Abstract:Chemical investigation on the 95% ethanol extract of green walnut husks of Juglans mandshurica Maxim. led to the isolation of two new diarylheptanoid compounds, including Juglanin K ( 1 ) and Myricananin I ( 2 ), together with ten known compounds ( 3–13 ). Their structures were elucidated by extensive analyses of comprehensive spectroscopic methods. The isolated compounds were evaluated for their antibacterial activities against Escherichia coli and Staphylococcus aureus . The results showed that compound 2 had moderate antibacterial activity with the MIC values of 0.313 and 0.156 mg/mL, respectively. compounds 1 , 3 – 7 and 13 showed weakly antibacterial activities against Escherichia coli and Staphylococcus aureus with the MIC values of 0.625–2.5 mg/mL. Abstract:Four new tetrahydrofuran lignans, schpropinrins A-D ( 1–4 ), together with five known ones, were isolated from the leaves and stems of Schisandra propinqua var. sinensis . Their structures, including absolute configurations, were characterized by means of spectroscopic analysis and ECD calculation. Compounds 1–4 featured a ketal or hemiketal substructure at C-7 and all of the isolates were tested for their anti-HIV integrase activity.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. The absolute configurations of the new compounds were further determined by ROESY and an empirical comparison of their experimental ECD spectra with literature.

Abstract:Three new diarylheptanoids, i.e., sodium(5S,2E)-1,7-bis(4-hydroxyphenyl)-1-hydroxy-2-hepten-5-sulfonate (1), sodium(5R,2E)-1,7-bis(4-hydroxyphenyl)-1-hydroxy-2-hepten-5-sulfonate (2) and 3,5-diacetoxy-1-(3-methoxy-4,5-dihydroxyphenyl)-7-(4-hydroxy-3-methoxyphenyl)heptane (3) were isolated from the rhizomes of Zingiber officinale. The structures of the new compounds were elucidated on the basis of spectroscopic methods. The antioxidant activities of 3 were assayed by in vitro models involving DPPH free radicals and superoxide anion radicals.

Abstract:Five new nortriterpenoids, schicagenins D-F (1-3, resp.) and negleschidilactones A and B (4 and 5, resp.), together with eleven known ones, were isolated from the stems of Schisandra neglecta. Their structures were established on the basis of extensive spectroscopic analyses. All the compounds were evaluated for their activities regarding insulin sensitivity in 3T3-L1 differentiated adipocytes. None of them showed a significant bioactivity at 10M concentration.

Abstract:Three new diarylheptanoids and one new monoterpenoid were isolated from the rhizomes of Zingiber officinale together with four known diarylheptanoids, 5-8. Their structures were elucidated mainly by spectroscopic methods, and they were deduced as 5-[4-hydroxy-6-(4-hydroxyphenethyl)tetrahydro-2H-pyran-2-yl]-3-methoxybenzene-1,2-diol (1), sodium (E)-7-hydroxy- 1,7-bis(4-hydroxyphenyl)hept-5-ene-3S-sulfonate (2), sodium (E)-7-hydroxy- 1,7-bis(4-hydroxyphenyl)hept-5-ene-3R-sulfonate (3), and hydroxycineole-10-O-beta-D-glucopyranoside (4), respectively. Among the isolated compounds, compounds 1, 5, and 8 exhibited strong superoxide anion radical scavenging activities in a phenazine methosulfate-NADH system. In a more biological system, these compounds were demonstrated to exhibit potent protection against lipid peroxidation in mouse liver microsomes exposed to oxidative conditions. These compounds were subsequently tested on primary cultures of rat hepatocytes exposed to oxidative damage, and definitive cytoprotective actions were found.

Abstract:A structurally novel 8,9-seco-eudesma-9,4:8,12-dilactone sesquiterpenoid, chlorajapodiolide (1), a new eudesmane sesquiterpenoid, chlorajapotriol (2), a new lindenane-type sesquiterpenoid dimer, chlorajaponilide I (3), and four known lindenane-type sesquiterpenoid dimers were isolated from the whole plant of Chloranthus japonicus. Their structures were established through extensive analysis of NMR spectroscopic data in combination with mass spectrometry. All compounds were evaluated for cytotoxicities against four human cancer cell lines A549, HCT116, MGC803, and HepG2.

Abstract:A new cyclic diarylheptanoid, 1,5-epoxy-3-hydroxy-1-(3-methoxy-4,5-dihydroxyphenyl)-7-(4-hydroxyphenyl)-heptane (1), as well as a new monoterpene, 10-O-β-d-glucopyranosyl-hydroxy cineole (2) were isolated from the rhizomes of Zingiber officinale. Abstract:Four previously unreported diarylheptanoids ( 1a / 1b − 2a / 2b ), one undescribed sesquiterpenoid ( 8 ), one new diterpenoid ( 12 ), and twelve known analogs were isolated from the fruits of Alpinia oxyphylla . The structural elucidation of these compounds was achieved through a comprehensive analysis of spectroscopic data, single-crystal X-ray diffraction, electronic circular dichroism (ECD), and modified Mosher's method. Enantiomeric mixtures ( 1a / 1b , 2a / 2b , 3a / 3b , 4a / 4b , and 5a / 5b ) were separated on a chiral column using acetonitrile-water mixtures as eluents. Among them, compounds 3a / 3b and 4a / 4b were isolated as optically pure enantiomers in the initial chiral separation. Furthermore, most of the isolates were evaluated for their inhibitory effects against the production of nitric oxide (NO) and interleukin-6 (IL-6) in lipopolysaccharide (LPS)-induced RAW264.7 macrophages. Interestingly, 2 and 4 showed significant inhibitory activities against NO production with IC 50 values of 33.65 and 9.88 μmol·L −1 (hydrocortisone: IC 50 34.26 μmol·L −1 ), respectively. Additionally, they also partially reduced the secretion of IL-6.

[Study on anti-tumor chemical constituents from pericarps of Juglans mandshurica]

Abstract:Objective: To study the anti-tumor chemical components of the pericarps of Juglans mandshurica. Methods: The chemical constituents were isolated and purified by AB-8 macroporous adsorption resin, silica gel, Sephadex LH-20 columns and recrystallization. The structures were elucidated on the basis of physicochemical properties and NMR spectral data analysis. Results: From the pericarps of Juglans mandshurica, twelve compounds were separated and identified as 3-methoxy juglone(1), 3-ethoxy juglone(2), 1,8-di-hydroxy anthraquinone (3), juglone (4), 2α, 3α, 19α-trihydroxy ursolic acid (5), 1α, 3β-dihydroxy-olean-18-ene (6), methyl gallate (7), pterocarine(8), quercetin(9), kaempferol(10), daucosterol(11), and β-sitosterol(12). Conclusion: Compounds 1 - 3 and 6 are isolated from the pericarps of Juglans mandshurica for the first time. Compounds 5 and 7 are isolated from Juglans genus for the first time.

Abstract:Two new triterpenoids, schisphendilactone A and B ( 1 and 2 ), together with three known triterpenoids, were isolated from the stems of Schisandra sphenanthera . Their structures were elucidated by spectroscopic methods, and the absolute configuration of 1 was determined by single-crystal X-ray diffraction. Compound 2 showed moderate inhibitory activity against SW480 cancer cell line, and compound 5 exhibited promising anti-HIV-1 activity with EC 50 value of 0.52 μg ml −1 and therapeutic index value of 117.12.

Abstract:Phytochemical study on 90% ethanol extract from the green walnut husks of Juglans mandshurica Maxim. resulted into the isolation of three undescribed triterpenoids, juglansmanoids A–C ( 1 – 3 ). Structural elucidation of all the compounds were performed by spectral methods such as 1D and 2D ( 1 H- 1 H COSY, HMQC, and HMBC) NMR spectroscopy, in addition to high resolution mass spectrometry. The isolated components were evaluated in vitro for anti-hyaluronidase activities. As a result, triterpenoid 1 exhibited potent anti-hyaluronidase activity (IC 50 = 9.78 μg/ml) three times more than the positive control drug oleanolic acid (IC 50 = 40.12 μg/ml).

Abstract:A new semiterpenoid glycoside, 3-methylbutan-1, 3-diol-1-O-β-d-glucopyranoside (1) and a new benzofuran derivative glycoside, 6-carboxylethyl-benzofuran-5-O-β-d-xylopyranosyl-(1→2)-β-d-glucopyranoside (2), together with seven known compounds (3-9) were isolated from the roots of Heracleum dissectum Ledeb. Their structures were elucidated on the basis of physicochemical properties and the detailed interpretation of various spectroscopic data. All the isolated compounds were screened for anti-inflammatory activity in vitro. And the result showed that compound 2 exhibited significantly inhibitory activity on nitric oxide production in RAW264.7 cells, which IC50 value was equivalent to that of the positive control indomethacin.

Abstract:As a traditional medicinal plant, Juglans mandshurica has been used for the treatment of cancer. Different organs of this plant showed anti-tumor activity in clinic and laboratory. Comparative identification of constituents in different plant organs is essential for investigation of the relationship between chemical constituents and pharmacological activities. For this aim, the roots, branches, and leaves of J. mandshurica were extracted with 50% v/v methanol and then subjected to ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry analysis conducted under low and high energy. As a result, we have to date identified 111 compounds consisting of 56 tannins, 29 flavonoids, 13 organic acids, 8 naphthalene derivatives, and 5 anthracenes. Five compounds, namely, diquercetin trihydroxy-truxinoyl-glucoside, two quercetin kaempferol dihydroxy-truxinoyl-glucosides, syringoyl-tri-galloyl-O-glucose, and dihydroxy-naphthalene syringoyl-glucoside, were tentatively identified as new compounds. Of the compounds identified, 76 were found in the root extract, 67 in the branch extract, and 37 in the leaf extract. Only six compounds including four organic acids and two tannins were found in all three extracts. We developed a rapid and sensitive ultra high performance liquid chromatography with quadrupole time-of-flight mass spectrometry approach to identify multiple constituents of complex extracts without separation and ion selection. The results presented provide useful information on further research of the bioactive compounds of J. mandshurica.
