Abstract:In order to determine the optimal conditions of polysaccharides extraction from seed shell of Juglans mandshurica,the content of polysaccharides was measured by the method of phenol-sulphate acid with the extraction yield of polysaccharides as evaluation index.Based on the single factor test,the extraction temperature,extraction time and solid-to-liquid ratio were selected as independent variables,and the extraction yield of polysaccharides was as response value,the extraction technology of polysaccharides from seed shell was optimized by response surface methodology (RSM).Results showed that,the variables were ranked in decreasing order as extraction time,solid-to-liquid ratio and extraction temperature.The optimized extraction conditions were found to be a extraction temperature of 82 ℃,extraction time of 3.7 h and a solid-to-liquid ratio of 1∶45(g/mL).The validation experiments indicated that,the extraction yield of polysaccharides from seed shell of Juglans mandshurica was 8.64 mg/g with RSD of 1.94％(n=5),and the experimental data could be well fitted with the developed regression equation.
