Abstract:In this study, polyphenol oxidase (PPO) from corn tassel was extracted and partially purified through (NH4)2SO4 precipitation and gel filtration chromatography. Optimal temperatures for subsrates catechol and 4-methyl catechol were 40 °C and 30 °C, respectively. The optimal pH values were 8.0 for catechol and 6.0 for 4-methyl catechol. Catechol was the most suitible substrate (Km: 3.48 mM, Vmax: 1.0 Abs./ min.). The moleculer mass of PPO was determined as 158 kDa. In this work, sodium azide, ethylenediaminetetraacetic acid (EDTA) and sodium dodecyl sulfate (SDS) were found to inhibit the enzyme activity as 26.6 %, 22.2 % and 12.2 % ratio, respectively. Besides, the effects of carbohydrates such as sucrose, fructose, ribose and glucose on PPO activity were investigated. The enzyme was found to be activated 17 % by fructose and ribose, 16 % by glucose and 4 % by sucrose.

Abstract:This study was designed to establish an efficient purification method for phenolic compounds from rice bran and to acquire insightful knowledge of purification impact on its chemical constituents, antioxidant and antidiabetic activity. Among the eight macroporous resins investigated, HPD-300 resin was verified to bosses superior adsorption and desorption qualities for the extract. Purification parameters were optimised. Eighteen phenolic compounds were identified using HPLC-PDA, the contents of ferulic acid and syringaldehyde were significantly (P < 0.05) higher than the other constituents. However, phenolic compounds concentrations varied significantly (P < 0.05) after HPD-300 resin purification. In addition, the extract exhibited a significant (P < 0.05) increase in total flavonoid contents, in vitro antioxidant (DPPH and FRAP) and antidiabetic (alpha-glucosidase and BSA-glycation inhibition) activity after HPD-300 resin treatment. These findings indicate that selection of resin and applying optimal purification condition is an appropriate economical approach for obtaining bioactive phenolic compounds from rice bran.
Isolation and Characterization of Thermo-Tolerant Polyphenol Oxidases in a Black Tea Infusion

Abstract:Polyphenols are important active components of tea. Tea polyphenol oxidase (PPO) catalyzes polyphenols oxidation, hence it is vitally important to the quality and function of black tea. The enzyme was believed to be mostly deactivated during the thermal processing. Here we report the macromolecular fraction of a Chinese black tea (Camellia. sinensis var. Lapsang souchong) infusion oxidized pyrocatechol even at the enzyme's deactivation temperature. Utilizing urea-assisted extraction and ion-exchange chromatography, the PPOs and/or co-enzymes were isolated from the tea infusion and the cold water extracts of fresh tea leaves. The isolates were found to exhibit PPO activities after removing urea by dialysis. A 66.0 kDa PPO with specific activity of 1200 U/mg (Km = 6.973 mM and Vmax = 15.79 U/min) was purified and characterized to be a glycoprotein with isoelectric point of 4.82. The efficiency of four inhibitors against this PPO are cysteine > EDTA > sodium sulfite > ascorbic acid. The PPOs isolated from black tea infusion reserved 40-80% of activities of their aliquots from fresh tea leaves. It reveals the PPOs can survive the thermal processing of the black tea, possibly in form of PPO-polyphenol complex. Exploring the biochemistry and oxidation mechanism of tea infusion, this study may empower the future development of tea and plant-based beverages.
Functional analysis of polyphenol oxidase 1 gene in common wheat

Abstract:Polyphenol oxidase (PPO) activity is a major cause of the undesirable brown color of wheat-based products. Ppo1 , a major gene for PPO activity, was cloned based on sequence homology in previous studies; however, its function and regulation mechanism remain unclear. In this study, the function and genetic regulation of Ppo1 were analyzed using RNA interference (RNAi) and Targeting Induced Local Lesions IN Genomes (TILLING) technology, and superior mutants were identified. Compared with the control, the level of Ppo1 transcript in RNAi transgenic lines was drastically decreased by 15.5%–60.9% during grain development, and PPO activity was significantly reduced by 12.9%–20.4%, confirming the role of Ppo1 in PPO activity. Thirty-two Ppo1 mutants were identified in the ethyl methanesulfonate (EMS)-mutagenized population, including eight missense mutations, 16 synonymous mutations, and eight intron mutations. The expression of Ppo1 was reduced significantly by 6.7%–37.1% and 10.1%–54.4% in mutants M092141 (G311S) and M091098 (G299R), respectively, in which PPO activity was decreased by 29.7% and 28.8%, respectively, indicating that mutation sites of two mutants have important effects on PPO1 function. Sequence and structure analysis revealed that the two sites were highly conserved among 74 plant species, where the frequency of glycine was 94.6% and 100%, respectively, and adjacent to the entrance of the hydrophobic pocket of the active site. The M092141 and M091098 mutants can be used as important germplasms to develop wheat cultivars with low grain PPO activity. This study provided important insights into the molecular mechanism of Ppo1 and the genetic improvement of wheat PPO activity.

Abstract:Two fractions of polysaccharides, named PTPS-1 and PTPS-2, was extracted and purified from puerh tea by Sephacryl S-300 column chromatography.The physicochemical properties of these two polysaccharides were investigated by high-performance liquid chromatography (HPLC), Fourier Transform IR spectra, scanning electron microscope (SEM) and atomic force microscope (AFM).Analysis of chemical compositions (protein, neutral sugars, uronic acid and monosaccharide composition) suggested that they were both kinds of acid heteropolysaccharides bound with protein, and contained seven monosaccharides with different molar ratio.Meanwhile, evaluation of antioxidant activities by in vitro assays of DPPH, ABTS and FRAP showed that PTPS-1 demonstrated stronger antioxidant ability than PTPS-2.Similarly, PTPS-1 exhibited remarkable inhibitory potential on α-glycosidase in vitro, significantly stronger than that of PTPS-2 and acarbose.Moreover, the results from animal test indicated PTPS-1 possessed significant inhibition on postprandial hyperglycemia in diabetic mice compared with the model group.

Abstract:BACKGROUND According to recent International Organization for Standardization (ISO) standards, the limit of detection (LoD) of an assay should be estimated taking both type I (alpha) and II (beta) errors into account. The suggested procedure, however, supposes gaussian distributions of both blank and sample measurements and a linear calibration curve. In clinical chemistry, asymmetric, nongaussian blank distributions are common, and the calibration curve may be nonlinear. We present a partly nonparametric procedure that takes these aspects into account. METHODS Using theoretical distribution models and simulation studies, we developed a LoD estimation procedure suitable for the field of clinical chemistry that is partly based on nonparametric statistics. RESULTS For sample size n, the nonparametrically determined 95th percentile of the blank measurements obtained as the value of the [n(95/100) + 0.5]th ordered observation defines the limit for results significantly exceeding zero [limit of blank (LoB)]. The LoD is the lowest value that is likely to yield a result exceeding the LoB. LoD is estimated as: LoB + cbeta x SDS, where SDS is the analytical SD of a sample with a low concentration; cbeta = z(1 - beta)/[1 - 1/(4 x f)]; z(1 - beta) is the standard normal deviate; and f is the number of degrees of freedom for estimation of SD(S). c(beta) is approximately equal to 1.65 for a type II error of 5%. Approaches and needed tabular values for calculation of confidence limits are presented as well as sample size. Worked examples are given to illustrate estimation and verification of the limit of detection. Simulation results are used to document performance. CONCLUSION The proposed procedure appears useful for application in the field of clinical chemistry and promotes a standardized approach for estimating LoDs of clinical chemistry assays.

Abstract:Wheat bran is a major by-product of wheat processing, but the structural characterization and antioxidant activity of the polysaccharide are not clear. The manuscript aimed to preliminarily investigate the physicochemical properties, glycoside linkages and antioxidant activity of wheat bran polysaccharide. In the present study, one novel polysaccharide (WBP) was extracted by hot water method, isolated and purified by Sephadex G-100 chromatography. The physiochemical properties and structure were primarily analyzed by HPLC chromatography, FT-IR spectrum, NMR spectra, SEM, monosaccharide composition and methylation. The results revealed that WBP (Mw = 2.99 ×  105 Da) was mainly consisted of arabinose, xylose, mannose, glucose, and galactose in the ratio of 5.90: 6.25: 1.00: 2.06: 4.74. In addition, wheat bran polysaccharides exhibited remarkable antioxidant activities according to DPPH, ABTS, hydroxyl and superoxide radical scavenging tests. The manuscript provides theoretical basis for the utilization of wheat bran in functional food or medicine industries.

Should women be treated with testosterone?

Abstract:The use of testosterone therapeutically in women is a controversial issue in contemporary endocrinology and gynaecology. Perhaps least debated is its administration to postmenopausal women, particularly those who have undergone bilateral oophorectomy, and who complain of loss of libido and impaired sexual function, despite adequate oestrogen replacement (Burgeret al., 1984, 1987; Sherwinet al., 1985; Davis et al., 1995; Davis & Burger, 1996). In these circumstances, combined oestrogentestosterone replacement results in substantial benefit, without significant adverse effects. The rationale for such treatment is based on one of the known consequences of bilateral oophorectomy, a 50% fall in circulating testosterone concentrations. Testosterone supplementation, particularly using a 50 mg implant of crystalline testosterone approximately every 6 months in combination with oestradiol 50 mg causes no adverse changes in circulating lipids. In fact the favourable effects of postmenopausal oestrogen replacement on total and LDL-cholesterol are not diminished by the addition of either oral or parenteral testosterone replacement (Watts et al., 1995; Daviset al., 1995). Even for this indication, however, testosterone therapy for women is not in widespread use: it is not officially approved of by most regulatory authorities. The dose of testosterone required for therapeutic efficacy in postmenopausal women has not been established rigorously, though published experience indicates that 50 mg every 6 months provides satisfactory benefit (Burgeret al., 1987). At this dose virilizing side-effects have not been encountered and it may be that 100 mg or more is in excess of therapeutic requirements. Administration of androgens to women in their reproductive years, with intact ovarian function, has been much less studied. Hence the paper by Bucklert al. (1998) in this issue of Clinical Endocrinology addressing the role of androgen replacement in the management of PMS tackles a very common and important women’s health issue. However, the design of the study and the conclusions reached merit further discussion. The study was small, 22 women in each group, and no data are given concerning the definition of premenstrual syndrome (PMS) nor how severity was ascertained. However, the major methodological flaw is that no baseline data are provided for any parameter. Therefore, not only do we not know whether the two groups were comparable pretherapy, we also do not know the effect of treatment from baseline within the group who received testosterone. There are no placebo treated controls. No indication is given regarding the number of women who discontinued testosterone therapy which had been initiated for PMS, and it is for the reader to assume that those who persisted with treatment for an average of 3·3 years experienced some benefit. Since we therefore have no evidence that the two groups of women are actually comparable it is impossible to be confident of the conclusions drawn. This is further underlined by the probability of selection bias and the possibility of a Type II error. The authors describe their testosterone treatment as ‘low dose’ though this description can be argued. In fact, reported mean testosterone levels were more than twice the upper limit of normal, at 4·5 nmol/l compared with 1·0 nmol in the control group. Clinically, it is somewhat surprising that none of the women complained of masculinizing side-effects but we are given no indication that such information was formally sought or possible changes evaluated. The only adverse metabolic effects were a 16% decrease in apolipoprotein A1 levels and a 13% decrease in high density lipoprotein cholesterol levels in comparison with controls. There was a doubling of very low lipoprotein cholesterol levels in the treated patients. There were, however, no changes of significance in other lipid markers nor markers of activation of the coagulation cascade. The significance of the lipid changes is difficult to assess critically. This group of women received treatment over an average three year period but it is unlikely that they would be treated for more than a total of 5–10 years given that their average age at study was already 39·6 years. Hence, whether these adverse lipid changes would really be of overall cardiovascular significance is uncertain but perhaps not great. A lower dose of testosterone could be expected to have a less adverse effect and might be therapeutically equivalent. The therapeutic use of testosterone in PMS has not been advocated widely and is to some degree a novel concept. It would be of great interest to establish whether it has a scientific rationale. Relevant information to take into account in this context is the study by Zumoff et al. (1995) which showed that 24 h mean plasma testosterone concentrations declined by 50% when women in their early 40 s were compared with 20 year olds. Abstract:Longan polyphenoloxidase (PPO) was extracted and partially purified using ammonium sulfate precipitation and dialysis. The PPO characterizations were compared using endogenous substrate (-)-epicatechin and exogenous substrate catechol. The optimal pH and optimal temperature for the PPO activity were different when reacting with both substrates. The addition of ethylenediaminetetraacetic acid disodium salt into both substrate-enzyme systems exhibited the same lowest inhibition of the PPO activity. L-ascorbic acid and L-cysteine were the best inhibitors to endogenous substrate-enzyme system, while L-ascorbic acid and glutathione were most effective inhibitors to exogenous substrate-enzyme system. Cupric (Cu2+), ferric (Fe3+), and ferrous (Fe2+) ions accelerated the enzymatic-catalyzed reactions of both substrates. Kinetic analysis indicated that longan PPO strongly bound endogenous substrate but possessed a higher catalytic efficiency to exogenous substrate, and moreover, (-)-epicatechin was determined as the optimal substrate for longan PPO. This study is useful to exactly illuminate the enzymatic-catalyzed browning mechanism of postharvest longan fruit.
Polyphenol Oxidase Dominates the Conversions of Flavonol Glycosides in Tea Leaves

Abstract:Flavonol glycosides are associated with astringency and bitterness of teas. To clarify the dominant enzymatic reaction of flavonol glycosides in tea leaves, the catalytic effects of polyphenol oxidase (PPO), peroxidase (POD) and beta-glucosidase were studied, with the maintaining rates of total flavonol glycosides (TFG) being 73.0%, 99.8% and 94.3%. PPO was selected for further investigations, including the effects of pH value (3.5 similar to 6.5), temperature (25 degrees C similar to 55 degrees C) and dosage (39 similar to 72 U/mL PPO and 36 U/mL PPO, 3 similar to 36 U/mL POD). The oxidation of flavonol glycosides were intensified at pH 6.5, with 51.8% and 15.4% of TFG maintained after PPO and PPO + POD treatments, suggesting an enhancement from POD. The sensitivity ranking to PPO was: myricetin glycosides > quercetin glycosides > kaempferol glycosides. The inhibitor treatment testified the leading role of PPO in catalyzing flavonol glycosides in tea leaves. Sugar moiety enhanced the docking affinity of flavonol glycosides for PPO. PPO shows the potential of modifying flavonol glycoside composition.
Simple and efficient method for the extraction, isolation, and purification of peroxidase enzyme from wheat bran

Abstract:Peroxidase is an industrially important enzyme with a broad range of applications. In this study, the extraction, isolation, and purification of peroxidase was conducted with wheat bran as a feedstock. Several commercially available buffers (K2HPO4, Na3PO4, CH3COONa, and Tris‐base) were explored for the extraction of peroxidase from wheat bran. Of the tested media, sodium acetate buffer gave an excellent performance and displayed maximum protein (0.44 mg mL−1) and enzyme activity (28 U mL−1). A process optimization study was also conducted to obtain the maximum protein concentration and enzyme activity. The intensified process provided 1.39 mg mL−1 protein concentration with 90 U mL−1 enzyme activity with specific enzyme activity of 135.8 U mg−1. Crude enzyme was purified using a three‐step process: (a) precipitation, (b) ultrafiltration, and (c) chromatographic purification. Commercially available strong and weak ion exchange resins, with different surface properties, were tested for purification. The weak anion exchange resin showed an excellent performance for maximum binding of peroxidase. Adsorption study investigated the best fit model of Freundlich isotherm with maximum binding capacity of 54 mg mL−1. Overall, purification provided peroxidase with a protein concentration of 1.24 mg mL−1 and 727.58 U mg −1 specific enzyme activity with purity increased by around ~6.6 fold. The reported process provides a simple and efficient method for the extraction and purification of peroxidase enzyme from wheat bran for its efficient valorization.

Abstract:There has been considerable interest in using natural polyphenol oxidase (PPO) inhibitors to control browning in fruit and vegetable products. p-Coumaric acid (pCA), a common secondary metabolite of plants, has been studied as an inhibitor of PPOs/tyrosinases from several foods (e.g., mushroom, apple, and potato). However, studies on the use of pCA for the inhibition of PPO-initiated browning in actual food systems are limited. Therefore, a study was carried out to ascertain the efficacy of using pCA to limit PPO-initiated browning in fresh potato juice. The extent of browning inhibition by pCA was shown to be reaction system-dependent. Browning in potato juice was unexpectedly enhanced by the addition of pCA. This was interpreted as pCA acting as an alternative substrate with significantly higher browning efficiency; extent of browning under this condition was higher than that observed in the native potato juice. The addition of pCA to any of the model reaction mixtures (i.e., those containing semi-purified enzymes and substrates) significantly inhibited browning. The discrepancy in pCA effects on browning inhibition in different reaction systems is postulated to be mainly due to non-enzyme and non-substrate components in potato juice that participate in the post-PPO reaction sequences, which ultimately lead to brown color formation.

Abstract:The polyphenol oxidase was extracted and purified from truffles (Terfezia arenaria) and it exhibited a molecular weight of 67 kDa. The truffle PPO was able to oxidize monophenolic, o-diphenolic and triphenolic substrates. Thus, the enzyme seems to be stable under wide range of pH and temperature. Best catalytic efficiency was observed for catechol as substrate (Kcat/km; 674.2 S-1 mM-1).The effect of detergents, chaotropic agents, metal ions and eleven different inhibitors on relative activity of Truffles PPO was also investigated. A latent form of enzyme was observed and its activity was stimulated using 4 mM of SDS. Likewise, the type of inhibition and the values of KI and IC50 were reported for L-cysteine, Sodium fluoride, sodium metabisulfite and kojic acid. Besides, the effect of four concentrations of kojic acid(0.05.,0.1.,0.2 and 0.3 mM) on thermal inactivation of PPO was performed in temperature range " 60-75° C". The use of Kojic acid increase the rate of inactivation process and disrupt enzymatic activity. Moreover, the combined effect of temperature and kojic acid prevent from enzymatic browning reaction and maintain high antioxidant activities including ABTS scavenging activity, FRAP, and total phenolic contents.
Realistic magnetohydrodynamical simulation of solar local supergranulation

Abstract:Three-dimensional numerical simulations of solar surface magnetoconvection using realistic model physics are conducted. The thermal structure of convective motions into the upper radiative layers of the photosphere, the main scales of convective cells and the penetration depths of convection are investigated. We take part of the solar photosphere with a size of 60×60 Mm2 in the horizontal direction and of depth 20 Mm from the level of the visible solar surface. We use a realistic initial model of the sun and apply the equation of state and opacities of stellar matter. The equations of fully compressible radiation magnetohydrodynamics (MHD) with dynamical viscosity and gravity are solved. We apply (i) the conservative total variation diminishing (TVD) difference scheme for MHD, (ii) the diffusion approximation for radiative transfer and (iii) dynamical viscosity from subgrid-scale modeling. In simulation, we take a uniform two-dimensional grid in the horizontal plane and a nonuniform grid in the vertical direction with the number of cells being 600×600×204. We use 512 processors with distributed memory multiprocessors on the supercomputer MVS-100k at the Joint Computational Centre of the Russian Academy of Sciences.

Abstract:Polyphenol oxidase (PPO) from Cucumeropsis edulis was extracted and partially purified through (NH4)2SO4 precipitation, dialysis, and ion-exchange chromatography on DEAE-Sephadex-A50. The spectrophotometric method was used to assay the enzyme activity in C. edulis using L-DOPA as substrate, the physicochemical properties such as the effect of pH and temperature, substrate specificity, kinetic constants - maximum enzyme velocity (Vmax), and Michaelis - Menten constant (Km) for three substrates namely, L-Dopa catechol and tyrosine were determined. The effects of inhibitors and metal ions on PPO activity were also investigated. The optimum pH and temperature values were found to be pH 6.5 and 50 °C, and the inhibitory effects of inhibitors such as ascorbic acid, EDTA, SDS, and metal ions were enhanced positively with increased concentration except with divalent metals such as Cu2+, Fe2+, and Zn2+ reflecting an activating effect on C. edulis PPO. Moreover, the enzyme solution showed both monophenolase and diphenolase activity with L-DOPA having the highest Vmax/Km value. However, the data obtained in this research provided a theoretical basis for the prevention of enzymatic browning of C. edulis during processing.

Abstract:Fennel seeds were recognized as a promising polyphenol oxidase (PPO) source upon investigating some edible green plants (carob, jujube, coriander, fennel, and licorice). The fennel PPO enzyme was purified by three-phase partitioning and biochemically characterized in detail for the first time. The purification fold and activity recovery values were determined as 20-fold and 120%, respectively. Its molecular weight was 27.8 kDa. The temperature for the selected substrates (catechol, 4-tert-butylcatechol, 4-methylcatechol, and pyrogallol) was 30 °C, while the optimum pH value varied from 5.0 to 7.0 depending on the substrate. The kcat/Km values exhibited that the enzyme presented the best activity towards catechol among the substrates used. Sodium metabisulfite, ascorbic acid, benzoic acid, l-cysteine, thiourea, β-mercaptoethanol, and glutathione prominently inhibited PPO activity. A remarkable decrease in PPO activity was observed at elevated concentrations of organic solvents, but in cases of the solvents with polarity indexes ≥5.1, the residual activity maintained more than 75% of its original activity up to 10% (v/v). Consequently, the current study suggested that fennel seeds could be used in various industrial sectors to produce low-cost polyphenol oxidase enzymes with an agricultural origin.

Abstract:Potato peel from food industrial waste is a good source of polyphenol oxidase (PPO). This work illustrates the application of an aqueous two-phase system (ATPS) for the extraction and purification of PPO from potato peel. ATPS was composed of polyethylene glycol (PEG) and potassium phosphate buffer. Effect of different process parameters, namely, PEG, potassium phosphate buffer, NaCl concentration, and pH of the system, on partition coefficient, purification factor, and yield of PPO enzyme were evaluated. Response surface methodology (RSM) was utilized as a statistical tool for the optimization of ATPS. Optimized experimental conditions were found to be PEG1500 17.62% (w/w), potassium phosphate buffer 15.11% (w/w), and NaCl 2.08 mM at pH 7. At optimized condition, maximum partition coefficient, purification factor, and yield were found to be 3.7, 4.5, and 77.8%, respectively. After partial purification of PPO from ATPS, further purification was done by gel chromatography where its purity was increased up to 12.6-fold. The purified PPO enzyme was characterized by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), followed by Km value 3.3 mM, and Vmax value 3333 U/mL, and enzyme stable ranges for temperature and pH of PPO were determined. These results revealed that ATPS would be an attractive option for obtaining purified PPO from waste potato peel.

Abstract:The activity of polyphenol oxidase(PPO) produced from Trametes Trogii was determined through using tea polyphenols as substrate.The pH value and the substrate concentration of strongest activity of PPO are 5.0 and 4 mg/mL,respectively.The activity of PPO processed by ultrahigh pressure of 100,300 and 500 MPa for 10 min accounted for 93%,87% and 86% of that of untreated one.The CD spectra of PPO processed by 100 and 300 MPa pressure respectively displayed folding shape and tillering shape of Cotton effect peak located in 190-210 nm,while the retention time of the first peak fraction(i.e.,the maximum molecular weight fraction) of the two processed PPO on HPGPC profiles were advanced to 1.514 and 2.296 min,respectively from 6.678 min.Compared with control,CD spectrum and HPGPC of PPO treaded by 500 MPa showed more significant difference with only one positive Cotton effect peaks on CD spectra and reduced peak area of main components on HPGPC profiles(ie,the response value of 280 nm decreased).The disparity of CD and HPGPC of PPO treated by various UHPP showed the corresponding difference in the degree
