Abstract:population may be identified by the expression of the cell surface marker CD133 and by absence of CD45 and CD31.CD133 expression has been reported in several tumor tissues, including colon and liver cancers.Aim of our study was to investigate if there is a CD133+/ CD45-/CD31-cell population in hepatic metastasis from colon cancer versus normal peritumoral samples harvested from patients subjected to hepatic resection.Methods: Tumor and normal samples were obtained from consenting patients diagnosed with hepatic metastasis from colon cancer.Samples were mechanically fragmented and, then, digested by collagenase I (0,2%) at 37°C, for 90 min.The cell suspensions (from tumor and normal tissue) were filtered through cell strainers and characterized by flow cytometry, using antibodies against CD133, CD45 and CD31.Results: We collected 5 hepatic metastasis from colon cancer samples (2 males/3females).Median age of the patients was 55,7 years.The CD133+/CD45-/ CD31-cell fraction resulted to be 12,2% (ranging from 6% to 16%) in metastasis specimens compared with 0,15% (ranging from 0% to 0,45%) in normal liver tissue.The difference between both groups was significant .Conclusions: Our preliminary data confirm a significant higher proportion of CD133+/CD45-/CD31-cells in metastasis samples respectively to healthy tissues.The high prevalence of CD133+ positive cells within metastasis reinforces the "CSC theory", a subpopulation of tumorigenic cells responsible for tumor aggressiveness.

Abstract:Chronic gastritis is very common in Chinese population,primarily as a result of widespread infection with Helicobacter pylori,leading to intestinal metaplasia,atrophy,dysplasia and malignancy if not treated effectively.A growing body of evidence derived from recent clinicopathologic research suggests that incomplete intestinal metaplasia and multifocal atrophy are associated with a greater potential for malignant progression.These premalignant lesions usually start at the incisure-antrum,and spread,mainly along the lesser curvature,to the body,cardia and fundus,together with parietal cell atrophy,decreased serum level of pepsinogenⅠ,and increased level of gastrin-17.As the loss of functional gastric glands intensifies and expands,dysplasia in epithelial cells emerges,which may progress to malignancy.Effective treatment of chronic gastritis to prevent development or alleviate progression of intestinal metaplasia and atrophy holds the key to defend against malignant progression.Therefore,it is essential that meticulous examination of gastric mucosa at gastroscopy and comprehensive histopathologic evaluation of biopsy tissues should be carried out in every patient for an accurate diagnosis and staging of the lesion to be served as the foundation for effective therapy of chronic gastritis and prevention of gastric malignancy.
Expressions of FATI0 and P53 Mutant in Human Gastric Cancer and Their Relations

Abstract:Objective To investigate the expressions of FAT10 and p53 mutant in gastric cancer tissues and their relations. Methods Immunohistochemistry and RT-PCR were used to detect the expressions of FAT10 and p53 in gastric cancer tissues (n=62), para-cancerous tissues (2-5 cm apart from cancer, n=62), and normal gastric tissues (7>5 cm apart from cancer, n=62). The association of FAT10 with p53 and clinical outcomes were analyzed by Spearman and Pearson correlation. Results The immunohistochemistry examination showed that expressions of FAT10 [51.61%(32/62)] and p53 [45.16% (28/62)] were significantly higher in cancerous tissues than in para-cancerous tissues [12.90%(8/62) and 14.51% (9/62), χ2=21.26 and 20.69, P<0.01] and normal tissues [6.45% (4/62) and 9.68% (6/62), χ2=13.91 and 19.61, P<0.01]. Overexpressions of FAT10 protein and mRNA in cancerous tissues were closely related to lymph node metastasis and TNM staging (both P value<0.05). There was a positive correlation between FAT10 and p53 in protein and mRNA expressions (protein r=0. 865, P<0.05; mRNA r=0.761, P< 0.01). Those with positive expression of FAT10 had lower survival rate compared to those with negative expression (P<0.05). Conclusions The positive relation between over-expression of FAT10 and p53 implicates that both are involved in the gastric carcinogenesis, and FAT10 is a novel gastric cancer marker with prognostic significance.
Clinicopathological Significance of Intestinal Metaplasia in Endoscopically Resected Early Gastric Carcinoma.

Abstract:ObjectiveTo investigate the clinicopathological and prognostic significance of intestinal metaplasia (IM) in endoscopically resected early gastric carcinoma (EGC).MethodsAltogether 136 consecutive cases with EGC resected by endoscopic submucosal dissection over 5 years were included and divided into the early gastric cardiac (EGCC; n = 60) and non‐cardiac carcinoma (EGNCC; n = 76) groups. Goblet cell IM and subtypes were determined with histology and immunostaining. Recurrence‐free survival (RFS) was compared among various IM groups.ResultsIM was identified in 128 (94.1%) EGC cases, including complete IM (n = 39), incomplete IM (n = 27), and mixed IM (n = 62). Incomplete IM was significantly more common in EGCC and exhibited a lower frequency of en bloc resection than the complete subtype. The frequency of synchronous or metachronous gastric tumor was significantly more common in EGCC with complete IM than in those with incomplete IM. Compared to EGC without IM, EGC with IM showed a significantly higher frequency of non‐poorly cohesive carcinoma, en bloc resection, and non‐eCuraC‐1 grade. EGNCC with IM was significantly associated with negative resection margins and en bloc resection. The 5‐year RFS was significantly lower in EGNCC patients with incomplete IM compared with those with mixed IM. The independent risk factors for RFS included tumor size >2 cm and eCuraC‐1 grade.ConclusionsSubtyping IM in EGC helped predict endoscopic resectability, prognosis, and risk of synchronous or metachronous gastric tumor. The significance of IM differed between EGCC and EGNCC. Large studies with longer follow‐up are warranted to validate our findings.

Abstract:OBJECTIVE:To study the expression of p53 protein, incidence of p53 gene mutation and microsatellite instability in ulcerative colitis, dysplasia of colonic mucosa and ulcerative colitis-associated colorectal cancer.METHODS:P53 protein expression was detected by immunohistochemistry in 70 specimens from 21 cases of ulcerative colitis and 25 colonic mucosa specimens from normal subjects. The specimens of ulcerative colitis were examined for the mutation in exon 5, 6, 7, 8 of p53 gene with microdissection-PCR-SSCP/HA-clone-sequencing technique and the alterations in 10 microsatellite loci with microdissection-PCR-SSLP-clone- sequencing technique.RESULT:None of 25 normal specimens was p53-positive immunohistochemically, while 4/21 of ulcerative colitis specimens were p53-positive. P53- positive rate in inflammatory mucosa of ulcerative colitis specimens was 0/5, while that was 1/7, 2/7 and 1/2 in low-grade displasia (LGD), high-grade displasia (HGD) and carcinoma, respectively. The abnormal exons were detected by SSCP and confirmed by sequencing in 2 out of 21 cases: one was exon 6 in a case with carcinoma and the other was exon 8 in a HGD case; both had positive P53 expression. Two cases showed positive in Bat26 locus by SSLP: one was a LGD case, the other was a case of carcinoma, who also had abnormal exon 6 of p53 gene. Other 9 microsatellite loci, (TGF beta RII(A)(10), IGFIIR(G)(8), IGFIIR(CT)(5), TGF beta RII(GT)(3), BAX(G)(8), hMSH3(A)8, hMSH6(C)(8), TCF4(A)(9) and DPC4 (CA)(17)) were showed negative in all cases.CONCLUSION:P53 gene mutations and microsatellite instability may be one of the mechanisms for higher risk of carcinogenesis in ulcerative colitis.

Abstract:Most gastric cancers arise in the setting of chronic inflammation which alters gland organization, such that acid-pumping parietal cells are lost, and remaining cells undergo metaplastic change in differentiation patterns. From a basic science perspective, recent progress has been made in understanding how atrophy and initial pyloric metaplasia occur. However, pathologists and cancer biologists have long been focused on the development of intestinal metaplasia patterns in this setting. Arguably, much less progress has been made in understanding the mechanisms that lead to the intestinalization seen in chronic atrophic gastritis and pyloric metaplasia. One plausible explanation for this disparity lies in the notable absence of reliable and reproducible small animal models within the field, which would facilitate the investigation of the mechanisms underlying the development of gastric intestinal metaplasia (GIM). This review offers an in-depth exploration of the current state of research in GIM, shedding light on its pivotal role in tumorigenesis. We delve into the histological subtypes of GIM and explore their respective associations with tumor formation. We present the current repertoire of biomarkers utilized to delineate the origins and progression of GIM and provide a comprehensive survey of the available, albeit limited, mouse lines employed for modeling GIM and engage in a discussion regarding potential cell lineages that serve as the origins of GIM. Finally, we expound upon the myriad signaling pathways recognized for their activity in GIM and posit on their potential overlap and interactions that contribute to the ultimate manifestation of the disease phenotype. Through our exhaustive review of the progression from gastric disease to GIM, we aim to establish the groundwork for future research endeavors dedicated to elucidating the etiology of GIM and developing strategies for its prevention and treatment, considering its potential precancerous nature.

Clinic-pathological Features of Epstein-barr Virus Infection, Microsatellite Instability, Tumor Mutation Burden and PD-L1 Status in 2504 Chinese Gastric Cancer Patients

Abstract:Objectives: Gastric cancer is the 4th most common cancer worldwide. Different subtype showed unique molecular features that could potentially guide therapeutic decisions. The aim of this study was to investigate the Epstein-Barr virus infection, microsatellite instability status, PD-L1 expression and gene mutation in surgically treated gastric cancer patients. Methods: We reviewed all GC patients who underwent potentially curative gastroectomy with lymphadenectomy at Peking University Cancer Hospital between 2013 and 2018 from a prospective collected medical database. We analyzed the clinic-pathological factors associated with immunohistochemistry profiles. We also analyzed gene changes through next-generation sequencing. Results: d-MMR gastric cancer patients are more likely to expression programmed death-ligand 1 (p<0.001, programmed death-ligand 1 cut off value 1%). EBV-positive and d-MMR patients were identified in 4% and 7.5% of the 2504 gastric cancer patients, respectively. The MLH1/PMS2 negative case number was 126. The MSH2/MSH6 negative case number was 14. d-MMR status was related to diffuse/mixed group (p<0.05), but not related to tumor differentiation. In our study, the microsatellite instability results detected by next generation sequencing and d-MMR gastric cancer results detected by immunohistochemistry were in high consistency. d-MMR gastric cancer patients had more microsatellite instability core. Many pathogenic genes were detected in microsatellite instability gastric cancer patients, such as POLE, ETV6, TP53, BRCA, RNF43 and other genes. Conclusion: Through immunohistochemistry and next generation sequencing, we got MSI status, protein expression, TMB and gene changes of GC, which provided a theoretical basis for future G clinical treatment.
Prognosis of intestinal metaplasia and expressions of biomarkers in high-risk populations of gastric cancer in Shangdong province

Abstract:AIM: To investigate the relationship between the expression of biomarkers and the prognosis of hyperproliferative intestinal metaplasia (HIM) and usual intestinal metaplasia (UIM) in Padova and Vienna international classification. METHODS: The risk of progression to dysplasia or gastric cancer in patients with UIM or HIM was estimated in a prospective study in Chinese populations at high risk area of gastric cancer from 1992 to 2001 in Shandong province. The cases included 2 groups: progressive group (n = 27) and non-progressive group (n = 60). Immu-nohistochemistry was used to detect the expres- sions of pS2, MUC5AC, MUC6 and Ki-67 in both groups. RESULTS: HIM had a higher risk of dysplasia or gastric cancer than UIM (OR = 7.23, 95%CI 2.59-20.20). The risk of dysplasia or gastric cancer was reduced in IM with the expression of MUC5 (OR = 0.27, 95%CI 0.10-0.73) or pS2 (OR = 0.33, 95%CI 0.12-0.90). Ki-67 labeling index was elevated in progressive IM in comparison with that in non-progressive one (28.5%±14.3% vs 16.4%±9.8%, P < 0.01). An elevated risk of dysplasia or gastric cancer was observed in patients with HIM and negative expression of MUC5AC (OR = 10.40, 95%CI 2.99-36.21). The expression of was also higher in progressive IM than that in non-progressive one (OR = 1.56,95%CI 0.53-4.60), but the difference was not significant. CONCLUSION: HIM has a higher risk of progression to dysplasia or gastric cancer than UIM. It may be helpful to evaluate the prognosis of IM by combining the histological classification and the detection of some biomarkers, such as Ki-67, MUC5AC and pS2.
Bio-characteristics of Intestinal Metaplasia in the Stomach: Hyperproliferative and Usual Type

Abstract:Background In Padova and Vienna International Classification, the usual intestinal metaplasia (UIM) of the stomach, including complete and incomplete type, is defined as negative for dysplasia, and hyperproliferative intestinal metaplasia (HIM) as indefinite for dysplasia, but the biological characteristics of these two types of intestinal metaplasia (IM)remain to be studied. Objective To investigate the biological differences between UIM, HIM and intestinal type gastric cancer (IGC), a panel of biomarkers were detected. Methods A total of 38 cases of IGC, 41 HIM and 56 UIM adjacent to gastric cancer were studied. Immunohistochemistry was used to detect the expressions of pS2, MUC2, MUC5AC, MUC6, Ki-67, EGFR, p53 and sulfo-Lewis a in UIM, HIM and IGC. Microsatellite instability (MSI) in UIM, HIM and IGC was detected by using Denaturing High Performance Liquid Chromatography (DHPLC). Results The pS2 antigen expression in UIM (78.6%) was significantly higher than in HIM and IGC (9.8%, 10.5%), p<0.01. The MUC6, sulfo-Lewis a and EGFR protein expressions were significant increased in HIM (24.4%, 82.9%, 48.7%) and IGC (34.2%, 75.0%, 42.1%) than in UIM (3.6%, 25.5%, 17.9%), p<0.01. A reversed pattern of expressions of MUC2 and MUC5AC was observed in UIM (96.4%, 50.0%) and HIM (82.9%, 36.6%) compared with IGC (52.6%, 13.2%), p<0.05; and the p53 gene expression was increased from UIM (1.8%) to HIM (19.5%) to IGC (57.9%), p<0.01. The Ki-67 labeling index was significantly different among three lesions (UIM: 16%±6%, HIM: 45%±9%, IGC: 63%±10%, p<0.01). Conclusion These findings suggest that there are different bio-characteristics among UIM, HIM and IGC, and HIM may have higher potential to progress to more advanced lesions in comparison with UIM.

Redefining aberrant P53 expression of gastric cancer and its distinct clinical significance among molecular-histologic subtypes

Abstract:Background: Numerous studies have demonstrated a correlation between p53 overexpression and diminished survival in gastric cancer patients. However, conflicting findings exist, and we hypothesize that these discrepancies arise from the cancer's complexity and heterogeneity, coupled with a lack of consensus on aberrant p53 expression. Methods: We enrolled a cohort of 187 patients with surgically resected gastric cancer. Patient categorization was based on Epstein-Barr virus (EBV), microsatellite instability (MSI), and Lauren classification (intestinal, diffuse and mixed). Utilizing an incremental algorithm, we evaluated p53 immunohistochemical (IHC) patterns in all 187 cases, while next-generation sequencing was successfully performed on 152 cases to identify TP53 mutations (mutTP53). Results: MutTP53 was identified in 32 % of the 152 cases, comprising 36 missense, 5 nonsense, and 7 frameshift alterations. Missense mutations predominantly correlated with p53 overexpression, while nonsense and frameshifting alterations related to null expression. Trial calculations indicated that null expression and a p53 IHC cutoff at >40 % offered the best prediction of mutTP53 (kappa coefficient, 0.427), with the highest agreement (0.524) observed in diffuse type and the lowest (0.269) in intestinal type. Null expression and a p53 IHC cutoff at >10 %, but not mutTP53 per se, provided the optimal prediction of survival outcome (p = 0.043), particularly in diffuse type (p = 0.044). Multivariate analysis showed that aberrant p53 IHC expression was not an independent prognostic factor. Conclusions: P53 IHC patterns are predictive biomarkers for mutTP53 and gastric cancer outcomes, where a prerequisite involves a nuanced approach considering cutoff values and molecular-histologic subtyping.
Clonal Analysis of Gastric Carcinoma and Precancerous Lesions and Its Relation to Ki-67 Protein Expression.

Abstract:The pathogenesis of intestinal carcinoma is characterized as progressing through multiple steps, which begin with atrophic gastritis followed by intestinal metaplasia, dysplasia and carcinoma. However, the clonal status of gastric precancerous lesions and its association with proliferative kinetics have not been fully understood. In this study, gastric lesions and normal epithelial cells were isolated from formalin-fixed paraffin embedded tissues using a laser capture microdissection (LCM) system, the clonality was analyzed with human androgen receptor gene (HUMARA) polymerase chain reaction (PCR), and the PCR products were examined using Applied Biosystems 3730 DNA Analyzer. The relationship between the clonal status and Ki-67 protein expression was also investigated. Ki-67 was detected by two-step immunohistochemical staining. 5/32 intestinal metaplasia lesions, 10/45 low grade intraepithelial neoplasia, 25/36 high grade intraepithelial neoplasia and 20/20 intestinal gastric carcinoma were of monoclonal origin. Similar to monoclonal inactivation, the expression rate of Ki-67 also increased along the multi-step gastric carcinogenesis. Clonal status was associated with the expression rate of Ki-67 to a certain extent, which may be useful in assessing susceptibility to gastric carcinoma. Relationship between nm23H1 genetic instability and clinical pathological characteristics in Chinese digestive system cancer patients

Abstract:Aim: To study the relationship between nm23H1 gene genetic instability and its clinical pathological characteristics in Chinese digestive system cancer patients. Methods: Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was used to analyze the microsatellite instability (MSI) and loss of heterozygosity (LOH). Immunohistochemistry was employed to detect the expression of nm23H1. Results: The MSI was higher in TNM stage I + II than in stage III + IV of gastric, colonic and gallbladder carcinomas. The LOH was higher in TNM stage III + IV than in stage I + II of gastric, colonic and hepatocellular carcinomas. Lymphatic metastasis was also observed. The expression of nm23H1 protein was lower in TNM stage III + IV than in stage I + II of these tumors and in patients with lymphatic metastasis.The nm23H1 protein expression was higher in the LOH negative group than in the LOH positive group. Conclusion: MSI and LOH may independently control the biological behaviors of digestive system cancers. MSI could serve as an early biological marker of digestive system cancers. Enhanced expression of nm23H1 protein could efficiently inhibit cancer metastasis and improve its prognosis. LOH mostly appears in late digestive system cancer.

Abstract:The development of intestinal gastric carcinoma involves several precancerous stages. The environmental factor plays an important role in gastric carcinogenesis, while the host's genetic makeup may influence the susceptibility to cancer. In this study we investigated correlations of the p53 variations at codon 72 and p21(WAF1/CIP1) haplotype with the risk of intestinal gastric carcinoma. Forty-eight intestinal gastric carcinoma cases (GC), 96 chronic atrophic gastritis (CAG), 96 intestinal metaplasia (IM) and 96 dysplasia (DYS) controls were enrolled in this study. The p53 codon 72 proline allele carriers were found to be more susceptible to progress to GC than to IM (OR = 2.22, 95%CI = 1.05-4.70, P = 0.038). Patients carrying homozygous p21(WAF1/CIP1) haplotype A, which contains the serine at codon 31, the cytidine at the 16th base of the second intron, and the cytidine at the 70th base of the exon 3 were more prone to develop GC than to reach the IM or DYS stage (IM versus GC, OR = 3.35, 95%CI = 1.11-10.15; DYS versus GC, OR = 3.27, 95%CI = 1.09-9.80, P = 0.035). The combination of p53 codon 72 variation with the p21(WAF1/CIP1) haplotype further distinguished the risk of GC from IM precancerous lesion (OR = 9.31, 95% CI = 1.77-48.85, P = 0.08). These results suggest that p53 and/or p21(WAF1/CIP1) genotype may influence the progression during gastric tumorigenesis.

Abstract:To explore the association of p53-MDM2 feedback loop related proteins with gastric pathologies in relation to Helicobacter pylori infection.Gastric biopsies were obtained from 157 H. pylori-negative and positive patients, including normal gastric mucosa (NGM), chronic gastritis (CG), intestinal metaplasia (IM), dysplasia (Dys), and gastric cancer (GC). The expression of mutant p53, MDM2, Bax and PUMA in gastric tissues was detected by immunohistochemistry.Overall expression of MDM2 and Bax is progressively increased from NGM to GC. PUMA expression is increased in CG but subsequently decreased after the development of IM. H. pylori infection is associated with increased mutant p53 and Bax expression but decreased PUMA expression in IM, and increased MDM2 expression in Dys.These results suggest that different p53-MDM2 feedback loop related proteins are distinctly expressed in the various stages of gastric carcinogenesis; their roles in gastric carcinogenesis in the presence of H. pylori infection need to be further investigated.
Studies on Apoptosis and the Expression of PCNA、P16 in Gastric Cancer

Abstract:Objective: The apoptosis and the expression of proliferating cell nuclear antigen(PCNA) and P16 gene in gastric carcinoma were examined were ,the biological and clinicpathologic significance were explained .Methods:The measures of terminal deoxynucleotidy transferasemediated dUTPbiotin nick end labeling(TUNEL)and streptavidin peroxidase immunohistochemical were used. Results:Among well or moderately differentiated adenocarcinomas,the AI(apoptoticindex)was (10.4±3.1%),While the poorly differentiated adenocarcinomas and mucous carcinomas was (4.12±1.4%) and (3.5±1.2%),respectively ,The AI of specimens with invasion depth exceeding muscularis(4.3±2.1%) was significantly lower than that of specimens with invasion depth not exceeding muscularis(imcluding muscularis).The subgroup with lymph node metastasis AI was (3.7±1.5%),while the subgroup without lymph node metastasis was (9.2±4.1%),The percentage of PCNA positive cells in well or moderately differentiated specimens was 41.7%.Among poorly differentiated specimens of carcinomas and mucos carcinomas ,the percentage of PCNA positive cells was 60% and 77.3%,respectively .The percentage of PCNA positive cells in the subgroup of specimens with lymph node metastasis was 72%,while the subgroup withou tlymphnode metastasis was 27.3%,The subgroup of specimens with invasion depth exceeding muscularis was 28.6% and the subgroup not exceeding muscularis was 77.2%. Among the PCNA positive subgroup,the AI was (3.4±1.2%),while in the PCNA negative subgroup was (7.6±3.1%).In the subgroup of well or moderately differentiated adenocarcinonas ,the positive percentage of P16 was 66.7%,while in the subgroup of poorly differentiated adenocarcinomas and mucous carcinomas was 33.3% and 44.4%,respectively .Conclusions:Apoptosis in gastric carcinoma was positively associated with the differentiation degree while negatively associated with invasion depth and lymph noed metastasis ,The expression of PCNA was positively associated with the invasion depth and lymph node metastasis ,while negatively associated with differentiation degree.Apoptosis index and expression of PCNA being examined at the same time can be useful to know the biological characteristics .In the gastric carcinomas tissues ,the express of P16 was positively associated with the differentiation degree.
Clonality and Ki-67 Protein Expression in Gastric Carcinoma and Precancerous Lesions

Abstract:OBJECTIVE:To study the clonality of gastric carcinoma and precancerous lesions and its relationship with Ki-67 protein expression.METHODS:Formalin-fixed paraffin embedded tissues were collected from 174 cases of gastric endoscopic biopsies and surgical removed specimens. The lesional tissues were isolated by Laser Capture Microdissection. Methylation sensitive restriction enzyme (HpaII) digestion and polymerase chain reaction (PCR) were used to detect the clonality at the polymorphic human androgen receptor gene locus on the X chromosome. PCR products were analyzed by capillary electrophoresis using applied Biosystems 3730 DNA Analyzer. In addition, a two-step immunohistochemical staining EnVision method was used to detect the expression of Ki-67 protein.RESULTS:The frequency of detection of monoclonality and expression rate of Ki-67 were found increased in a stepwise fashion from gastrointestinal metaplasia, low grade intraepithelial neoplasia, high grade intraepithelial neoplasia to intestinal carcinoma (15.63%, 5/32; 22.22%, 10/45; 69.44%, 25/36 and 100.0%, 20/20; respectively). The presence of clonal proliferation was correlated with Ki-67 expression in low grade intraepithelial neoplasia (P < 0.01).CONCLUSIONS:The presence of clonal proliferation and increased Ki-67 are increasingly detected in the lesions along the multi-step gastric carcinogenesis model. Clonal status is associated with the expression rate of Ki-67 to a certain extent, suggesting a combined application of both markers may be useful in assessing early stages of gastric carcinoma.

Microsatellite instability in gastric carcinoma and intestinal metaplasia

Abstract:Objective To investigate the role of microsatellite instability(MSI) in the carcinogenesis and development of gastric cancer (GC). Methods MSI was examined in 36 gastric cancer specimens obtained endoscopically and during surgery and in 30 intestinal metaplasia specimens using PCR SSCP methods. Results MSI was detected in 15 of 36 GC and 9 of 30 intestinal metaplasia specimens at one or more loci. MSI was positive in all three cases of early GC. The incidence of MSI in well differenciated GC was significantly higher than that in poorly differentiated GC (66.7% vs 26.3%, P 0.05). The frequency of MSI in intestinal type of GC was significantly higher than that in diffused type (64.7% vs 22.2%, P 0.05). No significant correlation was found between MSI and tumor size, location, stage and Helicobacter pylori infect ion. MSI was detected in 2 of 8(25.0%) type Ⅰ, 3 of 12(25.0%) type Ⅱ, and 4 of 10(40.0%) type Ⅲ intestinal metaplasia respectively. There was no significent difference among the three types. Conclusions MSI is an early molecular marker in the multistep cascade of gastric carcinogenesis and may play an important role in the development of GC.
