Abstract:On the basis of fluorescent resonance energy transfer, a new fluorophore dyad (L) bearing rhodamine B and naphthalimide was developed as fluorescent ratiometric chemosensor for Hg2+ in aqueous solution. L exhibited high selectivity and excellent sensitivity towards Hg2+ with a broad pH span (1.0-8.0) and the detection limit of L was 2.11 x 10(-8) M. Sensor L for the detection of Hg2+ was rapid and the recognizing event could complete in 2.5 min. A significant change in the color could be used for naked-eye detection. The selective fluorescence response of L to Hg2+ is due to the Hg2+-promoted ring opening of spirolactam of rhodamine moiety, leading to a cyclization reaction of thiourea moiety. Abstract:An effective Hg2+-specific probe (1) was designed and prepared based on a rhodamine–nitrobenzoxadiazole conjugate. The sensing behavior of this probe was studied by UV–visible and fluorescence spectroscopy. With Hg2+, the probe displayed both colorimetric responses and ratiometric changes. The ratio of the absorption intensities at 525 and 460nm for the two chromophores in the probe increased from 0.17 to 0.46. The probe exhibited excellent selectivity and sensitivity towards Hg2+ over other metal ions, which could be attributed to the Hg2+-triggered ring opening of the spirolactam of the rhodamine moiety and the formation of a 1:1 1-Hg2+ complex.

Abstract:Two novel rhodamine-pyrene conjugated chemosensors were successfully designed and synthesized, which exhibited high affinity to Hg2+ ions. As ratiometric chemosensors, each displayed highly selective and sensitive colorimetric and fluorogenic dual-responses toward Hg2+. The comparison of two chemosensors indicated that the spirothiolactone containing sensor was superior to the spirolactone analog in sensing behavior when detecting Hg2+, presumably due to the thiophilic nature of mercury and the different sensing mechanisms in operation. Upon interaction with Hg2+, the spirothiolactone showed a 1:1 stoichiometry for the Hg2+ complex, accompanied with a weakened fluorescence resonance energy transfer (FRET) behavior. However, the spirolactone was hydrolyzed and consequently induced the monomer-excimer switch of the resulted pyrene in the presence of Hg2+ ions. Crown Copyright (c) 2013 Published by Elsevier Ltd. Abstract:Inspired by dual-signaling ratiometric mechanism which could reduce the influence of the environmental change, a novel, convenient, and reliable method for the detection of mercury ions (Hg(2+)) based on Y-shaped DNA (Y-DNA) was developed. Firstly, the Y-DNA was formed via the simple annealing way of using two different redox probes simultaneously, omitting the multiple operation steps on the electrode. The Y-DNA was immobilized on the gold electrode surface and then an obvious ferrocene (Fc) signal and a weak methylene blue (MB) signal were observed. Upon addition of Hg(2+), the Y-DNA structure was transformed to hairpin structure based on the formation of T-Hg(2+)-T complex. During the transformation, the redox MB gets close to and the redox Fc gets far away from the electrode surface, respectively. This special design allows a reliable Hg(2+) detection with a detection range from 1 nM to 5 μM and a low detection limit down to 0.094 nM. Furthermore, this biosensor exhibits good selectivity and repeatability, and can be easily regenerated by using L-cysteine. This study offers a simple and effective method for designing ratiometric biosensors for detecting other ions and biomolecules.

Abstract:A ratiometric near-infrared (NIR) fluorescent probe L based on chromenylium-cyanine fluorophore with chelate unit NS2O2 for the detection of Hg2+ ions in aqueous media has been prepared. The probe L shows high selectivity for Hg2+ ions with immediately fluorescence intensity increasing at 663 nm and decreasing at 728 nm synchronously via a Hg2+-induced intramolecular charge transfer (ICT) mechanism, leading to both "naked-eye" and ratiometric detection of Hg2+ ions in real time. At the same time, the probe exhibits a high sensitivity to Hg2+ in a concentration of ppb range with detection limit of 9.59 nM. Moreover, the probe is cell permeable and low toxic, and can be used in living cells for ratiometric visual monitoring of Hg2+ ions by confocal laser scanning fluorescence microscopy.

Abstract:A highly selective fluorescence sensor was developed for Hg(II) ion detection in aqueous Solutions based on the selective binding of Hg(II) ions with a pair of thymine-thymine mismatch. The sensor consists of two DNA probes functionalized with a fluorophore (fluorescein, F) and a quencher (tetramethyl rhodamine, Q) moiety separately. This pair of DNA probes contains two pairs of thymine-thymine mismatches used to detect Hg(II) ions. In the presence of Hg(II) ions, thymine-Hg2+-thymine was formed between thymine residues of probes. From that, the interaction of the two DNA probes increased. Thus, the DNA probes formed a double-stranded structure. Both the fluorophore and quencher were brought close to each other leading to the fluorescence resonance energy transfer (FRET) between F and Q. Under the optimum conditions, the sensor was used to detect the Hg(II) ions from 50 to 1000 nmol.L-1 with a regression equation y=5281.13-1650.56 lg[Hg2+] (R-2=0.985). The linear range covers 100 to 500 nmol.L-1, and the limit of detection (LOD) is 79 nmol.L-1. The disturbance of some co-existing metal ions was explored, and no significant fluorescence quenching in the presence of 1.0 mu mol-L-1 other metal ions was observed. The fluorescence sensor has good sensitivity and selectivity for Hg(II) ions providing a rapid, simple and low cost method for the detection of memcury(II) ions in aqueous solutions.

Abstract:A novel fluorescent rhodamine based chemosensor (E)-3′,6′-bis(diethylamino)-2-((2-(pyridin-2-ylmethoxy)benzylidene)amino)spiro[isoindoline-1,9′-xanthen]-3-one, RSP, had been successfully developed and well characterized by NMR, FT-IR and Mass spectroscopy. The chemosensor exhibits high selectivity for Hg2+ over other ions (Ag+, Pb2+, Cu2+, Ni2+, Fe3+, Co2+, Zn2+ and Cd2+) with fluorescence enhancement in ethanol solution. More over the detection limit of the sensor is in the 10−6 M level. The binding ratio of RSP-Hg2+ complex was determined to be 1:1 according to the Job plot. Test strips based on RSP were fabricated, which showed the application of the sensor for detection of mercuric ions in water by naked eyes.

Abstract:A novel Hg2+-selective fluorescent sensor (Sensor-Hg) based on a rhodamine scaffold, with N-Ethylthioethyl-N-[N',N'-(2'-Diethylthioethylamino)-5'-methyl-Phenoxyethyl]-2-Methoxy Aniline (EDPMA) as a receptor, has been developed. Sensor-Hg exhibited similar to 35-fold enhancement in fluorescence emission upon binding with Hg2+, with a characteristic emission band of rhodamine at similar to 585 nm. It demonstrates high selectivity and sensitivity for detecting Hg2+ in aqueous solution and living cells.

Abstract:A new and simple fluorescent probe bearing 7-nitrobenz-2-oxa-1,3-diazole and N, N-bis(thiophen-2- ylmethyl) ethane-1,2-diamine was developed as a fluorescent chemosensor with high selectivity and sensitivity toward Hg2+ over other cations tested. The signaling is due to the coordination of S and N atoms of N, N-bis(thiophen-2-ylmethyl) ethane-1,2-diamine to Hg2+. Upon addition of 30 equivalents of Hg2+, a 45-fold increase in fluorescence emission intensity was observed. Moreover, the probe is further successfully used to image Hg2+ in MCF-7 cells.

Abstract:In this paper, we unveil a novel naphthalimide-porphyrin hybrid based fluorescence probe (1) for ratiometric detection of Hg2+ in aqueous solution and living cells. The ratiometric signal change of the probe is based on a carefully predesigned molecule containing two independent Hg2+-sensitive fluorophores with their maximal excitation wavelengths located at the same range, which shows reversibly specific ratiometric fluorescence responses induced by Hg2+. In the new developed sensing system, the emissions of the two fluorophores are well-resolved with a 125 nm difference between two emission maxima, which can avoid the emission spectra overlap problem generally met by spectra-shift type probes and is especially favorable for ratiometric imaging intracellular Hg2+. It also benefits from a large range of emission ratios and thereby a high sensitivity for Hg2+ detection. Under optimized experimental conditions, the probe exhibits a stable response for Hg2+ over a concentration range from 1.0 x 10(-7) to 5.0 x 10(-5) M, with a detection limit of 2.0 x 10(-8) M. The response of the probe toward Hg2+ is reversible and fast (response time less than 2 min). Most importantly, the ratiometric fluorescence changes of the probe are remarkably specific for Hg2+ in the presence of other abundant cellular metal ions (i.e., Na+, K+, Mg2+, and Ca2+), essential transition metal ions in celsl (such as Zn2+, Fe3+, Fe2+, Cu2+, Mn2+, Co2+, and Ni2+) and environmentally relevant heavy metal ions (Ag+, Pb2+, Cr3+, and Cd2+), which meets the selective requirements for biomedical and environmental monitoring application. The recovery test of Hg2+ in real water samples demonstrates the feasibility of the designed sensing system for Hg2+ assay in practical samples. It has also been used for ratiometric imaging of Hg2+ in living cells with satisfying resolution, which indicates that our novel designed probe has effectively avoided the general emission spectra overlap problem of other ratiometric probes.

Abstract:The detection of Hg2+ in biological systems and its imaging is of highly importance. In this work, a novel ratiometric fluorescence probe is developed based on through-bond energy transfer (TBET) with a 2-(2-hydroxyphenyl)benzoxazole (HBO) as donor and a Rhodamine derivative-Hg conjugate (RDM-Hg) as acceptor. Hg2+ weakens the fluorescence of HBO at 430 nm and meanwhile interacts with Rhodamine B derivative to form a fluorescent conjugate (RDM-Hg) giving rise to emission at 597 nm with a 167 nm red-shift. Further, the difference 282 nm between the donor absorption (315 nm) and the accepter emission (597 nm) for 1 + Hg2+ is comparable to the highest value of the Stokes shift (282 nm) reported earlier for other reported TBET-based cassette. Through-bond energy transfer from HBO to RDM-Hg is triggered by Hg2+ resulting in concentration dependent variation of fluorescence ratio I-597/I-430. A linear calibration of I-597/I-430 versus Hg2+ concentration is obtained within 0-5 mu M, along with the lowest detection limit being found to be as low as 1.31 x 10(-9) mol center dot l(-1) (similar to 0.26 ppb) for Hg2+. This feature is further demonstrated by colorimetric imaging of test strip and intracellular Hg2+. On the other hand, the HBO/RDM TBET sensing system is characterized by a combination of high sensitivity and selectivity. The present study provides an approach for further development of ratiometric probes dedicated to selective in vitro or in vivo sensing some species of biologically interest.

Abstract:A new multisignaling sensor based on rhodamine B with a ferrocene substituent (1) has been synthesized and has been shown to display extreme selectivity for Hg2+ over other metal ions. Multisignaling changes are observed through UV/vis absorption, fluorescence emission, and electrochemical measurements. Furthermore, by means of confocal laser scanning microscopy experiments, it is demonstrated that 1 can be used as a fluorescent probe for monitoring Hg2+ in living cells.

Abstract:Fluorescence analysis by means of a single fluorescence signal output usually leads to the signal fluctuation caused by various external factors. Ratiometric fluorescence probes that can significantly eliminate the external effects by self-calibration of two different emission bands are preferable for the detection of real samples. In this work, we designed a dual-emission quantum dots (QDs) nanocomposite as a ratiometric probe for the visual detection of Hg2+. The dual-emission QDs nanocomposite consists of two differently sized CdTe/CdS QDs. The red-emitting larger sized CdTe/CdS QDs embeded in silica nanoparticles are insensitive to Hg2+, while the green-emitting smaller sized ones are covalently conjugated onto the silica nanoparticles surface and sensitive to Hg2+. The addition of Hg2+ can only quench green fluorescence in the dual-emission QDs nanocomposites, which triggers the change of fluorescence intensity ratio of two different emission wavelengths and hence induces the evolution of fluorescence color of the probe solution with variation of Hg2+ concentration. Based on this feature, the dual-emission QDs nanocomposites can be used to develop a ratiometric fluorescence probe for the visual detection of Hg2+. Under the optimized conditions, the ratiometric fluorescence QDs probe shows a linear relationship between fluorescence intensity ratio and Hg2+ concentration in the range of 5–300nM. The detection limit of this probe was found to be 3.1nM. This ratiometric assay also exhibits a high selectivity and it has been successfully used in the determination of Hg2+ content in fetal bovine serum and human urine.

Abstract:A rhodamine-inspired fluorescence dye (2) bearing 7-diethylaminocoumarin fluorophore was designed as a platform for the construction of an NIR and ratiometric fluorescent probe. The ring-open form of 2 shows NIR absorption and emission; however, its ring-closed form displays the visible absorption and emission because an intact 7-diethylaminocoumarin fluorophore was involved in the structure, providing the basis for an NIR and ratiometric fluorescent platform based on the spirocyclization-induced fluorescence switching mechanism. With the platform, we developed a novel NIR and ratiometric fluorescent probe R1, a thiolactone of 2, for sensing Hg2+, an environmentally and biologically concerned species. R1 displays an emission peak with the maximum at 480 nm, which is the typical emission of 7-diethylaminocoumarin moiety; however, upon addition of Hg2+ ions, the emission intensity at 480 nm gradually decreased with the simultaneous appearance of a new NIR emission band centred at 695 nm. Thus, the Hg2+-promoted ratiometric fluorescence response can be realized. The high selectivity towards Hg2+ over various cations and the high stability in a wide pH range of 1-12 indicate its potential for applications in biological systems. The subsequent cell imaging experiment revealed that R1 is cell permeable, and could be employed for ratiometric fluorescence imaging of Hg2+ in living cells.

Abstract:A irreversible Hg2+ selective ratiometric fluorescence probe FR, a fluorescein fluorophore linked to a rhodamine B hydrazide by a thiourea spacer, was designed and synthesized. The developed probe FR exhibited great ratiometric fluorescence enhancement and remarkable yellow-magenta color change toward Hg2+ with excellent selectivity in aqueous acetone solution, and the ratiometric fluorescence response to Hg2+ was not interfered by other metal cations including Fe3+, Co2+, Ni2+, Cr3+, Zn2+, Pb2+, Cd2+, Ca2+, Mg2+, Ba2+ and Mn2+. The linear range and the detection limit of this supposed ratiometric fluorescence method for Hg2+ were 0.0-10.0 x 10(-6) and 5x10(-8) M, respectively.

The colorimetric and fluorescence responses, allowing naked-eye detections, are based on Hg 2+ -induced opening of the rhodamine spirocycle. Among all the testes ions, only Hg 2+ generated a significant fluorescence enhancement of up to 300-fold, with a bright yellow–green emission. This sensor was a low toxic compound, and was successfully applied in the in vivo imaging of Hg 2+ in Spill 2 cells and C. elegans. This approach provides a sensitive and accurate method for the estimation of Hg 2+ in environmental, tobacco and biological applications.
