the newly synthesized compounds were established by elemental and spectral analyses including IR, and 1HNMR in addition to 13C-NMR and mass spectra. Materials and methods: A novel substituted pyrazole, pyrimidine and pyrazolothienopyrimidine were obtained via Gewald synthesis of thiophene and fused thiophene and Mannich reactions of 5-amino-3-phenyl-1Hthieno[ 3,2-c]pyrazole-6-carbohydrazide. Results and discussion: A series of some newly azoles and azines were prepared via reaction of thieno[3,2- c]pyrazol-6-carbohydrazide derivative 3 as starting material with some electrophilic and nucleophilic reagents. The structures of target compounds were established by elemental analyses and spectral data. Conclusion: Pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the current investigation, we have developed new and efficient methods for the synthesis of thieno[3,2-c]pyrazol-6-carbohydrazide derivatives. Abstract:Fused imidazole and pyrimidine heterocycles represent two important classes of organic compounds. Numerous synthetic methods have been developed for the synthesis of these compounds due to their diverse biological activities and potential pharmaceutical applications. This review provides a comprehensive assessment of the cutting‐edge methods used in the synthesis of fused heterocycles from pyridin‐2‐amines and their aza derivatives involving reaction with 1,3‐dicarbonyl compounds or α‐halo carbonyl compounds. In addition, the Groebke‐Blackburn‐Bienaymé (GBB) reaction, metal‐mediated reactions and other related reaction systems have been considered. We summarize the pertinent literature on related synthetic methods in the period 2020 to 2023. The analysis provided can inform future developments directed at new cost‐effective approaches to the synthesis of fused imidazole and fused pyrimidine analogues.

Abstract:BACKGROUNDAnthranilic diamide insecticides containing pyridylpyrazole-5-carboxamide are extremely important in modern agriculture. New structurally modified compounds with high insecticidal activity were discovered by designing a series of novel pyridylpyrazole-4-carboxamides (9I to 9IV) and pyridylpyrazole-4-carboxamides (10I to 10IV), the latter designed by the cyclisation of two amides. The structure-activity relationship (SAR) between the two series is of interest.RESULTSTwo series of novel anthranilic diamides containing pyridylpyrazole-4-carboxamide were synthesised and characterised via melting point, H-1 NMR, C-13 NMR, MS and elemental analyses. The insecticidal activities of these compounds against Plutella xylostella were evaluated. At a concentration of 100mgL(-1), the compounds with unmodified amide moieties (9I to 9IV) exhibited much better larvicidal activities than the other derivative compounds (10I to 10IV). Most of the compounds 9I to 9IV showed over 90% larvicidal activity at 100mgL(-1). Furthermore, compounds 9IIIa, 9IIIc, 9IIId and 9IVd displayed significant insecticidal activity at 10mgL(-1). Density functional theory (DFT) calculation was carried out to provide more information regarding SAR.CONCLUSIONThirty-two new anthranlic diamides containing pyridylpyrazole-4-carboxamide were designed and obtained. SAR analysis and DFT calculation results revealed that the amide moiety had a very important effect on bioactivity.
