RNAi and its Applications in Crop Genetics and Breeding

Abstract:Fire found that double-stranded RNA could very successfully suppress the expression of a homological gene in 1998, and this phenomenon was termed RNA interference (RNAi). RNAi is a homology-dependent gene silencing phenomena that is triggered by the presence of a double-stranded RNA directed against mRNA of a gene or the promoter region of a gene. There are three types of transgenes that can trigger the RNA silencing of endogenous plant genes: inverted repeat RNA interference, antisense-RNA technology and sense over-expression transgenes. This technology, as a knockdown technology, has been applied to a wide range of species to silence the expression of both specific genes and genes of invading viruses for crop genetics breeding. The advances of its application in gene function, virus resistance, the improvement of crop traits by decreasing disproducts content or increasing desire products content, and the benefit to industry in crop genetics and breeding were reviewed in this paper. Learning more about metabolic regulation of crop and RNAi mechanism, researchers would make RNAi more widely available for crop genetics breeding.
