Abstract:The synthesis of retinoic acid ester by Wittig-Horner reaction was improved. Retinoic acid ester was obtained directly by the reaction of 3-methyl-5-(2,6,6-trimethyl-1-cyclohexen-1-yl)-1,3(or1,4)-pentadienyl phosphonate and C 5-aldehyde. One step of double bond rearrangement catalysted by base is curtailed . C 1 double bond phosphonate is used in Wittig-Horner reaction directly.

Abstract:A visible light-mediated approach for the preparation of α-bromo-α,β-unsaturated ketones and aldehydes was developed. In comparison to traditional methods that generally take two steps to afford the above compounds, this protocol was highlighted by its operational simplicity, avoiding using hazardous bromine and mild reaction conditions.

Abstract:Asymmetric Wittig-type olefination of 4-substituted cyclohexanones with chiral ligand-modified stable arsonium ylides has been examined. The 8-phenylmenthol-derived chiral arsonium ylide of 4 reacted with prochiral ketones 9a-d at -15 degrees C to give the 4-substituted cyclohexylideneacetates 11a-d in 58-69% yield and in up to 80% diastereomeric excess (de). Abstract:A facile synthesis of free-base and metalated stilbene-linked porphyrin dimers via the Wittig reaction is reported. 5-(4-chloromethylphenyl)-10,15,20-triphenyl porphyrin was prepared by direct chloromethylation of meso-tetraphenylporphyrin as the useful precursor for both the porphyrin-derived Wittig reagent and the monoformylporphyrin. It was found that the stilbene-linked porphyrin dimers were formed with a remarkable sterochemical preference in the cis-form. Their absorption and emission spectra were measured.

Abstract:Significant base and temperature effects on the Wittig reactions of cyclohexanones with (carbethoxymethylene)triphenylphosphorane under microwave irradiation were observed. For the Wittig reactions carried out in a domestic microwave oven under solvent-free conditions, the initially formed exo-olefin products isomerized into the thermodynamically more stable endo-olefins due to uncontrolled high reaction temperature. In sharp contrast, under controlled microwave heating, both exo- and endo-olefins have been selectively synthesized by accurately regulating the reaction temperature with or without a base. Selective α-Bromination of Functionalized Ketene Dithioacetals

Abstract:Functionalized ketene dithioacetals are important intermediates in organic synthesis.α-Functionalization reactions based on them provide potential precursors for the construction of usefully carbo-and heterocyclic compounds.In this paper,a versatileα-bromination of functionalized ketene dithioacetals was developed.In the presence of H2 O,the reactions of functionalized ketene dithioacetals(1) with CuBr 2 in THF at room temperature gave variousα-bromo ketene dithioacetals(2) in excellent yields with high regioselectivities.The mechanism of thisα-bromination is proposed to undergo an addition of bromine formed in situ to compound 1 and sequontial elimination of theα-proton.The simplicity of execution,mild conditions,inexpensive,easily usable brominating agent and high yields,make this synthetic method attractive for practical applications.

Abstract:The Wittig reaction, which is one of the most effective methods for synthesizing alkenes from carbonyl compounds, generally gives thermodynamically stable E-alkenes, and synthesis of trisubstituted Z-alkenes from ketones presents notable challenges. Here, we report what we refer to as Wittig/B─H insertion reactions, which innovatively combine a Wittig reaction with carbene insertion into a B─H bond and constitute a promising method for the synthesis of thermodynamically unstable trisubstituted Z-boryl alkenes. Combined with the easy transformations of boryl group, this methodology provides efficient access to a variety of previously unavailable trisubstituted Z-alkenes and thus provides a platform for discovery of pharmaceuticals. The unique Z-selectivity of the reaction is determined by the maximum overlap of the orbitals between the B─H bond of the borane adduct and the alkylidene carbene intermediate in the transition state.

Abstract:The Wittig reaction involving direct olefination of triphenylphosphonium ylides (Ph(3)P=CHR) with aldehydes is arguably the most often used method for alkene synthesis, but in general it yields mixtures of Z- and E-alkenes for semistabilized triphenylphosphonium ylides (R = aryl or vinyl). We have developed a simple and efficient protocol to improve the stereoselectivity significantly by replacing the aldehydes used in the Wittig reaction with N-sulfonyl imines, which possess distinct electronic and steric properties relative to aldehydes. A broad range of aromatic, alpha,beta-unsaturated, and aliphatic imines bearing appropriate N-sulfonyl groups smoothly undergo olefination reaction with various benzylidenetriphenylphosphoranes or allylidenetriphenylphosphoranes under mild reaction conditions to afford an array of both Z- and E-isomers of conjugated alkenes in good to excellent yields and with greater than 99:1 stereoselectivity. Moreover, this tunable protocol has been successfully applied to the highly stereoselective synthesis of two anticancer agents, DMU-212 and its Z-isomer.

Abstract:The Wittig reaction was widely used in the synthesis of natural products particularly in the area of insect pheromore synthesis. But unfortunately when a moderated triphenylphosphonium ylid is used, the resultant compound is a impractical E, Z, mixture. For instance, the title compound was once synthesized by the Wittig between butenylidene triphenylphosphorane and 8-oxo-octanoate, followed by hydrolysis and reduction. The product was a mixture containing 75% 8E,10E, isomer and 25% 8Z,10E isomer. Recently we have reported that a simple change in phosphorus substituents from Ph3P=CHR to Ph2(RCH2)P=CHR(R′=alkenyl or H) dramatically increases the proportion of Eolefin formed from moderated ylides(R=alkenyl) and aliphatic aldehydes. According to this fact, here we report a stereoselective synthesis of E,E,-8,10-dodecadien-1-ol from butenylidene methyl diphenylphosphorane and 8-oxo-octanoate in the absence of lithium salt(Schemel). After hydrolysis and reduction, the title compound containing 94.7% 8E, 10E isomer was obtained. This approach affords a convenient synthesis for insect pheromones bearing a E,E,conjugated diene moiety. This experimental fact once again illustrates the stereochemistry of allylic diphenylphosphonium ylid is quite different from the corresponding triphenylphosphonium one.

Abstract:A direct synthesis of 3-(bromomethylene)isobenzofuran-1(3H)-ones from 2-alkynylbenzoic acids is developed through a regioselective 5-exo-dig bromocyclization. The reaction proceeds smoothly with a broad substrate scope. The products can be further elaborated via a palladium-catalyze cross-coupling reaction, or can convert to other heterocycles through simple operation. Preliminary mechanistic studies reveal that this transformation undergoes a radical process.

Abstract:A concise and efficient photoinduced rearrangement of (E)-3-arylvinyl-4H-chromen-4-ones for the synthesis of benzoaryl-5-yl(2-hydroxyphenyl)methanones is described. Benzoaryl-5-yl-(2-hydroxyphenyl)methanones were obtained in 77-95% yields via the irradiation of (E)-3-arylvinyl-chromones in the 95% EtOH with a high-pressure mercury lamp at room temperature under Ar atmosphere. The reported method provides a novel procedure for the synthesis of α,α'-diaryl ketone derivatives without addition of any transition metals and oxidants or other additives. A plausible mechanism was proposed, and the rearrangement product was characterized by NMR, HRMS, and X-ray.

Abstract:We report the preparation of 1-(1-alkenyl)benzotriazoles from alkenyl (phenyl)iodonium salts and benzotriazole. The reaction offers a simple and convenient route for the stereoselective synthesis of 1-(1-alkenyl)benzotriazoles. In addition, present method is particularly efficient for preparing 1-(1-alkenyl)benzotriazoles with amino-, acyl- and alkoxycarbonyl substituents, which were difficult to prepare by conventional methods.
