Effect of Chronic Restraint on Behavioral Manifestation and Expression of Phosphorylated Extracellular Signal-Regulated Kinase in the Rat Prefrontal Cortex

Abstract:Objective To provide clues for understanding the molecular mechanism underlying chronic stress,the behavioral manifestation and changes of expression of phosphorylated ERK1/2 in the prefrontal cortex of the rat subjected to chronic restraint stress were investigated.Method Sixteen male SD rats were randomly divided into normal control group (NC) and restraint stress (RS) group.The rats in RS group were taken into the tailor-made restraint container for 6 hours everyday in 21 continuous days.The behavioral changes were examined with open-field test and Morris water maze,the levels of p-ERK1/2 in the prefrontal cortex with Western blotting and immunohistochemistry.Results As compared with controls,the rats in RS group showed reduced horizontal activity [ (4265±864) mm vs.(8562±502)mm ] and increased central time [(39.1±4.3)s vs.(24.6±1.6)s] (P0.01) in open-field test,and reduced time stayed at target quadrant [ (57.2±1.7) s vs.(70.7±3.6) s ] and frequency crossing the stage [ (2.0±0.8) times vs. (6.2±1.0)times] in Morris water maze as well.After stress,the level of p-ERK1/2 in prefrontal cortex PFC in RS group was lower than that in NC group (P0.05).However,there was no difference in the level of total ERK1/2 between two groups.The number of p-ERK1/2-IL neurons in PFC in RS group was less than that in ConNC group [ (76±5) times vs.(110±14) times ] and their dendrites were stained lightly.Conclusion Chronic psychological stress significantly influences rats activity and memory,and the reduction of p-ERK1/2 signaling pathway in PFC may be involved in the mechanism of chronic stress.
