Abstract:RNA interference (RNAi) is a naturally occurring biological process that regulates plant growth and development, defense against pathogens, and environmental stresses. It is a sequence-specific homology-based silencing mechanism in which the function of a gene is interfered or suppressed. Small interfering RNAs (siRNAs) and microRNAs (miRNAs) are produced inside the plant cell through the activation of RNAi machinery, which downregulates the expression of the target genes at transcriptional and translational levels. RNAi is more specific, precise in its action, and considered as a potential technology for functional genomics studies. In the last 15 years, it has emerged as a scientific breakthrough for crop improvement without affecting other agronomic traits. It has also been employed as a novel method in understanding the basic phenomenon of plant defense and metabolism. Several desirable traits have been improved in the crop varieties through RNAi, which include crop protection against biotic and abiotic stresses, enhancement of nutritional value, alteration in plant architecture for better adaptation to environmental conditions, overexpression or removal of secondary metabolites, enhancement of shelf life of fruits and vegetables, generation of male sterile lines, and development of seedless fruits. In this book chapter, we have discussed RNAi and its applications in crop improvement.

Abstract:RNA interference (RNAi) is a conserved molecular mechanism that plays a critical role in post-transcriptional gene silencing across diverse organisms. This review delves into the role of RNAi in plant functional genomics and its applications in crop improvement, highlighting its mechanistic insights and practical implications. The overview starts with the fundamental finding of the mechanism of RNAi, following its journey from petunias to its ubiquitous presence in a wide range of species. The discovery of several kinds of regulatory non-coding small RNAs, including as phasiRNAs, miRNAs, and siRNAs, has broadened our knowledge of RNAi-mediated gene regulation beyond what is previously thought. These RNA types have an impact on gene expression through complex post-transcriptional and epigenetic mechanisms. RNA interference (RNAi) has become a potent tool for comprehending gene activities in the context of crop improvement. It has shown to be useful in determining the functions of genes for metabolic pathways, stress tolerance, and more. Furthermore, RNAi-based strategies have potential for sustainable agriculture and integrated pest management, supporting international efforts to ensure food security. Abstract:RNA interference (RNAi) is a powerful tool for gene silencing in different organisms, including plants. It is being used in functional genomics to decipher the function of genes. This technology has also witnessed a variety of potential applications in agriculture for crop improvement, including the development of crops for resistance against biotic (weeds, pathogens, insect pests and nematode parasites) and abiotic stresses (drought, high and low temperature, etc.), nutritional quality improvement, healthier oils, delayed ripening, male sterility, modification of flowering time and flower colour, alteration of plant architecture, enhancement of secondary products, and removal of allergens and toxins. RNAi has several advantages over traditional transgenic approaches as genetically modified RNAi plants do not contain transgene protein, however the risk assessment of these plants should be examined to rule out any off-target effects.

Abstract:RNA interference (RNAi) is a gene silencing mechanism that occurs after transcription. It is induced by double stranded RNA which results in a sequence-specific gene regulation by small RNAs thus results in mRNA degradation. RNA silencing pathways in plants are small interfering RNA (siRNA), micro RNA (miRNA), short hairpin RNA (shRNA) that also involves enzymes, and protein complex for complete mechanism. RNAi applications have been used to increase biotic and abiotic resistance, alter plant architecture, improve fruit quality, enhanced nutritional values, enhanced secondary metabolites, seedless fruits, reduced anti-nutritional toxic compounds, reduced allergens and to develop high-value industrial products. Therefore, RNAi is a novel approach that can be exploited for functional analysis of target genes, regulation of gene expression, improved quality traits, alternative method for crop protection and production in vegetables. This review is an attempt to collect current information on the RNAi mechanism and its application in the improvement of vegetable crops.

Abstract:With the rapid population growth, there is an urgent need for innovative crop improvement approaches to meet the increasing demand for food. Classical crop improvement approaches involve, however, a backbreaking process that cannot equipoise with increasing crop demand. RNA-based approaches i.e., RNAi-mediated gene regulation and the site-specific nuclease-based CRISPR/Cas9 system for gene editing has made advances in the efficient targeted modification in many crops for the higher yield and resistance to diseases and different stresses. In functional genomics, RNA interference (RNAi) is a propitious gene regulatory approach that plays a significant role in crop improvement by permitting the downregulation of gene expression by small molecules of interfering RNA without affecting the expression of other genes. Gene editing technologies viz. the clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein (CRISPR/Cas) have appeared prominently as a powerful tool for precise targeted modification of nearly all crops’ genome sequences to generate variation and accelerate breeding efforts. In this regard, the review highlights the diverse roles and applications of RNAi and CRISPR/Cas9 system as powerful technologies to improve agronomically important plants to enhance crop yields and increase tolerance to environmental stress (biotic or abiotic). Ultimately, these technologies can prove to be important in view of global food security and sustainable agriculture.

Abstract:RNA interference (RNAi) is a powerful technique used to regulate gene expression and silence specific genes. RNAi has been widely adopted in agriculture to enhance plant resistance against biotic stresses and improve crop traits. The mechanism of RNAi involves the introduction of small RNA molecules into cells, which then bind to complementary mRNA molecules and prevent their translation into proteins. The small RNA molecules responsible for gene silencing include small interfering RNAs (siRNAs) and microRNAs (miRNAs). siRNAs can be introduced into host cells and combine with the RNA-induced silencing complex (RISC). The siRNA binds to the target mRNA molecule and leads to its degradation, preventing protein synthesis. miRNAs can also incorporated into the RISC and bind to their target mRNAs resulting in translational repression. To initiate targeted gene silencing using RNAi, siRNAs or artificial miRNAs are then introduced into plant cells using various methods. In the context of increasing plant resistance against biotic stresses, RNAi has been used to silence genes involved in plant-pathogen interactions. By targeting virulence or host susceptibility genes, RNAi can disrupt the infection process and enhance plant resistance. Also, RNAi could be employed as an efficient tool to improve crop traits, grain yield, and fruit quality characteristics. Generally, by silencing genes that negatively regulate plant defense responses or enhance resistance, researchers could be able to develop crops with enhanced resistance to pathogenes and diseases with enhanced attributes.

Abstract:Over the past two decades, genetic engineering and related techniques have demonstrated striking progress in manipulation of the genes for induction of the desired characteristics into transgenic organisms. The regulatory mechanism of RNA interference (RNAi) has been studied in many higher organisms. The accuracy and precision of this phenomenon assures a great success rate in plant improvement. The RNAi provides us with an explicit methodology for down-regulation of genes of interest without hampering the expression of any other gene in the plant. The examination of different RNAi pathways, along with their components, including microRNAs and small interfering RNAs, have provided us with more than one way to achieve gene manipulation for mediated crop improvement. The phenomenon of RNAi has been studied in different abiotic environments; such as salinity, drought, extreme temperatures, heavy metals, and nutrition deprivation and radiation in various crops. RNAi-mediated crop manipulations have been reported that incorporate the utilization of vital stress-responsive elements with their subsequent mRNA and/or protein targets. Hence, collectively, RNAi technology has proven to be a promising tool for abiotic stress improvement in crops. This chapter focuses on the potential and successful application of RNAi technology for crop improvement in response to various abiotic stress factors.

Abstract:Plant pathogens and other biological pests represent significant obstacles to crop Protection worldwide. Even though there are many effective conventional methods for controlling plant diseases, new methods that are also effective, environmentally safe, and cost-effective are required. While plant breeding has traditionally been used to manipulate the plant genome to develop resistant cultivars for controlling plant diseases, the emergence of genetic engineering has introduced a completely new approach to render plants resistant to bacteria, nematodes, fungi, and viruses. The RNA interference (RNAi) approach has recently emerged as a potentially useful tool for mitigating the inherent risks associated with the development of conventional transgenics. These risks include the use of specific transgenes, gene control sequences, or marker genes. Utilizing RNAi to silence certain genes is a promising solution to this dilemma as disease-resistant transgenic plants can be generated within a legislative structure. Recent investigations have shown that using target double stranded RNAs via an effective vector system can produce significant silencing effects. Both dsRNA-containing crop sprays and transgenic plants carrying RNAi vectors have proven effective in controlling plant diseases that threaten commercially significant crop species. This article discusses the methods and applications of the most recent RNAi technology for reducing plant diseases to ensure sustainable agricultural yields.

Abstract:RNA interference (RNAi) is a novel technique in the field of functional genomics. It has an immense potential for managing plant diseases by down regulating expression of phytopathogens’ genes (invader’s gene) and other negative regulators of resistance pathways. This technique has become a breakthrough in the field of managing plant diseases rather than implementing biological and chemical control measures. RNAi mechanism involves the silencing of specific genes responsible for infection in the host plant, in a homology-dependent manner, before their translation. Incorporation of RNAi over the time has become one of the most promising technology, which reduces the risks incurred in the production of transgenic plants. The idea of gene silencing has been successful under laboratory conditions, and it is now gaining importance for field applications as well. However, problem presently to solve include delivering RNAi gene silencing in the field, in a convenient way for managing fungal, bacterial and viral plant diseases, on host-pathogen related targeted sites. This chapter will give an insight on the strategies of delivering RNAi mediated gene silencing and managing plant diseases in a most practical way for the farmers.

Abstract:RNA interference (RNAi) is a naturally occurring gene silencing mechanism conserved across organisms with a clearly defined cell nucleus (eukaryotes). Gene silencing by RNAi through the degradation of a target messenger RNA (mRNA) has historically been used as a research tool to study the function of genes. Over the past two decades, silencing of vital genes through RNAi has been explored for agricultural applications, including managing plant insect pests and pathogens, improving plant agronomic traits, and increasing consumer desirability of food. Using RNAi for crop protection is especially attractive because of its high specificity, which minimizes unintended effects on non-target organisms and improves the safety profile of RNAi products. This paper describes how RNAi functions, its current applications in agriculture, the current regulatory views of RNAi-based pesticides, and concludes with a discussion of current challenges for the commercial application of RNAi in agriculture. The content presented is intended to serve as a resource for regulatory agencies, policy and lawmakers, private and public institutions, and the general public to inform regulatory assessments and consumer choice decisions.

Abstract:RNAs play a significant role in regulating gene expression and their principal areas have been exploited for the control of plant viruses by the discovery of RNA silencing mechanism. RNA silencing or RNA interference (RNAi) is an innovative mechanism that regulates and restricts the amount of transcripts either by suppressing transcription (TGS) or by the degradation of sequence-specific RNA. RNAi can be used effectively to study the role of genes in a variety of eukaryotic organisms by reverse genetics. The technology has been employed in several fields such as drug resistance, therapeutics, development of genetically modified animals for research and transgenic plants targeting plant viruses. In plants, small interfering RNAs (siRNA) are the characteristic of 21 to 22 bp long dsRNA, which has been recognized by the regulatory mechanism of RNAi and leads to the sequence-specific degradation of target mRNA. In addition to virus disease control, RNAi can also be used to control mycotoxins and plant diseases caused by other organisms. This review enhances our current knowledge of RNAi and its larger applications in agriculture, specifically in plant virus disease management.

Abstract:RNAi technology is a versatile, effective, safe, and eco-friendly alternative for crop protection. There is plenty of evidence of its use through host-induced gene silencing (HIGS) and emerging evidence that spray-induced gene silencing (SIGS) techniques can work as well to control viruses, bacteria, fungi, insects, and nematodes. For SIGS, its most significant challenge is achieving stability and avoiding premature degradation of RNAi in the environment or during its absorption by the target organism. One alternative is encapsulation in liposomes, virus-like particles, polyplex nanoparticles, and bioclay, which can be obtained through the recombinant production of RNAi in vectors, transgenesis, and micro/nanoencapsulation. The materials must be safe, biodegradable, and stable in multiple chemical environments, favoring the controlled release of RNAi. Most of the current research on encapsulated RNAi focuses primarily on oral delivery to control insects by silencing essential genes. The regulation of RNAi technology focuses on risk assessment using different approaches; however, this technology has positive economic, environmental, and human health implications for its use in agriculture. The emergence of alternatives combining RNAi gene silencing with the induction of resistance in crops by elicitation and metabolic control is expected, as well as multiple silencing and biotechnological optimization of its large-scale production.

Abstract:Insect pests rank among the major limiting factors in agricultural production worldwide. In addition to direct effect on crops, some phytophagous insects are efficient vectors for plant disease transmission. Large amounts of conventional insecticides are required to secure food production worldwide, with a high impact on the economy and environment, particularly when beneficial insects are also affected by chemicals that frequently lack the desired specificity. RNA interference (RNAi) is a natural mechanism gene expression regulation and protection against exogenous and endogenous genetic elements present in most eukaryotes, including insects. Molecules of double-stranded RNA (dsRNA) or highly structured RNA are the substrates of cellular enzymes to produce several types of small RNAs (sRNAs), which play a crucial role in targeting sequences for transcriptional or post-transcriptional gene silencing. The relatively simple rules that underlie RNAi regulation, mainly based in Watson–Crick complementarity, have facilitated biotechnological applications based on these cellular mechanisms. This includes the promise of using engineered dsRNA molecules, either endogenously produced in crop plants or exogenously synthesized and applied onto crops, as a new generation of highly specific, sustainable, and environmentally friendly insecticides. Fueled on this expectation, this article reviews current knowledge about the RNAi pathways in insects, and some other applied questions such as production and delivery of recombinant RNA, which are critical to establish RNAi as a reliable technology for insect control in crop plants.

Abstract:<p>RNA interference (RNAi) is a biological process in which small RNA (sRNA) molecules sequence-specifically silence gene expression at the transcriptional or post-transcriptional level, either by directing inhibitory chromatin modifications or by decreasing the stability or translation potential of the targeted mRNA. The trigger for gene silencing is double-stranded RNA (dsRNA) generated from an endogenous genomic locus or a foreign source, such as a transgene or virus. The process of gene silencing can be exploited in agriculture to control plant diseases and pests. Of the pests that impact crop yield (including nematodes, arthropods, rodents, snails, slugs and birds), insects constitute the largest and most diverse group. Here, we review the "pros" and "cons" of using RNAi technology mediated by dsRNA-expressing transgenic plants (host-induced gene silencing, HIGS) or direct application of chemically synthesized dsRNA to control plant-damaging insects. Rapid progress in elucidating RNAi mechanisms has led to the first commercial products on the market. Given the high potential of RNAi strategies, their use in agriculture, horticulture, and forestry will likely be extensive in the future. However, further studies are needed to improve the efficacy of RNAi-based plant protection strategies and to assess their associated safety risks.</p>

Abstract:Abstract RNA interference (RNAi) is being developed and exploited to improve plants by modifying endogenous gene expression as well as to target pest and pathogen genes both within plants (i.e. host-induced gene silencing) and/or as topical applications (e.g. spray-induced gene silencing). RNAi is a natural mechanism which can be exploited to make a major contribution towards integrated pest management and sustainable agricultural strategies needed worldwide to secure current and future food production. RNAi plants are being assessed and regulated using existing regulatory frameworks for GMO. However, there is an urgent need to develop appropriate science-based risk assessment procedures for topical RNAi applications within existing plant protection products legislation.

Abstract:The insect pests are big threat in meeting the food demands for future generation. The present pest control strategies, including the existing transgenic approaches show certain limitations and are not completely successful in limiting the insect pests. However, the sequence-specific gene silencing via RNA interference (RNAi) holds a great promise for effective management of agricultural pests. RNAi is naturally occurring conserved process responsible for gene regulation and defense against pathogens. The efficacy of RNAi varies among different insect orders and also depends upon various factors, including the target gene selection, method of dsRNAs delivery, expression of dsRNAs and presence of off-target effects. RNAi-mediated silencing of different insect genes involved in various physiological processes was found to be detrimental to insects growth, development and survival. In this article, we have reviewed the potential of RNAi-based strategies for effective management of insect pests. We have also discussed the various parameters, which are to be considered for host-induced RNAi-mediated control of insect pests without producing any effect on non-target organisms and environment.

Abstract:Introduction. Sustainable farming requires new tools for the control of pathogens, since there is a constant evolution to overcome the current biological and chemical strategies. The information provided by the transcriptomics allows creating new possibilities to tackle the pathogens. It is possible to interrupt the genetic expression of a pathogen and disable it using RNA interference (RNAi). Objective. To perform an analysis of an emerging technology useful for pest control, based on RNA interference. Development. Sustainable farming is measured based through social, economic, and environmental indicators. A key indicator of agriculture is the decrease in inputs for the control of pathogens and the increase in their specificity. Pest control mechanisms based on RNA interference meet both parameters. RNAi is known to have at least two functions, first for gene expression regulation, and secondly as a defense mechanism against pathogens. Consequently, RNAi can be used to protect crops from pathogens by developing genetically modified plants, or by the external application form of an aerosol. The RNAi aerosol is a tool that relies on inactivating the pathogen genes and can complement other agronomic tools available for this purpose. It is possible to design RNAi against tropical pests based on published transcriptomes, although it is necessary to overcome limitations regarding design, degradation, and stability. Conclusion. Interference RNA methods have the potential to be useful tools to control tropical pathogens as an alternative to achieve sustainable farming.

Regulatory Mechanisms in Biosystems

Abstract:Wheat is a strategic cereal crop of global importance and plays a leading role in the food supply of mankind. Despite the general trend to increase in its production, climatic changes leading to significant temperature changes, unpredictable precipitation or droughts and the appearance of new races of pathogens and pests significantly affect its yield. In order to prevent the negative impact of changes in climatic conditions on the productivity of this crop, it is necessary to develop innovative technologies for improving the resistance of wheat to environmental stresses. RNA interference (RNAi) represents a new potential tool for wheat breeding by introducing small non-coding RNA sequences with the ability to silence gene expression in a sequence-specific manner. A decrease in the expression of a certain gene determines the acquisition of a new characteristic through the elimination or accumulation of certain plant traits, which leads to biochemical or phenotypic changes that the original plants do not have. This literature review describes the progress achieved over the past decades in the application of RNAi to create wheat plants with improved economic and valuable traits. The main stages of the gene silencing mechanism mediated by short interfering RNA (siRNA) and microRNA (miRNA), features of their biogenesis, modes of action and distribution are presented. Examples of the use of various biotechnological approaches to wheat improvement using gene transformation, endogenous and exogenous double-stranded RNA molecules (dsRNA) are given. The possibility of using RNAi technology to increase the nutritional value and quality of grain, remove toxic compounds and allergens is highlighted. Considerable attention is paid to the practical results of various applications of RNAi to increase the resistance of wheat to biotic stress factors, in particular, viruses, bacteria, fungi, insect pests, and nematodes. Examples of the use of siRNA-mediated RNAi and the role of miRNA in improving wheat tolerance to abiotic stresses are summarized.
