Expression of TGF-β1,BMP-2 and Smad4 in the development of the thoracic vertebrae of rat embryo and its significance

Abstract:Objective: To explore the expression of the transforming growth factor-β1 (TGF-β1), bone morphogenetic protein 2 (BMP-2) and Smad4 in the rat thoracic vertebrae of developing embryo. Methods: The presence of TGF-β1, BMP-2 and Smad4 in the thoracic vertebrae tissue of 90-100 specimens of 11-19 embryonic day (E11-E19) was investigated using immunohistochemical methods and RT-PCR. Results: The location of each factor expression had no conspicuous difference in the same embryonic day. The positive expressions of TGF-β1, BMP-2 and Smad4 were found in notochord constantly and then in the cells of nucleus pulposus. The expressions of all factors were less pronounced in sclerotomal cells or none at E11-E12. At E13-E19, they appeared in the chondroblasts and cartilaginous cells of developing vertebrae as well as in the cells of perichondrium at E14-E19 and enhanced with age, but weakly in the cells of periosteum; the expression of the cartilaginous matrix was negative. In addition, the positive expressions of TGF-β1 and BMP-2 were observed in the extracellular matrix (ECM) at E11-E13.5. However, their expression abundances were significantly different (P<0.05). Conclusion: TGF-β1, BMP-2 and Smad4 may interact with other extracellular molecules to co-regulate the migration and adhesion of the sclerotome and the formation of the perichordal tube and the cartilage matrix, promote the functions of cartilaginous cells of vertebrae such as proliferation, differentiation and mature, and then induce the osteoblastic process.
