Role of ChREBP and Mlx in Regulating Glucose-Responsive Genes

Abstract:Carbohydrate responsive element binding protein(ChREBP) and Max-like protein X(Mlx) are two principal transcription factors regulating glucose-responsive genes involved in glucose utilization and energy storage in the liver.ChREBP and Mlx dimerize and function together as a glucose-responsive transcription factor to up-regulate the expression of target genes.All glucose-responsive genes contain a carbohydrate responsive element(ChoRE) consensus sequence in their promoters,and ChREBP·Mlx binds to the ChoRE and activates gene transcription.ChREBP·Mlx is able to regulate expression of hepatic glycolytic and lipogenic enzymes in response to excess carbohydrate in diet,and play an important role in disease states such as diabetes,obesity,nonalcoholic fatty liver and hypertension.In this review,we introduced the structures and functions of ChREBP and Mlx, and the mechanism of the regulation of their expression.
