Abstract:Four phosphonodipeptides containing 1-aminoalkylphosphonate were synthesized by applying four different coupling methods. One of them was then converted into monoacid, ethyl hydrogen 1-(N-acetyl-prolyl)amino-2-phenylethyl phosphonate. Three phosphonodipeptides containing ethyl 1w-amino-2-phenylethylphosphonamidate were also prepared by converting phosphono monoacid into phosphonochloridate and reacting it with amino acid ester. Benzyl ethoxyphosphonyl-glycyl-glycine ethyl ester also was synthesized from ethyl glycylglycinate and ethyl benzylphosphonochloridate derived from direct chlorination of diethyl benzylphosphonate with phosphorus oxychloride.

Abstract:Optically active 1-aminoalkylphosphonic acid derivatives were synthesized in moderate yields and optical purities via Mannich-type reactions of benzyl carbamate, aldehydes, and optically pure chlorophosphites. Side chain-functionalized depsiphosphonopeptides were also prepared in satisfactory yields directly from one-pot reactions of benzyl carbamate, aldehydes, and diethyl (R,R)-2-chloro-1,3,2-dioxaphospholane-4,5-dicarboxylate.

Abstract:Phosphonopeptides are important phosphorus analogs of natural peptides and have been widely applied as enzyme inhibitors and antibacterial agents. A series of phosphonodipeptides containing C-terminal α-aminoalkylphosphonic acids was synthesized in satisfactory to good yields conveniently from 2-(N-benzyloxycarbonylamino)alkanamides, aldehydes, and phosphorus trichloride via Mannich-type reaction and subsequent sequential hydrolysis. The reaction mechanism was proposed and verified by (31)P NMR tracing experiments. The current method is an efficient and convenient method for preparation of phosphonopeptides containing C-terminal α-aminoalkylphosphonic acids.

Origins of Life and Evolution of the Biosphere

Abstract:Nucleoside N-phosphoamino acids were synthesized through Atherton-Todd reaction of nucleoside H-phosphonate with amino acids, and their structures were confirmed by NMR and ESI-MS. After nucleoside N-phosphoamino acid was incubated in anhydrous methanol at 40 °C for 72 h, di- to tetra-peptide derivatives were detected by ESI-MS, and their structures were further identified by multistage mass spectrometry. These and previously published studies in aqueous solution suggest that nucleoside N-phosphoamino acids could have been prebiotic precursors of oligopeptides.

A series of model compounds, analogue of phosphoryl amino acids, were synthesized and their biomimic mechanism was also investigated by NMR and MS methods. The results indicated that the reactivity of phosphoryl biological small molecules was depended on the configuration, function groups and positions.

Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:Mannich-type condensation of benzyl carbamate, aldehydes, and chlorophosphite has been widely used in the synthesis of 1-(N-benzyloxycarbonylamino)arylmethyl-phosphonic derivatives. Reactions of benzyl carbamate, aldehydes, and cyclic chlorophosphite 2-chloro-1,3,2-dioxaphospholane were conducted in benzene to afford a mixture of benzyl [(2-oxido-1,3,2-dioxaphospholane-2-yl)arylmethyl]-carbamates and 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters. 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters were obtained exclusively after the addition of water under stirring. The reaction mechanism was discussed. Their mass spectrometric fragmentations were also described.

Abstract:The quinoline derivatives arouse interest due to their broad spectrum of activity. The phosphorus compounds under investigation, quinolinylphosphonic and -phosphinic acids and aminophenylphosphonic and -phosphinic acids, possess potent bioactive properties, mimicking amino acids, phosphate esters, anhydrides, or carboxylate groups in enzymes. Despite its potential value, there is no reported example of quinolinylphosphonic or - phosphinic acids with phosphonic or phosphinic functional groups connected directly to the benzene ring in quinoline constitution. The selected quinoline derivatives have been synthesized by adopting the Skraup-Doebner-Von Miller reaction. To this end, the syntheses of aminophenylphosphonic and -phosphinic acids were conducted and afforded the target products with high yield. All structures have been proven by the combination of NMR, IR, MS, and HRMS techniques and were rationalized based on DFT calculation. The structures of triphenylphosphane oxide (TPO), diphenylphosphosphinic acid (1c), ( tert -butyl)phenylphosphinic acid (1d) and bis(3-nitrophenyl)phosphinic acid (2c) were determined by single-crystal X-ray diffraction measurements. The Hirshfeld surface analyses for 1c, 1d and 2c were performed to analyze the intermolecular interactions in their crystal structures. According to our findings, the presence of numerous intermolecular PO•••H, NO•••H, and CH•••O contacts stabilizes the crystal structures. The NO•••H interactions manifest in the IR spectrum of 2c crystal as a narrow band with a maximum at 3088 cm -1 . The PO•••H intermolecular interactions are attributed to a weak experimental band at 1288 cm -1 .

Abstract:The X-ray photoelectron spectra of sixteen derivatives of N-beta-phenethyl amine and N-beta-phenethyl glycine have been studied. The effect of different structures on N1s binding energy and that of their Pauling's atomic charge density on the nitrogen atoms are investigated. The experimental binding energy of N1s is proportional to the calculated Pauling's atomic charge density. It shows that the phosphoryl group is the strongest nitrogen lone pair electron localizing group as compared with the corresponding sulfonyl and acyl derivatives. The XPS results agree with the data that there is no decarbonylation during the synthesis of N-dialkylphosphoryl-tetrahydro-3-benzazepin-1-one. In addition, since the N1s in P-N bond is smaller than that in S-N and C-N bonds, the phosphoryl group can be removed under much milder conditions. These results provide a semi-empirical evidence for the synthesis of the skeleton of the cephalotaxine. It also differentiates the reaction paths for these three different amino protecting groups.

Abstract:O-phosphorylated peptide and amino acid are important in living system. In this paper, Dialkyl-N,N-dialkyl phosphoramidites (DDPA) 3 were synthesized using two methods. The reaction of DDPA with the hydroxyl group of the corresponding amino acids in the presence of tetrazole, followed by oxidation, gave O-phosphoryl amino acid methyl esters in a good yield. A systematical study of the reactivity of DDPA was presented too.

Abstract:We have synthesized a series of gamma-aminoxy acids, including unsubstituted and gamma(4)- Ph-, gamma(4)-alkyl-, and gamma(3,4)-cyclohexyl-substituted systems. Coupling of these monomers to oligomers can be realized using EDCl/HOBt (or HOAt) as the coupling agent. gamma-Aminoxy peptides can form 10-membered-ring intramolecular hydrogen bonds-so-called "gamma N-O turns"-between adjacent residues, the extent of which is controlled by the nature of the side chain of each gamma-aminoxy acid residue, increasing from the unsubstituted gamma-aminoxy peptide to the gamma(4)-alkyl aminoxy peptides to the gamma(4)-phenyl- and gamma(3,4)-cyclohexyl-substituted aminoxy pepticles. The presence of two consecutive homochiral 10-membered-ring intramolecular hydrogen bonds leads to the formation of a novel helical structure. Theoretical studies on a series of model peptides rationalize very well the experimentally observed conformational features of these gamma-aminoxy peptides.

Abstract:Phosphonopeptides are phosphorus analogues of peptides and have been widely applied as enzyme inhibitors and antigens to induce catalytic antibodies. Phosphonopeptides generally contain one aminoalkylphosphonic acid residue and include phosphonopeptides with C-terminal aminoalkylphosphonic acids and phosphonopeptides with a phosphonamidate bond. The phosphonamidate bond in the phosphonopeptides is generally formed via phosphonylation with phosphonochloridates, condensation with coupling reagents and enzymes, and phosphinylation followed by oxidation. Pseudo four-component condensation reaction of amides, aldehydes, alkyl dichlorophosphites, and amino/peptide esters is an alternative, convergent, and efficient strategy for synthesis of phosphonopeptides through simultaneous construction of aminoalkylphosphonic acids and formation of the phosphonamidate bond. This review focuses on the synthetic methods of phosphonopeptides containing a phosphonamidate bond.
