Abstract:Metal ions are widely distributed in nature and they have key roles in the environment, bioscience and medical science. Some metal ions,such as Hg2+,Fe3+,Cu2+,Zn2+and Al3+,play important roles in the physiology and pathology of organisms. An excess or deficiency of these metal ions would lead to physiological dysfunction and cause a variety of diseases. Recently,reaction-based fluorescence probes have attracted considerable attention due to their high sensitivity and good selectivity. This review mainly focuses on reaction-based fluorescence probes for the detection of metal ions in the past five years. The design and synthesis of probes,their sensing mechanisms and their applications were summarized. Currently,the reaction-based fluorescence probes are still in their initial stages and hence many problems need to be solved. With the resolution of these problems, reaction-based fluorescence probes would show higher sensitivities with less reaction-time and possess wider applications. In the future,reaction-based fluorescence probes should show efficient development in biological detection and expand practical applications.

Abstract:Metal ions have a significant impact on the environment and the growth and development of organism,therefore,the recognition and detection of metal ions in the environment and organism is gaining widespread attention. Among different analytical methods,fluorescence analysis has the advantages of high sensitivity,good selectivity,real-time and in situ detection,etc.,which is a good tool to achieve the recognition and detection of metal ions in the environment and organism. This paper is aimed to review the coumarin-based fluorescent sensors for the recognition and detection of heavy metal ions (Hg2+、Pb2+、Cd2+、Ni2+、Ag+),biologically important transition metal ions (Cu2+、Zn2+、Fe3+) and their applications in recent five years,focusing on the design and synthesis of sensors, recognition mechanism,sensing characteristics and applications. With the higher requirement for metal ions detection , the design of coumarin-based fluorescent sensors will be developed for better performance with higher sensitivity,selectivity and stronger anti-interference in the future. In addition, applications of coumarin-based fluorescent sensors in biological detection are expected to be further developed.

Abstract:Good sense: A highly sensitive and selective ratiometric fluorescent probe for Fe2+ was developed by linking an Fe2+-insensitive fluorophore to an Fe2+-sensitive reporter–fluorophore fragment. The fluorescence intensity ratio increased linearly with the concentration of Fe2+ (see scheme). The probe was applied to the ratiometric imaging of intracellular and adscititious fluctuations of Fe2+ in live HL-7702 and HepG2 cells.

Abstract:The metalloprotein, CadR, was redesigned to optimize cadmium and mercury specificity of CadR-based E. coli biosensors. By truncating 10 and 21 amino acids from the C- terminal extension of CadR, CadR-TC10 and CadR-TC21 were obtained, respectively. The genes cadR , cadR - TC10 and cadR - TC21 were used as sensing elements to construct green fluorescent protein based E.coli biosensors. Induction at 30 °C for 4 h in supplemented M9 medium was the optimized condition for the biosensor. Compared with CadR-based biosensor, there was a clear decline in induction coefficient for CadR-TC21-based biosensor (decreased by 86 % in Zn(II), 44 % in Hg(II), and only 37 % in Cd(II)). While in CadR-TC10-based biosensor, the induction coefficient decreased by 95 % in Zn(II), 70 % in Hg(II), and 67 % in Cd(II). Improved performances of CadR mutants based E. coli biosensors indicated that truncating C- terminal extension of CadR could improve the specificity.

Abstract:The recognition and sensing of the biologically and environmentally important metal ions has emerged as a significant goal in the field of chemical sensors in recent years. Among the various analytical methods, fluorescence has been a powerful tool due to its simplicity, high detection limit and application to bioimaging. This review highlights the fluorescent chemosensors for metal ions, which have been recently developed from our laboratory. This review was categorized by target metal ions, such as Cu2+, Hg2+, Zn2+, Pb2+, Cd2+, Vanadate, Ag+ and Au3+. Selectivity and sensitivity for these metal ions were achieved by introducing various ligands to core fluorophores, such as, rhodamine, fluorescein, pyrene, anthracene, naphthalimide, coumarin, and BODIPY.

Abstract:Many different metal ions are involved in various biological functions including metallomics and trafficking, and yet there are currently effective sensors for only a few metal ions, despite the first report of metal sensors for calcium more than 40 years ago. To expand upon the number of metal ions that can be probed in biological systems, we and other laboratories employ the in vitro selection method to obtain metal-specific DNAzymes with high specificity for a metal ion and then convert these DNAzymes into fluorescent sensors for these metal ions using a catalytic beacon approach. In this Forum Article, we summarize recent progress made in developing these DNAzyme sensors to probe metal ions in living cells and in vivo, including several challenges that we were able to overcome for this application, such as DNAzyme delivery, spatiotemporal control, and signal amplification. Furthermore, we have identified a key remaining challenge for the quantitative detection of metal ions in living cells and present a new design and the results of a Förster resonance energy transfer (FRET)-based DNAzyme sensor for the ratiometric quantification of Zn2+ in HeLa cells. By converting existing DNAzyme sensors into a ratiometric readout without compromising the fundamental catalytic function of the DNAzymes, this FRET-based ratiometric DNAzyme design can readily be applied to other DNAzyme sensors as a major advance in the field to develop much more quantitative metal-ion probes for biological systems.

Abstract:Transition metals play essential roles in biology, where these nutrients regulate protein activity as active site cofactors or via metalloallostery. In contrast, dysregulation of transition metal homeostasis can lead to unique metal-dependent signaling pathways connected to aging and disease, such as cuproptosis and ferroptosis for copper- and iron-dependent cell death or cuproplasia and ferroplasia for copper- and iron-dependent cell growth and proliferation, respectively. New methods that enable detection of bioavailable transition metal pools with both metal and oxidation state specificity can help decipher their contributions to health and disease. Here we summarize recent advances in designing sensors for imaging transition metals and their applications to uncover new metal biology.

Fluorescent DNAzyme biosensors for metal ions based on catalytic molecular beacons.

Abstract:In this chapter, methods for designing metal ion sensors using fluorophore- and quencher-labeled DNAzymes are discussed. In contrast to the classical molecular beacon method based on binding, the methods described here utilize catalytic cleavage to release the fluorophore for detection and quantification, making it possible to take advantage of catalytic turnovers for signal amplification. Unlike classical molecular beacons that detect only nucleic acids, catalytic molecular beacons can be applied to different DNAzymes to detect a broad range of analytes. The methods described are based on the finding that almost all known trans-cleaving DNAzymes share a similar structure comprised of a catalytic DNAzyme core flanked by two substrate recognition arms. Using a typical DNAzyme called the "8-17" DNAzyme as an example, the design of highly sensitive and selective Pb2+ sensors is described in detail. The initial design employs a single fluorophore-quencher pair in close proximity, with the fluorophore on the 5'-end of the substrate and the quencher on the 3'-end of the enzyme. Although this sensor is highly sensitive and selective at 4 degrees C, high background fluorescence is observed at higher temperatures. Therefore a new design with an additional quencher attached to the 3'-end of the substrate is employed to suppress background fluorescence. The dual quencher method allows the sensor to perform at ambient temperatures with a high signal-to-noise ratio.

Abstract:Toxic cations, including heavy metals, pose significant environmental and health risks, necessitating the development of reliable detection methods. This review investigates the techniques and approaches used to strengthen the sensitivity and selectivity of Schiff base fluorescent chemosensors designed specifically to detect toxic and heavy metal cations. The paper explores a range of strategies, including functional group variations, structural modifications, and the integration of nanomaterials or auxiliary receptors, to amplify the efficiency of these chemosensors. By improving selectivity towards targeted cations and achieving heightened sensitivity and detection limits, consequently, these strategies contribute to the advancement of accurate and efficient detection methods while increasing the range of end-use applications. The findings discussed in this review offer valuable insights into the potential of leveraging Schiff base fluorescent chemosensors for the accurate and reliable detection and monitoring of heavy metal cations in various fields, including environmental monitoring, biomedical research, and industrial safety.

Abstract:Copper is an essential nutrient for sustaining life, and emerging data have expanded the roles of this metal in biology from its canonical functions as a static enzyme cofactor to dynamic functions as a transition metal signal. At the same time, loosely bound, labile copper pools can trigger oxidative stress and damaging events that are detrimental if misregulated. The signal/stress dichotomy of copper motivates the development of new chemical tools to study its spatial and temporal distributions in native biological contexts such as living cells. Here, we report a family of fluorescent copper sensors built upon carbon-, silicon-, and phosphorus-substituted rhodol dyes that enable systematic tuning of excitation/emission colors from orange to near-infrared. These probes can detect changes in labile copper levels in living cells upon copper supplementation and/or depletion. We demonstrate the ability of the carbon-rhodol based congener, Copper Carbo Fluor 1 (CCF1), to identify elevations in labile copper pools in the Atp7a-/- fibroblast cell model of the genetic copper disorder Menkes disease. Moreover, we showcase the utility of the red-emitting phosphorus-rhodol based dye Copper Phosphorus Fluor 1 (CPF1) in dual-color, dual-analyte imaging experiments with the green-emitting calcium indicator Calcium Green-1 to enable simultaneous detection of fluctuations in copper and calcium pools in living cells. The results provide a starting point for advancing tools to study the contributions of copper to health and disease and for exploiting the rapidly growing palette of heteroatom-substituted xanthene dyes to rationally tune the optical properties of fluorescent indicators for other biologically important analytes.

Abstract:Mercury exists naturally and mainly as a man-made pollutant in the environment, where it exerts adverse effects on local ecosystems and living organisms. It is important to develop an appropriate synthetic biological device that recognizes, detects and removes the bioavailable fraction of environmental mercury. Both single-signal and double-signal output mercury biosensors were assembled using a natural mer operon as a template. Selectivity and sensitivity of whole-cell biosensors based on artificial mer operons were determined. Three whole-cell biosensors were highly stable at very high concentrations of mercuric chloride, and could detect bioavailable Hg(II) in the concentration range of 6.25–200 μM HgCl 2 . A novel Hg(II) bioadsorption coupled with biosensing artificial mer operon was assembled. This would allow Hg(II)-induced Hg(II) binding protein cell surface display and green fluorescence emission to be achieved simultaneously while retaining the linear relationship between fluorescent signal and Hg(II) exposure concentration. The present study provides an innovative way to simultaneously detect, quantify, and remove bioavailable heavy metal ions using an artificially reconstructed heavy metal resistance operon.

Abstract:Mercury, as a highly toxic heavy metal, can cause very serious harm to human health and even death in severe cases. Therefore, we synthesized a novel ratiometric fluorescent probe for detecting mercury ions, with mercaptoethanol as the recognition receptor. Probe CMER could determine mercury ions in 0-1.6 μM and the detection limit is 7.6 nM. Moreover, CMER manifested a fast response for Hg2+ (within 5 s) and simultaneously observed that the color changed from light yellow to orange by naked eye. In addition to these preeminent spectral properties, the probe also had satisfactory bioimaging results in RAW 264.7 macrophage cells and zebrafish.

Abstract:Metal ions are ubiquitous elements within our environments. Their excessive accumulation poses a great threat to human health. With the development of biology and bioanalytical chemistry, the role played by metal ions in many biological and chemical processes is revealed. Since trace amounts of metal ions can lead to essential influences, there is a growing demand for the study of highly sensitive and selective sensors. Among them, functional nucleic acid (FNA)-based fluorescent probes are proposed as ideal options. The programmability of FNA is constantly integrated with various kinds of fluorophores and strategies for efficient sensing assays. However, a comprehensive review of their advancements in medical and chemical areas is still needed. This review aims to summarize the FNA-based fluorescent methods for the detection and imaging of various metal ions. We focus on the unique properties of metal ions and versatile design strategies. In principle, each signaling strategy can be employed for different FNA-based sensors. Besides, the applications of DNA-templated nanomaterials, conformation-specific fluorophores and isothermal amplification are discussed. This comprehensive review can inspire more advanced fluorescent methods for metal ion sensing and promote the development of FNA for biomedical applications.

Abstract:An easily prepared "turn-on" -type fluorescent chemosensor for mercury based on Rhodamine-B, which exhibits high sensitivity and selectivity over other metal ions in aqueous systems, was obtained. The distinctive wavelengths in the UV/ Vis absorption range can sense Hg 2+ , Cu 2+ , and Fe 3+ separately. Furthermore, this "turn-on"-type fluorescent sensor, upon the addition of Hg 2+ over other competitive species, was successfully applied to bioimaging in yeast and HeLa cells. The potential of these types of chemosensors for use in environmental and biological systems is great.

Abstract:Metal ions play a crucial role in many biochemical processes, and when in a state of scarcity or surplus, they can lead to various diseases. Therefore, the development of a selective, sensitive, cost-effective, and fast-responding sensor to detect metal ions is critical for in vitro medical diagnostics. In recent years, fluorescent sensors have been extensively investigated as potent kits for the effective assessment of metal ions in living systems due to their high sensitivity, selectivity, ability to perform real-time, non-invasive monitoring, and versatility. This review is an overview of recent advances in fluorescent sensors for the detection and imaging of metal ions in biosystems from 2018 to date. Specifically, we discuss their application in detecting essential metal ions and non-essential metal ions for in vitro diagnostics, living cell imaging, and in vivo imaging. Finally, we summarize remaining challenges and offer a future outlook on the above topics.
