PPO Activity Determination and Anti-browning Measure for Amorphophallus Albus

Abstract:Enzymatic browning normally occurs during dehydration of Amorphophallus albus with hot air, and the quality of the konjac flour is affected. In this study, the effects of temperature, pH and the concentration of the substrate on PPO (Polyphenol oxidase) activity were investigated and, based on the results, the optimum conditions for sulphur-free anti-browning were selected. PPO activity in A.albus was highest at a temperature of 35 ℃ and a pH of 5.5. With a pH of 2 or 8, or a temperature over 90 ℃, PPO activity was effectively suppressed. Ascorbic acid, citric acid, phytic acid and cysteine were found to be able to inhibit PPO activity.
